Abstract
Compuesto que comprende un soporte de silica funcionalizada con grupos vinilsulfona. Además, se refiere a su procedimiento de obtención y sus usos como inmovilizador de biomoléculas, y más concretamente de enzimas tales como invertasa, lactasa, peroxidasa ó tiorredoxina h1 y de otras biomoléculas tales como glutatión, nitrosoglutation ó biotina.

La presente invención se refiere a un compuesto de silica conteniendo grupos vinilsulfona de fórmula general (I), a su procedimiento de obtención y a sus usos. Más particularmente, el uso de los compuestos de silica en aplicaciones biotecnológicas.

ESTADO DE LA TÉCNICA ANTERIOR
La inmovilización de macromoléculas a soportes sólidos se remonta a comienzos del siglo XX con los trabajos de Nelson y Griffin (cfr. Nelson, J. M. et al., 1916 J. Am. Chem. Soc. vol. 38, pp. 1109-1115) sobre inmovilización de invertasa. El interés inicial por los enzimas inmovilizados y su actividad catalítica se ha ido extendiendo a otras macromoléculas. En Ia actualidad, Ia inmovilización de macromoléculas sobre un soporte sólido y el concepto de afinidad entre moléculas juegan un papel central en Io que se ha dado en denominar Ia era de las "omics" (cfr. Sheldon, R. A., 2007, Adv. Synth. CataL vol. 349, pp.1289-1307). En este contexto se han descrito numerosas aproximaciones para inmovilizar macromoléculas a soportes con el objetivo de fabricación de microarrays (cfr. Sun, H. et al., 2006, Anal. Bioanal. Chem. vol. 386, pp. 416-426; Sun, X-L, Stabler, C. L., Cazalis, CS. , Chaikif, E. L., 2006. Bioconiuαate Chem. vol. 17, pp. 52-57).
A pesar de los avances, aún persiste Ia necesidad de efectuar una "activación" para promover dicha inmovilización. Básicamente, existen dos métodos para llevar a cabo Ia mencionada activación: (a) activación de Ia macromolécula a inmovilizar; ó (b) activación del soporte.
La primera de las metodologías (activación de Ia macromolécula a inmovilizar) ha tenido una amplia difusión y uso. En este sentido, mediante técnicas de Biología Molecular las proteínas pueden ser "activadas" vía fusión a colas o péptidos/dominios proteicos que Ie confieren afinidad por el soporte. Esta estrategia ha dado lugar al desarrollo de distintos sistemas comerciales entre los que se encuentran: (a) Colas de histidina ("His-Tag") (cfr. Hengen, P., 1995, Trends Biochem Sci. vol. 20, pp. 285-286.) en combinación con soportes conteniendo Ni (Quiagen, IBA, Amershem- Bioscience, Clontech); (b) Fragmento de Ia haloalcano deshalogenasa ("Halo-Tag") en combinación con soportes con haloalcano (Promega); (c) SNAP: O6-alquilguanina- ADN alquiltransferasa (cfr. Keppler, A..et al. 2004 Methods vol. 32, pp. 337-344) en combinación con soportes conteniendo O6-bencilguanina (Covalys); (d) Colas conteniendo péptidos con afinidad por estreptavidina ("Strep-tag") (cfr. Schmidt, TGM, et al., 1996 J. Mol. Biol. vol. 255, pp. 753-766) en combinación con soportes conteniendo estreptavidina (Novagen, IBA, Quiagen, Stratagene); (e) Glutatión transferasa en combinación con soportes conteniendo glutatión (Novagen, Clontech o Amershan-Biosciences); (f) Proteína de unión a maltosa en combinación con soportes con maltosa, (New England Biolab); (g) Péptido de unión a calmodulina en combinación con soportes conteniendo calmodulina (Stratagene); (h) Dominio de unión a celulosa en combinación con soportes conteniendo celulosa (Novagen); (i) Péptido biotinilado in vivo (cfr. Cronan, J. E., 1990, J. Biol. Chem vol. 265, pp. 10327-10333) en combinación con soportes conteniendo avidina (Promega); (j) IMPACT Inteina en combinación con soportes conteniendo quitina (New Englad Biolabs). Esta aproximación está limitada a proteínas recombinantes y generalmente conduce a una inmovilización no covalente de Ia macromolécula, hecho que determina su aplicación en purificación de proteínas mediante cromatografía de afinidad.
