Description
本發明係關於一種載體,包含免疫球蛋白(Ig)之Fc區(Fc domain)及半胱胺酸富含區(cysteine-rich domain),特別是運用於接合Tn,sTn,或GM3等醣質抗原,可有效地提升其抗原性。
某些特定醣質抗原例如Tn(α-N
-acetylgalactosamine-O-Ser/Thr)、sialyl Tn(sTn)、GM2及Globo H常被發現在癌細胞中有過度表現之情形。此些抗原,特別是醣質抗原決定位(sugar epitope),係可以作為癌症診斷及免疫療法之標的物。
由於醣質抗原決定位通常具有低免疫性(low immunogenicity)之特性,通常需結合一載體以提升其免疫反應,例如結合鑰孔血藍蛋白(Keyhole limpet hemocyanin,KLH)或胎牛血清蛋白(bovine serum albumin,BSA)以增強其免疫反應。然而,此方法至少具有兩種缺點。第一,KLH/BSA與醣質抗原決定位的結合效率通常較低。第二,結合後的複合物引發的免疫反應對醣質抗原決定位的專一性不佳。
因此,亟待發展出一種可增強低抗原性分子之免疫性的方法。
本發明係一載體,可增強醣質抗原Tn,sTn或GM3免疫抗原性之蛋白質,該載體包含有免疫球蛋白(Ig)之Fc區(Fc domain)及半胱胺酸富含區(cysteine-rich domain),可有效地改善醣質之抗原性。
據此,本發明之一實施例係關於一種非自然生成的多胜肽,包括一抗原呈現細胞結合區(antigen presenting cell(APC)binding domain)以及半胱胺酸富含區。抗原呈現細胞結合區可以是一免疫球蛋白(Ig)Fc片段(例如人類免疫球蛋白之Fc片段)或是毒素(例如綠膿桿菌外毒素A(Pseudomona exotoxin A)、破傷風毒素(tetanus toxin)或霍亂毒素(cholera toxin))之一受體結合區。半胱胺酸富含區包括一或多個重複(例如2-30個重複片段)之10個胺基酸之片段,在該片段中,至少有3個(例如6個)胺基酸為半胱胺酸。在一實施例中,該片段序列為Pro-Cys-Cys-Gly-Cys-Cys-Gly-Cys-Gly-Cys(SEQ ID NO:1),且半胱胺酸富含區包括7個串聯重複之此片段。同時,此發明並揭露可轉譯出上述之多胜肽的核苷酸之任一核酸(例如一種表現載體),包括一編碼有前述多胜肽片段之核苷酸,以及一種包含有此核苷酸之宿主細胞。
本發明之另一實施例係為一種免疫結合複合物,該免疫結合複合物包括前述之多胜肽偶聯結合且接合上小分子抗原(例如sTn、Tn、TF、GM2、GM3(醣質抗原)、磷酸絲胺酸(phosphoserine)、磷酸蘇胺酸(phosphothreonine)、唾液酸(sialic acid)、N-acetylglucosamine、Globo H、路易斯寡糖x(Lewis x)、路易斯寡糖y(Lewis y)或類固醇荷爾蒙等)。在半胱胺酸富含區中至少有3個半胱胺酸可結合上此抗原。此免疫結合複合物可與一輔劑(adjuvant)混合以產生一免疫組成物。當此抗原為癌症相關之抗原時(例如sTn、Tn、TF、GM2、GM3、Globo H、路易斯寡糖x(Lewis x)、及路易斯寡糖y(Lewis y)),則此免疫組成物可用於治療癌症(例如前列腺癌、乳癌、子宮頸癌、肝癌、腎癌、直腸癌、肺癌或卵巢癌等)。
本發明之又一實施例中係為給予一個體(如人類或其他哺乳動物)中有效劑量的前述免疫組成物或免疫結合複合物以誘發免疫反應的方法。在實施例中,此免疫反應係指產生一專一性抗體以辨認免疫結合複合物上之抗原,且個體係為一非人類之哺乳動物,例如鼠類、兔、綿羊、山羊或馬。
本發明之範圍亦包括一種以前述之免疫複合物或免疫組成物引發免疫反應、治療疾病(例如癌症)或製備治療該疾病之藥劑(例如疫苗、抗體)之用途。

本發明所揭露之載體可與一抗原,特別是一低免疫性之小分子,以群集形式偶聯結合。此載體包括兩個部位:(1)抗原呈現細胞結合區及(2)半胱胺酸富含區。
抗原呈現細胞結合區係為一配體(ligand),可與抗原呈現細胞之受體結合(例如樹突細胞、巨噬細胞或B淋巴細胞,因而引發內噬作用)。抗原呈現細胞之受體及其同源配體皆為該領域所熟知,參閱Abbas et al.,Cellular and Molecular Immunology,4th
