Vorrichtung und Verfahren zur Strukturanalyse und etektion von komplexen Glykostrukturen
Die Erfindung betrif ft eine Vorrichtung und ein Ver- fahren zur Bestimmung und zur Analyse von komplexen Glykostrukturen .
Kohlenhydrate können in Monosaccharide, Disaccharide . Oligosaccharide und Polysaccharide unterteilt werden . Die Oligo- und Polysaccharide können wiederum in Homoglykane oder Heteroglykane gegliedert werden, wobei die Heteroglykane mit den Vertretern der Proteo- glykane , Peptidoglykane , Glykolipide und Glyko- proteine zu den biologisch wichtigen Strukturein- hei en gehören . Zusammengesetz e Kohlenhydrate , die konj ugierte Verbindungen umfassen, wie beispielsweise Glykolipide und Glykoproteine , werden auch als Glyko- konj ugate bezeichnet . Glykokonjugate gehören zu den zentralen Biomolekülen der Seile und sind an vielen physiologischen Prozessen beteiligt . Sie haben unter anderem Funktionen in der Immunabwehr , Seildifferenzierung, bei Entzündungsprozessen sowie allgemein bei Seil - Seil - Interaktionen . Aufgrund ihrer Eigenschaften und vielfältigen Funktionen im Organismus spielt die
Struktur der Glykokonjugate in den Bereichen der Grundlagenforschung, der Diagnostik, der Pharmafor- schung sowie für die Arzneimittelentwicklung eine große Rolle. Der Begriff Glykokonjugate bezeichnet Moleküle, bei denen ein Kohlenhydratanteil (Mono-, Oligo- oder Polysaccharid) kovalent an ein Protein oder ein ipid gebunden ist. Die Vielfalt innerhalb dieser Substanzklasse ist sehr groß, denn zu den Glykokonjugaten gehören neben den Glykoproteinen und Glykolipiden, bei denen der Protein- bzw. Lipidanteil überwiegt, auch die Lipopolysaccharide und die Pro- teoglykane, bei denen der Kohlenhydratanteil überwiegt. Der Kohlenhydratanteil besteht bei den Glykoproteinen und Glykolipiden typischerweise aus bis zu 20 Monosacchariden, bei Lipopolysacchariden und Pro- teoglykanen kann dieser erheblich größer sein. In der Biosynthese dieser komplexen Glykokonjugate nutzt die Natur neben der Auswahl an unterschiedlichen Mono- saccharid-Bausteinen die verschiedensten Ver- knüpfungs- und Verzweigungsmöglichkeiten, so dass eine gigantische Strukturvielfalt innerhalb dieser Substanzklasse vorliegt. Kohlenhydrate weisen innerhalb der biologischen Bindungspartner von Proteinen in einer kurzen Sequenz potentiell die größte Anzahl von Variationsmöglichkeiten auf. Unter Verwendung einer üblichen Bibliothek von 20 verschiedenen Monosacchariden ergeben sich für die Bildung eines Hexasaσcharids (einschließlich verzweigter Isomere) theoretisch 1015 verschiedene Strukturen.
Wenn die Oligoεaccharidketten verzweigt sind, spricht man von oligoantennären Verbindungen, wobei normalerweise di- bis pentaantennäre Strukturen vorliegen. Die dadurch gegebene "Multivalenz" der Moleküle in Bezug auf die Wechselwirkung mit einem Bindungs- partner ist ein wichtiger Aspekt für die Spezifität und Stärke der Bindung. An jeder glykosylierten Stelle eines Proteins können die Oligosacharid- strukturen variieren. Die unterschiedlich glyko- sylierten Varianten bezeichnet man als Glykoformen dieses Proteins oder auch als Mikroheterogenität .
Im Gegensatz zu Monosacchariden bzw. Disacchariden oder anderen einfach aufgebauten Molekülen ist die schnelle, einfache und sichere Strukturanalyse von Glykokonj ugaten mit bisherigen Vorrichtungen und Methoden nur bedingt möglich. In den genannten Bereichen, in denen an und mit Glykokonjugaten geforscht wird, sind analytische Verfahren zur Auf- klärung der Identität des BindungsVerhaltens und der Struktur dieser Komponenten jedoch essentiell. Die bei Mono- und Disacchariden mit Erfolg angewandten physikalisch-chemischen Analyseverfahren können nicht mit ausreichendem Erfolg auf den Bereich der Glyko- konjugate übertragen werden, da die Methoden aufgrund der Komplexität der Sielstrukturen sehr zeitaufwendig, kompliziert und wenig spezifisch sind. Aufgrund der großen Bedeutung der Glykokonjugate gab es
im Stand der Technik mehrere Bestrebungen, deren Struktur sicher und effektiv zu detektieren.
Es wurde beispielsweise versucht, über massenselek- tive Verfahren eine Aussage über die Struktur zu treffen. Jedoch geben solche Verfahren keine Auskunft über das biospezifische Bindungsverhalten einer Substanz. So erfolgt die Identifizierung und Quantifizierung der Glykosilierungsmuster von Glyko- proteinen durch massenselektive Verfahren. Dabei werden die Glykoproteine zunächst aufgespalten, das heißt, die zu untersuchende Struktur wird zunächst zerstört und die Glykane oder Glykopeptide werden nach chromatographischer Auftrennung mittels Massen- Spektroskopie charakterisiert. Bei einem v/eiteren Verfahren zur Charakterisierung der Glykokonjugate, der Affinitätschromatographie, werden mit Hilfe von Lektinen, die an Gelpartikel gebunden sind, Sacchari- de oder Glykokonjugate für den Einsatz weiterer Analyseschritte aufgereinigt . Um Substanzmischungen zu fraktionieren oder Strukturinformationen über Glykokonjugate zu erhalten, steht vor allem das Verfahren der seriellen Lektin-Affinitätschromatographie zur Verfügung, bei dem die Probenkomponenten im Durchflussverfahren über -eine Kombination von Lektin- Adsorbentien fraktioniert werden.
Bei den genannten Techniken ist es nachteilig, dass eine Aufreinigung der Glykokonjugate notwendig ist,
da jegliche Verunreinigungen die Analytik behindern. So ist es erforderlich, dass oft mit größeren Mengen des aufgereinigten Glykokonjugats - häufig im Milligramm-Bereich - gearbeitet werden muss, was einen großen präparativen Aufwand erfordert. Auch ist es bisher nicht möglich, Glykostrukturen in einem einheitlichen Verfahren zu detektieren, so dass die Analytik von Glykokonjugaten mit herkömmlichen Methoden immer aus mehreren Teilschritten besteht und daher sehr zeitintensiv ist. Ein zentraler Nachteil der herkömmlichen Analyseverfahren ist jedoch, dass die Glykostrukturen nicht zerstörungsfrei analysiert werden können, das heißt, die Probe befindet sich nicht mehr in ihrem nativen Sust nd, wodurch auch eine einmal analysierte Probe, die Glykostrukturen um- fasst, in der Regel nicht weiterverwendet werden kann. Komplexe Strukturen, wie z.B. Glykoproteine mit mehr als zwei oder drei mehr oder weniger verzweigten Sucker-Ketten (Glykanen) , werden von den bekannten Verfahren in der Regel nicht erfasst.
