Abstract
본 발명은 벌개미취(Aster koraiensis) 추출물, 벌개미취 분획물, 이로부터 분리한 짐나스테르코리아인(Gymnasterkoreaynes) 유도체 또는 이의 약학적으로 허용가능한 염을 유효성분으로 함유하는 발암 예방용 건강기능성 식품 조성물에 관한 것으로서, 특히 벌개미취 추출물로부터 분리한 단일화합물인 짐나스테르코리아인 B는 암 예방 지표효소인 퀴논 리덕타제(Quinone reductase)의 활성을 유도하므로, 본 발명에 따른 조성물은 암을 예방하는데 유용하게 사용될 수 있다.
Description
본 발명은 벌개미취 추출물, 벌개미취 분획물, 이로부터 분리한 짐나스테르코리아인 유도체 또는 이의 약학적으로 허용가능한 염을 유효성분으로 함유하는 발암 예방용 건강기능성 식품 조성물에 관한 것이다.
암에 대한 연구가 30년 이상 심도 있게 이루어진 현재에도 환경오염, 잘못된 식생활습관 등으로 인하여 암 발생률은 계속 증가하여 전 세계적으로 매년 1,000만 명 정도 발생하며 세계보건기구(WHO)는 사망의 주요원인 중 하나로 암을 꼽고 있다. 암은 중년층의 질환으로서 새로운 임상예의 70% 이상이 60세 이상에서 진단되어 세계인구가 고령화됨에 따라 암 발생률이 증가하게 되는 것은 피할 수 없는 현상이다. 국내에서도 보건복지부에 따르면 매년 10만 명 정도의 암 환자가 발생하는 것으로 알려져 있다.
일반적인 암의 치료법으로는 수술, 방사선 치료, 항암제 치료 등이 있다. 암의 확실한 치료를 위해서는 조기에 암을 진단하여 수술하고, 방사선 또는 항암제 치료를 병행하는 것이다. 그러나 암의 진단이 늦어지거나 암이 전이된 경우에는 치료 방법이 복잡해지며 치료에 소요되는 정신적, 경제적 부담이 훨씬 커지게 된다. 따라서 화학적 암 예방과 암을 초기에 발견하기 위한 진단방법 개발이 최근 암 연구의 중요 분야이다.
암 분야는 1) 기발생한 암세포를 사멸시키는 암 치료 용도와 2) 발암물질이 세포내 표적물과 반응하거나 목적물에 도달하는 것을 억제하거나 발암물질을 체외로 방출시킴으로써 암 발생 자체를 억제하는 암 예방 용도로 나눌 수 있다. 암 치료 용도를 갖는 것으로 알려진 물질의 경우 암 예방 용도는 갖지 않는 경우가 있으며, 암 예방 용도는 가지나 암 치료용도를 갖지 않는 물질이 있어, 최근에는 각각을 구체적으로 나누어 적용하고 있는 추세에 있다. 기존의 항암제는 암 치료용도로 사용되었는데, 이는 암세포뿐만 아니라 정상세포에도 손상을 입혀 심각한 독성 및 부작용을 나타내는 단점이 있다.
이에 반하여 암 예방 용도의 조성물은 비교적 독성이 없는 안전한 화학물질을 이용하여 발암과정을 인위적으로 억제하거나 지연 또는 역전시킴으로서 정상세포가 암세포로 전환하는 것을 예방하려는 암 연구의 새로운 전략이다. 화학적 암 예방 물질의 후보군이 되기 위해서는 독성이 없거나 미약해야 하며 다수에게 널리 보급될 수 있도록 천연물로부터 분리나 합성이 용이한 화학물질들이 이용되므로 몇몇 비타민 대사물질들을 제외하고는 주로 식물성 화합물이 이용되고 있다(Surh YJ, Nature Rev Cancer, 3, 768-780, 2003).
암은 개시단계(initiation), 촉진단계(promotion), 진행단계(progression), 전이단계(metastasis)와 같은 다단계로 되어있는데, 화학적 암 예방은 주로 개시단계와 촉진단계에서 암 발생을 억제하는 것을 목표로 하며 그 기전에 따라 크게 두 가지로 구분된다. 즉, 발암물질이 세포내 표적물과 반응하거나 목적물에의 도달을 억제하는 기전을 가진 블록킹 에이전트(blocking agent)와 발암물질에 노출된 세포가 신생물(neoplasia)로 나타나는 과정을 억제하는 서프레싱 에이전트(suppressing agent)이다(Surh YJ, Nature Rev Cancer, 3, 768-780, 2003). 블록킹 에이전트로는 1) 사이토크롬 P-450(cytochrome P-450) 효소의 억제물질인 디알릴 설파이드(diallyl sulfide), 이소티오시아네이트(isothiocyanate), 에라직 액시드(ellagic acid) 등이 알려져 있으며, 2) 2상 해독효소계의 유도제인 페닐에틸 이소티오시아네이트(phenylethyl isothiocyanate), 설포라판(sulforaphane), 올티프라즈(oltipraz) 등이 알려져 있다. 서프레싱 에이전트로는 1)폴리아민(polyamine) 대사 억제물질로 DFMO, 2) 세포분화촉진제로 레티노이드(retinoid) 계열, 3) 프로틴 키나아제 C(protein kinase C) 억제물질로 제니스테인(genistein), 4) 산화성 DNA 손상억제물질로 EGCG 등이 알려져 있다.
