Abstract
Description
本発明はこれらの知見を基にして完成されたものである。
(1)(S)-1-(4-クロロ-5-イソキノリンスルホニル)-3-アミノピロリジン及び薬理学的に許容されるその塩、並びに(S)-1-(4-クロロ-5-イソキノリンスルホニル)-3-(メチルアミノ)ピロリジン及び薬理学的に許容されるその塩からなる群から選択される1種又は2種以上の物質を有効成分として含有し、クエン酸又はその塩を含有しない水溶液組成物;
(2)(S)-1-(4-クロロ-5-イソキノリンスルホニル)-3-アミノピロリジン又は薬理学的に許容されるその塩を有効成分として含む上記(1)に記載の水溶液組成物;
(3)(S)-1-(4-クロロ-5-イソキノリンスルホニル)-3-(メチルアミノ)ピロリジン又は薬理学的に許容されるその塩を有効成分として含む上記(1)に記載の水溶液組成物;
(5)薬理学的に許容される塩が1塩酸塩である上記(4)に記載の水溶液組成物;
(6)水溶液のpHが5~9である上記(1)~(5)のいずれか1つに記載の水溶液組成物;
(7)さらに、クエン酸又はその塩以外のpH調整剤を含有する上記(6)に記載の水溶液組成物;
(8)クエン酸又はその塩以外のpH調整剤が、リン酸及びその塩、酢酸及びその塩、トリス(ヒドロキシメチル)アミノメタン及びその塩、並びにホウ酸及びその塩からなる群から選ばれる1種又は2種以上のpH調整剤である上記(7)に記載の水溶液組成物;
(10) 水溶液のpHが7~9である、(S)-1-(4-クロロ-5-イソキノリンスルホニル)-3-アミノピロリジン及び薬理学的に許容されるその塩、並びに(S)-1-(4-クロロ-5-イソキノリンスルホニル)-3-(メチルアミノ)ピロリジン及び薬理学的に許容されるその塩からなる群から選択される1種又は2種以上の物質を有効成分として含有する水溶液組成物;
(12)(S)-1-(4-クロロ-5-イソキノリンスルホニル)-3-アミノピロリジン及び薬理学的に許容されるその塩、並びに(S)-1-(4-クロロ-5-イソキノリンスルホニル)-3-(メチルアミノ)ピロリジン及び薬理学的に許容されるその塩からなる群から選択される1種又は2種以上の物質を有効成分として含む水溶液組成物の安定化方法であって、クエン酸又はその塩の非存在下において、クエン酸又はその塩以外のpH調整剤を用いてpHが5~9となるように調整する工程を含む方法;
(14)(S)-1-(4-クロロ-5-イソキノリンスルホニル)-3-アミノピロリジン及び薬理学的に許容されるその塩、並びに(S)-1-(4-クロロ-5-イソキノリンスルホニル)-3-(メチルアミノ)ピロリジン及び薬理学的に許容されるその塩からなる群から選択される1種又は2種以上の物質の眼圧降下作用が増強された医薬組成物の調製方法であって、該物質をpH7~9の水溶液組成物として調製する方法;
(16)(S)-1-(4-クロロ-5-イソキノリンスルホニル)-3-アミノピロリジン及び薬理学的に許容されるその塩、並びに(S)-1-(4-クロロ-5-イソキノリンスルホニル)-3-(メチルアミノ)ピロリジン及び薬理学的に許容されるその塩からなる群から選択される1種又は2種以上の物質を、クエン酸又はその塩の非存在下でpH調整剤を用いてpHが7~9となるように水溶液組成物として調製する工程を含む、眼圧降下作用が増強された該物質を含有する長期保存可能な水溶液組成物の製造方法;
(18)波長350nm以下の紫外線の透過率が40%以下の容器に充填され、及び/又は波長350nm以下の紫外線の透過率が20%以下の包装容器中の容器に充填された(S)-1-(4-クロロ-5-イソキノリンスルホニル)-3-アミノピロリジン又は薬理学的に許容されるその塩、及び、リン酸又はその塩、ホウ酸又はその塩、トリス(ヒドロキシメチル)アミノメタン又はその塩、あるいは酢酸又はその塩を含有し、且つ、クエン酸又はその塩を含有しない、pH7~9の点眼用水溶液組成物;
(20)波長350nm以下の紫外線の透過率が40%以下のプラスチック製容器に充填され、及び/又は波長350nm以下の紫外線の透過率が20%以下の包装容器中の容器に充填された(S)-1-(4-クロロ-5-イソキノリンスルホニル)-3-アミノピロリジン又は薬理学的に許容されるその塩、及び、リン酸又はその塩、ホウ酸又はその塩、トリス(ヒドロキシメチル)アミノメタン又はその塩、あるいは酢酸又はその塩を含有し、且つ、クエン酸又はその塩を含有しない、pH7~9の点眼用水溶液組成物;
