Abstract
Description
Description of the invention
The invention relates to medicine, in particular to pharmacology, and relates to the method of obtaining biologically active substance 2 (BAR), which is isolated from plant material and contains a mixture of flavonoids and amino acids and can be used for the manufacture of drugs of specific action for the treatment and prevention of pathological conditions, preferably, for the treatment of infections caused by the herpes virus, for the complex treatment of hepatitis B and hepatitis C, as well as for the treatment of patients with immunodeficiency.
А61КЗ5/78, 1999).There is a known method of obtaining BAR with antitumor activity, in which the green parts of plants of the Riyozrogacea family, mainly plants of the genus Riyozrogyte, are used as plant raw material 70, and which consists in the fact that the extraction of components from raw materials is carried out by maceration in an organic solvent, shaking the solution with another solvent, treatment of each phase with another organic liquid, separation and collection of separated components, and before each stage of the process, they are drained and cleaned. Ethyl alcohol is used as an organic solvent. At the same time, the ratio of vegetable 12 raw materials to the extractant is 1:10 - 10:11. Processing time - from 5 minutes. up to 3 months, and the processing temperature - from 5 to 1002C (patent of Ukraine Mo26321, class A61KZ5/78, 1999).
The biologically active substance of plant origin, obtained by this method, has a fairly high activity and low toxicity compared to other medicinal preparations, however, the method of their preparation described above is very complex, material-intensive and time-consuming due to the presence of many operations and the use of special reagents.
A known method of obtaining BAR for the prevention and treatment of pathological conditions (international application UMO 91/11191, cl. IPC Ab1KZ5/78, 1991), in which the green parts of grass plants of the Sgatipeaea family, collected at the stage of their development, are used as plant raw materials, and which includes the selection of liquid water- and fat-soluble fractions in the form of juice from this raw material and their purification from solid impurities by centrifugation with subsequent high-quality fine filtration of the particles remaining after centrifugation. Extraction of juice from plant raw materials is mainly carried out by squeezing, crushing or crushing plants. The obtained biologically active substance is stabilized by either thickening with the help of spray (sublimation) drying, or by introducing a special preservative - stabilizer, and BAR stabilization is recommended to be carried out within two hours after its extraction. 30 The disadvantage of this method of obtaining a biologically active substance is also the complexity of the technological process, (Se) which consists of the sum of time-consuming, material- and energy-intensive operations, due to the use of such energy-intensive equipment as a centrifuge, crusher, press, and the use of special reagents, as well as about performing an extract purification operation in two stages: centrifugation and fine filtration. In addition, one of the disadvantages of this method is the duration of the technological process.
«In addition, the use of valuable grain crops as raw materials, in particular, wheat, rye, barley, oats, rice and other similar cereals, for obtaining BAR is not economically beneficial. The mention of the possibility of using for these purposes the wild herbs of the Sgatipeae family is not specific, because this family is represented in Ukraine by 334 species (98 genera) of plants. "
The BAR obtained in this way cannot be used independently as a drug for the treatment of З7З 70 pathological conditions, but needs to be mixed with a pharmacologically acceptable liquid carrier. In addition, this BAR has a relatively small specific antitumor activity and insufficiently high antiviral activity. "with the closest to the claimed method, according to the set of features, is the method of obtaining BAR for the prevention and treatment of pathological conditions (patent of Ukraine Mo22549 A, AbIK 35/78, 1997) according to which the green parts of cereal plants of the Sgatipeae family are used as plant raw materials in in the form of a mixture of plants of the genus Saiatadgovziiz Adapz and the genus Oezspatrzia Veashm, collected after sweeping and before the beginning of flowering on the spikelets, which includes separate drying of the plants to a moisture content of 8-2095, mixing them at a weight ratio of Saiatadgoviya Adapz 40-6095 and Osezspatrzia Veaiim 60-4095, separation of liquid water-soluble fractions from this raw material by extracting them with 92-99.896 ethyl alcohol at a mass ratio of raw materials to alcohol as 1:3-1:8 by the method of maceration at a temperature of 18-47 "Sb with simultaneous (22) 50 periodic control of the refractive index of the extract, and after this index reaches a value of "from 1.362-1.364, water-soluble fractions are purified from solid impurities by filtering them through a large-pore filter to obtain the final product.
This method of obtaining BAR is simpler than the method described in the prototype, because it excludes the use of a number of such time-consuming and energy-intensive operations as squeezing juice from raw materials, its purification by centrifugation and high-efficiency fine filtration, as well as stabilization of the extract.
GF) But this method of obtaining BAR involves the use of a mixture of wild cereal plants of the genus Saliatadgovii 7 Adapz and the genus Oezspatrzia Veashu, which are taken in a narrow range of component ratios, which does not allow obtaining BAR with a wide spectrum of specific action. Due to this, medicinal preparations manufactured at this BAR will have a narrow therapeutic capacity. In addition, the collection of these plants is also carried out at a narrow stage of their 60 development: after the sweeping of the stems and before the beginning of the flowering spikes, which leads to a decrease in the amount of extractive substances in the raw materials. All this leads to a narrowing of the degree of use of the specified raw material base in one season.
The disadvantage of this known method is also the need to measure the refractive index of the extract in the process of extracting water-soluble fractions, which makes the technological process more complicated and energy-intensive. Until 69, it is also necessary to have an analytical device - a refractometer.
The invention is based on the task of creating a simple and effective method of obtaining a stable