Otra opción para Ia activación de Ia macromolécula a inmovilizar es Ia introducción en su estructura de grupos químicos que Ie confieran reactividad o sobre los que se pueda llevar a cabo posteriores transformaciones químicas. Para proteínas, el grupo de elección es el
grupo tiol (-SH) y Ia aproximación experimental usada es Ia mutagénesis dirigida o Ia fusión a una inteina (cfr. Girish, A., et al., 2005 Bioorg. Med. Chem. Lett.. vol. 15, pp. 2447-2451 ).
La segunda de las metodologías ("activación" de los soportes) supone Ia preparación de soportes sólidos conteniendo grupos funcionales reactivos para que reaccionen y formen un enlace covalente con las macromoléculas a inmovilizar. Los soportes que tradicionalmente se vienen empleando son Ia agarosa y el poliestireno. Los procedimientos clásicos de activación del soporte son: (a) el método de aminación reductiva (Pierce: agarosa y poliestireno funcionalizados con grupos tosil, tresil, epoxi, alquilamina, aldehido, hidrazida); (b) el método de cabonildiimidazol (Quiagen: poliestireno activado para reaccionar con SH, carboxi para ser activado con grupos succinimida o carbodiimida); (c) el método de Ia N-hidroxisuccinimida éster (Hispanagar: agarosa funcionalizada con grupos glioxal ó aminoetil); (d) el método de Ia maleimida (Polygenetics: poliestireno funcionalizado con grupos amina ó CN); (e) el método de Ia hidrazida (Bangs Laboratories: poliestireno funcionalizado con grupos carboxi para ser activado con carbodiimida ó grupos amino). Los grupos diana de Ia macromolécula son las aminas para los métodos (a), (b) y (c), los grupos tiol para el método (d) y los grupos aldehido, obtenidos por oxidación de carbohidratos, para el método (e).
El uso de Ia silica/vidrio como soporte para Ia inmovilización de macromoléculas ha cobrado recientemente interés. En Ia actualidad existen en el mercado (a) silica con Concanavalina A (Biotech Support Group) que interacciona con carbohidratos, (b) silica con ácidos grasos (Nimbus Biotechnology) para incorporación de proteínas de membrana y (c) silica con albúmina sérica humana (Nimbus Biotechnology). Las estrategias de inmovilización para este soporte sólido son las mismas que las indicadas para los otros soportes: (a) fusión de Ia proteína a inmovilizar con colas de Histidina (HisLink Resin de Promega en combinación con silica con Ni de Biotech Support Group), con GST (silica con glutatión de Biotech Support Group) con péptidos que interaccionan con estreptavidina
(silicas con estreptavidina de Bangs Laboratories y de Polysciences), con péptidos que interacciona directamente con Ia silica (Arg-Tag14, Si-Tag15) b) empleo de soportes funcionalizados que permitan su fácil activación y reacción con Ia macromolécula.
Por otro lado, las sulfonas α,β-insaturadas (vinil sulfonas) son reconocidas como intermediarios sintéticos de gran utilidad debido fundamentalmente a su capacidad para participar en reacciones de adición 1 ,4 (aceptores de Michael) y en reacciones de cicloadición (como donores 2π). Adicionalmente, las vinilsulfonas son fáciles de preparar, a través de una amplia variedad de procesos sintéticos, y de manipular (Simphinks, N. S., 1990, Tetrahedron vol. 282, pp. 6951-6984; Meadows, D. C, et al., 2006, Med. Res. Rev. vol. 26(&) pp. 793-814). Estas características han encontrado recientemente utilidad en el diseño de fármacos y en química médica cuando se demostró su capacidad para inhibir de forma potente y reversible una variedad de procesos enzimáticos, fundamentalmente aquellos en los que están implicados cistein proteasas a las que se unen a través de reacciones de adición con el grupo tiol presente en el residuo de cisteína del sitio activo de estas enzimas, (cfr. Simphinks, N. S., 1990, Tetrahedron vol. 282, pp. 6951-6984; Meadows, D. C, et al., 2006 Med. Res. Rev. vol. 26(6), pp. 793-814)
La alta reactividad del grupo vinilsulfona ha sido explotada previamente en Ia preparación de distintos materiales conteniendo esta funcionalización: (i) poliestireno (comercializados por Aldrich) (ii) agarosa (comercializada por Gentaur) y (iii) sefarosa (comercializada por Affiland). Estos materiales han sido utilizados para el acoplamiento covalente con compuestos conteniendo grupos amina, tioles e hidroxilo habiendo encontrado aplicabilidad en Ia inmovilización de biomoléculas portadoras de estos grupos funcionales.