(22)波長350nm以下の紫外線の透過率が40%以下の容器に充填され、及び/又は波長350nm以下の紫外線の透過率が20%以下、好ましくは10%以下の包装容器中の容器に充填された上記(1)~(21)のいずれか1つに記載の水溶液組成物;
(24)水溶液組成物を充填する容器が点眼用容器である上記(22)に記載の水溶液組成物;
(25)水溶液組成物を充填する容器が、プラスチック製である上記(22)、(23)、又は(24)に記載の水溶液組成物;
(27)上記(1)~(11)のいずれか1つに記載の水溶液組成物を、波長350nm以下の紫外線の透過率が40%以下の容器に充填する工程、及び/又は波長350nm以下の紫外線の透過率が20%以下、好ましくは10%以下の包装容器中の容器に充填する工程を含む前記水溶液組成物の光安定化方法。
(29)(S)-1-(4-クロロ-5-イソキノリンスルホニル)-3-(メチルアミノ)ピロリジンまたは薬理学的に許容されるその塩、及び、リン酸又はその塩、ホウ酸又はその塩、トリス(ヒドロキシメチル)アミノメタン又はその塩、あるいは酢酸又はその塩を含有し、且つ、クエン酸又はその塩を含有しない、pH7~9の点眼用水溶液組成物を、波長350nm以下の紫外線の透過率が40%以下の容器に充填する工程、及び/又は波長350nm以下の紫外線の透過率が20%以下の包装容器中の容器に充填する工程を含む該水溶液組成物の光安定化方法;
(31)(S)-1-(4-クロロ-5-イソキノリンスルホニル)-3-(メチルアミノ)ピロリジンまたは薬理学的に許容されるその塩、及び、リン酸又はその塩、ホウ酸又はその塩、トリス(ヒドロキシメチル)アミノメタン又はその塩、あるいは酢酸又はその塩を含有し、且つ、クエン酸又はその塩を含有しない、pH7~9の点眼用水溶液組成物を、波長350nm以下の紫外線の透過率が40%以下のプラスチック製容器に充填する工程、及び/又は波長350nm以下の紫外線の透過率が20%以下の包装容器中の容器に充填する工程を含む該水溶液組成物の光安定化方法;
本発明の水溶液組成物における別の1態様としては、(S)-1-(4-クロロ-5-イソキノリンスルホニル)-3-アミノピロリジン及び薬理学的に許容されるその塩、並びに(S)-1-(4-クロロ-5-イソキノリンスルホニル)-3-(メチルアミノ)ピロリジン及び薬理学的に許容されるその塩からなる群から選択される1種又は2種以上の物質のモル濃度の総和の下限値が、0.03mM以上である態様が好ましく、0.08mM以上である態様がより好ましく、0.14mM以上である態様が特に好ましい。該総和の上限値としては、該化合物の該水溶液液に対する飽和溶解度以下である態様が挙げられ、140mM以下である態様が好ましく、14mM以下である態様が特に好ましい。
本発明の水溶液組成物におけるpH調整剤のモル濃度と、(S)-1-(4-クロロ-5-イソキノリンスルホニル)-3-アミノピロリジン及び薬理学的に許容されるその塩、並びに(S)-1-(4-クロロ-5-イソキノリンスルホニル)-3-(メチルアミノ)ピロリジン及び薬理学的に許容されるその塩からなる群から選択される1種又は2種以上の物質のモル濃度の総和との比が1:10~5:1付近では、所望のpHよりも酸性側に片寄る可能性があるが、そういう場合は、モル濃度比をそれ以上に挙げるか、あるいはアルカリを用いてpH調整を行えばよい。該アルカリとしては固体か濃い濃度の水溶液を用いることが好ましい。
等張化剤の例としては、塩化ナトリウム、塩化カリウム等が挙げられる。粘稠剤の例としては、メチルセルロース、カルボキシメチルセルロースナトリウム等が挙げられる。また、防腐剤の例としては、パラオキシ安息香酸メチル、パラオキシ安息香酸エチル、ベンジルアルコール、クロロブタノール等が挙げられる。
本発明水溶液の浸透圧は特に限定されないが、通常200~700mOsm/Kg、好ましくは200~600mOsm/Kgであり、生理食塩水に対する浸透圧比が0.6~3、特に0.6~2であることが好ましい。
本発明で水溶液組成物を充填した容器を格納する包装容器としては、350nm以下の紫外線透過率が20%以下である包装容器が好ましく、これらのうち350nm以下の紫外線透過率が10%以下である包装容器がさらに好ましく、5%以下であることが特に好ましく、1%以下であることが最も好ましい。該包装容器の形状は特に限定されないが、箱状、袋状等が好ましい例として挙げられる。