Aufgabe der Erfindung ist es daher, eine Vorrichtung bereitzustellen, mit der komplexe Kohlenhydrats ruk- turen, wie Glykokonj gate, schnell, einfach, sicher und effizient detektiert und in ihrer Struktur bestimmt werden können.
Die Erfindung löst dieses technische Problem durch die Bereitstellung eines Mikroarrays, umfassend min-
destens zwei voneinander verschiedene immobilisierte, gegen die Grundstruktur eines Glykokonjugates gerichtete Lektine, wobei die Grundstruktur mindestens ein Glykan, enthaltend mindestens zwei glykosidisch ver- knüpfte Monosaccharide, umfasst.
Der Erfindung liegt demgemäß die überraschende Lehre zugrunde, dass komplexe Glykokonjugate, wie Glykopro- teine oder Glykolipide, nicht nur mit einem Lektin spezifisch wechselwirken, sondern dass sie in der Lage sind, so mit mehreren Lektinen in messbare Interaktion zu treten, dass eine Analyse der Struktur der Glykokonjugate - des Analyten - möglich ist. Durch diese Wechselwirkung ein und desselben Analy- ten, mit mehreren verschiedenen Lektinen, entstehen auf dem Array mehrere Bereiche oder Orte mit einer speziellen Lektin-Sucker-Wechselwirkung, die mit verschiedenen, dem Fachmann bekannten, Methoden detek- tiert werden können. Diese Orte der spezifischen Wechselwirkung ergeben auf dem Mikroarray ein bestimmtes Muster, insbesondere wenn die Lektine auf dem Mikroarray in einer bestimmten geometrischen Ordnung - beispielsweise in mehreren Linien oder in einem Raster - immobilisiert sind. Durch bekannte Algorithmen der Bioinformatik und Strukturanalytik ist es möglich, von diesem Muster auf die Struktur und das Bindeverhalten des Glykokonjugates bzw. auf die Glykosylierung einer Struktur zu schließen. Es kann beispielsweise vorgesehen sein, dass zwei, drei,
vier oder mehrere Dutzend verschiedene Lektine auf einem geordneten Raster - immobolisiert sind. Wenn die Bereiche, in denen Lektin-Kohlenhydrat Wechselwirkungen oder Lektin-Sucker Bindungsereignisse stattfinden, mit einem Marker detektiert werden bzw. wenn durch diese Wechselwirkung zum Beispiel ein elektrischer Impuls freigesetzt wird, der gemessen werden kann, bildet sich für jedes zu untersuchende Glykokonjugat ein spezifisches Muster oder ein Impulsalgorithmus heraus, anhand dessen analoge bzw. homologe Strukturen bestimmt werden können. Während Monosaccharide, Disacchariden sowie einfach aufgebaute Oligosaccharide und einfache, nicht komplexe Polysaccharide mit physikalischen oder chemischen Me- thoden ausreichend charakterisiert werden können, ist dies bei komplexen Glykokonjugaten nicht möglich. So ist es aber mit Vorteil möglich, komplexe Glykokonjugate mit dem erfindungsgemäßen Mikroarray einfach, sicher und schnell zu detektieren und zu analysieren.
Unter Immobilisierung im Sinne der Erfindung sind alle Methoden zur Einschränkung der Beweglichkeit und Löslichkeit von Lektinen auf chemischen, biologischen und/oder physikalischen Wegen zu verstehen. Die Immo- bilisierung kann durch unterschiedliche Methoden erfolgen, wie der Bindung der Lektine untereinander oder an Träger, durch Festhalten im Netzwerk einer polymeren Matrix 'oder die Immobilisierung auf einer Membran. Durch die Immobilisierung wird ein
Auswaschen der Lektine bei den verschiedenen Waschschritten verhindert. Weiterhin kann nach dem Prozess der Interaktion die biologische Probe leicht wieder vom immobilisierten Lektin abgetrennt werden. Die Bindung bzw. die Immobilisierung der Lektine an den Träger bzw. Mikroarray kann durch direkte Trägerverbindung und durch Quervernetzung erfolgen. Die Trägerbindung bzw. Quervernetzung erfolgt gemäß der Erfindung insbesondere ionisch/adsorptiv oder durch kovalente Bindung. Die Quervernetzung im Sinne der Erfindung ist eine Vernetzung der Detektionsmole- küle - das heißt der Lektine - untereinander oder mit anderen Polymeren. Bei der Immobilisierung durch Ein- schluss werden die Lektine in Gelstrukturen bzw. in Membranen so eingeschlossen, dass eine Interaktion mit den Glykokonjugaten möglich ist.
Auf den Mikroarray können Lektine z.B. durch Auf- tropfen von runden Spots oder durch Aufdrucken von Linien immobilisiert werden, wobei als Mikroarray planare wie auch nicht planare Gebilde wie Kugeln, Quader oder Fibrillen und Stäbe eingesetzt werden können, die mit unterschiedlichen Lektinen imprägniert oder immobilisiert und so in eine räumliche An- Ordnung gebracht werden.
Dem Fachmann sind zahlreiche Möglichkeiten bekannt, die Lektine auf dem Mikroarray zu immobilisieren. Die Immobilisierung sollte hierbei so erfolgen, (i) dass
jedem Lektin eine definierte Position auf dem Träger oder Mikroarray zugeordnet und (ii) dass jede Position auf dem Mikroarray oder Träger unabhängig ausgewertet werden kann. Es kann aber auch vorteilhaft sein, dass sich die Auftragungsorte verschiedener Lektine teilweise oder vollständig überlappen oder dass Lektingemische in Form eines Detektionsspots aufgetragen werden. Die Immobilisierung kann beispielsweise mit einem an die Halbleitertechnik an- gelehnten Verfahren erfolgen. Auch Druckverfahren ermöglichen es, die Lektine in definierten Flächen oder Orten an oder auf der Oberfläche des Mikroarrays abzulegen, wodurch eine stabile Bindung mit hoher Kopplungseffizienz erfolgen kann. Alle dem Fachmann bekannten Immobilisierungsmaßnahmen von Biomolekülen - z.B. an Säulenmaterialien - können ebenfalls verwandt werden, um die Lektine auf dem Array zu immobilisieren.
Ausgewählte Verfahren zum Immobilisieren sind beispielsweise das Contact Tip Printing, Ring and Pin Printing, Nanoelectric Printing and Nanopipetting, Buble Jet Printing, TopSpot Printing, Micro Contact Printing, Micro Fluidic Networks-Methoden, Photo- lithographic Activation-Verfahren, Photoresist Li- thography, Electroche ical Focusing und Micro Wet Printing. Es können aber auch vergleichsweise einfache Probenauftragsgeräte aus der Dünnschichtchromatographie oder Autosampier für die HPLC sowie Pipet-
ten und Mikropipetten (Kolbenhubpipetten) eingesetzt werden. Mit Hilfe dieser Verfahren kann eine Lektin- Spot-Größe von ca. 1 μm bis über 6 mm erzeugt werden. Die Lektindichte pro cm2 kann erfindungsgemäß bei- spielsweise im Bereich von ca. 0,01 bis 1000 pmol/cm2 liegen. Die genannten Verfahren ermöglichen eine einfache Handhabung und eine große Präzision der angelegten Volumina sowie sehr homogene Lektinspots, die mit einer hohen Parallelisierung auftragen werden. Die Verfahren, die eine direkte Synthese der Lektine auf dem Träger ermöglichen, weisen insbesondere eine hohe Integrationsdichte und eine einfache Kombination von Immobilisierung und Kopplung auf. Es ist möglich, die Lektine, die auch als Sonden bezeichnet werden können, mit weiteren Verfahren vorzubereiten, um den Anteil unspezifisch synthetisierter bzw. gebundener Lektine gering zu halten.