Además, el grupo vinilsulfona se usa en Ia preparación de intermedios de síntesis para Ia posterior obtención de materiales más elaborados, que contienen soportes de silica. Estos materiales se suelen denominar resinas tiofílicas o "T-GeI, (cfr. Schwarz A. et al., 1994 Reactive Polvmers vol. 22,
pp. 256-266; GB 2 230 003 A; US 4,696,980; DE 10330204 A1 ). En este tipo de materiales el concepto clave es el de adsorción tiofílica descrito por Porath (cfr. Porath et al., 1984, Phvsical Chemistrv of Colloids and Macromolecules. pp 137-142; Porath J. et al., 1985, FEBS vol. 185 (2), pp. 306-310). La adsorción tiofílica se basa en Ia afinidad de macromoléculas por resinas del tipo SOPORTE-O-CH2-CH2-SO2-CH2-CH2-S-R (R= un residuo alifático pequeño) y en Ia actualidad es usada como una técnica alternativa para Ia inmovilización no covalente y purificación de moléculas biológicas (cfr. Hutchens T.W. et al., 1987, Biochemistrv vol. 26, pp. 7199- 7204). Para Ia preparación de estas resinas tiofílicas Ia estrategia utilizada supone Ia reacción de divinilsulfona, como reactivo difuncional, con un compuesto de silica conteniendo grupos hidroxilo, silica recubierta con dextrano ó grupos tiol obteniéndose así un material activado que se hace reaccionar normalmente in situ con tiol derivados sencillos.
EXPLICACIÓN DE LA INVENCIÓN
En Ia presente invención se proporciona un nuevo compuesto que comprende silica funcionalizada con grupos vinilsulfona a través de una cadena conteniendo un grupo funcional del tipo tiol o amina. Además de proporcionar el procedimiento de obtención de los mismos y sus usos, en particular en Ia inmovilización de biomoléculas dentro del campo de Ia biotecnología.
El nuevo compuesto proporciona una técnica de inmovilización que no requiere ninguna estrategia de activación y que combina las propiedades de Ia silica como soporte con Ia reactividad de Ia vinilsulfona con grupos presentes de forma natural en las biomoléculas en condiciones de reacción suaves compatibles con su naturaleza biológica.
El uso de silica como material para soportar grupos vinilsulfona supone una serie de importantes ventajas con respecto a otros materiales poliméricos como, por ejemplo, polisacáridos o poliestireno:
o Estabilidad mecánica. El material puede ser sometido a agitación
mecánica sin sufrir alteraciones en su tamaño Io que permite llevar a cabo procesos en "baten". Otro aspecto importante es Ia estabilidad a alta presión de Ia silica Io que permite su uso en HPLC.
o Estabilidad química y biológica. El material no es susceptible de sufrir degradaciones por Ia acción de microorganismos y agentes químicos (por ejemplo ácidos).
o Estabilidad térmica. Es posible utilizar un amplio rango de temperaturas sin que el material sufra descomposiciones térmicas. El material es también susceptible de ser usado en procesos asistidos con microondas.
o Independencia del disolvente. El material no sufre ni encogimiento ni hinchado como consecuencia de Ia acción de los disolventes. Además, no es soluble en ningún disolvente Io que facilita Ia operatividad.
o Facilidad de uso en Io que respecta, fundamentalmente, al manejo y pesado. No se requieren lavados extensos. El material no se pega ni al vidrio ni al plástico. Por estas razones, el material es susceptible de ser usado en procesos automatizados.
o Flexibilidad de formatos. Es posible preparar fácilmente silicas funcionalizadas con un amplio rango de tamaños de partícula. Al no sufrir hinchamiento en contacto con disolventes estos materiales pueden ser empaquetados en una variedad de flujos a través de formatos tales como columnas de HPLC, cartuchos flash.
o Reproducibilidad. Permiten cargas de material exactas incrementando el grado de reproducibilidad en sus aplicaciones.
o Optimización de los recursos de partida: silica, silanoles y divinilsulfona (DVS) que son productos de partida baratos. Optimización de las condiciones de reacción: temperatura ambiente, disolventes baratos, y
tiempo de reacción. Ambos factores permiten obtener un producto competitivo en el mercado.