ed,W.B. Saunders,pp 500-514。在此所述之抗原呈現細胞結合區可以是一免疫球蛋白(Ig)的Fc片段,例如人類免疫球蛋白(例如IgA,IgE,IgD,IgG,或IgM)或者是非哺乳內動物的免疫球蛋白(例如鼠類(mouse,rat)、兔、綿羊、山羊或馬)。抗原呈現細胞結合區也是一毒素(toxin)的受體結合區。在此所使用的「毒素」係指會與同源細胞受體作用而致病的微生物多胜肽。其受體結合區係為其中一片段,可結合至該毒素的同源受體結合區。在許多毒素的同源受體結合區,例如綠膿桿菌外毒素A(Pseudomonas exotoxin A)、白喉毒素(diptheria toxin)或破傷風毒素(tenatus toxin)等,都是該領域所習知的(可參閱美國專利6,838,553號)。
半胱胺酸富含區包括一或多個重複之10個胺基酸長之片段,在該片段中至少有3個胺基酸為半胱胺酸。在多個片段重複的情況下,這些重複可以是隨機性重複。在一實施例中,該半胱胺酸富含區包括有42個半胱胺酸。這些半胱胺酸的硫基(thio-group)可用以結合抗原。就半胱胺酸富含區的硫基的多重性而言,其可群集地偶聯結合多個抗原分子。(參閱第1圖)
本發明所描述之載體,可利用傳統的重組技術(recombinant technology)製備而成。舉例來說,可利用習知的重複模版聚合酶鏈鎖反應(Template-repeat PCR)技術製備得到編碼有該半胱胺酸富含區的核苷酸序列。再將該核苷酸序列連結至另一編碼有抗原細胞結合區的核苷酸序列,較佳地連結於其3’端,以得到編碼有該載體之核苷酸序列。將此核苷酸序列選殖於一表現載體(expression vector),並送入一適當的宿主細胞中表現該蛋白質。該表現載體又包括編碼含有一蛋白質標記之核苷酸,例如組胺酸標記(His-tag)或穀胱甘肽硫轉移酶(glutathione-S-transferase,GST),融合於蛋白質載體,用於幫助純化。若需要的話,亦可將該蛋白質標記除去,例如可利用酵素切除的方式。
就抗原而言,特別是具有低免疫性的抗原,其可偶聯於前述之載體,係藉由共價鍵結於半胱胺酸富含區之複數個半胱胺酸,以形成一免疫結合複合物。實施例中包括有下列的抗原,但並不僅限於此,例如:醣體(glycotopes)(例如Th,sTn,GM2,及GM3)、醣脂(glycolipids)(例如Globo H及SSEA3)、糖鞘脂(glycophingolipids)(例如B-I及B-II);該些抗原亦可以為類固醇荷爾蒙(steroid hormone),例如糖質皮質固醇(glucocorticoid)(例如prednisone,dexamethasone,或triamcinolone),鹽皮質素(mineralocorticoid)(例如fludrocortisone),維他命D(vitamin D)(例如dihydrotachysterol),雄激素(androgen)(例如oxandrolone或nandrolone),雌激素(estrogen)(例如diethylstilbestrol),或是孕激素(progestins)(例如norethindrone或medroxyprogesterone acetate),其中雄激素及雌激素就是所謂的性荷爾蒙。
抗原係透過一連結子偶聯於該蛋白質載體(例如馬來醯亞胺連結子可與胺基及硫基兩者發生反應)。參閱第1、2圖。連結子例如N-e-馬來醯亞胺基己酸(N-e-Maleimidocaproic acid)、m-馬來醯亞胺苯甲醯-N-烴基琥珀醯亞胺酯(m-Maleimidobenzoyl-N-hydroxysuccinimide ester)、N-e-[馬來醯亞胺己醯基氧]琥珀醯亞胺酯(N-e-Maleimidocaproyloxy]succinimide ester)以及琥珀醯亞胺-6-[β-馬來醯亞氨基丙胺]-己酸(succinimidyl-6-[β-maleimidopropionamido]-hexanoate)等。
