Abstract

代理親魚技法により移植したドナー魚由来の生殖細胞が異種レシピエント魚生殖巣内で増殖・成熟しているかを調べるためには、生殖細胞に特異的に発現しており、なおかつ、レシピエント魚とドナー魚を判別できる形質を指標にする必要がある。 　生殖細胞特異的遺伝子であるＶａｓａ遺伝子は、始原生殖細胞および精原／卵原細胞に特異的であり、体細胞には発現が認められない。本発明では、マグロ、マサバ、ゴマサバ、スマ及びニベのＶａｓａ遺伝子配列を決定し、その遺伝子発現を生殖細胞のマーカーとして用いることとした。さらに、本発明によると、魚類で高度に保存されたＶａｓａ遺伝子配列の中から、シークエンス解析をすることなく、マグロＶａｓａ遺伝子のみを特異的に検出することが可能であるため、レシピエント魚生殖腺中のマグロ由来生殖細胞を確実かつ簡便に識別することができる結果、マグロの増殖や育種を効率よく行うことが可能となる。また、上記の知見を応用することにより、マグロのみならず、他のスズキ目魚類をドナーとして用いた場合にも、ドナー魚由来の生殖細胞を異種レシピエント魚の生殖腺内から効率よく検出できる。