Los materiales que se describen en el estado de Ia técnica, denominadas resinas tiofílicas o "T-GeI", han sido utilizados como soportes de cromatografía de afinidad explotándose las interacciones tiofílicas reversibles de este tipo de materiales con biomoléculas. La función vinilsulfona, en estos materiales, es utilizada para posibilitar Ia formación de grupos éter ó tioeter en Ia construcción de las cadenas de tipo 0-CH2- CH2-SO2-CH2-CH2-S-R unidas a los soportes. Sin embargo, en estas resinas tiofílicas, Ia función vinilsulfona ha sido utilizada para que los materiales que Ia contienen puedan llevar a cabo Ia unión no covalente en Ia inmovilización de biomoléculas. Mientras que en Ia invención se produce unión covalente en Ia inmovilización de biomoléculas con el uso del compuesto de silica-vinilsulfona.
Además, se ha comprobado, en Ia presente invención, que Ia incorporación de Ia función vinilsulfona a Ia silica se puede realizar en más de una etapa usando bisvinilsulfonas a partir de silica comercial. Se requiere el uso de un compuesto de silica funcionalizada con grupos que tengan un carácter nucleófilo, preferentemente grupos tioles o aminos, con capacidad para reaccionar fácilmente con bisvinilsulfonas. Estos grupos tioles o aminos pueden estar unidos al soporte inorgánico a través de un espaciador de longitud y naturaleza variable. Sin embargo, Ia utilización de silicas funcionalizadas conteniendo aminas primarias no es factible dado que en Ia reacción de estos compuestos con bisvinilsulfonas se produce una segunda delación intramolecular con formación de compuestos cíclicos. La solución a este inconveniente supone Ia preparación de un compuesto de silica conteniendo aminas secundarias que en Ia reacción con bisvinilsulfonas permiten Ia obtención de los compuestos de silica de Ia invención.

Por "alquilo" se refiere en Ia presente invención a cadenas alifáticas, lineales o ramificadas, que tienen de 1 a 10 átomos de carbonos, por ejemplo, metilo, etilo, n-propilo, i-propilo, n-butilo, t-butilo, n-pentilo, etc.
Por "alquenilo" se refiere en Ia presente invención a un radical alquilo que tiene entre 1 y 10 átomo de carbono y que tiene uno o más enlaces insaturados. Los radicales alquenilo pueden estar opcionalmente sustituidos por uno o más sustituyentes tales como un arilo, halógeno, hidroxilo, alcoxilo, carboxilo, ciano, carbonilo, acilo, alcoxicarbonilo, amino, nitro, mercapto, etc.

Otro aspecto de Ia presente invención se refiere al procedimiento de obtención del compuesto de fórmula general (I) que comprende: a. Obtención de silica funcionalizada con grupos tiol o amina. b. Reacción del compuesto de silica obtenido en el paso (a) con divinilsulfona en presencia de una base orgánica.
Una realización preferida del procedimiento de Ia invención comprende una amina terciaria como base orgánica que se puede seleccionar del grupo que comprende, sin limitarse a, trietilamina ó diisopropiletilamina
Otra realización más preferida del procedimiento de Ia invención, comprende en Ia reacción del paso (b) el uso de disolventes que se pueden seleccionar entre mezclas de THF con un alcohol como, por ejemplo isopropanol o terc-butanol, preferiblemente isopropanol.
A continuación se describe esquemáticamente el procedimiento de Ia invención:

Aún otro aspecto de Ia presente invención se refiere al uso del compuesto de fórmula general (I) para inmovilizar biomoléculas. Esta inmovilización se produce mediante Ia unión covalente entre Ia biomolécula y el grupo vinilsulfona presente en el compuesto de Ia invención. Este método de inmovilización covalente se lleva a cabo en condiciones de reacción suaves que son compatibles con Ia naturaleza biológica de las biomoléculas a través de grupos funcionales ya presentes en las mismas macromoléculas en un número considerable y accesible, por Io que no hay que realizar modificaciones químicas o mutagénesis dirigida para funcionalizarlas. La alta reactividad de Ia función vinilsulfona, en el compuesto de Ia invención, permite inmovilizar una amplia variedad de biomoléculas. Se describen varios casos en los que se pone de manifiesto Ia capacidad de inmovilización covalente de diferentes biomoléculas en términos de peso molecular, punto isoeléctrico y función. Los ejemplos se refieren, pero sin limitarse, a Ia posibilidad de unir el compuesto de Ia invención a enzimas (invertasa, lactasa y peroxidasa), para poner de manifiesto que tras Ia inmovilización mantienen Ia actividad catalítica, y a péptidos/proteínas (glutatión, nitroso glutatión y tiorredoxina h del guisante), para demostrar que Ia inmovilización transforma Ia silica- vinilsulfona en un soporte de cromatografía de afinidad apto para Ia purificación y experimentos de "pull-down".