화학적 암 예방에 의한 해독효소계의 유전자 조절에는 안티옥시던트 레스폰스 엘리먼트(Antioxidant response element;ARE)와 제노바이오틱스 레스폰시브 엘리먼트(Xenobiotics-responsive element;XRE)의 두 개 조절인자가 관여하는 것으로 보고되었다(Okey AB et al., Toxicol Lett, 70, 1-22). 그 중 ARE는 2상 해독효소계의 활성유도를 조절하기 때문에 단일기능 유도제의 효과 검정에 이용될 수 있다.
현재 해독효소계의 활성을 유도하는 발암 예방용 조성물을 찾아내기 위한 시도가 진행중이며, 대표적으로 식물로부터 천연 조성물을 분리하는 연구가 진행중이다. 본 발명자 역시 종래 전혀 알려진 바 없는 식물로부터 발암 예방 용도를 갖는 물질을 개발하고자 노력하였으며, 벌개미취라는 식물에 관심을 갖게 되었다.
벌개미취는 국화과 개미취속(자원속)에 속하는 낙엽성 여러해살이 풀이다. 학명(Aster koraiensis Nakai)에서 보듯이 한국 특산식물로서 영명 역시 Korean daisy(starwork)라 불리우며, 별개미취, 고려쑥부쟁이라고 부르기도 하며, 경기도 이남의 산과 들의 물기가 있는 곳에 분포하는데, 특히 강원도에 많은 군락을 이루며 자라고 있다.
키는 50~60 cm, 꽃대까지 칠 경우 1 m 내외이고, 옆으로 뻗는 뿌리줄기에서 원줄기가 곧게 자라고, 홈과 줄이 있다. 꽃은 8∼10월에 피고, 두상화(頭狀花)는 연한 자주색, 연한 보라색이며 지름 4∼5 cm로서 줄기와 가지 끝에 1송이씩 달린다.
벌개미취는 인공적으로 대량번식이 가능하고 경제성이 뛰어나며, 뿌리가 대단히 튼튼하고 성장이 왕성하여 노출된 절개사면, 척박지 등에 식재하면 토양고정 능력이 뛰어나 토사유출 방지 효과가 크기 때문에 오늘날에 와서는 각 지방자치단체의 사방공사용 소재, 도로 주변의 화단 식재용 소재로도 많이 쓰이고 있다.
하지만, 종래 벌개미취의 세포독성에 의한 암 치료 용도는 알려진 바 있으나, 암 예방용도는 전혀 알려진 바 없으며, 또한 구체적으로 어떤 성분이 발암 예방 활성을 갖는지에 대해서도 전혀 개시된 바 없다. 이에 본 발명자들은 인체 독성이 없거나 미약한 화학적 발암 예방제를 개발하기 위하여 식용 가능한 천연물에서 단일기능 유도체의 활성을 검색하던 중, 벌개미취에서 분리한 짐나스테르코리아인 유도체가 암 예방 지표효소인 퀴논 리덕타제의 활성을 유도하며 세포독성이 없고, 해독효소계의 활성을 유도함으로서 화학적 암 예방 효과를 나타내어 이를 유효성분으로 함유하는 조성물은 발암 예방을 위한 건강기능식품으로 사용될 수 있음을 밝힘으로써 본 발명을 완성하였다.
본 발명의 목적은 벌개미취 추출물, 벌개미취 분획물, 이로부터 분리한 짐나스테르코리아인 유도체 또는 이의 약학적으로 허용가능한 염을 유효성분으로 함유하는 발암 예방용 건강기능성 식품 조성물을 제공하는 데 있다.
상기의 목적을 달성하기 위하여, 본 발명은 벌개미취 추출물, 벌개미취 분획물, 이로부터 분리한 짐나스테르코리아인 유도체 또는 이의 약학적으로 허용가능한 염을 유효성분으로 함유하는 발암 예방용 건강기능성 식품 조성물을 제공한다.
본 발명은 벌개미취 추출물, 벌개미취 분획물, 이로부터 분리한 짐나스테르코리아인 유도체 또는 이의 약학적으로 허용가능한 염을 유효성분으로 함유하는 발암 예방용 건강기능성 식품 조성물에 관한 것으로, 본 발명의 추출물, 분획물 또는 화합물은 발암 예방 지표효소인 퀴논 리덕타제(Quinone reductase)의 활성을 유도하므로 암을 예방하는데 유용하게 사용될 수 있다.

본 발명은 퀴논리덕타제(QR) 효소의 활성을 증가시켜 발암물질을 해독함으로써 발암 예방 활성을 갖는 벌개미취 추출물을 유효성분으로 함유하는 발암 예방용 건강기능성 식품 조성물을 제공한다.
상기 벌개미취는 채취한 것, 양식한 것 또는 시판되는 것 등 제한 없이 사용할 수 있으며, 상기 벌개미취 추출물은 아스테르 코라이엔시스(Aster koraiensis), 짐나스테르 코라이엔시스(Gymnaster koraiensis) 또는 이들의 혼합으로부터 추출된 것이 바람직하고, 더욱 바람직하게는 아스테르 코라이엔시스로부터 추출된 것일 수 있다. 상기 벌개미취 추출물을 제조하는 방법은 초임계추출, 아임계추출, 고온추출, 고압추출 또는 초음파추출법 등의 추출장치를 이용한 방법 또는 XAD 및 HP-20을 포함한 흡착수지를 이용하는 방법 등 당업계의 통상적인 추출방법을 사용할 수 있다. 가온하며 환류 추출 또는 상온에서 추출하는 것이 바람직하나 이에 한정하는 것은 아니다. 아울러 추출 회수는 1 내지 5회인 것이 바람직하며, 3회 반복 추출하는 것이 더욱 바람직하나 이에 한정되는 것은 아니다.
