Description
Изобретение относится к медицине, в частности к экспериментальной стоматологии, и предназначено для получения модели предобтурационного состояния корневого канала моляра. Для этого у взрослых крысы линии CD корневые каналы моляров обрабатывают химикомеханическим способом с однократным облучением коротковолновым ультрафиолетовым светом 255±5 нм в течение 20 секунд. Способ позволяет получить адекватную модель предобтурационного состояния корневых каналов зубов, необходимую для разработки методов лечения осложненных форм кариеса без повреждения структурного комплекса тканей пародонта.The invention relates to medicine, in particular to experimental dentistry, and is intended to obtain a model of the pre-obturation state of the root canal of a molar. For this purpose, in adult rats of the CD line the root canals of molars are treated with a chemical-mechanical method with single irradiation with short-wave ultraviolet light of 255 ± 5 nm for 20 seconds. The method allows to obtain an adequate model of the pre-obturator state of the root canals of the teeth, which is necessary for developing methods for treating complicated forms of caries without damaging the structural complex of periodontal tissues.

Технический результат достигается тем, что способ моделирования предобтурационного состояния корневого канала моляра у крысы линии CD включает этапы химикомеханической обработки корневого канала моляра, заключающиеся в том, что укорачивают коронковую часть моляра, определяют вход в корневой канал и его проходимость, затем троекратно проходят магистральный корневой канал с помощью ручных К-файлов №06/02, №08/02, №010/02, чередуя с промыванием корневого канала после каждого инструмента, при этом промывание корневого канала осуществляют с помощью эндодонтических шприцов растворами: 17-ти процентным этилендиаминтетрауксусной кислоты в количестве 1,0 мл;, 2-х процентным гипохлорита натрия в количестве 1,0 мл и дистиллированной водой в количестве 2,0 мл, после чего однократно облучают корневой канал моляра коротковолновым ультрафиолетовым излучением с длиной волны 255±2 нм в течение 20 секунд путем введения одноразового световода в магистральный корневой канал моляра до упора.The technical result is achieved in that the method of modeling the pre-obturator state of the molar root canal in a CD rat includes the steps of chemomechanical treatment of the molar root canal, which consists in shortening the coronal part of the molar, determining the entrance to the root canal and its permeability, then passing through the main root canal three times using manual K-files №06 / 02, №08 / 02, №010 / 02, alternating with washing the root canal after each instrument, while washing the root canal is carried out with using endodontic syringes with solutions: 1.0% ethylenediaminetetraacetic acid in the amount of 1.0 ml ;, 2% sodium hypochlorite in the amount of 1.0 ml and distilled water in the amount of 2.0 ml, after which the root channel of the molar is irradiated once with short-wave ultraviolet radiation with a wavelength of 255 ± 2 nm for 20 seconds by inserting a disposable fiber into the main root channel of the molar to the stop.
Реализацию способа моделирования предобтурационного состояния корневого канала моляра у крысы линии CD осуществляют следующим образом: Эксперимент выполнен в острой фазе. Эндодонтическая работа проводилась под микроскопом. У наркотизированных животных фиссурным алмазным бором путем поперечного среза укорачивается коронковая часть моляра на 2/3 ее длины и маленьким шаровидным твердосплавным бором достигается доступ к устью корневого канала. Определяют вход в корневой канал с помощью стоматологического зонда, далее определяется проходимость корневого канала с использованием К-файла 06 с конусностью 02 до дентинно-цементного соединения, контроль границы проверяется апекслокатором. Далее корневой канал обрабатываем химикомеханическим способом: с помощью ручных К-файлов №06/02, №08/02, №010/02 проходят магистральный корневой канал, чередуя с промыванием корневого канала после каждого инструмента. Промывание корневого канала зуба осуществляется последовательно после прохождения каждого инструмента (ручного К-файла) с помощью эндодонтических шприцов. Последовательность растворов: 17%-й раствор этилендиаминтетрауксусной кислоты (ЭДТА) в количестве 1,0 мл, дистиллированная вода в количестве 2,0 мл, далее 2%-й раствор гипохлорита натрия (NaClO) в количестве 1,0 мл и дистиллированная