350nm以下の紫外線の透過率が40%以下、好ましくは30%以下の容器に本発明の水溶液組成物を充填し、かつ該容器を50nm以下の紫外線透過率が20%以下、好ましくは10%以下の包装容器に格納する態様は本発明の特に好ましい態様である。
[例1](S)-1-(4-クロロ-5-イソキノリンスルホニル)-3-(メチルアミノ)ピロリジン1塩酸塩の製造
国際特許公開第WO2007/026664号パンフレットに記載の方法に従って(S)-1-(4-クロロ-5-イソキノリンスルホニル)-3-(メチルアミノ)ピロリジンを塩酸塩(1.50g)として得、これを水(52mL)に溶解した。この溶液を激しく撹拌し、氷冷下、2mol/L水酸化ナトリウム水溶液(4.13mL、和光純薬社製)をゆっくり滴下した。得られた懸濁液を室温でさらに1時間撹拌した後、塩化メチレン(30mL)を加え、有機層を分離した。さらに水層を塩化メチレン(30mL)で抽出し、併せた有機層を水(50mL)で洗浄し、無水硫酸マグネシウムで乾燥した。減圧下で溶媒を留去し、残渣に酢酸エチル(10mL)とn-ヘキサン(20mL)を加えた。析出した固体をろ取し、減圧下、50℃で20時間加熱乾燥することにより、(S)-1-(4-クロロ-5-イソキノリンスルホニル)-3-(メチルアミノ)ピロリジン(845mg)を得た。また、ろ液を減圧下で溶媒を留去し、残渣に酢酸エチル(3mL)とn-ヘキサン(6mL)を加えた。析出した固体をろ取し、減圧下50℃で終夜加熱乾燥することにより、(S)-1-(4-クロロ-5-イソキノリンスルホニル)-3-(メチルアミノ)ピロリジン(195mg)を得た。
(1)生理食塩水溶液
塩化ナトリウム18gを蒸留水2Lに溶解し、これを溶媒とした。(S)-1-(4-クロロ-5-イソキノリンスルホニル)-3-(メチルアミノ)ピロリジン1塩酸塩12mgに溶媒60mLを加えて溶解し、pHを測定した。
(2)10mMリン酸緩衝剤(pH6)溶液
リン酸二水素ナトリウムニ水和物0.78gを例2(1)の溶媒500mLに溶解した。この液50mLに、リン酸水素二ナトリウム十二水和物1.432gを例2(1)の溶媒400mLに溶解した溶液を添加してpH6.0に合わせ、これを10mMリン酸緩衝剤(pH6)溶媒とした。(S)-1-(4-クロロ-5-イソキノリンスルホニル)-3-(メチルアミノ)ピロリジン1塩酸塩12mgに10mMリン酸緩衝剤(pH6)溶媒60mLを加えて溶解し、pHが6付近であることを確認した。該溶液を約0.55mM(S)-1-(4-クロロ-5-イソキノリンスルホニル)-3-(メチルアミノ)ピロリジン1塩酸塩/10mMリン酸緩衝剤(pH6)溶液とみなした。
リン酸二水素ナトリウムニ水和物0.78gを例2(1)の溶媒500mLに溶解した。この液50mLに、リン酸水素二ナトリウム十二水和物1.432gを例2(1)の溶媒400mLに溶解した溶液を添加してpH7.0に合わせ、これを10mMリン酸緩衝剤(pH7)溶媒とした。(S)-1-(4-クロロ-5-イソキノリンスルホニル)-3-(メチルアミノ)ピロリジン1塩酸塩12mgに10mMリン酸緩衝剤(pH7)溶媒60mLを加えて溶解し、pHが7付近であることを確認した。該溶液を約0.55mM(S)-1-(4-クロロ-5-イソキノリンスルホニル)-3-(メチルアミノ)ピロリジン1塩酸塩/10mMリン酸緩衝剤(pH7)溶液とみなした。
(4)10mMリン酸緩衝剤(pH8)溶液
リン酸二水素ナトリウムニ水和物0.78gを例2(1)の溶媒500mLに溶解した。この液50mLに、リン酸水素二ナトリウム十二水和物1.432gを例2(1)の溶媒400mLに溶解した溶液を添加してpH8.0に合わせ、これを10mMリン酸緩衝剤(pH8)溶媒とした。(S)-1-(4-クロロ-5-イソキノリンスルホニル)-3-(メチルアミノ)ピロリジン1塩酸塩12mgに10mMリン酸緩衝剤(pH8)溶媒60mLを加えて溶解し、pHが8付近であることを確認した。該溶液を約0.55mM(S)-1-(4-クロロ-5-イソキノリンスルホニル)-3-(メチルアミノ)ピロリジン1塩酸塩/10mMリン酸緩衝剤(pH8)溶液とみなした。
