Chia seed was added to a mixture of 1,3-butylene glycol and purified water and stirred until the fibrous component comes out. Then, the gel portion was separated by centrifugation and filtered to obtain chia seed gel. Thus prepared chia seed gel had an average molecular weight of 150,000 to 250,000 Da.
2. Preparation of agave syrup
Agave was crushed, heated, extracted and then cooled to prepare agave syrup. Thus prepared agave syrup had an average molecular weight of 1,000 to 20,000 Da.
3. Preparation of cosmetic composition
Chia seed gel and agave syrup, which have moisturizing effect and a relatively low molecular weight, were used together with poly-γ-glutamic acid, glucan and hyaluronic acid to accomplish moisturization without stickiness. Moisturizers were prepared with various contents as shown in Table 1.
Each ingredient (chia seed gel, poly-γ-glutamic acid, glucan and hyaluronic acid) was diluted with purified water. Specifically, chia seed gel was diluted to 3 wt%, poly-γ-glutamic acid to 5 wt%, glucan to 0.4 wt%, and hyaluronic acid to 1 wt%.
The average molecular weights of the ingredients are as follows: poly-γ-glutamic acid = 1,500,000 to 2,500,000 Da; hyaluronic acid = 800,000 to 1,500,000 Da; glucan = 700,000 to 1,300,000 Da; chia seed gel = 150,000 to 250,000 Da; and agave syrup = 1,000 to 10,000 Da. The molecular weight was measured using 120C Series HPLC/GPC (Agilent Technologies), using Alltech 3300 ELSD as a detector and using a pL-aquagel OH-30 8 ㎛ column. Gas flow rate was 1.4 L/min and purified water was flown at 0.7 mL/min. Sample injection volume was 20 ㎕.
4. Comparison of feeling after use
After use of the compositions given in Table 1, feeling after use and moisturizing effect were evaluated.
Feeling after use was evaluated by a panel of 30 trainees. Evaluation was made from ‘-’(none at all) to ‘+++’(a great deal). Between the two extremes were ‘+’(slightly) and ‘++’(to some extent). Preference for each composition was evaluated as overall rating.

Overall rating(feeling after use) | 27%(n = 8) | 60%(n = 18) | - | - | 13 %(n = 4) |
As seen in Table 1, Samples 1 and 2, which contained 45% of 3% chia seed gel, showed less stickiness than Samples 3, 4 and 5. Of Samples 1 and 2, Sample 2 containing 5% of agave syrup showed better preference with neat feeling. Sample 3, which contained 5% of agave syrup, 35% of 3% chia seed gel, 20% of 0.4% glucan and 20% of 1% hyaluronic acid, were sticker and less neat than Sample 2. Sample 4, which contained 10% less 3% chia seed gel and 5% less agave syrup than Sample 3, were sticker and less neat than Sample 3. And, Sample 5, which contained the same amount of 3% chia seed gel but 5% less agave syrup than Sample 3, showed similar stickiness but less neatness.
Concluding the test result, the composition of Sample 2, which contained 45% of 3% chia seed gel, 30% of 5% poly-γ-glutamic acid, 10% of 0.4% glucan, 10% of 1% hyaluronic acid and 5% of agave syrup, exhibited the best neatness and less stickiness.
5. Comparison of moisturizing effect
Moisturizing effect was tested for the samples. 2.0 mg/cm2 of each sample was applied on inside the left forearm and moisturizing effect was measured. Measurement was made using WSK-P500U (Inforward Inc.) in a room maintained at 25℃ and relative humidity 45% for 3.5 seconds. The result is shown in Fig. 2.
As seen in Fig. 2, Sample 2 again showed the best result.
