Abstract

L'invention permet de déterminer le statut de méthylation des bases de cytosine dans un polynucléotide avec un degré de sensibilité plus élevé, grâce à la mise en contact du polynucléotide avec un agent qui modifie la cytosine méthylée ou non méthylée, l'amplification du polynucléotide modifié au moyen d'un ou plusieurs nucléotides 'poids lourds' puis la détermination de la masse du produit amplifié. La masse du produit amplifié est déterminée, par exemple par la spectrométrie de masse, et mesurée par rapport à la masse d'un échantillon de contrôle ayant une séquence de base identique à celle de la séquence nucléotidique étudiée. La présence ou l'absence d'une différence de masse indique la présence ou l'absence de la méthylation du polynucléotide.