Die Lektine können jedoch auf dem Mikroarray auch durch ein Blot-Verfahren immobilisiert werden. Hierzu werden die Lektine oder sie umfassende Zellen oder Gewebe mittels einer Elektrophorese aufgetrennt. Die elektrophoretische Auftrennung kann z.B. mittels einer eindimensionalen oder mehrdimensionalen Elektro- phorese, insbesondere einer 2D-Elektrophorese, erfolgen.
Die elektrophoretisch aufgetrennten bzw. separierten Lektine werden dann entweder auf den Träger bzw.
Mikroarray übertragen bzw. auf eine Membran, die in einem nächsten Schritt auf den Träger oder Mikroarray aufgebracht wird.
In einer bevorzugten Ausführungsform der Erfindung umfasst der Mikroarray mindestens zwei voneinander verschiedene immobilisierte Lektine, wobei die Lektine isolierbar sind, ausgewählt aus der Gruppe bestehend aus Abrus precatorius, Adenia digitata, Agaricus bisporus, Aleuria aurantia, Amaranthus caudatus, Amphicarpaea biacteata, Anguilla anguilla, Arachis hypogaea, Artocarpus integrifolia, Bauhinia purpurea alba, Brachypodium sylvaticum, Canavalia ensiformis, Carcino scorpius rotunda canda, Cicer arietinum, Codium fragile, Crotalaria juncea, Cytisus sessilifolius, Datura stramonium, Dioclea grandiflora, Dolichos biflorus, Erythrina coralldendron, Erythrina cristagalli, Euonymos europaea, Galanthus nivalis, Glycine max, Griffonia (Bandeiraea) simplicifolia, Helix aspersa, Helix pomatia, Hippeaεtrum hybrid, Hordeum vulgäre, Hura crepitaus, Latyrus odoratus, Latyrus sativuε, Latyrus tingitanus, Lens culinaris, Lima; glavus , Limulus polyphemus, Lotus tetragonolobus , Lycopersicon esculentum, Maackia amurensis, Maclura po ifera, Macrotyloma axillare, Momordica charantia, Narcissus pseudonarcissus, Oryza εativa, Phaseolus coccineus, Phaseolus limensis, Phaseolus vulgaris, Phytolacca americana, Pisum sativum, Phosphocarpus

Aus den genannten Organismen können mit Vorteil spezifische Lektine isoliert werden, die dazu geeignet sind, komplexe Glykostrukturen bzw. Glykokonjugate so zu binden, dass eine Detektion und Strukturanalyse möglich ist. So kann beispielsweise aus Agaricus bisporus das Lektin ABA, -aus Aleuria aurantia AAL, aus Amaranthus caudatus ACL (ACA) , aus Anguilla (Eel) anguilla AAA, aus Arachis hypogaea PNA, aus Artocarpus integrifolia AIL (Jacalin) , aus Griffonia

In einer weiteren bevorzugten Ausführungsform der Erfindung umfaεst der Mikroarray mindestens zwei voneinander verschiedene immobilisierte Lektine, wobei die Lektine zu den zu detektierenden Glykostrukturen eine Bindungsaffinität von Kd ≤ 10~s mol/1 aufweisen. Mit Vorteil ist es bei einer Bindungεaffinität von Kd ≤ 10"* mol/1 möglich, die Interaktion zwischen Glykokonj ugat und immobilisierten Lektinen so genau
zu analysieren und zu detektieren, dass ein qualitativer Nachweis des Glykokonjugates und eine Strukturanalyse effizient möglich ist.
In einer weiteren vorteilhaften Ausführungsform der Erfindung umfassen die immobilisierten Lektine einen Bindungslinker. Bindungslinker im Sinne der Erfindung sind alle Strukturen, die so mit den Lektinen assoziiert sind, dass diese kovalent oder komplex auf einem Träger des Mikroarrays bzw. auf dem Mikroarray selbst immobilisiert werden können. Die Linker können beispielsweise Peptidfragmente im Bereich von 0 bis 50 Aminosäuren sein. Die Lektine, die Linker umfassen, werden demgemäß vorteilhafterweise nicht über eine unspezifische Adsorption auf dem Mikroarray immobilisiert. Die Linker können auch so ausgebildet sein, dass eine Interaktion zwischen den einzelnen Lektinen bzw. Lektinen und den Trägern so ausgeschlossen ist, dass Störungen bzw. unspezifische Interaktionen nicht oder nur in sehr geringem Umfang auftreten. In solch einem Fall können die Linker beispielsweiεe als Abstandshalter eingesetzt werden. Linker im Sinne der Erfindung können demgemäß auch kurze synthetische DNA-Doppelstränge sein. Sie können auch dazu einge- setzt werden, bestimmte Lektine zunächst zu immobilisieren, um sie dann mit Hilfe spezifischer, beispielsweise DNA-Doppelstrang abbauender Enzyme, wieder abzuspalten. So ist es beispielsweise möglich, die komplexen Glykokonjugatstrukturen in einem ersten
Schritt mit mehreren spezifischen Lektinen inter- agieren zu lassen, wobei ausgewählte Lektine dann durch einen Abspaltungsschritt mittels spaltbarer Linker vom Mikroarray gelöst werden, so dass die Glykokonjugatstrukturen in einer Probe nun mit anderen Lektinen wechselwirken können, die z.B. eine andere Bindungsaffinität als die abgespaltenen Lektine aufweisen.
In einer weiteren bevorzugten Ausführungsform der Erfindung ist der Linker insbesondere Streptavidin und/oder Biotin. Streptavidin im Sinne der Erfindung ist ein Protein aus Streptomyciε avidinii, das mit einer Affinität von 10"15 M"1 an Biotin bindet und das ein Molekulargewicht von zirka 60.000 Da aufweist. Es weist vorteilhafterweise eine geringere unspezifische P-eaktion als Avidin auf. Biotin im Sinne der Erfindung sind Coenzyme der Enzyme, die Carboxylierungen katalysieren. Die sehr hohe Affinität von Biotin für Streptavidin als auch für Avidin kann vorteilhafterweise zur Immobilisierung von biotinylierten Reagenzien genutzt werden.
