Abstract

Настоящее изобретение относится к контролю массы тела животных с помощью белков ожирения (Ож) и нуклеиновых кислот, экспрессирующих эти белки. В более широком контексте настоящее изобретение относится к выявлению нуклеотидных последовательностей генов, кодирующих такие белки. Эти нуклеотидные последовательности соответствуют мышиному и человеческому генам Ож, которые играют критическую роль в регуляции массы тела и ожирения. Предварительные данные свидетельствуют о том, что полипептиды, кодируемые указанными генами, функционируют подобно гормонам. В настоящем изобретении также описаны молекулярные зонды и праймеры для амплификации в полимеразной цепной реакции (ПЦР). Описан также клонирующий вектор, содержащий указанные последовательности, связанные с последовательностями, контролирующими его экспрессию. Далее описаны клетки бактерий или млекопитающих трансфицированные соответствующим экспрессирующим вектором и возможное применение указанных генных конструкций. Описаны также антитела к полипептиду гена Ож. Предложен метод, позволяющий контролировать массу тела животных. Приведены последовательности генов, кодирующих две изоформы соответствующих полипептидов мыши и человека.