또한, 상기 벌개미취 추출물은 바람직하게는 벌개미취 건조물을 물, C1~C4의 알코올, 초산에틸, 염화메틸렌 및 클로로포름으로 이루어지는 군으로부터 선택되는 용매로 추출한 추출 또는 이들의 혼합용매로 추출한 추출물일 수 있으며, 보다 바람직하게는 C1~C4의 알코올로 추출한 추출물일 수 있고, 가장 바람직하게는 메탄올 또는 에탄올로 추출한 추출물일 수 있다. 일례로 벌개미취 건조물을 적당한 크기로 분쇄하여 추출용기에 넣고 C1~C4의 저급 알코올 또는 이들의 혼합용매, 바람직하게는 메탄올 또는 에탄올을 넣고 용액을 끓이며 환류 추출한 후, 일정시간 방치한 다음 거름종이 등으로 여과하여 알코올 추출물을 얻을 수 있다. 추출 시간은 2 내지 12시간인 것이 바람직하며, 3~5시간인 것이 가장 바람직하다. 이후에 농축 또는 동결건조 등의 방법을 추가적으로 거칠 수 있다.
또한, 본 발명은 퀴논리덕타제(QR) 효소의 활성을 증가시켜 발암물질을 해독함으로써 발암 예방 활성을 갖는 벌개미취 분획물을 유효성분으로 함유하는 발암 예방용 건강기능성 식품 조성물을 제공한다.
바람직하게는 상기 벌개미취 분획물은 벌개미취 추출물을 물에 현탁시킨 후 유출 용매로 헥산/초산에틸(10:1(v/v)), 헥산/초산에틸(5:1(v/v)), 헥산/초산에틸(2:1(v/v)), 헥산/초산에틸(1:1(v/v)), 초산에틸 및 메탄올을 사용하여 컬럼 크로마토그래피를 수행하여 얻을 수 있다.
나아가, 본 발명은 퀴논리덕타제(QR) 효소의 활성을 증가시켜 발암물질을 해독함으로써 발암 예방 활성을 갖는 하기 화학식 1로 표시되는 짐나스테르코리아인 유도체 중 어느 하나, 이들의 혼합물 또는 약학적으로 허용가능한 이의 염을 유효성분으로 함유하는 발암 예방용 건강기능성 식품 조성물을 제공한다.
상기 식에서, R1 및 R2는 각각 수소, 산소, 히드록시, C1~C4의 알킬에스테로 및 C1~C4의 알콕시로 이루어지는 군으로부터 선택되는 어느 하나이고, R1 및 R2 중 어느 하나가 산소이고 다른 하나가 수소인 경우, 상기 R1 또는 R2는 이들과 인접한 두 개의 탄소와 결합하는 에폭사이드이며, 는 단일 또는 이중결합이다.
상기 화학식 1의 짐나스테르코리아인 유도체는 벌개미취 추출물 또는 벌개미취 분획물로부터 분리하거나, 유기화학적 합성법으로 합성하여 사용할 수 있다.
상기 화학식 1의 짐나스테르코리아인 유도체 중에서도 특히, 본 발명은 하기 화학식 2의 짐나스테르코리아인 B, 하기 화학식 3의 짐나스테르코리아인 C, 하기 화학식 4의 짐나스테르코리아인 E, 하기 화학식 5의 1,8,15-헵타데카트리엔-11,13-디인-10-올 또는 이들의 혼합물을 유효성분으로 함유하는 발암 예방용 건강기능성 식품 조성물을 제공한다.
본 발명에 따른 상기 화합물은 벌개미취에 존재하는 대표적인 활성성분으로, 상기 화합물의 분리는 하기의 단계 1 내지 3을 포함하여 이루어지는 방법에 의해 수행될 수 있다.
벌개미취 건조물을 물, C1~C4의 알코올, 초산에틸, 염화메틸렌 및 클로로포름으로 이루어지는 군으로부터 선택되는 용매로 추출하여 벌개미취 추출물을 얻는 단계(단계 1);
상기 단계 1에서 얻은 벌개미취 추출물을 물에 현탁시킨 후, 유출 용매로 헥산/초산에틸(10:1(v/v)), 헥산/초산에틸(5:1(v/v)), 헥산/초산에틸(2:1(v/v)), 헥산/초산에틸(1:1(v/v)), 초산에틸 및 메탄올을 사용하여 컬럼 크로마토그래피를 수행하여 벌개미취 분획물을 얻는 단계(단계 2); 및
상기 단계 2에서 얻은 분획물을 고속액체크로마토그래피를 수행하여 짐나스테르코리아인 유도체를 분리, 정제하는 단계(단계 3)
본 발명에 따른 단계 1은 벌개미취 건조물을 물, C1~C4의 알코올, 초산에틸, 염화메틸렌 및 클로로포름으로 이루어지는 군으로부터 선택되는 용매로 추출하여 벌개미취 추출물을 얻는 단계이다. 상기 알코올은 메탄올, 에탄올, 프로판올, 부탄올 등의 저급 알코올을 사용할 수 있으며, 바람직하게는 메탄올 또는 에탄올이고, 가장 바람직하게는 에탄올이다.
본 발명에 따른 단계 1에 있어서, 상기 알코올 추출물을 얻는 방법은 건조된 벌개미취를 적당한 크기로 분쇄하여 추출용기에 넣고 적당한 양의 알코올을 첨가하여 가열하여 추출액을 얻는다. 벌개미취의 추출 효율을 높이기 위하여 상기 과정을 수회 이상 반복 수행할 수 있다. 상기 추출액을 여과하고, 감압 농축하여 벌개미취의 알코올 추출물을 제조할 수 있다.
본 발명에 따른 단계 2는 상기 단계 1에서 얻은 벌개미취 추출물을 헥산/초산에틸, 초산에틸 및 메탄올로 분획하여 벌개미취 분획물을 얻는 단계이다.