In einer weiteren vorteilhaften Ausführungsform des Mikroarrays werden die Lektine mit einer Packungs- dichte von > 50 ng/cm2 immobilisiert. Eine daraus resultierende Oberflächehbelegung von > 10 % ermöglicht eine optimale Wechselwirkung mit den Glykostrukturen. Die Varianten der möglichen Funktionali-
sierungen von Trägermaterialien sind selbstverständlich vielfältig. Voraussetzung für die Anbindung von Lektinen ist das Vorhandensein von entsprechenden funktionellen Gruppen auf dem Trägermaterial, mit denen das Lektin reagieren kann. Es können unter anderem folgende Varianten eingesetzt werden.- Funk ionaliεierung mit Carboxylgruppen, Funktionali- sierung mit Aminogruppen, Funktionalisierung mit Aldehydgruppen, Funktionalisierung mit Phenylgruppen, Funktionalisierung mit Thiolgruppen, Funktiona- lisierung mit Divinylsulphone (entstehende Bindung: Ether, Thioether, sekundäre Amine) , Funk ionaliεierung mit Carbonyldiimidazol, Funktionalisierung mit Epichlorhydrine (entstehende Bindung: Ether, Thioether, sekundäre Amine) , Funktionalisierung mit

(entstehende Bindung.- Amid-Bindung) , Funktionalisierung mit Periodat (entstehende Bindung.- Alkylamin) , Funktionalisierung mit Diazonium (entstehende Bindung: Azo) , Funktionalisierung mit Tri- chloro-S-triazine (entstehende Bindung: Triazinyl) , Bisoxirane (entstehende Bindung-. Alkyla ide, Ether, Thioether) , Cyanbromid (entstehende Bindung: Isoharnstoff, Imid, (carbamat) , Funktionalisierung mit 4-Nitropheny-chloroformate (entstehende Bindung: Anhydrid, Carbamat, Urethan) , Funktionalisierung mit Epoxidgruppen und die Funktionalisierung mit orga-
nischen Sulfonylchloriden (Tosylchlorid, Tresyl- chlorid) (entstehende Bindung: Alkylamin) .
Mit Vorteil ist die Funktionalisierung in einer schnellen Reaktion möglich, wobei die Immobilisierung der Lektine über die beschriebenen Methoden besonders vorteilhaft ist, da εich hier eine kovalente Bindung ausbildet . Das verhindert die Auswaschung der Lektine und dadurch einen Verlust an Bindungsaktivität oder beim erwünschten Ablösen der Probe eine Kontamination dieser durch Lektin-Moleküle .
Die Immobilisierung erfolgt bei individuellen Reaktionsbedingungen und ist häufig unter Verwendung weiterer Chemikalien durchzuführen. Die Immobilisierung an Epoxidgruppen erfordert beispielεweise 8 - 48 h bei pH-Werten von pH 8,5 - 12. Die Anbindung von Proteinen an mit Carboxylgruppen aktivierte Matrices erfolgt bei pH 4 - 6 in 24 - 48 h. Dabei ist die Verwendung von Carbodiimid für die Pveaktion vorteilhaft. Die Immobilisierung von Proteinen an mit Aminogruppen substituierte Träger erfolgt ebenfalls unter Verwendung weiterer Chemikalien wie Carbodiimid oder NaCNBH3.
Vorteilhafterweise wird eine Funktionalisierung mit Tresylchlorid (2,2, 2-Trifluorethan-sulfonylchlorid) verwendet. Die Funktionalisierung von Trägermaterialien mit Tresylchlorid eignet sich besonderε
für die Anbindung von Proteinen, da die Reaktion unter milden Bedingungen stattfindet (pH 7-9) und die Proteine nicht schädigt. Die Bindung erfolgt über primäre Aminogruppen oder Thiolgruppen der Aminosäuren des Proteins. Die Reaktion ist vergleichsweise schnell (wenige Stunden) und läuft ohne den Einsatz weiterer Chemikalien ab. Die Tresylaktivierung ist bei trockenem Material oder bei Lagerung in verdünnter Salzsäure einige Monate stabil.
Mitunter ist es sinnvoll, den Träger unter Einbindung eines so genannten Spacers, eines Abs andεhalters , zu funktionalisieren. Dieser Spacer vergrößert die Distanz zwischen Trägeroberfläche und Ligand und kann daher zu einer besseren Zugänglichkeit des Liganden beitragen. Übliche Spacer bestehen aus zwei ter- inalen funktioneilen Gruppen und einer in der Länge wählbaren C-Kette wie z.B. 6-Aminohexansäure . Der verwendete Linker, das heißt die verwendete Träger- funktionalisierung, kann auch bereits selbst einen Spacer darstellen.
In einer weiteren vorteilhaften Ausführungsform der Erfindung umfasst der Mikroarray Metall, Polypropy- len, Teflon, Silizium, Polyethylen, Polyester, Poly- styren, Nitrid, Keramik, Quarz und/oder Glas. Metalle im Sinne der Erfindung sind alle Verbindungen, deren Zusammenhalt durch ein Kristallgitter entsteht. Die Grenze zwischen Metallen und Nichtmetallen ist flie-
ßend, so dass auch die Elemente Ce, Sn, As und Sb im Sinne der Erfindung Metalle sind. Zu den Metallen gemäß der Erfindung gehören auch die metallischen Gläser, das heißt Werkstoffe, die sich in einem meta- stabilen, weitgehend amorphen Zustand befinden. Selbstverständlich sind auch metallisch leitfähige Polymere Metalle im Sinne der Erfindung. Vorteilhafterweise weisen Metalle im Sinne der Erfindung insbesondere eine gute Festigkeit, eine gute Härte und Verschleißbeständigkeit, eine hohe Zähigkeit und eine gute elektrische und thermische Leitfähigkeit auf. Polypropylene im Sinne der Erfindung sind thermoplastische Polymere des Propylens . Polypropylene zeichnen sich insbesondere durch eine hohe Härte, Rückstellfähigkeit, Steifheit und Wärmebestandigkeit aus. Der Mikroarray kann jedoch auch Teflon umfassen. Teflon gemäß der Erfindung sind Polytetrafluoro- ethylene, die vorteilhafterweise gute thermoplastische Eigenschaften aufweisen. Polyethylene entstehen insbesondere durch eine Polymerisation von Ethylen nach im Wesentlichen zwei unterschiedlichen Methoden, dem Hochdruck- und dem Niederdruckverfahren. Polyethylene, die im Hochdruckverfahren hergestellt werden, weisen vorteilhafterweise eine geringe Dichte auf. Die Eigenschaften von Mikroarrays, die Polypropylen umfassen, werden im Wesentlichen durch den Charakter des Polyethylen als partiell kristallinem Kohlenwasserstoff bestimmt. Vorteilhafterweise sind Polyethylene bis zu 60° in allen üblichen Lösungs-
mittein praktisch unlöslich. Vorteilhafterweise bewirken polare Flüssigkeiten wie Alkohol, Ester und Ketone bei Zimmertemperatur kaum eine Quellung von Polyethylenen. Gegen Wasser, Laugen und Salzlösungen sowie anorganische Säuren verhalten sich Polyethylene vorteilhafterweise völlig indifferent. Mikroarrays oder Träger, die Polyethylene umfassen, haben z.B. eine sehr geringe Wasserdampfdurchlässigkeit . Der Mikroarray kann jedoch zweckmäßigerweise auch Polyester umfassen. Polyester im Sinne der Erfindung sind Verbindungen, die durch Ringöffnungspolymerisation von Lactonen oder durch Polykondensation von Hydroxycarbonsäuren bzw. von Diolen und Dicarbon- säuren bzw. Dicarbonsäurederivaten hergestellt wer- den. Polyester im Sinne der Erfindung umfassen auch Polyesterharze, Polyesterimide, Polyesterkautschuke, Polyesterpolyole und Polyesterpolyuretane . Polyester sind vorteilhafterweise Thermoplaste und besitzen ausgesprochenen WerkstoffCharakter . Sie zeichnen sich beispielsweise durch eine hohe Thermostabilität aus und können zu Legierungen mit Metallen, wie beispielsweise Kupfer, Aluminium und Magnesium, verarbeitet werden. Es kann jedoch auch vorgesehen sein, dass der Mikroarray Keramik umfaεst. Keramik im Sinne der Erfindung ist eine Sammelbezeichnung für aus insbesondere anorganischen und überwiegend nicht- metallischen Verbindungen und Elementen aufgebaute und insbesondere zu mehr als 30 Vol.% kristalline Materialien umfassende Stoffklasse. Dem Fachmann sind