リン酸二水素ナトリウム二水和物0.78gを例2(1)の溶媒500mLに溶解した。この液50mLに、リン酸水素二ナトリウム十二水和物1.43gを例2(1)の溶媒400mLに溶解した溶液を添加してpH5.0に合わせ、これを10mMリン酸緩衝剤(pH5)溶媒とした。(S)-1-(4-クロロ-5-イソキノリンスルホニル)-3-(メチルアミノ)ピロリジン1塩酸塩10mgに溶媒10mMリン酸緩衝剤(pH5)50mLを加えて溶解し、pHが5付近であることを確認した。該溶液を約0.55mM(S)-1-(4-クロロ-5-イソキノリンスルホニル)-3-(メチルアミノ)ピロリジン1塩酸塩/10mMリン酸緩衝剤(pH5)溶液とみなした。
(6)10mM酢酸緩衝剤(pH5)溶液
酢酸ナトリウム三水和物0.48gを例2(1)の溶媒500mLに溶解した。この液50mLに、酢酸(100)0.30gを例2(1)の溶媒500mLに溶解した溶液を添加してpH5.0に合わせ、これを10mM酢酸緩衝剤(pH5)溶媒とした。(S)-1-(4-クロロ-5-イソキノリンスルホニル)-3-(メチルアミノ)ピロリジン1塩酸塩10mgに10mM酢酸緩衝剤(pH5)溶媒50mLを加えて溶解し、pHが5付近であることを確認した。該溶液を約0.55mM(S)-1-(4-クロロ-5-イソキノリンスルホニル)-3-(メチルアミノ)ピロリジン1塩酸塩/10mM酢酸緩衝剤(pH5)溶液とみなした。
ホウ酸0.31gを例2(1)の溶媒500mLに溶解した。この液50mLに、四ホウ酸ナトリウム10水和物0.48gを例2(1)の溶媒500mLに溶解した溶液を添加してpH8.0に合わせ、これを10mMホウ酸緩衝剤(pH8)溶媒とした。(S)-1-(4-クロロ-5-イソキノリンスルホニル)-3-(メチルアミノ)ピロリジン1塩酸塩10mgに10mMホウ酸緩衝剤(pH8)溶媒50mLを加えて溶解し、pHが8付近であることを確認した。該溶液を約0.55mM(S)-1-(4-クロロ-5-イソキノリンスルホニル)-3-(メチルアミノ)ピロリジン1塩酸塩/10mMホウ酸緩衝剤(pH8)溶液とみなした。
(8)10mMホウ酸緩衝剤(pH9)溶液
四ホウ酸ナトリウム10水和物0.48gを例2(1)の溶媒500mLに溶解した。この液50mLに、ホウ酸0.31gを例2(1)の溶媒500mLに溶解した溶液を添加してpH9.0に合わせ、これを10mMホウ酸緩衝剤(pH9)溶媒とした。(S)-1-(4-クロロ-5-イソキノリンスルホニル)-3-(メチルアミノ)ピロリジン1塩酸塩10mgに溶媒10mMホウ酸緩衝剤(pH9)50mLを加えて溶解、pHが9付近であることを確認した。該溶液を約0.55mM(S)-1-(4-クロロ-5-イソキノリンスルホニル)-3-(メチルアミノ)ピロリジン1塩酸塩/10mMホウ酸緩衝剤(pH9)溶液とみなした。
トリス(ヒドロキシメチル)アミノメタン0.12gを例2(1)の溶媒80mLに溶解した。この液に、2mol/L塩酸2mLを正確に量り例2(1)の溶液を加えて正確に20mLとした溶液を添加してpHを8.0に合わせた後に、例2(1)の溶媒を加えて100mLとし、これを10mMトリス(ヒドロキシメチル)アミノメタン緩衝剤(pH8)溶媒とした。(S)-1-(4-クロロ-5-イソキノリンスルホニル)-3-(メチルアミノ)ピロリジン1塩酸塩10mgに10mMトリス(ヒドロキシメチル)アミノメタン緩衝剤(pH8)溶媒50mLを加えて溶解し、pHが8付近であることを確認した。該溶液を約0.55mM(S)-1-(4-クロロ-5-イソキノリンスルホニル)-3-(メチルアミノ)ピロリジン1塩酸塩/10mMトリス(ヒドロキシメチル)アミノメタン緩衝剤(pH8)溶液とみなした。
(1)10mMクエン酸緩衝剤(pH4)溶液
クエン酸水素二ナトリウム1.5水和物1.432gを例2(1)の溶媒400mLに溶解した。この液50mLに、クエン酸一水和物0.84gを例2(1)の溶媒400mLに溶解した溶液を添加してpH4.0に合わせ、これを10mMクエン酸緩衝剤(pH4)溶媒とした。(S)-1-(4-クロロ-5-イソキノリンスルホニル)-3-(メチルアミノ)ピロリジン1塩酸塩12mgに10mMクエン酸緩衝剤(pH4)溶媒60mLを加えて溶解し、pHが4付近であることを確認した。該溶液を約0.55mM(S)-1-(4-クロロ-5-イソキノリンスルホニル)-3-(メチルアミノ)ピロリジン1塩酸塩/10mMクエン酸緩衝剤(pH4)溶液とみなした。