Además, el compuesto de fórmula general (I) puede inmovilizar cualquier biomolécula que tenga grupos amina y/o tioles y usarse como soportes cromatográficos para cromatografía de afinidad según Ia necesidad en cada momento. Los resultados son extrapolables a lípidos y carbohidratos y otros sistemas que reaccionen con Ia vinilsulfona.
A Io largo de Ia descripción y las reivindicaciones Ia palabra "comprende" y sus variantes no pretenden excluir otras características técnicas, aditivos, componentes o pasos. Para los expertos en Ia materia, otros objetos,
ventajas y características de Ia invención se desprenderán en parte de Ia descripción y en parte de Ia práctica de Ia invención. Los siguientes ejemplos y dibujos se proporcionan a modo de ilustración, y no se pretende que sean limitativos de Ia presente invención.

EXPOSICIÓN DETALLADA DE MODOS DE REALIZACIÓN
A continuación se ilustrará Ia invención mediante unos ensayos realizados por los inventores, que ponen de manifiesto Ia especificidad y efectividad de los compuestos de Ia invención.
EJEMPLO 1.- Preparación del compuesto de silica-vinilsulfona de fórmula (II).
Primeramente, se transformó silica en el derivado mercaptopropil-sílica (IV). Este derivado es un compuesto conocido (cfr. Howard, A. G et al., (1987), Analvst vol. 112, pp. 159) que se obtiene por reacción de silica con 3-mercaptopropiltrimetoxisilano y que también se puede obtener comercialmente (Silicycle).
El compuesto de fórmula (II) se obtuvo por reacción del compuesto (IV) con divinilsulfona según el siguiente procedimiento: La mercaptopropil- silica (1 g) se suspendió en una mezcla de THF-isopropanol (1 :2, 10 ml_) que se burbujea con argón para posteriormente añadir DVS (0.515 ml_, 5 eq) y trietilamina (0.028 ml_, 0.2 eq). La mezcla de reacción se mantuvo a temperatura ambiente durante 16 h. Tras ese tiempo, se filtró el compuesto resultante de fórmula (II) que fue lavado con acetona y secado en una estufa de vacío a 50 0C.
EJEMPLO 2.- Preparación del compuesto de silica-vinilsulfona de fórmula (III).
El compuesto de formula (III) se obtuvo en un procedimiento de dos etapas:
a) Primero se obtuvo el compuesto 3-(metilamino)-propil-silica, de fórmula (V) (X=N R1=Me), por silanización de silica: Se suspendió silica (5 g) activada previamente por tratamiento térmico (120 0C a vacío durante 24 h) en tolueno (25 mL) y se adicionó 3-(metilamino)-propil- trimetoxisilano (1 ,250 g) calentándose a reflujo durante 2 h. Se evaporó un 20% del disolvente (5 mL) y se volvió a poner a reflujo durante 1 h adicional. El compuesto resultante de fórmula (V) se filtra y seca. b) Reacción del compuesto 3-(metilamino)-propil-silica (V, X=N R1=Me) con DVS: 1 .53 g del compuesto de fórmula (V) fueron suspendidos en una mezcla de THF-isopropanol (1 :2, 10 mL) y se Ie añadió DVS (0.640 mL, 5 eq). La mezcla de reacción se mantuvo a temperatura ambiente
durante 16 h. Tras ese tiempo, el compuesto resultante de fórmula (III) se filtró y lavó sucesivamente con metanol y cloruro de metileno, secándose posteriormente en una estufa de vacio a 50 0C.