Solving the given task allows not only to expand the limits of the use of the known raw material base for obtaining BAR of plant origin, but also to create a BAR of such a composition that provides a wide spectrum of 70 selective specific pharmacological effects of it on the human body and, accordingly, a high therapeutic effect with low toxicity.
9о:The set task is achieved by the fact that in the method of obtaining BAR of specific action for the prevention and treatment of pathological conditions from plant raw materials, in which the green parts of wild cereal plants of the family Sgatipeae of the genus Saiatadgoviiz Adapz and the genus Oezspatrzia Veashm, collected at stage 7/5 of their development, are used as raw materials , which includes separate drying of these plants to a moisture content of 8-2095, mixing them to obtain raw materials, separating liquid water-soluble fractions from these raw materials by extracting them with ethyl alcohol and cleaning them from solid impurities by filtration to obtain the final product, according to the invention, wild cereals plants are harvested after the stems are exhausted and before the flowering of the spikelets, the separation of liquid water-soluble fractions from raw materials is carried out at a mass ratio of raw materials to 2% alcohol as 1:1 - 1:10 with simultaneous periodic control of the dry residue mass indicator and when this indicator reaches a value equal to 0.5-1.596, conducted and their cleaning, and wild plants are mixed with the following ratio of components, mass. 9 o'clock:

Extraction of liquid fractions from vegetable raw materials with ethyl alcohol is carried out by the method of maceration and/or remaceration at a temperature of 18-472C. Moreover, to increase the efficiency of extraction of extractive substances from a mixture of herbs, 90-99,895% ethyl alcohol is used. The essence of the claimed method of obtaining BAR is that using a mixture of cereal plants of the family Sgatipeae of the genus SaYatadgovziz Adapz and the genus Yuezspatrzia Veacsm in the specified ratio of "0 components, increases the content of the extractive substances obtained, expanding the spectrum of selective and specific pharmacological action on the body a person
-In addition, the qualitative and quantitative composition of BAR, obtained by the claimed method, is significantly influenced by the time of harvesting plants of the genus Saiatagoziis Adapz and the genus Oezspatrzia Veheim, because in the specified specific period of their vegetation - after sweeping the stems and before the end of flowering spikelets, the green parts of these plants contain the maximum amount of flavonoid compounds that have the maximum selective specific activity. In addition, the component composition of the mixture of plants is equally important, because the content of extractive substances in the extract depends on it, and this will affect the specificity of the BAR action. Moreover, "the quantitative content of herbs in the mixture is directly dependent on the weather conditions and the seasonality of their collection. -
In addition to the selection parameters of specific component ratios of plants in the mixture, the development of the conditions for carrying out operations of the technological process is no less important for the method of obtaining BAR from this plant raw material.
According to the present invention, the biologically active substance is obtained in the following way.

The obtained biologically active substance is a highly stable agent, suitable for use as a medicinal product or as an active ingredient for the preparation of medicinal products.
The indicated humidity interval of the plants to which the raw materials are dried is optimal, because 60 drying the plants to a humidity lower than 895 will lead to the loss of bound water and, as a result, to the destruction of active extractive substances, and drying the plant raw materials to a humidity , which will be more than 2095, will lead to the fact that the content of free water in the extract will exceed the amount of free water necessary for its dilution of extractive substances found in plants, and this will lead to a decrease in the content of the active principle in the extract. because the mass ratio of raw materials to the extractant, which is 1:1-1:10, is also optimal, because the ratio of the specified components is more than 1:11, that is, the content of the extractant by mass will be less than the raw materials, the amount of extractive substances, in extract will be more than 1.595, which means that coagulation of the extractant will occur, and the drug will be ineffective for treating patients. When the mass ratio of the raw material to the extractant is less than 1:10, that is, when the mass of the extractant exceeds the mass of the raw material by more than 10 times, the dry residue will be less than 0.595, which will also lead to a decrease in the effectiveness of the medicinal product.
Determination of the dry residue mass index is carried out in order to establish the end of the process of extracting plant raw materials with an organic solvent. The selected interval of this indicator is 7/0. 9.5-1.596, allows you to stop the extraction process in time to get a highly efficient final product.
Moreover, when the dry residue mass indicator is less than 0.595, the activity of the biologically active substance, which will slow down under the action of inhibitors, will be below the globally accepted recommendations for the selection of medicinal products, i.e. below 2 logarithms of the infectious dose, as a result of which the drug will be ineffective. When the mass index of the dry residue of the extract will be more than 1.595, a precipitate will form in the obtained 7/5 biologically active substance, which means that it will also be ineffective as a medicinal product.
Thus, only with a mass ratio of raw materials and alcohol in the range of 1:1-1:10 and with a value of the dry residue indicator in the range of 0.5-1.5906 will the obtained BAR be a highly effective therapeutic drug of specific action.
The specified temperature interval of maceration and/or remaceration of the alcoholic extract will allow the process of extracting BAR from plants to be carried out most completely, because at a temperature below 1892, the percentage of the target product yield decreases, and at a temperature above 472C, the decomposition of extractive substances occurs by coagulation of plant amino acids.
To ensure the maximum release of extractive substances from dried plants of the genus Saiatadgovii Ha
It is very important to perform extraction with 90-99.8906% ethyl alcohol of Adapz and the genus Oezspatrzia Veatsm, because with an alcohol concentration below 90905, the completeness of the release of active buds from plants is significantly reduced, and with an alcohol concentration of more than 99.895, the yield of the target product does not increase, but there is an overconsumption of alcohol.