(3)10mMクエン酸・リン酸緩衝剤(pH6)溶液
リン酸二水素ナトリウムニ水和物0.78g及びクエン酸一水和物1.05gを例2(1)の溶媒500mLに溶解した。この液50mLに、リン酸水素二ナトリウム十二水和物1.43g及びクエン酸ナトリウム2水和物1.18gを例2(1)の溶媒400mLに溶解した溶液を添加してpHを6.0に合わせ、これを10mMクエン酸・リン酸緩衝剤(pH6)溶媒とした。(S)-1-(4-クロロ-5-イソキノリンスルホニル)-3-(メチルアミノ)ピロリジン1塩酸塩10mgに10mMクエン酸・リン酸緩衝剤(pH6)溶媒50mLを加えて溶解し、pHが6付近であることを確認した。該溶液を約0.55mM(S)-1-(4-クロロ-5-イソキノリンスルホニル)-3-(メチルアミノ)ピロリジン1塩酸塩/10mMクエン酸・リン酸緩衝剤(pH6)溶液とみなした。
以下の試験において40℃の恒温器はIN801(ヤマト科学株式会社)、50~80℃の恒温器はIS600(ヤマト科学株式会社)を使用した。
例2の(1)~(4)、並びに比較例1の(1)及び(2)の溶液を孔径0.22μmのメンブランフィルターろ過し、それぞれ18本の褐色ガラスアンプルに2mLずつ充填し、融閉した。これらを40~80℃で表1の期間保存した。
例2の(1)~(4)、及び比較例1の(1)及び(2)の溶液の保存前後にpHの測定を実施した後、以下の条件の高速液体クロマトグラフィーにて、(S)-1-(4-クロロ-5-イソキノリンスルホニル)-3-(メチルアミノ)ピロリジン純度の測定を実施した。(S)-1-(4-クロロ-5-イソキノリンスルホニル)-3-(メチルアミノ)ピロリジン純度は面積百分率法で求めた。

(S)-1-(4-クロロ-5-イソキノリンスルホニル)-3-(メチルアミノ)ピロリジン1塩酸塩の生理食塩水溶液(例2(1))は下記の条件において経時的な純度の低下は見られなかったが、pHの経時的な上昇が見られた。
pH調整剤としてリン酸又はその塩を加えた(S)-1-(4-クロロ-5-イソキノリンスルホニル)-3-(メチルアミノ)ピロリジン1塩酸塩を含有する水溶液(例2(2)~(4))の中ではpH6(上記(4))が最も安定で、pH7(上記(5))、pH8(上記(6))の順で安定であった。ただし、pH8においても、少なくとも40℃6箇月保存後に(S)-1-(4-クロロ-5-イソキノリンスルホニル)-3-(メチルアミノ)ピロリジン純度が99%以上であった。
また、pH調整剤を加えた(S)-1-(4-クロロ-5-イソキノリンスルホニル)-3-(メチルアミノ)ピロリジン1塩酸塩を含有する水溶液(例2(2)~(4)、並びに比較例1(1)及び(2))は、全ての条件でpHの大きな変動は見られなかった。
結果を表3に示す。但し、表3には、各温度条件における開始時と終了時の結果のみ収載した。
例2(5)~(8)及び比較例1(3)の溶液を孔径0.22μmのメンブランフィルターろ過し、それぞれ5本の褐色ガラスアンプルに2mLずつ充填し、融閉した。また、例2(9)の溶液を孔径0.22μmのメンブランフィルターろ過し、褐色ガラスバイアル3本に充填し、密栓した。これらを80℃で例2(5)~(8)、及び比較例1(3)の溶液は2箇月、例2(9)の溶液は2週間保存した。測定方法は試験例1と同様で測定した。
クエン酸又はその塩を含有する比較例1(3)の溶液は80℃2箇月の保存により、pH6においても純度の低下が見られた。一方、クエン酸又はその塩を含有しない例2(5)、(6)、(7)及び(8)の溶液は80℃2箇月の保存においても大きな純度の低下は見られなかった。また、例2(9)の溶液は80℃2週間のみの保存であるが、その純度の変化から80℃2週間の保存においても大きな純度の低下は見られないものと思われる。結果を表4に示す。
国際特許公開第WO2007/026664号パンフレットに記載の方法に従って得られた(S)-1-(4-クロロ-5-イソキノリンスルホニル)-3-アミノピロリジンの塩酸塩(5.500g)を水(50mL)に溶解した。この溶液を激しく撹拌し、氷冷下、1mol/L水酸化ナトリウム水溶液(29.32mL、和光純薬社製)をゆっくり滴下した。酢酸エチル(300mL)及び水(20mL)を加え、有機層を留去した。さらに水層を塩化メチレンで抽出し、併せた有機層を水で洗浄し、無水硫酸マグネシウムで乾燥した。減圧下で溶媒を留去し、残渣に酢酸エチル(35mL)とn-ヘキサン(105mL)を加えた。析出した固体をろ取し、n-ヘキサン/酢酸エチル混液(4:1)で洗浄した後に減圧下、50℃で真空乾燥することにより、(S)-1-(4-クロロ-5-イソキノリンスルホニル)-3-アミノピロリジン(4.24g)を得た。
