Abstract

Die vorliegende Erfindung betrifft rekombinante Nukleinsäuremoleküle, die eine DNA-Sequenz enthalten, die ein Protein mit der enzymatischen Aktivität einer Palatinose-Hydrolase, auch als Palatinase bezeichnet, bzw. einer Trehalulose-Hydrolase, auch als Trehalulase bezeichnet, kodiert, und die Verwendung dieser Nukleinsäuremoleküle als Selektionsmarker in transformierten Zellen. Insbesondere betrifft die Erfindung rekombinante Nukleinsäuremoleküle, die eine DNA-Sequenz enthalten, die ein Protein mit der enzymatischen Aktivität einer Palatinase bzw. einer Trehalulase kodiert, und worin diese DNA-Sequenz unter Kontrolle von regulatorischen Sequenzen eines in Pflanzen aktiven Promotors steht. Weiter betrifft die Erfindung Vektoren und Wirtszellen, die die erfindungsgemäßen rekombinanten Nukleinsäuremoleküle enthalten. Weiter werden Verfahren zur Herstellung transformierter Pflanzenzellen und Pflanzen unter Verwendung der rekombinanten Nukleinsäuremoleküle und Vektoren bereitgestellt. Weiter betrifft die Erfindung transgene Pflanzen, deren Ernteprodukte und Vermehrungsmaterial sowie transgene Pflanzenzellen, die die erfindungsgemäßen Nukleinsäuremoleküle oder Vektoren enthalten oder durch das erfindungsgemäßen Verfahren hergestellt werden. Die Erfindung betrifft insbesondere auch die Verwendung von DNA-Sequenze, die ein Protein mit der enzymatischen Aktivität einer Palatinose-Hydrolase bzw. einer Trehalulose-Hydrolase kodieren, als nutritiver Selektionsmarker in Pflanzenzellen zur Selektion transformierter Pflanzenzellen und Pflanzen. Des weiteren betrifft die Erfindung ein Verfahren zur Selektion von Mikroorganismen auf Palatinose- bzw. Trehalulose-haltigem Medium.