바람직하게는 상기 벌개미취 추출물을 물에 현탁시킨 후 유출 용매로 헥산/초산에틸(10:1(v/v)), 헥산/초산에틸(5:1(v/v)), 헥산/초산에틸(2:1(v/v)), 헥산/초산에틸(1:1(v/v)), 초산에틸 및 메탄올을 사용하여 컬럼 크로마토그래피를 수행하여 6개의 분획물을 얻을 수 있다.
본 발명자들은 벌개미취의 에탄올 추출물 5.8 g을 C-18 역상 수지로 충진된 감압 컬럼 크로마토그래피를 이용하여 6개의 분획물로 나누었으며, 용리조건은 헥산/초산에틸(10:1(v/v)) 400 mL로 시작하여 헥산/초산에틸(5:1(v/v)) 400 mL, 헥산/초산에틸(2:1(v/v)) 400 mL, 헥산/초산에틸(1:1(v/v)) 400 mL, 초산에틸 400 mL 및 메탄올 400 mL를 유출용매로 사용하였다.
본 발명에 따른 단계 3은 상기 단계 2에서 얻은 벌개미취 분획물을 고속액체크로마토그래피를 수행하여 짐나스테르코리아인 유도체를 분리, 정제하는 단계이다.
상기 단계는 65% 아세토니트릴/물을 유출용매로 사용하여 수행하는 것이 바람직한데, 본 발명에서는 단계 2로부터 얻은 분획물들 중 헥산/초산에틸(5:1) 분획 및 헥산/초산에틸(2:1) 분획을 합하여 400 mg의 분획물 중 200 mg을 상기 단계 3에 적용하여 4종의 짐나스테르코리아인 유도체를 분리, 정제할 수 있었다.
본 발명에 따른 짐나스테르코리아인 유도체들의 암 예방 지표효소인 퀴논 리덕타제 활성 유도 효과를 측정하기 위하여 흰쥐의 간암세포주(Hepa1clc7)를 대상으로 실험을 수행한 결과, 짐나스테르코리아인 B 및 짐나스테르코리아인 E가 높은 퀴논 리덕타제 활성화 작용을 나타내었다. 특히 짐나스테르코리아인 B는 4종의 화합물들 중 가장 현저한 퀴논 리덕타제 활성화 작용을 나타내었다.
짐나스테르코리아인 B는 1.25, 2.5, 5 μM의 농도에서 무처리의 대조구에 비하여 퀴논 리덕타제 활성을 각각 2.6배, 3.3배, 3.9 배 증가시켰다(도 1 참조).
나아가, 본 발명의 벌개미취로부터 분리된 짐나스테르코리아인 B의 2상 해독효소계 활성에 대한 효과를 측정하기 위하여 인간의 대장암세포주(Caco-2)를 대상으로 하는 실험을 수행하였다. 본 발명의 짐나스테르코리아인 B는 10, 25, 50 μg/ml 의 농도에서 2상 해독효소계 활성을 유도하는 항산화반응인자 (ARE) 활성에 농도 의존적으로 영향을 주는 것으로 확인하였다(도 3 참조). 이는 본 발명의 짐나스테르코리아인 B가 2상 해독효소계의 활성을 증가시키는 화학적 발암 예방 효과를 설명하는 것이다 .
또한, 본 발명의 벌개미취로부터 분리된 짐나스테르코리아인 B의 2상 해독효소계 효소중 하나인 퀴논 리덕타제의 단백질 발현을 증가시키는 효과를 측정하기 위한 실험에서 짐나스테르코리아인 B는 1, 6, 24 시간에서 처리농도가 높아질수록 퀴논 리덕타제 단백질(NQO1)의 양을 현저하게 증가시킴을 알 수 있었다 (도 4 참조). 이는 본 발명의 짐나스테르코리아인 B가 2상 해독효소계인 퀴논 리덕타제의 활성을 증가시키는 화학적 발암 예방 효과를 설명하는 것이다 .
본 발명의 발암 예방은 후두암, 폐암, 식도암, 췌장암, 대장암, 간암, 위암, 설암, 피부암, 뇌종양, 자궁암, 유방암, 자궁경부암, 난소암, 신장암, 담낭암, 구강암, 결장암, 간암, 방광암 및 대장암으로 이루어진 군으로부터 선택되는 것에 대한 예방일 수 있으나, 이에 한정되는 것은 아니다.
본 발명은 상기 화학식 1로 표시되는 짐나스테르코리아인 유도체 및 이의 약학적으로 허용되는 염뿐만 아니라, 이로부터 제조될 수 있는 가능한 용매화물, 수화물, 라세미체 또는 입체이성질체를 모두 포함한다.
