Abstract

Die vorliegende Erfindung betrifft ein Verfahren zur Erzeugung eines Endoinulinase produzierenden Mikroorganismus. Erfindungsgemäß sind folgende Verfahrensschritte vorgesehen: Sammeln von Erdproben und/oder Gewebeproben aus dem Wurzelbereich inulinspeichernder Pflanzen, Selektion von Mikroorganismen, die Inulin als einzige Kohlenstoffquelle nutzen, durch Zufügen von inulinhaltigem Medium über einen definierten Zeitraum, Vereinzeln und Kultivieren der selektierten Mikroorganismen. Die vorliegende Erfindung betrifft ferner einen Mikroorganismus mit reiner Endoinulinase-Aktivität sowie ein Verfahren zur Detektion von Inulinase-Aktivität mit folgenden erfindungsgemäßen Verfahrensschritten: Markieren von Inulin mit einer Markierungssubstanz, Inkubation des markierten Inulins mit Inulinase über einen definierten Zeitraum, Abtrennung des nicht umgesetzten markierten Inulins, Bestimmung der Extinktion des Ansatzes bei 592 nm.