EJEMPLO 3. Protocolo general de inmovilización en discontinuo ("baten")
A una suspensión de sílica-vinilsulfona de fómula (II) ó (III) (0.5 g) en un tampón que no contenga aminas libres, como por ejemplo fosfato o HEPES, de fuerza iónica moderada (50 - 200 mM) y pH básico (7,5 -8,7) (10 ml_) se adicionó Ia molécula a inmovilizar (0.6 micromoles) y se dejaron reaccionar durante tiempo suficiente (entre 6 horas y 10 horas) con agitación. Posteriormente se adicionó mercaptoetanol (8 micromoles) y se agitó durante 1 hora. Los excesos de reactivos se eliminaron mediante sucesivos lavados con una solución de NaCI 2M y Ia resina (proteína inmovilizada en Ia silica-vinilsulfona) se equilibró con el tampón en el que se realizaron los experimentos posteriores.
EJEMPLO 4. Protocolo general de inmovilización en columna.
La inmovilización de una biomolécula puede ser llevada a cabo directamente sobre silica-vinilsulfona empacada en una columna haciendo recircular una solución de Ia biomolécula de interés en un tampón que no contenga aminas libres, como por ejemplo fosfato o HEPES, de fuerza iónica moderada (50 - 200 mM) y pH básico (7,5 -8,7) durante un tiempo suficiente (en función del flujo de Ia columna y del volumen de Ia solución de macromolécula. A modo de ejemplo, para 49 mi de solución de macromolécula y un flujo de de 0.5 ml/min, el tiempo de reacción fue de 14 horas). El empacado se llevó a cabo suspendiendo Ia silica vinil-sulfona en el tampón anteriormente mencionado y de forma general se utilizaron 3 g de silica vinil sulfona/micromol de biomolécula. Para bloquear los grupos vinilsulfona que no hayan reaccionado se trató el material con una disolución de β-mercaptoetanol (16 micromoles por gramo de vinil-sílica empacada) durante tiempo suficiente (en función del flujo de Ia columna y del volumen de Ia solución) a temperatura ambiente. Este protocolo
permite Ia transformación directa de vinil-sílica previamente empacada en una columna cromatográfica y/o reactor enzimático basado en Ia macromolécula que se inmoviliza.
EJEMPLO 5. Inmovilización de Ia invertasa.
La invertasa (beta-fructofuranosidasa [EC3.2.1.26]), enzima que cataliza Ia hidrólisis de sacarosa en fructosa y glucosa, es elegida como enzima modelo teniendo en cuenta que (a) Ia proteína es comercial, (b) el sustrato es barato, (c) Ia actividad puede ser fácilmente medida en el laboratorio y (d) tiene aplicación industrial en el sector alimentario, donde se prefiere Ia fructosa sobre Ia glucosa por ser más dulce y no cristalizar tan fácilmente.
La inmovilización de invertasa de levadura comercial (10 mg, Sigma I- 4504) se realizó según el protocolo general mediante una reacción en discontinuo ("baten"). El estudio de Ia influencia de Ia temperatura sobre Ia reacción (FIG. 1 ) muestra que a las 10 horas Ia reacción es completa, independientemente de Ia temperatura utilizada (temperatura ambiente ó
40C). El análisis de Ia funcionalidad de Ia invertasa inmovilizada (FIG. 2) demuestra que el enzima mantiene su actividad.
EJEMPLO 6. Inmovilización de Ia lactasa
La lactasa (glicosidasa hidrolasa [EC3.2.1.23]), enzima que cataliza Ia hidrólisis de lactosa en glucosa y galactosa, es elegida como enzima a inmovilizar teniendo en cuenta el alto interés que ello tiene tanto en Ia industria alimentaria, dado que Ia lactosa es poco soluble y su capacidad edulcorante es insuficiente, como en medicina, debido a Ia intolerancia a este disacárido.
La inmovilización de lactasa de Kluyveromyces lactis (Sigma G-3665) se llevó a cabo en columna, siguiendo el protocolo general arriba indicado, empacando Ia silica-vinil sulfona (2.5 g) y haciendo recircular una solución de lactasa (49 mL) a temperatura ambiente a un flujo de 0.5 mL/min.
Para estudiar Ia actividad de Ia enzima inmovilizada se midió el poder rotatorio específico de Ia muestra de lactosa a distintos intervalos de tiempo sobre Ia base de que Ia hidrólisis de Ia lactosa en glucosa y galactosa origina un incremento del poder rotatorio (FIG. 3). El estudio muestra que Ia inmovilización de lactasa sobre silica-vinilsulfona previamente empacada es factible y que el material así obtenido mantiene Ia actividad del enzima.
EJEMPLO 7. Inmovilización de peroxidasa.
