Abstract
Description
The invention relates to veterinary medicine, in particular to veterinary obstetrics, and can be used for the treatment of subclinical mastitis in lactating cows.
2001. - С. 105-113.].The relevance of the proposed method is determined by the high incidence of lactating cows with mastitis, which recently tends to increase and is 80% [Parikov V.A., Slobodyanik V.I., Klimov N.T. Etiological and pathogenetic aspects of mastitis in cows, methods and means of its prevention and therapy // Ecological and adaptive strategy of animal health and productivity in modern conditions: Monograph-Voronezh. 2001. - S. 105-113.].
И. №1-2. - С. 135-144 (2007)].In the occurrence and spread of inflammatory diseases of the mammary gland in cows, factors that reduce the resistance of the mammary gland and the body as a whole play an important role [Slobodyanik V.I. Veterinary Bulletin. No. 1-2. - S. 135-144 (2007)].
- 2016. - №6. - С. 867-874].Modern pharmaceuticals offer a wide range of veterinary drugs and treatments for mastitis in cows. The main active ingredients of these drugs are antibiotics. However, their widespread and often unsystematic use has led to a decrease in efficiency due to the development of resistant strains of microorganisms [Artemyeva OA, et al., Agricultural biology. - 2016. - Volume 51. - No. 6. - S. 867-874].
Currently, an alternative to the use of antibiotics in the treatment and prevention of mastitis in cows is the use of immunostimulating drugs obtained using recombinant protein and cryofractionation technologies.
An analogue is known - a method for the treatment of mastitis in cows [RU 2320325 C1, A61K 31/00, 03/27/2008], involving the introduction of a complex preparation containing a 2% solution of novocaine, an antiseptic stimulant Dorogov ASD fraction-2, dimexide and 40% glucose solution, with the following ratio of components, wt. %: novocaine solution 2% - 20-40; antiseptic stimulant Dorogov fraction-2 - 1-3, dimexide - 13-15 and glucose solution 40% - the rest. The drug is heated to 37-38°C and injected intraperitoneally from the side of the right hungry fossa at a dose of 100-150 ml once a day 2-3 times in 48 hours until the cow recovers.
The disadvantage of this method of treating mastitis in cows is the multicomponent nature of the proposed complex preparation and, therefore, the complexity in preparation, as well as the complexity of administration.

Клетки этого органа вырабатывают опсонины (тафтсин, фибронектин и др.,) большой комплекс цитокинов (Ил-1, Ил-2, Ил-3, Ил-4, Ил-5, Ил-6, Ил-7, Ил-8, Ил-10, Ил-18, ФНО, гранулоцитарно-макрофагальный колониестимулирующий фактор, IFN-α, IFN-γ) вазоактивный интестинальный пептид, фактор роста гепатоцитов или рассеивающий фактор - HGF/SF, селезеночно-производный фактор роста - SDGF (Патент RU 2538721).Aminoseleton is a tissue preparation obtained from the spleen by cryofractionation. This drug is used in immunodeficiency states of various etiologies. It is known that biologically active substances that increase cellular and humoral immunity are contained in the organs of the immune system of animals, including the spleen. The cells of this organ produce opsonins (tuftsin, fibronectin, etc.), a large complex of cytokines (IL-1, IL-2, IL-3, IL-4, IL-5, IL-6, IL-7, IL-8, IL-10, IL-18, TNF, granulocyte-macrophage colony-stimulating factor, IFN-α, IFN-γ) vasoactive intestinal peptide, hepatocyte growth factor or scattering factor - HGF/SF, spleen-derived growth factor - SD (Patent RUGF 2538721 ).
М., 2005].Recombinant bovine interferon is a species-specific drug that exhibits immunostimulating and antiviral activity in cattle. The effect of the drug is determined by the total effect of the exogenous protein on the affected cells and the rapid induction of the endogenous interferon system, cellular and humoral immunity. Recombinant bovine interferon acts as an inducer of lysozyme and bactericidal activity of blood serum. It has an anti-inflammatory effect. Increases the resistance of the animal organism to the effects of DNA and RNA-containing viruses and pathogenic microorganisms [Instruction for the use of the veterinary drug "Interferon bovine recombinant IBR. Ershov F.I., Kiselev O.I. Interferons and their inductors (from molecules to drugs). M., 2005].
Testing the effectiveness of the method of treating mastitis in lactating cows was carried out on black-and-white cows of 2-3 lactation owned by LLC "SP Vyaznovatovka" of the Voronezh region.
The essence of the technical solution is explained by examples.
Tests were performed on 28 cows with subclinical mastitis. The diagnosis of subclinical mastitis was made comprehensively, taking into account the anamnesis, clinical examination and the results of milk tests. The reaction with the diagnostic reagent was carried out using a milk-control plate MKP-2. For this purpose, 1 ml of milk was milked from each lobe of the mammary gland into the corresponding wells, then 1 ml of the reagent - Masttest-AF was added using a dispenser, mixed by rotating the plate in a horizontal plane for 10-15 seconds. Accounting for the reaction was carried out for 10-15 seconds by the formation of a jelly-like clot or a change in the color of the mixture.
Animals of the first group (n=14) were treated with aminoseleton and bovine recombinant a- and y-interferons (basic method): on days 1-3-5 subcutaneously aminoseleton in increasing doses of 40-45-50 ml, and on days 2- 4-6 days - intramuscularly bovine recombinant α- and γ-interferons at a dose of 10 ml.
Animals of the second group (n=14) on days 1, 2 and 3 of treatment were intramuscularly injected with a complex preparation based on aminoselton containing bovine recombinant α- and γ-interferons with a total activity of 1 × 10 4 IU/ml on days 1, 2 and 3 of treatment at a dose of 10.0 ml per animal.
Evaluation of the effectiveness of the use of drugs was carried out on the basis of a study of milk from the affected lobes of the mammary gland with a 2% masttest solution, its bacteriological studies and determination of the content of somatic cells in it, selected before treatment and 7 days after its completion.
The results of clinical studies are presented in table 1, from which it follows that the effectiveness of the basic treatment method was 78.5%. The best therapeutic effect was achieved when using the proposed method - 85.7%, which is 7.2% more than when using the basic option.
The results of a bacteriological study of the udder secretion of cows before and after the treatment of subclinical mastitis (Table 2) indicate that 7 days after the end of treatment with the basic method, the degree of microbial contamination decreased by 9.3 times, infection - by 2.5 times. When using the proposed method, the degree of microbial contamination decreased at the end of the experience by 19.4 times, infection by 5 times. The content of somatic cells in the animals of both groups 7 days after the end of treatment was approximately the same and corresponded to the indicators of healthy animals.
Thus, the developed method for the treatment of cows with subclinical mastitis, involving the use of a complex preparation based on aminoselton containing bovine recombinant α- and γ-interferons with a total activity of 1 × 10 4 IU / ml on days 1, 2 and 3 of treatment at a dose of 10, 0 ml per animal ensures the recovery of 85.7% of cows with subclinical mastitis and helps to reduce microbial contamination of milk and normalize the content of somatic cells in it.