본 발명의 화학식 1의 짐나스테르코리아인 유도체는 약학적으로 허용 가능한 염의 형태로 사용할 수 있으며, 염으로는 약학적으로 허용 가능한 유리산(free acid)에 의해 형성된 산 부가염이 유용하다. 산 부가염은 염산, 질산, 인산, 황산, 브롬화수소산, 요드화수소산, 아질산 또는 아인산과 같은 무기산류와 지방족 모노 및 디카르복실레이트, 페닐-치환된 알카노에이트, 하이드록시 알카노에이트 및 알칸디오에이트, 방향족 산류, 지방족 및 방향족 설폰산류와 같은 무독성 유기산으로부터 얻는다. 이러한 약학적으로 무독한 염류로는 설페이트, 피로설페이트, 바이설페이트, 설파이트, 바이설파이트, 니트레이트, 포스페이트, 모노하이드로겐 포스페이트, 디하이드로겐 포스페이트, 메타포스페이트, 피로포스페이트 클로라이드, 브로마이드, 아이오다이드, 플루오라이드, 아세테이트, 프로피오네이트, 데카노에이트, 카프릴레이트, 아크릴레이트, 포메이트, 이소부티레이트, 카프레이트, 헵타노에이트, 프로피올레이트, 옥살레이트, 말로네이트, 석시네이트, 수베레이트, 세바케이트, 푸마레이트, 말리에이트, 부틴-1,4-디오에이트, 헥산-1,6-디오에이트, 벤조에이트, 클로로벤조에이트, 메틸벤조에이트, 디니트로 벤조에이트, 하이드록시벤조에이트, 메톡시벤조에이트, 프탈레이트, 테레프탈레이트, 벤젠설포네이트, 톨루엔설포네이트, 클로로벤젠설포네이트, 크실렌설포네이트, 페닐아세테이트, 페닐프로피오네이트, 페닐부티레이트, 시트레이트, 락테이트, β-하이드록시부티레이트, 글리콜레이트, 말레이트, 타트레이트, 메탄설포네이트, 프로판설포네이트, 나프탈렌-1-설포네이트, 나프탈렌-2-설포네이트 또는 만델레이트를 포함한다.
본 발명에 따른 산 부가염은 통상의 방법, 예를 들면, 화학식 1의 유도체를 과량의 산 수용액 중에 용해시키고, 이 염을 수혼화성 유기 용매, 예를 들면 메탄올, 에탄올, 아세톤 또는 아세토니트릴을 사용하여 침전시켜서 제조할 수 있다.
동량의 화학식 1의 유도체 및 물 중의 산 또는 알코올을 가열하고, 이어서 이 혼합물을 증발시켜서 건조시키거나 또는 석출된 염을 흡입 여과시켜 제조할 수도 있다.
또한, 염기를 사용하여 약학적으로 허용 가능한 금속염을 만들 수 있다. 알칼리 금속 또는 알칼리 토금속 염은 예를 들면 화합물을 과량의 알칼리 금속 수산화물 또는 알칼리 토금속 수산화물 용액 중에 용해하고, 비용해 화합물 염을 여과하고, 여액을 증발, 건조시켜 얻는다. 이때, 금속 염으로는 나트륨, 칼륨 또는 칼슘염을 제조하는 것이 제약상 적합하다. 또한, 이에 대응하는 은 염은 알칼리 금속 또는 알칼리 토금속 염을 적당한 음염(예, 질산은)과 반응시켜 얻는다.
본 발명에 따른 조성물은 발암의 예방을 목적으로 벌개미취 추출물, 벌개미취 분획물, 이로부터 분리한 짐나스테르코리아인 유도체를 식품, 음료 등의 건강보조 또는 건강기능성 식품에 첨가할 수 있다. 이 경우, 벌개미취 추출물, 벌개미취 분획물, 이로부터 분리한 짐나스테르코리아인 유도체를 식품 첨가물로 사용시에 원료에 대하여 0.01~20 중량%, 바람직하게는 0.1~5 중량%의 양으로 첨가될 수 있다. 유효 성분의 혼합양은 사용목적(예방, 건강 또는 치료적 처치)에 따라 적합하게 결정될 수 있다. 그러나, 건강 및 위생을 목적으로 하거나 건강 조절을 목적으로 하는 장기간의 섭취의 경우에 상기 양은 상기 범위 이하일 수 있으며, 안정성 면에서 아무런 문제가 없기 때문에 유효성분은 상기 범위 이상의 양으로도 사용될 수 있다. 상기 추출물 또는 이로부터 분리한 화합물을 다른 식품 또는 식품 성분과 함께 사용될 수 있고, 통상적인 방법에 따라 적절하게 사용될 수 있다.
상기 식품의 종류에는 특별한 제한이 없다. 상기 추출물, 이로부터 분리한 짐나스테르코리아인 B 또는 이의 약학적으로 허용가능한 염을 첨가할 수 있는 식품의 예로는 육류, 소세지, 빵, 쵸컬릿, 캔디류, 스넥류, 과자류, 피자, 라면, 기타 면류, 껌류, 아이스크림류를 포함하는 낙농제품, 각종 스프, 음료수, 차, 드링크제, 알코올 음료 및 비타민 복합제 등이 있으며, 통상적인 의미에서의 건강식품을 모두 포함한다.
본 발명의 식품 보조 첨가제는 여러 가지 향미제 또는 천연 탄수화물 등을 사용할 수 있다. 상기 천연 탄수화물은 포도당, 과당과 같은 모노사카라이드, 말토오스, 수크로오스 등과 같은 디사카라이드 및 덱스트린, 시클로덱스트린과 같은 폴리사카라이드, 자일리톨, 솔비톨, 에르트리톨 등의 당알코올이다. 감미제로서는 타우마틴, 스테비아 추출물과 같은 천연 감미제나, 사카린, 아스파르탐과 같은 합성 감미제 등을 사용할 수 있다. 상기 천연 탄수화물의 비율은 본 발명에 따른 조성물 100 중량부당 일반적으로 약 0.01~0.04 중량부, 바람직하게는 0.02~0.03 중량부이다.
상기 외에 본 발명에 따른 조성물은 여러 가지 영양제, 비타민, 전해질, 풍미제, 착색제, 펙트산 및 그의 염, 알긴산 및 그의 염, 유기산, 보호성 콜로이드 중점제, pH 조절제, 안정화제, 방부제, 글리세린, 알코올, 탄산 음료에 사용되는 탄산화제 등을 함유할 수 있다. 그밖에 본 발명에 따른 조성물은 천연 과일쥬스 및 야채 음료의 제조를 위한 과육을 함유할 수 있다. 이러한 성분은 독립적으로 또는 조합하여 사용할 수 있으며, 첨가제의 비율은 크게 중요하진 않지만 본 발명에 따른 조성물 100 중량부당 0.01~0.1 중량부의 범위에서 선택되는 것이 일반적이다.