verschiedene Keramiken bzw. keramische Werkstoffe bekannt, die er als Mikroarray einsetzen kann. Es kann sich beispielsweise um so genannte Töpferware, Steingutgeschirr, Spaltplatten, Laborporzellan, Ge- schirrporzellan, Knochenpσrzellan, Aluminiumoxidkeramik, Dauermagnetwerkstoffe, Silicasteine und Magnesiasteine handeln. Bei tonkeramischen Werkstoffen wird im Sinne der Erfindung in grobe und feine Werkstoffe unterschieden, wobei feine tonkeramische Werkstoffe Irdengut, Steingut, Steinzeug und Porzellan umfassen. Es können jedoch mit Vorteil auch sonderkeramische Werkstoffe wie Glas-, Oxidkeramik, SiC-Steine und schmelzflüssig gegossene Steine als Mikroarray eingesetzt werden. Bevorzugt kann der Mikroarray auch Glas umfassen. Glas im Sinne der Erfindung sind Stoffe im amorphen, nichtkristallinen Festzustand, das heißt, der Glaszustand lässt sich im Sinne der Erfindung als eingefrorene unterkühlte Flüssigkeit bzw. Schmelze auffasεen. Gläser sind daher anorganische oder organische, meist oxidische Schmelzprodukte, die durch einen Einführvorgang ohne Auskristallisation der Schmelzphasenkomponenten in einen festen Zustand überführt wurden. Selbstverständlich sind im Sinne der Erfindung auch Kristalle, Quarz, Schmelzen und unterkühlte Schmelzen als Gläser aufzufassen. Die Gläser können zum Beispiel Flachglas, Behälterglas, Wirtschaftsglas, Laborgeräteglas, Bleikristallglas, Faserglas, optische Glasfasern und andere sein. Selbstverständlich ist es auch möglich,
daεs Silicat-freie Gläser, beispielsweise Phosphatgläser, eingesetzt werden. Der Mikroarray kann j doch auch so beschaffen sein, dass optische Gläser, das heißt, z.B. Gläser mit besonderen optischen Bre- chungsindexen, eingesetzt werden.
Die Erfindung betrifft auch einen Bindungsassay zur Bestimmung von Glykostrukturen in einer Probe, wobei die Probe mit mindestens zwei voneinander verschiede- nen immobilisierten Lektinen unter Ausbildung von Lektin-Ξucker-Komplexen in Kontakt gebracht wird und die Lektin-Sucker-Komplexe in an sich bekannter Weise detektiert werden und die Lektine isolierbar sind, ausgewählt aus der Gruppe umfassend Abrus precatorius, Adenia digitata, Agaricus bisporuε, Aleuria aurantia, Amaranthus caudatus, Amphicarpaea biacteata, Anguilla anguilla, Arachis hypogaea, Artocarpus integrifolia, Bauhinia purpurea alba, Brachypodium εylvaticum, Canavalia ensiformiε, Carcino scorpius rotunda canda, Cicer arietinum, Codium fragile, Crotalaria juncea, Cytisus sessilifolius, Datura stramonium, Dioclea grandiflora, Dolichos biflorus, Erythrina coralldendron, Erythrina cristagalli, Euonymos europaea, Galanthus nivalis, Glycine max, Griffonia

Bestimmung von Glykostrukturen durch den erfindungs- gemäßen Bindungsassay betrifft zum einen die Detek- tion, das heißt den qualitativen Nachweis von in einer Probe befindlichen Glykostrukturen, als auch die Detektion von bestimmten Pattern auf dem Mikroarray, die Rückschlüsse darüber zulassen, mit welchen immobilisierten unterschiedlichen Lektinen ein und dieselbe Glykostruktur wechselwirkt, um so die Struktur des komplexen Oligo- oder Poly- saccharides oder des Glykokonjugates zu bestimmen. Eine Probe im Sinne der Erfindung ist die Bezeichnung für ein durch Probenentnahme entnommenes biologisches
oder chemisches Gut oder eines Teiles bzw. einer kleinen Menge eines solchen, dessen Beschaffenheit chemisch, biologisch, klinisch oder ähnlich geprüft werden soll . Die Probenentnahme erfolgt insbesondere so, dass die entnommene Teilmenge einem Durchschnitt der gesamten Menge entspricht. Die durch Untersuchung der Probe ermittelten Merkmale dienen der Beurteilung der durch die Probe e fassten Menge, die RUCKSchlüεse auf die Gesamtmenge, z.B. Trinkwasser, Lebensmittel, Gewebe, Zellen, Zellkulturen, Sellkultur-Überstände, transplantierte Organe und anderes , zulässt. Für die Untersuchung können die Proben durch Mischen, Serteilen, Separieren, Vorfraktionieren, Zerkleinern, Sugabe von Enzymen oder Markern - bzw. anders vorbehandelt werden. Dem Fachmann sind verschiedene Möglichkeiten der Vorbehandlung der Proben bekannt. Selbstverständlich kann es auch vorgesehen sein, dass die Probe so entnommen wird, dass sie keinem Durchschnitt der gesamten Menge entspricht . Eine Probe können alle biologischen und nichtbiologischen Materialien sein, wie biologische Gewebe und Flüssigkeiten, z.B. Blut, Lymphe, Urin, Gehirnflüssigkeit und andere, sowie Umweltabwässer, Bioreaktorenflüssigkeiten, Lebensmittelinhaltsstoffe, Gefahren- Stoffe, Aerosole, Lipid ischungen u.v.a. mehr.
In einer vorteilhaften Ausführung form des Bindungs- assays werden die Lektine physikalisch, chemisch und/oder biologisch durch in situ Synthese oder durch
Ablegen zuvor synthetisierter Lektine auf dem Mikroarray immobilisiert. Für den Bindungsassay können unabhängig von der Art der immobilisierten Lektine die Träger oder Mikroarrays auf zwei prinzipiell ver- schiedenen Wegen hergestellt werden:
1. Ablegen und Immobilisieren von zuvor synthetisierten oder aus Bibliotheken stammenden Lektinen an definierten Positionen eines funktionali- sierten Trägermaterials . Hierfür können sowohl Spotting als auch Druckverfahren eingesetzt werden. Unter Spotting versteht man Verfahren, bei denen Flüssigkeitεtropfen abgelegt werden, wobei durch Oberflächenwechselwirkung und Trocknen im Wesentlichen runde Spots entstehen. Andere Druckverfahren ermöglichen das Substrat in definierten Flächen auf der Oberfläche abzulegen.
2. In si u-Synthese der Lektine an definierten Po- sitionen des Trägers oder des Mikroarrays durch sukzessive Kopplung monomerer Synthesebausteine.
In einer weiteren vorteilhaften Ausführungsform werden die Lektine durch Contacc Tip Printing, Ring and Pin Printing and Nanopipetting, Buble Jet Printing, TopSpot Printing, Micro Contact Printing, Micro Fluidic Networks-Methoden, Photolithographic Activa- tion-Verfahren, Photoresist Lithography, Electroche-
mical Focusing und/oder Micro Wet Printing immobilisiert .
