Description
A useful model belongs to medicine, in particular to microbiology, and can be used in determining the sensitivity of Musobasiegisht Shbegsytsiov (M. yIshbegstsiov) to secondary metabolites of streptomycetes.
The emergence and spread of M. Shregsioviz creates a global threat to the control of tuberculosis patients not only in Ukraine, but also in the whole world. Modern schemes for studying the sensitivity of isolates of M. yshbregsciovie to antituberculosis drugs testify to an important aspect of controlling the disease. Observation and analysis of indicators of resistance to anti-tuberculosis drugs helps in qualitative detection and monitoring of the scale of spread of multi-resistant strains. M. iiregstiov5ie has properties that cause natural resistance to anti-tuberculosis drugs. The use of modern schemes for the specific treatment of tuberculosis can cause the development of chemoresistance, which is determined by a change in chromosomal genes, the transfer of resistance gene plasmids, which prompted intensive research into the extrachromosomal inheritance of antibiotic-producing streptomycetes.
Streptomycete strains can simultaneously produce metabolites with antibiotic properties of different chemical nature and mechanism of action. Further, their use in the microbiological industry as producers of antibiotics found wide application in medicine.
One of the main research methods of M. (Shregscio5zize) remains the study of antibiotic sensitivity.
їШбегсиЇовіє (Наказ М 45 від 06.02.2002 МОЗA known method of determining the sensitivity of M. yShbegsiYovie (Order M 45 dated 06.02.2002 of the Ministry of Health
України "Про затвердження Інструкції з бактеріологічної діагностики туберкульозної інфекції").of Ukraine "On approval of the Instructions for bacteriological diagnosis of tuberculosis infection").
Levenstein-Jensen nutrient medium with different concentrations of antimycobacterial drugs is poured into test tubes of approximately 5.0 ml and coagulated in a slanted form at a temperature of 85 "C for 30 min. in an apparatus for coagulation of blood serum. Test tubes with prepared media containing medical preparations, and one control tube with a medium that does not contain drugs, inoculate 0.2 ml of the prepared culture suspension in each tube. The tubes are placed in an inclined state in a thermostat at a temperature of 37 "C.
From Crops is reviewed every week. After 2-3 weeks after sowing, the obtained results are recorded. In the case of insignificant growth of the samples in the control, incubation is continued for another week in the thermostat, after which the final indicators regarding the presence or absence of growth of the tested isolates are recorded.
According to the protocol for the sensitivity study of M. Shvregsiiov5 and 5, during the introduction of test extracts of secondary metabolites of streptomycetes, atypical changes in the nutrient medium were observed, namely the folding of the protein that is part of the medium. When diluting the drug in the Levenstein-Jensen liquid medium, uneven distribution of the drug occurred, which led to false results.
The basis of a useful model is the task of creating a method of studying the sensitivity of M. imbetgstsovziz to secondary metabolites of streptomycetes by achieving a uniform distribution of the studied extracts to avoid false results.
їирегсиціовів до вторинних метаболітів стрептоміцетів, що включає введення досліджуваних екстрактів у пробірку з поживним середовищем Левенштейна-Йєнсена, яке знаходиться у нахиленому стані, згідно з корисною моделлю, досліджуваний екстракт вторинних метаболітів стрептоміцетів вводять у пробірку на верхній край нахиленої поверхні поживного середовища Левенштейна-The task is solved by the fact that in the method of studying the sensitivity of M. irregsitsiov to secondary metabolites of streptomycetes, which includes the introduction of the studied extracts into a test tube with nutrient medium of Levenstein-Jensen, which is in an inclined state, according to a useful model, the studied extract of secondary metabolites of streptomycetes is introduced into test tube to the upper edge of the inclined surface of the Levenshtein nutrient medium-
Jensen with subsequent inoculation of bacterial suspension M. (Shbegsytsov.
The advantages of the proposed method are the introduction of studied extracts of secondary metabolites of streptomycetes on the upper edge of the inclined surface of the nutrient medium followed by the addition of the studied strain, which ensures wetting of the entire surface of the nutrient medium and eliminates the possibility of uneven distribution of the studied extracts.