이하, 실시예 및 실험예를 통하여 본 발명을 상세히 설명한다. 단, 하기의 실시예는 본 발명을 예시하는 것일 뿐, 본 발명의 내용이 하기의 실시예에 의해 한정되는 것은 아니다.
실시예 1: 벌개미취 추출물의 제조
건조된 벌개미취(강원도 평창 등지에서 채취)를 세절한 후 107 g에 2 L의 에탄올을 가하고 3시간동안 가온 환류 추출한 후 추출액을 여과하였다. 추출액을 여과한 후 남은 잔사에 다시 2 L의 에탄올을 가하고 3시간동안 가온 환류 추출하는 방법을 추가로 2회 반복하여 총 추출액 6 L를 수득하였다. 상기 추출액 6 L를 35 ℃에서 감압 농축하여 에탄올 추출물 5.8 g을 수득하였다.
실시예 2: 벌개미취 분획물의 제조
상기 실시예 1에서 수득한 벌개미취의 에탄올 추출물 5.8 g을 85 g의 C-18 역상 수지로 충진된 감압 컬럼 크로마토그래피를 이용하여 여섯 개의 분획물을 수득하였다. 용리 조건은 헥산과 초산에틸 및 메탄올을 사용하였으며 헥산-초산에틸(10:1(v/v)) 400 mL로 시작하여 헥산-초산에틸(5:1(v/v)) 400 mL, 헥산-초산에틸(2:1(v/v)) 400 mL, 헥산-초산에틸(1:1(v/v)) 400 mL, 초산에틸 400 mL 및 메탄올 400 mL를 유출용매로 사용하였다.
실시예 3: 짐나스테르코리아인을 포함한 화합물들의 분리 및 동정
상기 실시예 2에서 수득한 분획물들 중 헥산-초산에틸(5:1) 분획물 및 헥산-초산에틸 (2:1) 분획물을 합하여 400 mg의 혼합분획물을 제조하였고, 이 혼합분획물 중 200mg으로 65% 아세토니트릴/물을 유출용매로 사용한 고속액체크로마토그래피법을 이용하여(컬럼 21 mm x 250 mm; 유속 10 mL/min) 주성분을 분리한 결과, 화학식 2로 표시되는 짐나스테르코리아인 B(9 mg), 화학식 3로 표시되는 짐나스테르코리아인 C(2 mg), 화학식 4로 표시되는 짐나스테르코리아인 E(0.5 mg) 및 화학식 5로 표시되는 1,8,15-헵타데카트리엔-11,13-디인-10-올(1 mg)을 수득하였다.
상기 화합물들의 구조 분석을 위하여, 하기와 같이 NMR 분석 및 질량 분석을 실시하였다. 분자량 및 분자식은 Agilent 1100 고속유체크로마토그래피-질량 분광계(HPLC-ESI-MS)를 이용한 MS 측정을 통하여 결정하였으며, 핵자기공명기(Varian 500 ㎒ NMR)를 이용한 1H, 13C-NMR 스펙트럼(spectrum) 분석을 통하여 그 구조를 하기 화학식 2 내지 5와 같이 동정하였다. 분석 결과는 다음과 같다.

실험예 1: 벌개미취 추출물로부터 분리한 4종의 화합물들의 퀴논 리덕타제 활성 유도 효과 측정
실시예 3에서 수득한 4종의 화합물들의 암 예방 지표효소인 퀴논 리덕타제 활성 유도 효과를 측정하기 위하여 흰쥐의 간암세포주(Hepa1c1c7)를 대상으로 하는 실험을 수행하였다. 먼저, a-MEM(최소 필수 배지; minimum essential medium)/10% FBS(우태아혈청; fetal bovine serum) 용액을 간암세포배양액과 혼합하여 세포의 수를 1×105 cells/mL로 조제한 후에 96-웰 플레이트에 100 μL씩 처리하여 24시간 동안 5% 이산화탄소 및 37 ℃의 조건에서 배양하였다. 세포가 안정화 된 후에, 상기 실시예 3에서 수득한 4종의 화합물들을 2배 농도구배로 1.5625-25 μM의 5개의 농도로 처리하고 24시간 동안 5% 이산화탄소 및 37 ℃의 조건에서 배양하였다. 배양 종료 후 세포를 인산버퍼식염수(PBS; phosphate buffered saline) 용액으로 씻어낸 후 0.08% 디지토닌(digitonin)과 2 mM EDTA를 포함한 용액 80 μL로 세포막을 용해시키고 단백질 용액을 얻었다. 상기 단백질 용액 50 μL와 반응용액A(49 mL의 25 mM Tris 버퍼, 34 mg의 BSA, 0.34 mL의 1.5% 트윈-20 용액, 0.34 mL의 조효소용액, 100 유닛의 글루코스-6-포스페이트 디하이드로게나아제, 15 mg MTT, 50 μL의 50 mM 메나디온) 200 μL를 혼합하여 마이크로플레이트 리더로 610 nm에서 흡광도 증가율을 측정하였으며, 단백질의 양은 브래드포드(Bradford) 용액을 사용하여 595 nm에서 측정하여 구하였다. 상기의 반응용액A 중 조효소용액은 150 mM 글루코우즈-6-포스페이트(glucose-6-phosphate), 4.5 mM NADP, 0.75 mM FAD로 구성되었으며 상기의 MTT는 3-(4,5-dimethylthiazo-2-yl)-2,5-diphenyltetrazolium bromide의 약어이다. 퀴논 리덕타제의 활성은 하기 수학식 1에 의해 계산하였다.