In einer weiteren bevorzugten Ausführungsform der Er- findung ist die Oberfläche des Mikroarrays mit Poly- L-Lysinen, Aminosilanen, Aldehydsilanen, Epoxy-Grup- pen, Gold, Streptavidin, Polylysinen, Silanen, reaktiven Gruppen, Polyacrylamid-Pads , immobilisierter Nitrocellulose, aktivierten Aldehyden, Agarose- Aldehyd-Gruppen und/oder Tresyl-Gruppen beschichtet. Durch derartige Substratoberflächenbehandlungen ist es vorteilhafterweise möglich, die Haltbarkeit und die Bindungskapazität der Oberfläche der Mikroarrays so zu verbessern, dass die Lektine sehr gut über einen längeren Zeitraum immobilisiert bleiben können. Selbstverständlich kann die Oberfläche des Trägers ganz allgemein, insbesondere durch Einwirken von biologischen, physikalischen und/oder chemischen Einflüssen erfolgen. Physikalisches Einwirken sind beispielsweise das Polieren, Ätzen, Beizen, Sandstrahlen, aber auch physikalische Verfahren, die zu einem Härten, Beschichten, Vergüten, Überziehen mit Schutzhäuten und ähnlichem fuhren. Eine Oberflächenbehandlung durch biologische Einwirkung kann bei- spielsweise das Bewachsen durch Mikroorganismen umfassen. Eine chemische Modifikation der Oberfläche der Träger beinhaltet beispielsweise die Behandlung mit Säuren, Basen, Metalloxiden und anderen. Die Oberfläche der Träger kann so modifiziert werden,
dass die Detektionsmolekule bzw. die Lektine auf dem Träger besonders gut haften bzw. so haften, dass sie in ihrer Aktivität nicht nachteilig modifiziert werden. Eine Oberflächenmodifizierung der Träger umfasst selbstverständlich auch eine Behandlung, die zu einer erhöhten Stabilität und Bruchfestigkeit insbesondere des Mikroarrays führt. Eε können selbstverständlich auch klassische Oberflächenmodifizierungen aus der Histologie vorgenommen werden, wie daε Beschichten mit z.B. Eiweißglycerin, Polylysin, aktivierten Dextranen und/oder Chromgelatine.
In einer weiteren bevorzugten Ausführungsform der Erfindung werden vor oder nach der Ausbildung der Zucker-Lektin-Komplexe ein markiertes oder unmarkiertes Lektin, eine markierte oder unmarkierte Glykostruktur und/oder ein markierter oder unmarkierter Antikörper zugesetzt.
Mit Vorteil können folgende Detektionssysteme eingesetzt werden:
Direkt : Die Probenmoleküle selbst werden mit einem Marker versehen (Enzym (A) , Fluoreszenz- oder Vis- Farbstoff (B) , Biotin (C) ... )
Nach dem Bindungsereignis und dem Waschen kann entweder :
direkt ausgelesen werden (B) oder nach Umsetzung eines Enzy substrates und Auslesung des entstehenden Farbsignals (A) oder (C) nach Zugabe eines weiteren Markers (Enzym, Fluoreszenz- oder Vis-Farbstoff) , der mit Avidin/Streptavidin gekoppelt ist.
Mit Vorteil wird ein einfaches, schnelles, direktes Ergebnis erreicht, da die Probe selbst markiert wird.
Indirekt: Nach Adsorption der Probenmoleküle werden diese mit einem für das zu bestimmende Glykokonjugat spezifischen Antikörper oder Lektin versehen. Dieser ist wiederum markiert. Nach erfolgter Bindung kann das Ereignis ausgelesen werden.
Durch die "doppelte" Spezifität wird vorteilhafterweise sehr gezielt eine bestimmte Substanz nachge- wieεen.
Sandwich: Nach Adsorption der Probenmoleküle werden diese mit dem selben Lektin versehen, welches auf der Oberfläche immobiliεiert ist (siehe Ausführungs- beiεpiel) . Dieses Lektin bindet an das zu bestimmende Glykokonjugat spezifisch von der gegenüberliegenden Seite und ist selbst wiederum markiert. Nach erfolgter Bindung kann das Ereignis ausgelesen werden. Mit Vorteil wird die selbe Zuckerstruktur "doppelt" erkannt und detektiert. Es kann die eine Zuckerspezi-
fität bestimmt werden. Es werden nur komplexe Systeme mit mindestens zwei Glykosylierungssteilen erfasst. Es werden nur die ohnehin vorhandenen Lektine benötigt, keine weiteren Antikörper o.a..
Kompetitiv: Nach Adsorption der Probenmoleküle werden die wells mit einem markierten Modelladsorptiv versehen. Dieses bindet bekanntermaßen das immobilisierte Lektin. Das Modelladsorptiv kann nur an noch freie Bindungs teilen andocken bzw. konkurriert mit dem Probenmolekül um diese Bindungsstellen. Es wird ein positives Signal erhalten, wenn keine oder nur wenige Probenmoleküle angebunden sind bzw. nach dem Verhältnis, das sich in dieser KonkurrenzSituation einstellt.
Vorteilhafterweise ist der Nachweis von Systemen mit nur einer komplexen Zuckerstruktur möglich. Es werden bekannte Modellsyεteme eingesetzt. Die ko petitive Situation gibt auch Auskunft über die Bindungs- konstanten des Komplexes mit dem Probenmolekül.
In einer weiteren vorteilhaften Ausführungsform der Erfindung wird zur Markierung ein Enzym, ein Farb- stoff, ein Fluoreszenzmarker, ein Radioisotop, ein Metallkolloid und/oder ein Chelator eingesetzt. Dem Fachmann sind Methoden bekannt, wie er Marker insbesondere durch Verstärkungsmechanismen sensitiver ausbilden kann.
In einer weiteren Ausführungsform der Erfindung werden die Lektine an Mikrotestplatten, Glasplättchen, Membranen, Geflechte, Fibrillen, insbesondere aus Po- lypropylen, Nitrozellulose, Glas und/oder PVDF immobilisiert. Als Mikrotestplatten können z.B. Mikroti- terplatten eingesetzt werden. Vorteilhafterweise weisen Mikrotestplatten Maße auf, die einen Einsatz in zahlreichen Laborroutinen gestatten. Beispielsweise sind zahlreiche Fluoreszenzmessgeräte, Fluoreszenz- spektrometer und -photometer wie Fluoreszenz - mikroskope und ähnliches so ausgebildet, dass Mikrotestplatten als Standard verwandt werden können. Die Immobilisierung der Moleküle auf spezielle Labor- gefäße, beispielsweise Mikrotiterplatten, Petri- schalen, Vielf chschalen, Multischalen und andere Kulturgefäße und Objektträger, ermöglicht daher vorteilhafterweise auch die Verwendung der vorhandenen Labormittel und -gerate zum Inkubieren, Einfrieren, Lyophilisieren und ähnlicher Laborgeräte in klinischen oder Forschungslaboratorien. Als Mikrotestplatten können beispielsweise bevorzugt Mikrotiterplatten mit transparentem, nicht fluoreszierendem Flachboden verwandt werden oder weißem oder εchwarze , undurch- sichtigem Flachboden.