Las peroxidasas [EC1.11.1.x] son enzimas que catalizan Ia reducción de peróxido de hidrógeno con Ia ayuda de un sustrato que pierde dos átomos de hidrógeno. Los enzimas con capacidad redox son capaces de provocar Ia polimerización de compuestos fenólicos en presencia de peróxido de hidrógeno. Estos polímeros fenólicos son inocuos o menos tóxicos e insolubles en agua, facilitando su eliminación. En este contexto Ia inmovilización de peroxidasa es interesante. Además, al tratarse de una glicoproteína es un buen modelo para demostrar Ia capacidad de Ia silica vinil-sulfona en Ia inmovilización de este tipo de biomoléculas. En concreto se seleccionaron las peroxidasas de rábano picante y alcachofa.
La inmovilización de las peroxidasa (10.9 mg) fue llevada a cabo usando el protocolo general mediante una reacción en discontinuo ("batch").
Para demostrar que Ia inmovilización de peroxidasa de alcachofa ha tenido lugar se midió Ia actividad enzimática. El enzima inmovilizado es activo.
EJEMPLO 8. Inmovilización de glutatión
El glutatión es un tripéptido de interés en biotecnología como elemento presente en resinas comerciales de uso en purificación de proteínas de fusión con glutatión-S- transferasa (GST) basadas en Ia afinidad de Ia GST por el glutatión inmovilizado (cfr Smith, D. B. and Johnson, K. S. ,1988, Gene vol. 67, pp. 31-40). Las proteína diana se purifican casi a homogeneidad en un solo paso y las condiciones de elución con glutatión
reducido son suaves, evitando desnaturalización de Ia proteína.
La inmovilización de glutatión comercial (2.4 mg Sigma G4251 ) se realizo según el protocolo general mediante una reacción en "baten" usando 1 g de silica-vinil-sulfona. Los grupos vinilsulfona que no han reaccionado se bloquean con β-mercaptoetanol (563 μL).
La silica-glutation fue empacada en una columna y empleada para purificar una proteína de fusión con GSH. Un lisado de un cultivo de bacterias que expresaron Ia proteína de fusión se pasó a través de Ia columna de silica- glutatión. Se lavó Ia columna con 200 mi de HEPES 100 mM pH 7,5 y NaCI 200 mM. La elución se realizó con 50 mM de glutatión en 50 mM Tris-HCI pH 7,5.
EJEMPLO 9. Inmovilización de nitrosoglutatión
El S-nitrosoglutatión (GSNO) es un tripéptido que transfiere NO a proteínas con grupos tiol. Dada Ia importancia del óxido nítrico como molécula implicada en aspectos claves de Ia regulación de los sistemas inmune, cardiovascular y nervioso, Ia inmovilización de GSNO se llevó a cabo para poder obtener un soporte de afinidad cromatográfica basado en este péptido y poder realizar experimentos de "pull-down" para identificar proteínas susceptibles de ser nitrosadas (proteínas diana).
La preparación de este material se llevó a cabo en un proceso en tres etapas: a) Inmovilización de L-glutatión oxidado (GSSG) comercial (2.5 mg Sigma G4501 ). Se lleva a cabo según el protocolo general en "baten" usando 1 g de resina. Los grupos vinilsulfona que no han reaccionado se bloquean por tratamiento con β-mercaptoetanol (563 μL). b) Reducción del GSSG y desprotección del grupo -SH del L-glutatión (GSH) inmovilizado. Se lleva a cabo por tratamiento del material obtenido en Ia anterior etapa con DTT (14 mg) durante 2 h. a 20 0C c) Nitrosación del GSH inmovilizado y obtención de silica-GSNO. Se lleva a cabo según el procedimiento de Lamas y col. (Biochem J. 2000, 349,
567) por tratamiento con HCI 10 mM y NaNO2 10 mM (10 mL) durante 1 h a 20 0C. Se filtra y repite el tratamiento durante 30 min. Se filtra y lava con HEPES 40 mM pH 7.5 y NaCI 50 mM.
