Die Fläschchen sind jeweils mit einer Gummimembran verschlossen und für anaerobe Bakterien mit einer Schutzgasatmosphäre gefüllt. Zu den angegebenen Zeiten wird jeweils 0,1 ml der Mischung entnommen auf Agarplatten, welche für den betreffenden Mikroorganismus geeignet sind, ausgestrichen und bei 37° bebrütet.