本發明之免疫複合物可用於製備免疫組成物(例如疫苗),其方法皆為該領域所熟知。可參閱美國專利4,601,903、4,599,231、4,599,230及4,596,792號。此免疫複合物可與一輔劑混合(例如霍亂毒素(cholera toxin)、大腸桿菌熱敏性腸毒素(Escherichia coli heat-labile enterotoxin)、微脂體(liposome)、免疫刺激複合物(immune-stimulating complex)、免疫刺激序列寡去氧核苷酸(immunostimulatory sequences oligodeoxynucleotides)、或氫氧化鋁(aluminum hydroxide)或磷酸鹽(phosphate)),以及選擇性地為一種藥學上可接受載體(pharmaceutically acceptable carrier)(例如磷酸鹽緩衝液(phosphate buffered saline)或重碳酸溶液(bicarbonate solution)),或是一種賦形劑(excipient),(例如水、食鹽水、葡萄糖(dextrose)、甘油(glycerol)、乙醇(ethanol)及其組合),以形成一免疫組成物。此免疫組成物可以配製成一種可注射物、液態溶液或是一乳狀液。其中更可包含有少量的輔助成分,例如保溼或乳化成分及/或一pH緩衝成分,用以增進免疫反應之功效性。在該成分中亦可包括一聚合物(polymer),以幫助其在體內的輸送。可參閱Audran R. et al. Vaccine 21:1250-5,2003;及Denis-Mize et al. Cell Immunol.,225:12-20,2003。
本發明之免疫組成物可用於治療不同疾病,係取決於所結合之抗原(偶聯於蛋白質載體)。例如,當抗原為癌症相關抗原時(例如sTn,Th,或GM3),可引起抗癌免疫反應。本發明之免疫組成物亦可用於治療感染性疾病,例如該抗原係取自一相關之致病原時。本發明所使用的「治療」係指將包括一或多種活性成分的組成物應用或施加於一個體,該個體係患有標的疾病、具有該疾病之症狀,或是有罹患該疾病之傾向,其目的在於治癒、治療、減輕、緩和、改變、改善或影響該疾病、該疾病之症狀或罹患該疾病之傾向。
在治療標的疾病時,將本發明之免疫組成物,透過一適當路徑,取有效劑量給予一個體(例如人類)。其施加方式可以是腸胃外方式,例如靜脈、皮下、肌肉內注射,或透過栓劑或口服施用。栓劑可包含黏合劑及載體,例如聚烯烴基二醇(polyalkalene glycols)或三酸甘油脂(triglycerides)。口服劑型可包括使用例如醫藥等級的糖精(saccharine)、纖維素(cellulose)、碳酸鎂(magnesium carbonate)及其相似物。該些組成物可以具有下列形式,如溶液、懸浮液、藥丸、錠片、膠囊、緩釋形式或粉末等,並且包含10-95%的本發明所述之免疫組成物。
該免疫組成物係以製劑適合地形式施用,並且其劑量係為藥學上有效、保護性、及可產生免疫性的劑量。其施用的量主要係取決於所治療的個體,包括例如個體免疫系統所製造的抗體量,並且在需要時,造成一細胞中介之免疫反應。其中所需活性成分之精確劑量係由醫生判斷。然而,適當劑量的範圍係由該領域熟習此技術者決定,且該免疫組成物的量可能以微克(microgram)之等級。適當規律的生活對於初次施用或施用增效劑量都是有必要的,但也可能包括在一序列施用後的初次施用。該免疫組成物的施用劑量可視施用路徑及施用對象的個體大小而改變。
本發明之免疫複合物可用於在非人類哺乳動物或在人類中產生抗體,在非人類哺乳動物係為了製造抗體,而用在人類係為了治療疾病。在動物中製造單株抗體或多株抗體及其片段係為該領域所習知的技術。可參閱Harlow and Lane,(1988)Antibodies:A Laboratory Manual,Cold Spring Harbor Laboratory,New YorK等相關資料。本發明所使用的「抗體」包含完整分子或其片段,例如Fab,F(ab')2
,Fv,scFv(single chain antibody,單鏈抗體)以及區域抗體dAb(domain antibody);Ward,et. al. (1989)Nature,341,544)。該些抗體可用於偵測一種與疾病相關的抗原,並用於疾病診斷,或進一步地用於疾病治療。