例3で得られた化合物について以下のイオン交換クロマトグラフィー条件で塩化物の塩数を確認したところ、(S)-1-(4-クロロ-5-イソキノリンスルホニル)-3-アミノピロリジン1分子当たり1.0個の塩化物イオンを認め、この物質が(S)-1-(4-クロロ-5-イソキノリンスルホニル)-3-アミノピロリジンに1個の塩酸が付加した塩であることが確認された。
(1)生理食塩水溶液
(S)-1-(4-クロロ-5-イソキノリンスルホニル)-3-アミノピロリジン1塩酸塩12mgに例2(1)の溶媒60mLを加えて溶解し、pHを測定した。
(2)10mMリン酸緩衝剤(pH6)溶液
(S)-1-(4-クロロ-5-イソキノリンスルホニル)-3-アミノピロリジン1塩酸塩12mgに例2(2)の10mMリン酸緩衝剤(pH6)溶媒60mLを加えて溶解し、pHが6付近であることを確認した。該溶液を約0.55mM(S)-1-(4-クロロ-5-イソキノリンスルホニル)-3-アミノピロリジン1塩酸塩/10mMリン酸緩衝剤(pH6)溶液とみなした。
(S)-1-(4-クロロ-5-イソキノリンスルホニル)-3-アミノピロリジン1塩酸塩12mgに例2(3)の10mMリン酸緩衝剤(pH7)溶媒60mLを加えて溶解し、pHが7付近であることを確認した。該溶液を約0.55mM(S)-1-(4-クロロ-5-イソキノリンスルホニル)-3-アミノピロリジン1塩酸塩/10mMリン酸緩衝剤(pH7)溶液とみなした。
(4)10mMリン酸緩衝液(pH8)溶液
(S)-1-(4-クロロ-5-イソキノリンスルホニル)-3-アミノピロリジン1塩酸塩12mgに例2(4)の10mMリン酸緩衝液(pH8)溶媒60mLを加えて溶解し、pHが8付近であることを確認した。該溶液を約0.55mM(S)-1-(4-クロロ-5-イソキノリンスルホニル)-3-アミノピロリジン1塩酸塩/10mMリン酸緩衝液(pH8)溶液とみなした。
(1)10mMクエン酸緩衝剤(pH4)溶液
(S)-1-(4-クロロ-5-イソキノリンスルホニル)-3-アミノピロリジン1塩酸塩12mgに比較例2(1)の10mMクエン酸緩衝剤(pH4)溶媒60mLを加えて溶解し、pHが4付近であることを確認した。該溶液を約0.55mM(S)-1-(4-クロロ-5-イソキノリンスルホニル)-3-アミノピロリジン1塩酸塩/10mMクエン酸緩衝剤(pH4)溶液とみなした。
(2)10mMクエン酸緩衝剤(pH5)溶液
(S)-1-(4-クロロ-5-イソキノリンスルホニル)-3-アミノピロリジン1塩酸塩12mgに比較例1(2)の10mMクエン酸緩衝剤(pH5)溶媒60mLを加えて溶解し、pHが5付近であることを確認した。該溶液を約0.55mM(S)-1-(4-クロロ-5-イソキノリンスルホニル)-3-アミノピロリジン1塩酸塩/10mMクエン酸緩衝剤(pH5)溶液とみなした。
試験例1の(S)-1-(4-クロロ-5-イソキノリンスルホニル)-3-(メチルアミノ)ピロリジン1塩酸塩の代わりに(S)-1-(4-クロロ-5-イソキノリンスルホニル)-3-アミノピロリジン1塩酸塩を用いて、同様な実験を実施した。
pH調整剤としてクエン酸又はその塩を用いたpH4及びpH5は(S)-1-(4-クロロ-5-イソキノリンスルホニル)-3-(メチルアミノ)ピロリジン1塩酸塩と同様に純度の低下が見られた(比較例2(1)及び(2)の溶液)。一方、pH調整剤としてリン酸又はその塩を用いて、pHを6~8に合わせた溶媒(等張化剤として0.9%の塩化ナトリウムを含有する)に溶解することにより、60~70℃で4週間、80℃で2週間は安定であることを確認した(例4(1)~(4)の溶液)。
結果を表5に示す。
(S)-1-(4-クロロ-5-イソキノリンスルホニル)-3-(メチルアミノ)ピロリジン溶液のpHの違いによる眼圧降下効果の違いを見るために、実験動物として日本白色ウサギを使い、以下に示した試験を行った。
