- Different stock solutions of the substrate are made in DMSO in the presence of Tween® 20 or Tween ® 80 (Polyoxyethylenated sorbitan monooleate) or Tween® 60 (Polyoxyethylenated sorbitan monostearate) or Tween® 65 (Polyoxyethylenated sorbitan tristearate). Then a volume corresponding to a final concentration of 500 mg / 1 of substrate is added to a gelled medium by supercooling: Mac Conkey medium. Table 11 shows the composition of the media tested.