多株抗體,抗體分子的異源族群,係表現於免疫個體的血清中。單株抗體,對本發明之多肽之抗體分子的同源族群,係可利用標準融合瘤技術製備而成,(可參閱例如Kohler et al.(1975)Nature 256,495;Kohler et al.(1976)Eur. J. Immunol. 6,511;Kohler et al.(1976)Eur J Immunol 6,292;and Hammerling et al.(1981)Monoclonal Antibodies and T Cell Hybridomas,Elsevier,N.Y.)。特別是,單株抗體可藉由在培養基中所培養的連續細胞株所獲得,例如參閱Kohler et al.(1975)Nature 256,495及U.S. Patent No. 4,376,110;the human B-cell hybridoma technique(Kosbor et al.(1983)Immunol Today 4,72;Cole et al.(1983)Proc. Natl. Acad. Sci. USA 80,2026,以及EBV-hybridoma technique(Cole et al.(1983)Monoclonal Antibodies and Cancer Therapy,Alan R. Liss,Inc.,pp. 77-96)。該些抗體之免疫球蛋白類別包括IgG,IgM,IgE,IgA,IgD,以及任何其他次類別。利用融合瘤產生本發明之單株抗體時,可以利用體內(in vitro)或體外(in vivo)培養的方式。利用體內(in vitro)培養的方式可產生高力價(titers)的單株抗體,是一種相當有效的方式。另外,也可以使用生產嵌合抗體(chimeric antibodies)的技術進行生產,可參閱Morrison et al.(1984)Proc.Natl.Acad.Sci.USA 81,6851;Neuberger et al.(1984)Nature 312,604;及Takeda et al.(1984)Nature 314:452等資料。嵌合抗體係為一種利用源自不同動物的不同片段所組成的抗體分子,例如源自鼠科動物之單株抗體的可變區(variable region)的及人類免疫球蛋白的恆定區(constant region)。另外,用於產生單鏈抗體的技術(參閱美國專利4,946,778及4,704,692號)可應用於製備單鏈Fv抗體的噬菌體胜肽庫(phage library)。單鏈抗體的形成係藉由一胺基酸橋(amino acid bridge)連結Fv區域的重鏈和輕鏈片段。進一步地,抗體片段可利用已知技術產生。舉例來說,此些片段包含但不限於:可利用胃蛋白脢(pepsin)切除抗體分子的方式產生F(ab')2
片段,而Fab片段可利用還原F(ab')2
之雙硫鍵的方式而得到。抗體亦可藉由習知技術進行擬人化(humanize)。例如,將帶有特定結合專一性之單株抗體擬人化(Scotgene,Scotland;及Oxford Molecular,Palo Alto,Calif.)。完全的人類抗體,例如該些表現於轉殖動物的抗體在本發明亦有說明(參閱Green et al.(1994)Nature Genetics 7,13;and U.S.Patent Nos.5,545,806 and 5,569,825)。
利用重複模版聚合酶鏈鎖反應之技術(Template-repeat PCR,可參閱Hsu et al.,Cancer Research 60:3701-3705,2000)製備一DNA片段,該DNA片段編碼有富含半胱氨酸之Pro-Cys-Cys-Gly-Cys-Cys-Gly-Cys-Gly-Cys(SEQ ID NO:1)之7個重複胜肽序列。該DNA片段經過進一步修飾,以分別在其3’及5’端具有Bam HI及Hind III之限制酶切點。再將其選殖插入一表現載體中,該表現載體具有編碼有一組胺酸標記(His-tag)的片段,以及一兔子IgG之Fc部位或是一老鼠IgG之Fc部位之片段。該表現載體可表現融合蛋白,包括其N端至C端具有兔子或老鼠之Fc部位、富含半胱氨酸胜肽之7個重複序列,以及一組胺酸標記(His-tag)的二個複本,可表示為“rFc-Cys-rich7

將前述之表現載體送入E. coli
