Abstract

本発明は、標的ＤＮＡのセンス鎖とアンチセンス鎖の同時解析により、簡便、高精度に標的ＤＮＡを検出する方法、及びそのためのキットを提供する。本発明の標的ＤＮＡを検出する方法は、標的塩基配列を有するセンス鎖と、これに相補的なアンチセンス鎖とからなる標的ＤＮＡの検出方法であり、センス鎖とハイブリダイズする第１のオリゴヌクレオチドと、アンチセンス鎖とハイブリダイズする第２のオリゴヌクレオチドとを用い、アンチセンス鎖のうち、第２のオリゴヌクレオチドとハイブリダイズする領域の少なくとも一部分が、センス鎖の第１のオリゴヌクレオチドとハイブリダイズする領域の少なくとも一部と相補的であり、第１のオリゴヌクレオチドと第２のオリゴヌクレオチドとを、一の反応溶液に同時添加して、標的ＤＮＡとそれぞれハイブリダイズさせた後、ライゲーション反応及び／又はＰＣＲを行い、産物を検出する。