A : nmol MTT/min (시험 시료를 처리한 세포내 퀴논 리덕타제의 610 nm에서의 흡광도 증가율)
B : mg protein (시험 시료를 처리한 세포 전체의 단백질 양)
측정 결과, 4종의 화합물들 중 화학식 3의 짐나스테르코리아인 C와 화학식 5의 1,8,15-헵타데카트리엔-11,13-디인-10-올은 효과가 없는 반면, 화학식 2의 짐나스테르코리아인 B와 화학식 4의 짐나스테르코리아인 E는 퀴논 리덕타제 활성화 작용을 나타내었다. 특히, 도 1에 나타난 바와 같이 화학식 2의 짐나스테르코리아인 B는 1.25, 2.5, 5 μM의 농도에서 무처리의 대조구에 비하여 퀴논 리덕타제 활성을 각각 2.6배, 3.3배, 3.9 배 증가시킴을 확인하였으며 짐나스테르코리아인 E는 같은 농도에서 2.5배, 3.2배, 3.8배 증가시킴을 확인하였다.
한편, 하기 식을 이용하여 상기 짐나스테르코리아인 B의 암예방지표 (Chemoprevention Index, CI)를 계산하였다. 계산결과는 표 1에 나타내었다.

상기 표 1에 나타낸 바와 같이, 짐나스테르코리아인 B의 암예방지표 (CI)는 15.974로 기존의 암예방 기능성 식품소재로 알려진 설포라판(Sulforaphane, CI = 12.833)과 비교하였을 때 매우 좋은 효능을 가짐을 확인하였으며, 도 2에도 나타나 있듯이 퀴논리덕타아제 활성을 두 배 이상 증가시키는 농도에서 독성이 거의 없음을 확인하였다.
실험예 2: 짐나스테르코리아인
B의 항산화반응인자(ARE)에 대한 효과 측정
본 발명의 짐나스테르코리아인 B의 2상 해독효소계의 활성을 유도하는 항산화반응인자(ARE; antioxidant response element)에 대한 효과를 측정하기 위하여 인간의 대장암세포주(Caco-2)를 대상으로 항산화반응인자의 리포터 진 어세이(reporter gene assay)를 수행하였다. 먼저, MEM/10% FBS 용액을 간암세포배양액과 혼합하여 세포의 수를 1×105 cells/mL로 조제한 후에 24-웰 플레이트에 1 mL씩 처리하여 5% CO2/37 ℃의 조건에서 배양하였다. 세포가 안정화 된 후에 항산화반응인자를 포함한 프로모터와 루시퍼라제(luciferase) 단백질 발현 유전자가 삽입된 벡터를 Fugene6(Roche Biochemicals)를 이용하여 형질전환 시켰다. 세포가 안정화 된 후에 상기 실시예 3에서 수득한 짐나스테르코리아인 B를 10, 25, 50 μM 의 농도로 처리하고 24시간 동안 5% 이산화탄소 및 37 ℃의 조건에서 배양하였다. 배양 종료 후 루시퍼라제 분석 키트(Luciferase assay Kit; Promega)를 사용하여 세포 용해물(cell lysate)을 분석하였다.
분석 결과 도 3에 나타난 바와 같이, 짐나스테르코리아인 B의 처리 농도가 증가할수록 항산화반응인자의 활성이 농도 의존적으로 증가함을 확인할 수 있었다.
따라서, 본 발명의 짐나스테르코리아인 B는 해독효소계의 활성을 증가시킴으로써 화학적 발암 예방 효과를 가지는 것으로 판단된다.
실험예 3: 짐나스테르코리아인 B의 NQO1 발현 증가 활성 측정
본 발명자들은 본 발명의 짐나스테르코리아인 B의 2상 해독효소계의 하나인 NAD(P)H:Quinone Oxidoreductase 1(NQO1)의 단백질 발현에 미치는 영향을 측정하기 위하여 인간의 대장암세포주(Caco-2)를 대상으로 웨스턴블라팅 어세이를 수행하였다. 먼저, MEM/10% FBS 용액을 대장암세포 배양액과 혼합하여 세포의 수를 1×105 cells/mL로 조제한 후에 60 mm 페트리 디쉬에 5 mL씩 처리하여 5% 이산화탄소 및 37 ℃의 조건에서 배양하였다. 세포가 안정화 된 후에 세포가 안정화 된 후에 상기 실시예 3에서 수득한 짐나스테르코리아인 B를 0, 10, 25, 50 μM 의 농도로 처리하고 1, 6, 24시간 동안 5% 이산화탄소 및 37 ℃의 조건에서 배양하였다. 배양 종료 후 세포 용해물(cell lysate)로부터 단백질을 추출하여 NQO1의 항원(Santacruz)을 붙여 분석하였다.
분석 결과 도 4에 나타난 바와 같이, 상기 짐나스테르코리아인 B의 처리 농도가 증가할수록 또한 처리 시간이 증가할수록 NQO1의 단백질발현이 농도 의존적으로 증가함을 확인하였다.
이로부터 벌개미취에서 분리한 짐나스테르코리아인 B가 해독효소계의 활성을 유도함으로서 화학적 발암 예방 효과를 나타내는 것으로 판단되었다.