BL21(DE3)pLysS宿主細胞中以表現該蛋白質。將成功送入的菌株以適當的培養基在37℃下培養,當OD595值達到0.3時,加入isopropyl β-D-1-thiogalactopyranoside以誘導蛋白質表現。將菌株再培養1.5小時,之後以離心方式收集該些菌株。以裂解緩沖液(lysis buffer)將菌體打破,進行離心並將上清液(supernatant)以鎳樹脂(nickel resins)進行純化。將純化後取得之蛋白質進行膠體濃度為12.5%之SDS膠體電泳(SDS-PAGE)。接著進行考馬斯藍染色法(Coomassie blue staining),得到該蛋白質的分子量約為37kD,此數值接近於rFc-Cys-rich7

蛋白質載體之理論值大小。利用抗組胺酸標記(anti-His-tag)抗體或抗Fc(anti-Fc)之抗體進行免疫墨點法(Immunoblotting),以確認該純化蛋白質就是所要的蛋白質載體。
sTn抗原係利用第3圖所示的合成路徑所合成。簡言之,神經氨酸(neuraminic acid)製成亞磷酸-N-乙醯神經氨酸(phosphite N-acetylneuraminic acid)(14),脫去保護基並與丙酮化合物(acetonide)(15)偶合,得到混合比例為3:2(α:β)之1,6-鍵結-唾液酸(1,6-linked sialoside)(16)混合物,在一般條件下所得產率為83%。以適當的丙酮化合物(acetonides)進行酸性脫保護基反應以分離α/β異構物。其中一異構物(17)利用NaOMe於MeOH中以及0.1M NaOH溶液在室溫下,進一步脫去保護基並得到化合物(18)。為了將sTn結合於載體蛋白質上,利用半胱胺鹽酸鹽(Cysteamine hydrochloride)進行光化學活化作用,將胺基結合至(18)的烯基上,以得到產物(19)(90%),且該產物係利用Biogel P2管柱純化所得。
藉由前述方法得到sTn抗原,並偶聯於一具有馬來醯亞胺官能基的連結子(參第2(B)圖),並將其分別以不同的重量比例(1:1,2.5:1及5:1),與實施例1所述之mFc-Cys-rich7
-H2
蛋白質載體混合,以經由連結子的馬來醯亞胺官能基與半胱氨酸的硫基發生反應,產生mFc-Cys-rich7
-sTn免疫複合物(參第1(C)圖)。
蛋白質載體與sTn間的偶聯反應係經由位移測定(mobility shift assay)進行確認。簡言之,將複合物進行膠體濃度為12.5%之SDS膠體電泳(SDS-PAGE),並利用抗組胺酸標記(anti-His-tag)抗體進行西方墨點分析。蛋白質載體與sTn抗原偶聯後,在SDS電泳移動較緩慢,顯示其分子量的增加係因結合了sTn之故。如同預期,一個蛋白質載體結合了多個sTn分子。利用高碘酸鹽-硫代巴比妥酸法(periodiate-thiobarbituric acid method)測定,其偶聯反應之效率約為40%,其反應效率比sTn與KLH或BSA間都來得更高。
將Balb/c小鼠(8週大)皮下注射以mFc-Cys-rich7
-sTn免疫結合複合物(包含20μg的sTn),每兩個星期注射1次共3次。每2個星期收集免疫小鼠的血清,並以墨點印漬法(dot blotting)測試anti-sTn抗體的存在。
將Balb/c小鼠(6-8週大)皮下注射以包含有mFc-Cys-rich7

蛋白質載體結合GM3之免疫複合物。首先將該免疫複合物懸浮於佛氏完全輔劑(complete Freund’s adjuvant),該佛氏完全輔劑將於兩個星期後轉變成佛氏不完全輔劑(incomplete Freund’s adjuvant)。每2個星期收集免疫小鼠的血清,並以墨點印漬法(dot blotting)測試anti-GM3抗體的存在。
各個取自注射mFc-Cys-rich7
-sTn免疫複合物之免疫小鼠之血清樣本,以TBS緩衝液稀釋1000倍(25mM Tris-HCl pH8.0,125mM NaCl),並進行免疫墨點分析(immunoblotting)以測定anti-sTn抗體之力價,方法如下所示。將不同量(即30、10、3、1、0.3、及0.1μg)的sTn點置於PVDF膜上(Millipore Immobion-PSQ ISEQ00010)。在適當環境下,將膜處理以稀釋血清樣本,以使血清樣本之抗體與sTn抗原結合,洗去未與抗原結合之抗體,並處理以一已標定之二級抗體(專一於老鼠IgG)。由結果顯示免疫小鼠之血清中所得之anti-sTn抗體,具有相當高的力價。
