Abstract
Description
The invention relates to veterinary medicine, in particular to veterinary obstetrics, and can be used to prevent mastitis in lactating cows.
2001. - С. 105-113.].The relevance of the proposed method is determined by the high incidence of lactating cows with mastitis, which recently has a tendency to increase and amounts to 80% [Parikov V.A., Slobodyanik V.I., Klimov N.T. et al. Etiological and pathogenetic aspects of mastitis in cows, methods and means of its prevention and therapy // Ecological and adaptation strategy of animal health and productivity in modern conditions: Monograph-Voronezh. 2001. - S. 105-113.].
- 1998. - С. 69.].The main factors contributing to the emergence and development of mastitis are the technical imperfection of milking machines, violation of the technology and rules of machine milking of cows. Initially, irritation occurs, which, when repeated, passes into latent aseptic inflammation. The general and local resistance decreases, conditions are created for the penetration and multiplication in the tissues of the mammary gland of various conditionally pathogenic and pathogenic microflora, which causes inflammation of various forms from subclinical, occurring independently during one or two lactations and leading to a share of atrophy, or transforming into a clinically expressed (serous, catarrhal, purulent-catarrhal, etc.) mastitis. To date, more than 100 different types of mi are isolated from the secretion of the mammary gland of patients with mastitis cows the microorganisms which [Ivashura, AI Improvement of diagnostic and therapeutic drugs to combat mastitis of cows / A.I. Ivashura, A.V. Heirs // Actual problems and achievements in the field of reproduction and biotechnology. Sat scientific tr Stavropol State Agricultural Academy, Stavropol. - 1998. - S. 69.].
Currently, for the prevention of mastitis in lactating cows, vaccines are used, mainly of foreign production (Mastivak, Vakolin, Startvak). But they do not always give the desired effect, since the proposed vaccines are specific to only 2-4 of the microorganisms of etiological importance and if this pathogen is not identical to the strain from which the vaccine is made, then the use of the vaccine becomes ineffective.
Due to the increased demands on the quality of milk and the widespread use of drug-resistant strains of microorganisms that cause the development of mastitis, further study of new drugs that stimulate the general non-specific resistance of the body, factors of local protection of the mammary gland of cows, and thereby eliminate the inflammatory process in the mammary gland, is necessary to prevent its occurrence in clinically healthy animals.
Such drugs include immunomodulators - drugs that increase the activity of the immune system, and in accordance with existing classifications five groups of immunomodulators are distinguished: drugs of microbial origin (sodium nucleinate, ribomunyl, imudon, etc.), peptide drugs (T-activin, myelopid, thymogen etc.), cytokines and preparations based on them (interferons, interleukins), synthetic preparations (polyoxidonium, lycopid, etc.), preparations based on natural factors (immunoglobulin preparations, plant extracts ).
Недостатком данного способа профилактики мастита у коров является то, что препарат применяется только для сухостойных коров (за два месяца до отела).A known method for the prevention and treatment of mastitis in cows [RF patent No. 2201756, A61K 33/00, A61K 35/78, 04/10/2003 Bull. No. 10] including intracisternal administration of an antimicrobial substance with oxygen, characterized in that as an antimicrobial substance with oxygen use ozonated refined deodorized vegetable oil in a dose of 10-20 ml, the introduction is carried out in each quarter of the udder on the 4th day of launch and for 1- 15 days before calving. The disadvantage of this method for the prevention of mastitis in cows is that the drug is used only for dry cows (two months before calving).
A known method for the prevention of mastitis in cows [RF patent No. 2173515, A01J 7/04, A61H 23/00, 09/20/2001 Bull. No. 26], including the use of ultrasonic vibrations with a frequency of 15-100 kHz and an intensity of 0.1-0.5 W / cm 2 , using a specially designed device containing a specially designed cup-shaped housing with a working vibrating organ, a channel for supplying compressed air and mounting accessories, in the form of a rack with a variable height, on the top of which a cup-shaped housing is installed.
The disadvantage of this method for the prevention of mastitis in cows is the complexity of the structures, the need for multiple (2-3 times a week) and long-term (up to 10 minutes) use, which creates inconvenience in use and the high cost of time for veterinary specialists to carry out these procedures.