제제예 1: 건강식품의 제조
1-1. 음료의 제조
꿀 522 ㎎
치옥토산아미드 5 ㎎
니코틴산아미드 10 ㎎
염산리보플라빈나트륨 3 ㎎
염산피리독신 2 ㎎
이노시톨 30 ㎎
오르트산 50 ㎎
벌개미취 추출물, 분획물, 짐나스테르코리아인 유도체 또는 염 0.48~1.28 ㎎
물 200 ㎖
상기 조성 및 함량으로 하여 통상적인 방법을 사용하여 음료를 제조하였다.
1-2. 츄잉껌의 제조
껌베이스 20 %
설탕 76.36~76.76 %
벌개미취 추출물, 분획물, 짐나스테르코리아인 유도체 또는 염 0.24~0.64 %
후르츠향 1 %
물 2 %
상기 조성 및 함량으로 하여 통상적인 방법을 사용하여 츄잉껌을 제조하였다.
1-3. 캔디의 제조
설탕 50~60 %
물엿 39.26~49.66 %
벌개미취 추출물, 분획물, 짐나스테르코리아인 유도체 또는 염 0.24~0.64 %
오렌지향 0.1 %
상기 조성 및 함량으로 하여 통상적인 방법을 사용하여 캔디를 제조하였다.
1-4. 밀가루 식품의 제조
본 발명에 따른 벌개미취 추출물, 분획물, 짐나스테르코리아인 유도체 또는 염 0.5 내지 5 중량부를 밀가루 100 중량부에 첨가하고, 이 혼합물을 이용하여 빵, 케이크, 쿠키, 크래커 및 면류를 제조하여 건강 증진용 식품을 제조하였다.
1-5. 유제품(dairy products)의 제조
본 발명에 따른 벌개미취 추출물, 분획물, 짐나스테르코리아인 유도체 또는 염 5 내지 10 중량부를 우유 100 중량부에 첨가하고, 상기 우유를 이용하여 버터 및 아이스크림과 같은 다양한 유제품을 제조하였다.
1-6. 선식의 제조
현미, 보리, 찹쌀, 율무를 공지의 방법으로 알파화시켜 건조시킨 것을 배전한 후 분쇄기로 입고 60 메쉬의 분말로 제조하였다.
검은콩, 검정깨, 들깨도 공지의 방법으로 쪄서 건조시킨 것을 배전한 후 분쇄기로 입도 60 메쉬의 분말로 제조하였다.
상기에서 제조한 곡물류 및 종실류와 본 발명에 따른 벌개미취 추출물, 분획물, 짐나스테르코리아인 유도체 또는 염을 다음과 같은 비율로 배합하여 제조하였다.

Claims (7)
- 퀴논리덕타제 효소 활성을 증가시켜 발암물질을 해독함으로써 발암 예방 활성을 갖는 하기 화학식 1로 표시되는 짐나스테르코리아인 유도체 중 어느 하나, 이들의 혼합물 또는 약학적으로 허용가능한 이의 염을 유효성분으로 함유하는 발암예방용 건강기능성 식품 조성물.[화학식 1](상기 식에서, R1 및 R2는 각각 수소, 산소, 히드록시, C1~C4의 알킬에스테로 및 C1~C4의 알콕시로 이루어지는 군으로부터 선택되는 어느 하나이고, R1 및 R2 중 어느 하나가 산소이고 다른 하나가 수소인 경우, 상기 R1 또는 R2는 이들과 인접한 두 개의 탄소와 결합하는 에폭사이드이며, 는 단일 또는 이중결합이다.)
- 제1항에 있어서, 상기 짐나스테르코리아인 유도체는,벌개미취 건조물을 물, C1~C4의 알코올, 초산에틸, 염화메틸렌 및 클로로포름으로 이루어지는 군으로부터 선택되는 용매로 추출하여 벌개미취 추출물을 얻는 단계(단계 1);상기 단계 1에서 얻은 벌개미취 추출물을 물에 현탁시킨 후, 유출 용매로 헥산/초산에틸(10:1(v/v)), 헥산/초산에틸(5:1(v/v)), 헥산/초산에틸(2:1(v/v)), 헥산/초산에틸(1:1(v/v)), 초산에틸 및 메탄올을 사용하여 컬럼 크로마토그래피를 수행하여 벌개미취 분획물을 얻는 단계(단계 2); 및상기 단계 2에서 얻은 분획물을 고속액체크로마토그래피를 수행하여 짐나스테르코리아인 유도체를 분리, 정제하는 단계(단계 3)를 포함하는 방법에 의해 분리되는 것을 특징으로 하는 발암예방용 건강기능성 식품 조성물.
- 제3항에 있어서, 상기 단계 1의 알코올은 메탄올 또는 에탄올인 것을 특징으로 하는 발암예방용 건강기능성 식품 조성물.
- 제3항에 있어서, 상기 단계 2의 벌개미취 분획물은 헥산-초산에틸(5:1) 분획 및 헥산-초산에틸(2:1) 분획을 합한 분획물인 것을 특징으로 하는 발암예방용 건강기능성 식품 조성물.
- 제3항에 있어서, 상기 단계 3은 65% 아세토니트릴/물을 유출용매로 하여 고속액체크로마토그래피를 수행하는 것을 특징으로 하는 발암예방용 건강기능성 식품 조성물.
- 제1항 내지 제6항 중 어느 한 항에 있어서, 상기 발암은 후두암, 폐암, 식도암, 췌장암, 대장암, 간암, 위암, 설암, 피부암, 뇌종양, 자궁암, 유방암, 자궁경부암, 난소암, 신장암, 담낭암, 구강암, 결장암, 간암, 방광암 및 대장암으로 이루어진 군으로부터 선택되는 것에 대한 예방인 것을 특징으로 하는 발암예방용 건강기능성 식품 조성물.