1.被験化合物水溶液の調製
(S)-1-(4-クロロ-5-イソキノリンスルホニル)-3-(メチルアミノ)ピロリジン溶液の調製
国際特許公開WO2007/026664号パンフレットに記載の方法に従って得られた(S)-1-(4-クロロ-5-イソキノリンスルホニル)-3-(メチルアミノ)ピロリジン-の塩酸塩を生理食塩水に溶解した後、水酸化ナトリウムを加えて溶液のpHを4,5,6,7,8,9に調整し、終濃度をそれぞれ0.5mMに合わせた。これらを各pHが異なる(S)-1-(4-クロロ-5-イソキノリンスルホニル)-3-(メチルアミノ)ピロリジン溶液とした。
使用したウサギは、充分眼圧測定試験に馴化したものを用い、一群6匹で使用した。ベノキシール0.4%液(表面麻酔用点眼液、参天製薬)をウサギの両眼に点眼後、眼圧測定器(Model 30 Classic、solan社)を使用し、点眼前眼圧値(左右眼とも)を測定した(初期眼圧値)。被験溶液を左眼に点眼し、右眼は無処置とした。点眼2,4,6時間後に表面麻酔点眼液で麻酔後、左右の眼圧値を測定した。各測定時間における測定値の左右眼の眼圧差を眼圧降下値(平均値±標準誤差)とした。
結果を図1に示す。図1から明らかなように、(S)-1-(4-クロロ-5-イソキノリンスルホニル)-3-(メチルアミノ)ピロリジン溶液の眼圧降下作用は、pH4~9の間では溶液のpHが高くなるに従って効果が強く発揮されることが明確になり、特にpHが7~9でその効果が強く発揮されることがわかった。
以下の実施例において、遮光容器白としては万国0号原色(馬野化学容器株式会社)、遮光容器黄としては万国1号黄体色(馬野化学容器株式会社)、遮光容器青としては万国1号ルリ(馬野化学容器株式会社)、紫外線遮光透明袋としてはUV合掌背貼袋透明(岡田紙業株式会社)、紫外線遮光橙色袋としてはUV合掌背貼袋透明(岡田紙業株式会社)、紫外線遮光灰色袋としてはスモークSF-003(株式会社東洋マーク製作所)を袋状にしたもの、紫外線遮光濃灰色袋としてはダークスモークSF-002(株式会社東洋マーク製作所)を袋状にしたものを使用した。
塩化ナトリウム1.8gを蒸留水200mLに溶解し、これを溶媒とした。(S)-1-(4-クロロ-5-イソキノリンスルホニル)-3-(メチルアミノ)ピロリジン1塩酸塩50mgに溶媒100mLを加えて溶解した。この液2mLを遮光容器白に充填した。
(2)遮光容器黄
塩化ナトリウム1.8gを蒸留水200mLに溶解し、これを溶媒とした。(S)-1-(4-クロロ-5-イソキノリンスルホニル)-3-(メチルアミノ)ピロリジン1塩酸塩20mgに溶媒100mLを加えて溶解した。この液4mLを遮光容器黄に入れ、蓋をした。
塩化ナトリウム1.8gを蒸留水200mLに溶解し、これを溶媒とした。(S)-1-(4-クロロ-5-イソキノリンスルホニル)-3-(メチルアミノ)ピロリジン1塩酸塩20mgに溶媒100mLを加えて溶解した。この液4mLを遮光容器青に入れ、蓋をした。
(4)透明ガラス瓶及び紫外線遮光透明袋
塩化ナトリウム1.8gを蒸留水200mLに溶解し、これを溶媒とした。(S)-1-(4-クロロ-5-イソキノリンスルホニル)-3-(メチルアミノ)ピロリジン1塩酸塩50mgに溶媒100mLを加えて溶解した。この液5mLを透明ガラス瓶に入れて蓋をし、紫外線遮光透明袋に入れて密封した。
例5(1)で(S)-1-(4-クロロ-5-イソキノリンスルホニル)-3-(メチルアミノ)ピロリジン1塩酸塩溶液を充填した遮光容器白を、紫外線遮光透明袋に入れて密封した。
(6)透明ガラス瓶及び紫外線遮光橙色袋
塩化ナトリウム1.8gを蒸留水200mLに溶解し、これを溶媒とした。(S)-1-(4-クロロ-5-イソキノリンスルホニル)-3-(メチルアミノ)ピロリジン1塩酸塩50mgに溶媒100mLを加えて溶解した。この液5mLを透明ガラス瓶に入れて蓋をし、紫外線遮光橙色袋に入れて密封した。
例5(1)で(S)-1-(4-クロロ-5-イソキノリンスルホニル)-3-(メチルアミノ)ピロリジン1塩酸塩溶液を充填した遮光容器白を、紫外線遮光橙色袋に入れて密封した。
(8)透明ガラス瓶及び紫外線遮光灰色袋
塩化ナトリウム1.8gを蒸留水200mLに溶解し、これを溶媒とした。(S)-1-(4-クロロ-5-イソキノリンスルホニル)-3-(メチルアミノ)ピロリジン1塩酸塩50mgに溶媒100mLを加えて溶解した。この液5mLを透明ガラス瓶に入れて蓋をし、紫外線遮光灰色袋に入れて密封した。