In einer weiteren vorteilhaften Ausführungε orm der Erfindung wird eine weitgehende planare Oberfläche des Mikroarrays mit einem Polymer beschichtet und
folgend werden Lektine mittels des Polymers auf der Oberfläche immobilisiert. Vorteilhafterweise erfolgt in dieser Ausführungsform die Immobilisierung der Lektine direkt über das Polymer.
Bevorzugt iεt das Polymer so ausgewählt, dass es eine hydrophobe Oberfläche ausbildet und die Oberfläche gegenüber einem wässrigen Medium elektrisch isoliert. Mit Vorteil kann diese hydrophobe glatte Oberfläche unter anderem einen Selbstreinigungsef ekt zeigen.
In einer weiteren bevorzugten Ausführungsform der Erfindung ist das Polymer ein Polyimid oder Polystyren. Wählt man das Polymer geeignet aus, z.B. Polyimide, lassen sich auch weitere Mikroarrays oder sogar zusätzliche Microsysteme mit derselben anorganischen Oberfläche, z.B. einer Halbleiterschalung, industriell zu einem System verbinden. Polyimide sind insbeεondere hochtemperaturbeständige Polymere; vor- teilhaft weisen sie ausgezeichnete mechanische, thermische und elektrische Eigenschaften auf. Die Anwendungen des Polyimids umfassen insbesondere Pufferschichten, Passivierungsschichten, Bindeschichten und dielektrische Zwischenschichten auf dem Träger. Poly- imide werden insbesondere flüssig aufgebracht und dann ausgehärtet. Bei diesem Aushärtungsεchritt erhält daε Polyimid mit Vorteil die gewünschten Eigenschaften. Für die Anwendungen kann das Polyimid li-
thographisch strukturiert werden. Polyimid kann selbstverständlich auch als Haftungsvermittler für Vergussmaterial und als Pufferschicht eingesetzt werden. Die Polyimidschicht reduziert beispielsweise den Stress im Silicium, der durch die Kapselung hervorgerufen wird und verhindert Risse an den Kanten. Silicium kann in Leitbahnen auf dem Mikroaray aufgebracht sein. Das Polyimid muεs insbesondere unter sehr gleichmäßigen Temperaturbedingungen gehärtet werden, um Risεbildung im Polyimid und Farbungleich- mäßigkeiten zu verhindern. Niedrige Sauerεtoffwerte sind z.B. vorteilhaft, um eine gute Haftung zu erreichen. Polystyren oder Polystyrol ist ein thermoplastischer Kunststoff, der vor allem durch radi- kaiische Polymerisation von Styrol gewonnen wird. Das radikalische Ende einer wachsenden Polymerkette qreift nie eine Doppelbindung im Ring an, da der Ben- ∑olring eine außerordentlich stabile Struktur ist. Hieraus leiten sich mehrere Vorteile bei der Ver- wendung von Polystyrol ab, beispielsweise iεt Polystyrol be tändig gegenüber Säuren, Laugen und Alkohol .
Bevorzugt werden UV reaktive Moleküle durch die Bestrahlung mit UV Licht kovalent immobilisiert. Bevor- zugt ist weiterhin, dass das Polymer nur in vordefinierten Bereichen auf die Oberfläche des Mikroarrays aufgetragen wird. Hierdurch können vorteilhafterweise, ähnlich wie bei Schaltkreisen, bestimmte
Strukturen, eine definierte Anordnung der Lektin- Spotε oder Muster der Immobilisierung der Lektine vorgegeben v/erden. Somit sind bestimmte Reaktionen oder Wechselwirkungen zwischen den Lektinen und Glykostrukturen steuerbar.
In einer weiteren vorteilhaften Ausführungsform der Erfindung wird die Oberfläche, auf der die Lektine direkt gebunden werden sollen, durch Plasmabehandlung positiv und/oder negativ elektrisch geladen. Das heißt, die Oberflächen εind an den unterschiedlichsten Stellen unterschiedlich geladen. Polymere Werkstoffe liegen insbesondere in verschiedenen Formen vor. Die einzelnen Formen stellen unterschied- liehe Anforderungen an den Bearbeitungsprozess . Je nach Ausformung der Oberfläche ist diese z.B. den Plasmen in unterschiedlicher Weise zugänglich. Eine Plasmabehandlung der Polymeroberfläche kann vorteilhafterweise die Oberflächenenergie stark erhöhen und andere Verarbeitungsverfahren ermöglichen. Bei einer Plasmabehandlung reagieren vor allem die Ionen und Radikale des Plasmas mit der Polymeroberfläche und erzeugen dort funktionale Gruppen, welche die Oberflächeneigenschaften des Polymers mit Vorteil beεtim- men. Durch die positive oder negative Ladung wird insbesondere eine bessere Benetzbarkeit und/oder eine bessere Bindung der Biomoleküle erreicht.
Die Erfindung betrifft auch die Verwendung des Mikroarrays und/oder des erfindungsgemäßen Bindungsassays zum Nachweis eines Analyten und/oder zu einer Pattern Analyse . Neben den genannten Verfahren kann auch die Ellipsometrie eingesetzt werden. Bei dieser Technik werden adsorbierte Schichten (Monolayer und Submono- layer) aus einer Oberfläche mittels eines polarisierten Laserstrahls vermessen. Der reflektierte Laserstrahl mit veränderter Polarisation kann vermes- sen werden.
Die Technik der SPR (Surface Plasmon Resonance) arbeitet mit einem Sensorchip, der aus einer goldbeschichteten Glasplatte, überschichtet mit einem Polymer, besteht. Ein Prisma ist auf der Glasoberfläche installiert. Ein Signal entsteht durch die Änderung des Brechungsindex durch das Prisma, wenn aus der vorbeifließenden Lösung Moleküle an den Sensor adsorbieren. Das Signal zeigt eine Massen- änderung an, die auf der Goldoberfläche stattfindet.