вода в количестве 2,0 мл. После чего, в магистральный корневой канал моляра до упора вводится кончик одноразового световода, проводящего коротковолновый ультрафиолетовый луч с длиной волны (255±2 нм), и проводится однократное облучение в течение 20 секунд. Эндодонтический аппарат для окончательной предобтурационной стерилизации системы корневого канала зуба содержит корпус с блоком управления с таймером. Далее через 2 часа животное выводится из эксперимента путем эвтаназии. Проводится скелетирование нижней челюсти, подготовка биологического материала, изготовление гистологических препаратов с дальнейшим анализом.The implementation of the method of modeling the pre-obturator state of the molar root canal in a CD rat is as follows: The experiment was performed in the acute phase. The sequence of solutions: 17% ethylenediaminetetraacetic acid solution (EDTA) in an amount of 1.0 ml, distilled water in an amount of 2.0 ml, then 2% sodium hypochlorite solution (NaClO) in an amount of 1.0 ml and distilled water in the amount of 2.0 ml. After that, the tip of a disposable light guide conducting a short-wave ultraviolet ray with a wavelength (255 ± 2 nm) is inserted into the trunk root canal of a molar up to the stop, and a single irradiation is carried out for 20 seconds. The endodontic apparatus for the final pre-obstruction sterilization of the root canal system of the tooth includes a housing with a control unit with a timer. After 2 hours, the animal is removed from the experiment by euthanasia. The skeletonization of the lower jaw, the preparation of biological material, the manufacture of histological preparations with further analysis are carried out.

Заявляемый способ подтвержден в эксперименте на 13 половозрелых крысах линии CD (5 самок и 7 самцов) в филиале Института биоорганической химии им. академиков М.М. Шемякина и Ю.А. Овчинникова РАН (ФИБХ). Проведен микроскопический анализ тканей пародонта после однократного воздействия коротковолнового ультрафиолетового облучения в течение 20 секунд в область первого нижнего моляра слева. По истечению 2-х часов после воздействия крысы были подвергнуты эвтаназии. В качестве контрольных образцов были использованы соответствующие ткани у этих же животных, но с противоположной, - правой стороны.The inventive method was confirmed in an experiment on 13 adult rats of the CD line (5 females and 7 males) in the branch of the Institute of Bioorganic Chemistry. After 2 hours after exposure, the rats were euthanized. Corresponding tissues from the same animals, but from the opposite, right side, were used as control samples.
В процессе проведения микроскопического анализа тканей пародонта у всех животных со стороны экспериментальных образцов микроскопических отклонений от нормы не обнаружено. Микроскопическая структура эпителия и собственной пластинки слизистой оболочки десны, а также соединительной ткани периодонта экспериментах и контрольных образцов идентична и соответствует общепринятой норме. Слизистая десны представлена многослойным плоским ороговевающим эпителием, включающим следующие слои: роговой, зернистый, шиповатый и базальный. Соотношение толщины слоев эпителия соответствует общепринятой норме. Во всех образцах со стороны рогового и зернистого слоя признаков гиперплазии не отмечено. Клеточный состав собственной пластинки слизистой соответствует общепринятой норме. Структура соединительной ткани периодонта и его клеточный состав без особенностей и также соответствует общепринятой норме. Для проведения микроскопического анализа использовали микроскоп проходящего света DMLA Leica (Германия), видеокамеру Photometries Cool SNAP cf (США), программное обеспечение «Мекос» (Россия).In the process of conducting a microscopic analysis of periodontal tissues in all animals from the side of experimental samples, no microscopic abnormalities were found. The ratio of the thickness of the epithelial layers corresponds to the generally accepted norm. In all samples from the side of the horny and granular layer signs of hyperplasia are not observed. The cellular composition of the mucous plate of its own plate corresponds to the generally accepted norm. The structure of the connective tissue of the periodontium and its cellular composition without features and also complies with the generally accepted norm. For microscopic analysis, a passing light microscope DMLA Leica (Germany), a Photometries Cool SNAP cf video camera (USA), and Mekos software (Russia) were used.
