Abstract
本發明屬於保健品領域,本發明提供了一種粉劑,由低聚木糖、異麥芽低聚糖、甘露糖醇、菊粉、黨參、猴頭菇多糖、山藥、茯苓、白扁豆、陳皮、砂仁和白術製備而成,原料全部來源於天然中草藥,無輔料添加,成分天然,標籤清潔,符合人們追求食品天然健康的理念趨勢,且服用劑量小,服食方便,可直接口服或溶於水中服用,冷水可溶,入口即化,口感好,吸收快。本發明所述粉劑製備方法簡單,適合大規模生產攜帶方便,製得的粉劑穩定性好,存放時間長。實驗表明本發明所述粉劑對胃黏膜損傷有改善作用,可有效降低無水乙醇造成的急性胃黏膜損傷,且對腸道菌群具有良好的調節作用,可用於製備具有調節胃腸道、保護胃黏膜功能的保健食品。
Description
本發明屬於保健品領域,具體涉及一種具有調節腸道菌群和保護胃黏膜功能的粉劑及其製備方法,尤其是以天然中草藥為主要原料製備的具有調節腸道菌群和保護胃黏膜功能的粉劑及其製備方法。
胃腸道為人體消化系統的主要器官,一般指胃、十二指腸、小腸、大腸和直腸。胃腸道功能:主要有消化、吸收和排泄。消化功能既釘胃酸、各種消化液及其消化酶等生化作用,亦需胃腸蠕動的機械物理運動,還有胃腸道正常腸道菌群的生理作用共同完成。吸收功能則以胃腸黏膜細胞完成為主;排泄則是直腸及其括約肌的功能。保持良好的胃腸功能既要有良好的飲食衛生、飲食習慣、進餐環境,也要及時防治胃腸道疾患,如消化不良、潰瘍病、腹瀉、便秘及痔瘡等症。
隨著現代生活節奏的加快、工作壓力的不斷加大、飲食睡眠等日常行為無規律化逐漸加劇,人們的胃腸功能也越來越低下,常出現打嗝、口乾、口苦、胸悶、反酸、厭食、噁心、嘔吐、少食後飽脹、腹痛、腹脹、腹瀉或便秘等症狀,進而出現疲倦、乏力、嗜睡、精神不集中等表現,使人長期處於一種亞健康的狀態、對人們的日常生活、工作、學習等造成嚴重的影響。
近年來人民生活水準明顯提高人們的消費觀念、健康觀念發生了較大變化。為規避不健康帶來的各種不利影響,使人們越來越重視營養保健品的使用。目前改善胃腸道功能類保健食品大多以口服液的形態存在,功能主要集中在調節腸道內有益菌群、潤腸通便,改善便秘,修復胃黏膜等方
面。口服液劑攜帶不便,穩定性差,存放時間短。片劑在壓片時大都需要加入澱粉、羥甲基纖維素等大劑量輔料,崩解時間長。同時對吞咽有困難的人群服用不方便,如老人和兒童。
有鑑於此,本發明的目的在於針對習知技術存在的缺陷提供一種具有調節腸道菌群和保護胃黏膜功能的粉劑及其製備方法。本發明所述保健粉劑以天然中草藥為主要原料製備,具有調節腸道菌群和保護胃黏膜功能,同時不含有澱粉、羥甲基纖維素等輔料,對有效成分的溶出和生物利用度基本沒有影響,且粉劑的性狀良好。
為實現本發明的目的,本發明採用如下技術方案:
一種具有調節腸道菌群功能和保護胃黏膜的保健粉劑,由菊粉、低聚木糖、異麥芽低聚糖、甘露糖醇、黨參、猴頭菇多糖、山藥、茯苓、白扁豆、陳皮、砂仁和白術製備而成。
進一步地,作為優選,所述菊粉、低聚木糖、異麥芽低聚糖、甘露糖醇、黨參、猴頭菇多糖、山藥、茯苓、白扁豆、陳皮、砂仁和白術的品質比為(10~30):(10~30):(10~20):(10~20):(3~10):(3~10):(3~8):(3~8):(2~5):(2~5):(1~5):(1~5)。
在一些實施方案中,所述菊粉、低聚木糖、異麥芽低聚糖、甘露糖醇、黨參、猴頭菇多糖、山藥、茯苓、白扁豆、陳皮、砂仁和白術的品質比為10:20:10:10:5:10:5:8:3:5:2:2。
在一些實施方案中,所述菊粉、低聚木糖、異麥芽低聚糖、甘露糖醇、黨參、猴頭菇多糖、山藥、茯苓、白扁豆、陳皮、砂仁和白術的品質比為20:10:20:20:4:4:4:4:3:3:3:3。
在另一些實施方案中,所述菊粉、低聚木糖、異麥芽低聚糖、甘露糖醇、黨參、猴頭菇多糖、山藥、茯苓、白扁豆、陳皮、砂仁和白術的品質比為20:15:14:10:7.5:7.5:7.5:6:5:5:3.5:3.5。
本發明還提供了所述粉劑的製備方法,將黨參、山藥、茯苓、白扁豆、陳皮、砂仁和白術加水提取,收集提取液,加入菊粉乾燥、低聚木糖、異