注射mFc-Cys-rich7
-GM3免疫複合物之免疫小鼠之血清中所得之anti-GM3抗體,其抗體力價係以前述的方法進行測定。所有血清中所得之anti-GM3抗體皆具有相當高的力價。
anti-sTn抗體之抗原決定位專一性,係以墨點印漬法(dot blotting)進行分析,方法如下所述:將不同量的GM3、一醣脂質混合物、一醣磷脂混合物,以及α-Tn,β-Tn,sTn,及N-acetylglucosamine之醣質抗原,將其點置於PVDF膜。在適當環境下,將膜以前述之稀釋血清樣本(1000 x)處理,該樣本包含有anti-sTn抗體,以使抗體與其同源抗原結合。結果顯示,anti-sTn抗體只會與sTn抗原結合,並不會與其他抗原結合,顯示該抗體對於sTn抗原具有高度專一性。
Tn,sTn及GM3抗原皆是與癌症有關的抗原分子,該些抗原藉由前述之方法偶聯於mFc-Cys-rich7

蛋白質載體以形成免疫複合物。該些免疫複合物係用以在小鼠或兔子中引發免疫反應,以製備得到小鼠的anti-Tn,anti-sTn及anti-GM3抗體,以及兔子的anti-Tn抗體。
前述之抗體係利用免疫組織化學染色法(immunohistochemical(IHC)staining assays),以檢測腫瘤組織中Tn,sTn或GM3之表現。簡言之,固定乳癌組織樣本於玻片上,以二甲苯(xylene)進行脫臘(dewax)並以酒精進行復水(rehydrate)。將玻片以0.01mM檸檬酸鹽溶液(pH 6.0)在95℃下處理40分鐘使抗原修復(antigen retrieval),並置於0.3%過氧化氫30分鐘以阻斷內生性過氧化酶(peroxidase),最後於室溫下置於含5%正常馬血清之PBS溶液30分鐘,以阻斷非專一性抗體反應。在以TBS溶液加上0.1% Tween 20(即TBST)清洗後,將玻片以老鼠之anti-sTn抗體在4℃下處理40分鐘。再將玻片以TBST洗滌以除去未結合之抗體,並以HRP-conjugated anti-mouse IgG(SUPERPICTURE POLYMER KIT Zymed Laboratories,Inc.)在室溫下處理10分鐘。再以TBST洗滌一次以除去未結合之抗體,以DAB顯色劑(DAB chromogen)進行顯色,並以蘇木精(Mayer's hematoxylin)進行復染。據此,組織為去水並固定在玻片上。由結果顯示出,小鼠之抗sTn抗體(mouse anti-sTn antibody)會與乳癌組織中的sTn反應,但免疫前血清(pre-immune serum)則無反應。
兔子抗Th抗體(rabbit anti-Tn antibody),係以前述方法製備,並以如同前述的免疫組織化學染色法(IHC),檢測由26位罹患不同期別前列腺癌的患者所取出的組織中的Tn表現量。由癌症組織中可得到陽性染色反應之結果,但在正常的組織中則無陽性染色反應。且由其結果觀察到訊號強度與癌症之惡化程度成正比。
將前述的小鼠抗GM3抗體用於免疫組織化學染色(IHC),並藉由該抗體之辨識,顯示腫瘤組織中的GM3表現。
含有mFc-Cys-rich7
-Tn之免疫複合物之抗Th疫苗以及輔劑(adjuvant)之抗癌效果,係在前列腺癌小鼠模型腫瘤(Transgenic Adenocarcinoma of the Mouse Prostate(TRAMP))中進行測試。雄性TRAMP小鼠(10週大)係以疫苗使其免疫(每2周1次共4次),而未引發免疫反應之小鼠係作為對照組。由結果顯示出對照組產生肝及腎腫瘤轉移,然而在免疫小鼠則無腫瘤的產生。進一步地,免疫小鼠存活時間亦較控制組小鼠更長。而利用免疫組織化學染色檢測(IHC assay),在免疫小鼠的前列腺中亦未發現Tn的表現。
在本發明之說明書中所揭露的所有技術特徵皆可任意組合。所揭露之技術特徵可以被相同功能、等效、具有相同目的的技術特徵所取代。因此,除非有明確地闡明,否則,各實施例中的技術特徵僅是等效的上位概念中的其中一種實施方式。經由以上實施例之說明,凡精於此項技藝者當可依據上述之說明而作其它種種之改良,惟這些改變仍屬於本發明之發明精神及以下所界定之專利範圍中。

- 一種單離之核苷酸,其係編碼如申請專利範圍第1項所述之多肽。