The closest analogue to the prototype is a method for the prevention and treatment of mastitis in cows [RU 2201758 C2, A61K 35/45, 04/10/2003 Bull. No. 10], which provides for the introduction of a tissue preparation with an immunomodulating effect, which is used as a filtrate of the extract of the breast of healthy animals stabilized by mannitol. For prophylactic purposes, the drug is administered twice for 20-30 and 10 days before calving, subcutaneously at a dose of 0.1-0.2 ml per kg of body weight.
The disadvantage of this method for the prevention of mastitis in cows is that the industrial production of this drug is not established, so it is prepared independently (artisanally) in laboratories and meat processing plants, as a result of which the drug differs in biological activity and, of course, effectiveness.
The technical result of the invention is improving the effectiveness of the prevention of subclinical mastitis in cows.
The technical result is achieved by the fact that in the method of preventing mastitis in lactating cows, including the use of an immunomodulator, according to the invention, bovine recombinant α and γ interferons are used as an immunomodulator, which are administered monthly, from the first day after calving, intramuscularly in a dose of 2, 5 ml with an interval of 24 hours.
Bovine recombinant interferon is a species-specific drug that exhibits immunostimulating and antiviral activity in cattle. The effect of the drug is determined by the total effect of exogenous protein on the affected cells and the rapid induction of the endogenous interferon system, cellular and humoral immunity. Recombinant bovine interferon acts as an inducer of the serum lysozyme and bactericidal activity. It has anti-inflammatory effect. Increases the resistance of the animal organism to the effects of DNA and RNA-containing viruses and pathogenic microorganisms (Instructions for use of the veterinary drug “Interferon bovine recombinant IDB).
Testing the effectiveness of the method of treating mastitis in cows during lactation was carried out on cows belonging to OOO SP Vyaznovatovka, Voronezh Region.
The essence of the technical solution is illustrated by examples.
The experiment used 32 clinically healthy lactating cows, divided by the principle of analogues into three groups. The cows of the first group (n = 8) were not subjected to treatment, the animals of the second group (n = 8) from the first day after calving, twice a month every 24 hours, intramuscularly injected with recombinant α and γ interferon, 1.5 ml each, animals of the third groups (n = 8) - recombinant α- and γ-interferon in 2.5 ml, cows of the fourth group (n = 8) α- and γ-interferon in 5.0 ml, the drugs were administered for 4 months.
The effectiveness of the use of bovine recombinant α- and γ-interferons for the prevention of the incidence of mastitis was assessed weekly using a 2% masttest solution, in addition, blood and milk samples were taken monthly for laboratory tests from 5 clinically healthy cows in each group.
The results of clinical data on the prevention of mastitis in lactating cows are presented in table 1, from which it follows that in the control group, 50.0% (4 goals) of animals, including subclinical - 25.0% (2 goals) fell ill with mastitis during the experiment period clinically expressed - 25.0% (2 goals). The use of α- and γ-interferons at a dose of 1.5 ml and 5.0 ml prevented the disease in 75.0% of animals (6 goals), at a dose of 2.5 ml in 87.5% of animals.
Therefore, the monthly double use of recombinant α- and γ-interferons at a dose of 2.5 ml / goal prevents the disease in 50.0% of the animals.
Monitoring of the immunobiochemical status of animals showed (Table 2) that in the blood of cows treated with recombinant α- and γ-interferons in a dose of 2.5 ml, a decrease in the content of circulating immune complexes by 15.4 62.6% (P <0.05), with an increase in the number of lymphocytes by 7.8-22.3%, the content of total immunoglobulins by 8.6-14.3% (P <0.05), bactericidal and phagocytic activity blood serum - by 15.5-23.1% (P <0.05), 13.7-17.4% and 7.5-10.9%, respectively.
P <0.05 with respect to the beginning of the experiment
In animals of the control group with respect to cows treated with recombinant α- and γ-interferons of 2.5 ml, at the end of the experiment, a higher content of circulating immune complexes by 56.4% and a lower content of total immunoglobulins by 23.0% bactericidal and phagocytic activity of blood serum - by 18.4% and 11.9%, respectively.
Table 3 presents the indicators of cellular and humoral immunity of cows in the control group
P <0.05 in relation to animals of the experimental group
Thus, the administration of recombinant α- and γ recombinant interferons α-and γ twice a month at a dose of 2.5 ml prevents mastitis in 50.0% of lactating cows, is accompanied by normalization of the immune status of the organism of lactating cows due to both humoral and cellular link (GPR, lymphocytes, BASK, general immunoglobulins, CEC).