Para comprobar Ia eficacia de Ia inmovilización y Ia aplicabilidad de Ia silica-GSNO, se procedió a realizar un experimento de "pull-down" a partir de un extracto de girasol según el procedimiento que se indica: Ia silica- GSNO se empacó usando 250 mi de HEPES 50 mM pH 7.5 y NaCI 50 mM. Se cargó un extracto de girasol (150 mi), se lavó con NaCI 200 mM para despegar las proteínas unidas a Ia columna mediante interacciones no específicas y se eluyó selectivamente con fuerza iónica (1.5 M NaCI) y con poder reductor (10 mM DTT en 1.5 M NaCI). El cromatograma obtenido (FIG.4) muestra que los picos de proteína eluida (Absorbancia a 280) presentaban Ia señal característica del enlace S-NO (Absorbancia a 340), es decir, que las proteínas retenidas por Ia resina son y/o se han nitrosado, por Io que constituyen potenciales dianas del GSNO.
EJEMPLO 10. Inmovilización de tiorredoxina h1.
Las tiorredoxinas, enzimas redox que actúan como interruptores que modulan Ia actividad de numerosas proteínas vía el intercambio de puentes disulfuro, han sido elegidas como proteínas de interés para demostrar Ia eficacia de Ia silica vinil-sulfona para Ia inmovilización de proteínas. La inmovilización de Ia tiorredoxina h1 de guisante no es conocida y experimentos de "pull-down" son una estrategia viable para identificarla.
La inmovilización de Ia tiorredoxina h1 de guisante recombinante con cola de His expresada en E. coli (6.08 mg) fue llevada a cabo usando el protocolo general mediante una reacción en "batch" y Ia alta reactividad de los restos de histidinas presentes en Ia cola de esa proteína hacia los grupos vinil-sulfona es aprovechada para favorecer Ia orientación de Ia molécula (Ia molécula reaccionará preferentemente con Ia cola de polihistidina, que está en Ia cara opuesta al centro activo, por Io que éste queda accesible).
Para demostrar Ia inmovilización de Ia tiorredoxina h1 y Ia aplicabilidad del material obtenido se llevaron a cabo experimentos de "pull-down" siguiendo el esquema de elución expuesto en el ejemplo anterior: se pasa el extracto, se lava Ia columna con 200 mM NaCI para despegar las proteínas unidas a Ia columna mediante interacciones no específicas y se eluye selectivamente con fuerza iónica (1.5 M NaCI) y con poder reductor (10 mM DTT en 1.5 M NaCI). Una electroforesis bidimensional (FIG. 5) de las fracciones recogidas en el experimento revela que sólo unas pocas proteínas han sido retenidas por Ia columna, Io cual apoya Ia especificidad de Ia interacción y Ia viabilidad de Ia silica-vinilsulfona como resina sobre Ia que llevar a cabo experimentos de "pull-down".
EJEMPLO 11. Inmovilización de biotina
La inmovilización de aminobiotina comercial (0.1 g de biotina Merck) se llevó a cabo usando el protocolo general mediante una reacción en "baten" con las siguientes modificaciones: 1.25 g de resina, una mezcla de THF- isopropanol (2:1 , 10 mi) como disolvente y trietil amina (38 mg) como base. Tras un periodo de reacción de 14 h, se filtró y lavó con cloruro de metileno:metanol (1 :1 ).
Para demostrar que Ia inmovilización de biotina ha tenido lugar se llevó a cabo una cromatografía de afinidad para el aislamiento y purificación de avidina.

9. Método de obtención de un compuesto según cualquiera de las reivindicaciones 1 a 8, que comprende: a. obtención de silica funcionalizada con grupos tiol o amina; b. reacción del compuesto de silica obtenida en el paso (a) con divinilsulfona en presencia de una base orgánica.
10. Método según Ia reivindicación 9, donde Ia base orgánica usada en el paso (b) es trietilamina
11. Método según cualquiera de las reivindicaciones 9 ó 10, que además comprende el uso de THF-isopropanol como disolvente de Ia reacción del paso (b).
12. Compuesto de fórmula general (V):
donde n y R1 están descritos en Ia reivindicación 1.
13. Compuesto según Ia reivindicación 12, donde n es 3.
14. Compuesto según cualquiera de las reivindicaciones 12 ó 13, donde R1 es metilo.
15. Uso de un compuesto según cualquiera de las reivindicaciones 1 a 8, como inmovilizador de biomoléculas.
16. Uso del compuesto según Ia reivindicación 15, donde las biomoléculas son enzimas, glutatión, nitrosoglutatión o biotina..
17. Uso del compuesto según Ia reivindicación 16, donde los enzimas se seleccionan de entre invertasa, lactasa, peroxidasa ó, tiorredoxina h1.