Mit dem erfindungsgemäßen Mikroarray bzw. dem erfindungsgemäßen Bindungsassay können unbekannte Substanzen aus der Gruppe der Glykokonjugate ge- screent, detektiert und analysiert werden. Durch die Verwendung von immobilisierten Lektinen ist es vorteilhafterweise möglich, dass co- und poεttranεla- tionale Veränderungen der Proteine-, wie die Glykosi- lierung, nachgewiesen und analysiert werden. Dies ist
von besonderer Bedeutung, da gerade die Gly- kosilierung von Proteinen deren Bioaktivität moduliert und einen entscheidenden Einfluss auf die Funktion, die Verfügbarkeit, die Wechselwirkung, die Kom- partimentierung und die Lebensdauer dieser Substanz im Organismus hat. Ebenso spielt die Glykosilierung der Lipide eine große Rolle für deren Funktion als Rezeptoren in der Signalübertragung. Die zu untersuchenden Glykostrukturen können beispielsweise in Pro- ben enthalten sein, die natürliche oder synthetische Glykostrukturen umfassen sowie deren Derivate und Abbauprodukte, definierte Stoffmischungen, synthetische und natürliche Vielstoffmischungen, wie zum Beispiel Zellen, Zeilüberstände, Nährlösungen, Extrakte und/oder Seren. Die Screeningfunktiσn des beschriebenen Mikroarrays und des Bindungsassays beruht insbesondere auf dem parallelen Einsatz vieler verschiedener, mindestens aber von zwei Lektinen oder Lektin- systemen. Die gezielte Auswahl kohlenhydratbindender Lektine bietet mit Vorteil ein breites Spektrum an Bindungsaffinitäten zu definierten Zuckerstrukturen. Somit macht die Verwendung des Mikroarrays und des Bindungsassays den Einsatz von Lektinen in Form eines Arrays für die Analyse, insbesondere Strukturanalyse, von Glykokonjugaten möglich. Es ist vorteilhafterweise im Unterschied zu den bekannten Screening- verfahren auch möglich, Glykokonjugate anhand ihrer Glykosylierung nachzuweisen und zu analysieren. Es werden damit die co- und posttranεlatiσnalen Modi-
fikationen, wie Glykosylierungen, der Proteine berücksichtigt und detektiert, die einen entscheidenden Einfluss auf die physiologischen Eigenschaften der Substanzen haben. Durch die hohe Spezifität der aus- gewählten Lektine können auch geringfügig veränderte Zuckerstrukturen voneinander unterschieden werden und dadurch StrukturInformationen vor allem über unbekannte Glykokonjugate gewonnen werden. Durch den Einsatz verschiedener Lektine mit sich ergänzenden Bin- dungεspezifitäten ist mit Vorteil auch eine Pattern- Analyεe der Probe möglich. Dies ermöglicht auch den Vergleich von Proben - wie z.B. von Zelllinien - untereinander, welches eine häufige Problematik in Grundlagenforschung, Medizin und Klinik ist. Die Er- findung ermöglicht so den Nachweis und die Analyse neuer unbekannter Glykostrukturen in komplexen Vielstoffgemisehen.
Im Folgenden soll die Erfindung anhand eines Bei- spiels anhand der zugehörigen Zeichnungen erläutert werden, ohne auf dieses Beispiel beschränkt zu sein.

Die Figur 1 zeigt die schematische Darstellung eines Ausführungsbeispiels des Lektin-Mikroarrays . Ein Lek- tin wird an die Oberfläche einer Mikrotiterplatten- Vertiefung immobilisiert, indem sich der hochaffine Blomplex Streptavidin-Biotin ausbildet. An die Saccharidbindungsstellen des Lektins können anschließend komplexe Glykane von Glykoproteinen binden.
Die Figur 2 zeigt die Anordnung von immobilisierten Lektinen auf der Oberfläche im Arrayformat.
Die Figur 3 zeigt den Vorteil dieses Anwendungsbei- spiels, der in der Information liegt, dass nur komplexe Glykokonjugate mit mindestens zwei Glykanen nachgewiesen werden, die sowohl mit dem immobilisierten Lektin als auch mit dem Detektions-Lektin gleichzeitig wechselwirken können. Durch diesen Nachweis in Form einer Sandwich-Technik ist die Spezifität und Aussagekraft des Analysenergebnisses besonders hoch.

Herstellung des Lektin-Arrays :
Die biotinylierten Lektine Con A, RCA. und WGA werden jeweils zu folgenden Konzentrationen in Puffer A gelöst: Con A: 5,5 μg/mL
RCA: 3, 0 μg/mL
WGA : 4,5 μg/mL
Je 100 μL der Lektinlösungen werden in ein Well der Mikrotiterplatte pipettiert und für 4 h bei 8 °C inkubiert. Pipettierschema: A 1 : ConA A 2: RCA A 3 : WGA
Die erzielte Belegung beträgt dann 100 - 400 ng Lektin/c 2 bzw. 70 - 300 ng Lektin/well (entsprechend 1 - 4 pmol/well) .
Die Platte v/ird anschließend dreimal mit Puffer A gewaschen. Danach wird mit 100 μL Puffer B pro well für weitere 4 h bei S °C inkubiert. Die Platte wird anschließend wieder dreimal mit Puffer A gewaschen. Die so hergestellte Vorrichtung besteht aus der An- Ordnung von immobilisierten Lektinen auf einer Oberfläche im Arrayformat (Fig. 2) .
Einsatz des Lektin-Arrays :
100 μL des 1:500 verdünnten fetalen Rinderserums werden auf die Wells AI, A2 und A3 gegeben (pipettiert) und für 4 h bei 8 °C inkubiert. Die Platte wird anschließend dreimal mit Puffer A gewaschen.
Die mit FITC markierten Lektine Con A, RCA und WGA werden jeweils zu folgenden Konzentrationen in Puffer A gelöst: Con A: 10 μg/mL RCA: 10 μg/mL WGA: 10 μg/mL
Je 100 μL dieser Lektinlösungen werden in die Wells der Mikrotiterplatte pipettiert und für 4 h bei 8 °C inkubiert. Pipettierschema: A I: FITC-ConA
A 2: FITC-RCA
A 3 : FITC-WGA
Die Platte wird anschließend dreimal mit Puffer A. gewaschen.
Messung und Auswertung :
Die Auswertung erfolgt im Fluoreszenzspektrometer
(Mikrotiterplatten-Reader) . Dabei wird die relative
Fluoreszenzaktivität in den Wells bestimmt . Die An- regungs ellenlänge beträgt 485 nm . Die Emission wird gemessen bei 535 nm .
Ergebnis :
In Well A 2 und A 3 wird jeweils eine positive Fluoreszenzaktivität vermessen. Well A 1 zeigt keine Fluoreszenz. Dies deutet auf die Bindung einer komplexen Glykostruktur aus dem Serum an die Lektine RCA und WGA hin. Entsprechend den Bindungsaffinitäten von RCA und WGA kann hiermit auf folgende Strukturen geschlossen werden.- die Glykostruktur (en) im Serum besitzen je zwei Glykane (zwei unterschiedliche Glykoεylierungs- stellen) die Glykane weisen ß-D-Galactosen (entεprechend der Affinität von RCA s.u.) und terminale N-Acetylglucoεamine oder Sialinsäuren (entsprechend der Affinität von WGA s.u.) auf. - Mannosehaltige Strukturen entsprechend der Affinität von Con A (s.u.) treten nicht auf.
Die genannten Eigenschaften treffen auf das im Serum vorliegende Glykoprotein Fetuin bekanntermaßen zu. P-inder-Fetuin besitzt einen Kohlenhydratanteil von etwa 30 % . Es besitzt insgesamt 6 Glykane, von denen
-. bi- und triantennäre N-Glykane mit endständigen Sialinsäuren sind.
Bindungsspezifitäten der verwendteten Lektine:
RCA: Generell werden Glykane mit terminalen Galac- tosen gebunden. Die Glykostruktur ß-D-Gal-1, 4-ß-D- GlcNAc-l,0-R (terminales Lactosamin) gilt als Voraussetzung für die Bindung eines Glykans, wobei feste Bindungen hochspezifisch nur mit verzweigten, multi- valenten N-Glykanen zustande kommen.
WGA ist ein chitinbindendes Lektin, das Bindungsstellen für N-Acetylglucosamin (GlcNAc) und N-Acetyl- neuraminsäure (NeuNAc) besitzt. Die Bindungsstärke für Oligosaccharide nimmt mit der Anzahl der GlcNAc- Reste stark zu. Das Disaccharid N, N'-Diacetyl- cliitobiose wird 131-mal stärker gebunden als das Monomer und das Trisaccharid N,N' ,N' '-Triacetyl- chitotriose 3700-mal stärker. WGA bindet sialinsäure- haltige Glykoproteine .
