  • the eluate was then analyzed using a Hewlett-Packard Diode- Array Ultraviolet Detector set at 205 nm. Two large peaks appeared in the eluate that corresponded to those obtained in the analytical column in Example 1. These peaks had no overlap with each other. About 1.5 mg were collected from the first peak, and about 3.5 mg were collected from the second peak.
  • the collected portions of the peaks were each analyzed by electrospray tandem mass spectrometric characterization.
  • the peaks were dissolved in 10 mM ammonium acetate to form sample solutions containing 10 ⁇ g of triglycerides per ml of solution.
  • Approximately 4 ⁇ l per minute of each sample solution was fed into a Sciex API-Ill ES/MS/MS mass spectrometer (Toronto, Canada) using a multiplier of -6000 volts, an ionspray voltage of 5000 volts, and a 60 volt orifice.
  • the triglyceride in the first peak had a total molecular weight matching that of a triglyceride having a DHA residue, a DPA residue, and a palmitic acid residue. It also showed that the triglyceride in this peak contained a constituent having a molecular weight matching that of a palmitic acid residue, a constituent having a molecular weight matching that of a DPA residue, and a constituent having a molecular weight matching that of DHA.
  • the triglyceride in the second peak had a total molecular weight matching that of a triglyceride having two DHA residues and a palmitic acid residue. The triglyceride also contained a constituent having a molecular weight matching that of a palmitic acid residue and a constituent having a molecular weight matching that of a DHA residue .
  • approximately 100 mg of the Kelco DHA-rich acid was incubated in 2.0 ml of 4% HC1 in methanol for 2 hours at 100°C to transesterify the triglycerides in the oil (i.e., the fatty acid residues of the triglycerides were cleaved from the glycerol backbones of the triglycerides to form methyl esters of the fatty acid residues).
  • Approximately 1.0 ml of toluene was added, and the mixture was allowed to stand at 100°C for another hour. Next, the mixture was partitioned between 2 ml of H 2 0 and 2 ml hexane. The hexane fraction yielded 80 mg of yellow oil.

Abstract

La présente invention concerne un procédé permettant de séparer un premier triglycéride comprenant une fraction d'acide docosahexaenoïque d'un deuxième triglycéride. Le procédé consiste à prendre un mélange à traiter comprenant le premier triglycéride et le second triglycéride, et à l'introduire dans une zone de séparation chromatographique, puis à isoler une fraction du mélange à traiter dans la zone de séparation où le rapport massique premier triglycéride / second triglycéride est supérieur à celui du mélange à traiter. La zone de séparation comprend une phase stationnaire qui comprend des ions de métal capables de se coordonner avec une double liaison d'une fraction d'acide gras du premier triglycéride de façon à former avec la fraction d'acide gras un complexe métallique.