例5(1)で(S)-1-(4-クロロ-5-イソキノリンスルホニル)-3-(メチルアミノ)ピロリジン1塩酸塩溶液を充填した遮光容器白を、紫外線遮光灰色袋に入れ、密封した。
(10)透明ガラス瓶及び紫外線遮光濃灰色袋
塩化ナトリウム1.8gを蒸留水200mLに溶解し、これを溶媒とした。(S)-1-(4-クロロ-5-イソキノリンスルホニル)-3-(メチルアミノ)ピロリジン1塩酸塩50mgに溶媒100mLを加えて溶解した。この液5mLを透明ガラス瓶に入れ、蓋をし、紫外線遮光濃灰色袋に入れ、密封した。
(11)遮光容器白及び紫外線遮光濃灰色袋
例5(1)で(S)-1-(4-クロロ-5-イソキノリンスルホニル)-3-(メチルアミノ)ピロリジン1塩酸塩溶液を充填した遮光容器白を、紫外線遮光濃灰色袋に入れ、密封した。
塩化ナトリウム1.8gを蒸留水200mLに溶解し、これを溶媒とした。(S)-1-(4-クロロ-5-イソキノリンスルホニル)-3-(メチルアミノ)ピロリジン1塩酸塩20mgに溶媒100mLを加えて溶解した。この液5mLを透明ガラス瓶に入れ、蓋をした。
以下の試験において、光安定性試験器はLT-120D3CJ(ナガノサイエンス株式会社)を使用した。また、実施例及び比較例の(S)-1-(4-クロロ-5-イソキノリンスルホニル)-3-(メチルアミノ)ピロリジン純度の測定方法は、試験例2の液体クロマトグラフ法にて実施した。結果を表6に示す。
例5及び比較例3のサンプルに対し、25℃、5,000lux、10日間(1,200,000lux)の光照射を実施した。試料への光照射は光安定性試験器でD65昼光色蛍光ランプを用いて実施した。光安定性試験器は、テーブルが回転して満遍なく試料に光を照射できるような装置を使用した。照度は5,000luxで、10日間、計1,200,000lux・hrを照射した。光安定性試験後の積算近紫外エネルギーは474~488W・h/m2であった。2)光照射をしたサンプルにおける溶液の(S)-1-(4-クロロ-5-イソキノリンスルホニル)-3-(メチルアミノ)ピロリジンの純度を5℃で暗所保存したサンプルと比較した。結果を表6に示す。

- (S)-1-(4-クロロ-5-イソキノリンスルホニル)-3-アミノピロリジン及び薬理学的に許容されるその塩、並びに(S)-1-(4-クロロ-5-イソキノリンスルホニル)-3-(メチルアミノ)ピロリジン及び薬理学的に許容されるその塩からなる群から選択される1種又は2種以上の物質を有効成分として含有し、クエン酸又はその塩を含有しない水溶液組成物。
- (S)-1-(4-クロロ-5-イソキノリンスルホニル)-3-アミノピロリジン又は薬理学的に許容されるその塩を有効成分として含む請求項1に記載の水溶液組成物。
- (S)-1-(4-クロロ-5-イソキノリンスルホニル)-3-(メチルアミノ)ピロリジン又は薬理学的に許容されるその塩を有効成分として含む請求項1に記載の水溶液組成物。

- 波長350nm以下の紫外線の透過率が40%以下の容器に充填され、及び/又は波長350nm以下の紫外線の透過率が20%以下の包装容器中の容器に充填された請求項1ないし17のいずれか1項に記載の水溶液組成物。
- 水溶液組成物を充填する容器が点眼用容器である請求項18に記載の水溶液組成物。
- 水溶液組成物を充填する容器が、プラスチック製である請求項18又は19に記載の水溶液組成物。
- (S)-1-(4-クロロ-5-イソキノリンスルホニル)-3-アミノピロリジン及び薬理学的に許容されるその塩、並びに(S)-1-(4-クロロ-5-イソキノリンスルホニル)-3-(メチルアミノ)ピロリジン及び薬理学的に許容されるその塩からなる群から選択される1種又は2種以上の物質を有効成分として含有する水溶液組成物を、波長350nm以下の紫外線の透過率が40%以下の容器に充填する工程、及び/又は波長350nm以下の紫外線の透過率が20%以下の包装容器中の容器に充填する工程を含む該水溶液組成物の光安定化方法。
- 請求項1ないし11のいずれか1項に記載の水溶液組成物を、波長350nm以下の紫外線の透過率が40%以下の容器に充填する工程、及び/又は波長350nm以下の紫外線の透過率が20%以下の包装容器中の容器に充填する工程を含む該水溶液組成物の光安定化方法。