麥芽低聚糖、甘露糖醇、猴頭菇多糖混合後乾燥粉碎。
其中,所述的加水提取優選為加水煎煮提取。
進一步地,所述的加水煎煮提取優選為加水煎煮提取1-2次,每次加3~20倍的水提取0.5~4小時。
在一些實施方案中,所述加水煎煮提取為加20倍水煎煮提取3小時。
在一些實施方案中,所述加水煎煮提取為第一次加5倍~20倍的水提取0.5~3小時,第二次加3倍到15倍的水提取0.5~2小時。在一些實施例中,第一次加入15倍水提取2小時,第二次加入10倍水,提取1小時。在另一些實施例中,第一次加5倍水,提取1小時,第二次加5倍的水提取0.5小時。
本發明所述製備方法在加水提取後收集提取液,進行濃縮。所述濃縮優選為真空濃縮。
進一步地,本發明所述製備方法在濃縮後加入菊粉乾燥優選為噴霧乾燥、冷凍乾燥、帶式乾燥、微波乾燥或真空乾燥。
本發明所述製備方法所述粉碎細微性優選控制在40目到80目。
在某一具體實施方案中,本發明通過灌胃給予本發明所述粉劑,觀察本發明所述粉劑對胃黏膜損傷的影響,結果顯示,本發明所述粉劑對胃黏膜損傷有改善作用,可有效降低無水乙醇造成的急性胃黏膜損傷。
在某一具體實施方案中,本發明通過灌胃給予本發明所述粉劑,觀察本發明所述粉劑對腸道菌群的影響,結果顯示本發明所述粉劑對腸道菌群具有良好的調節作用。
因此,本發明提供了所述粉劑在製備具有調節胃腸道、保護胃黏膜功能的保健食品中的應用。
由上述技術方案可知,本發明提供了一種粉劑由低聚木糖、異麥芽低聚糖、甘露糖醇、菊粉、黨參、猴頭菇多糖、山藥、茯苓、白扁豆、陳皮、砂仁和白術製備而成,原料全部來源於天然中草藥,無輔料添加,成分天然,標籤清潔,符合人們追求食品天然健康的理念趨勢,且服用劑量小,服食方便,可直接口服或溶於水中服用,冷水可溶,入口即化,口感好,吸收快。本發明所述粉劑製備方法簡單,適合大規模生產攜帶方便,製得的粉劑穩定性好,存放時間長。實驗表明本發明所述粉劑對胃黏膜損傷有

本發明公開了一種具有調節腸道菌群和保護胃黏膜功能的粉劑及其製備方法與應用。本領域具有通常知識者可以借鑒本文內容,適當改進工藝參數實現。特別需要指出的是,所有類似的替換和改動對本領域具有通常知識者來說是顯而易見的,它們都被視為包括在本發明。本發明的方法及產品已經通過較佳實施例進行了描述,相關具有通常知識者明顯能在不脫離本發明內容、精神和範圍內對本文所述的方法進行改動或適當變更與組合,來實現和應用本發明技術。

如無特殊說明,本發明實施例中所涉及的試劑均為市售產品,均可以通過商業管道購買獲得。其中所述猴頭菇多糖為猴頭菇經水提醇沉獲得,猴頭菇多糖含量>15%。
配方:
製備方法:將黨參、山藥、茯苓、白扁豆、陳皮、砂仁和白術加水煎煮提取兩次,第一次加13倍的水,提取3小時,第二次加15倍水提取,提取3.5小時,合併兩次提取液,真空濃縮;濃縮液加入菊粉、猴頭菇多糖、低聚木糖、異麥芽低聚糖、甘露糖醇進行噴霧乾燥,得中草藥提取乾粉,細微性控制在40目到80目。

2、試驗動物:SD大鼠,雄性,160-180g。
3、分組及受試樣品給予時間:
試驗動物共分5組,分別為3個劑量組、模型對照組和空白對照組。3個劑量以人體推薦量的5倍(166.67mg/kg)、10倍(333.33mg/kg)和30倍(1000mg/kg)設置。受試樣品給予時間為4周。
4、檢測指標:
胃黏膜損傷狀況、胃黏膜病理損傷及積分。
5、試驗結果:
5.1、急性胃黏膜損傷情況分析
無水乙醇損傷1h後,解剖各大鼠胃部,肉眼可見空白組大鼠胃黏膜光滑完整,沒有出血現象。其餘各組大鼠均出現了黏膜出血、組織壞死。模型組動物損傷最為嚴重,表現為大塊的淤血,呈鮮紅色。高、中、低劑量給藥組則表現為點狀或條狀出血,出血面積明顯縮小。根據評分標準計算各組動物損傷評分如表1。脾產品可劑量依賴性地減輕無水乙醇所致的胃黏膜損傷。
5.2病理組織學結果
病理學檢查結果與肉眼大體觀察結果一致,模型組動物胃黏膜損傷最為嚴重。各給藥組對胃黏膜損傷均有改善作用,且呈現劑量依賴性。
6、結論
(1)觀察指標:除了空白對照組大鼠,各組的胃黏膜損傷發生率均為100%,該產品各個劑量組損傷抑制率分別為低劑量組37%,中劑量組42.9%,高劑量組51.3%,結果證實該產品對胃黏膜損傷的保護具有劑量依賴性。
(2)病理組織學觀察及積分:通過出血點計算損傷抑制率及病理組織學分析可發現各個劑量組對胃黏膜損傷的保護效果均顯著,可有效降低無水乙醇造成的急性胃黏膜損傷,並且作用具有劑量依賴性。
按照上述方法對實施例1和實施例3所述粉劑進行檢驗,結果與實施例2結果相當。
1、受試樣品:採用實施例2製得粉劑樣品
2、試驗動物:Balb/C小鼠,雄性,18-22g。
3、分組及受試樣品給予時間:
試驗動物共分4組,3個劑量組和空白對照組。3個劑量以人體推薦量的5倍(166.67mg/kg)、10倍(333.33mg/kg)和20倍(666.66mg/kg)設置。受試樣品給予時間為4周。
4、觀察指標:體重、雙歧桿菌、乳桿菌、腸球菌、腸桿菌、梭菌、梭桿菌、擬桿菌。
5、試驗方法:
在給予受試樣品之前,無菌採取小鼠糞便0.1g,10倍系列稀釋,選擇合適的稀釋度分別接種在各培養基上。培養後,以菌落形態、革蘭氏染色鏡檢、生化反應等鑒定計數菌落,計算出每克濕便中的菌落,取對數後進行統計處理,檢測腸道菌群。然後分別給予受試樣品,14天中均給予飼料和飲水。,在最後一次給予受試樣品之後24小時,與按照同樣方式取直腸糞便,檢測腸道菌群,方法同上。比較各組試驗前後及組間雙歧桿菌、乳桿菌、腸球菌、腸桿菌、梭菌、梭桿菌、擬桿菌的變化情況。
6、資料處理:
兩組間差異顯著性檢驗採用兩獨立樣本的t檢驗,多組均數間顯著性比較用單因素方差分析。
7、結果與分析
7.1臨床表現及體重增長曲線
給藥4周以後,空白對照組小鼠皮膚較蓬鬆,活動力減弱。與空白對照組比較,各藥物組小鼠毛色明亮光澤、表現活躍。
如表2顯示,空白對照組最後一周的體重有所下降,可能為給藥時間過長,出現了生理性老化及生長停滯。各劑量組小鼠體重均有增加,其中低劑量組及高劑量組與正常組比較,具有統計學差異。
7.2腸道菌群變化情況分析
給藥3周後,各組間腸桿菌數量無統計學差異外,其他菌種均出現了上升或下降。與空白對照組相比,益生菌雙歧桿菌(低、中劑量給藥組)、乳桿菌(高劑量給藥組)及擬桿菌(中、高劑量給藥組)數量均上升(表3-7)。而非益生菌腸球菌(中、高劑量組)則出現明顯下降,梭桿菌(低、高劑量組)及梭菌(低劑量組)略有下降。上述結果說明該產品對腸道菌群具有良好的調節作用。
8結論
體重增長情況:給藥4周以後,空白對照組最後一周的體重有所下降,可能為給藥時間過長,出現了生理性生長停滯。該產品各劑量組小鼠體重均有增加,其中低劑量組及高劑量組與空白對照組比較,具有統計學差異。
給藥3周後,各組間腸桿菌數量無統計學差異外,其他菌種均出現了上升或下降。除與空白對照組相比,益生菌雙歧桿菌(低、中劑量給藥組)、乳桿菌(高劑量給藥組)及擬桿菌(中、高劑量給藥組)數量均上升。而非益生菌腸球菌(中、高劑量組)則出現明顯下降,梭桿菌(低、高劑量組)及梭菌(低劑量組)略有下降。上述結果說明該產品對腸道菌群具有良好的調節作用。
按照上述方法對實施例1和實施例3所述粉劑進行檢驗,結果與實施例2結果相當。

- 一種粉劑,其係由低聚木糖、異麥芽低聚糖、甘露糖醇、菊粉、黨參、猴頭菇多糖、山藥、茯苓、白扁豆、陳皮、砂仁和白术製備而成;其中所述菊粉、低聚木糖、異麥芽低聚糖、甘露糖醇、黨參、猴頭菇多糖、山藥、茯苓、白扁豆、陳皮、砂仁和白术的質量比為(10~30):(10~30):(10~20):(10~20):(3~10):(3~10):(3~8):(3~8):(2~5):(2~5):(1~5):(1~5);所述粉劑由如下方法製備:將黨參、山藥、茯苓、白扁豆、陳皮、砂仁和白术加水煎煮提取,收集提取液進行濃縮,加入菊粉、低聚木糖、異麥芽低聚糖、甘露糖醇、猴頭菇多糖混合後乾燥粉碎;所述粉劑具有調節胃腸道、保護胃黏膜功能。
