Описана биологически активная добавка для лечебных и косметических целей (Патент РФ 2153325) и способ ее получения. Она представляет собой надосадочную жидкость экстракта из гомогената медицинских пиявок или их частей, или секрета слюнных желез или крови из пищеварительного канала медицинских пиявок, обладает антмстрессорной и антиоксидантной активностью. Способ ее получения заключается в ТОМ, что пиявки или их части гомогенизируют водой, полученный гомогенат экстрагируют водой, центрифугируют, фильтруют, собирают надосадочную жидкость. Однако добавка, полученная таким способом, недостаточно стабильна, не известно ее влияние на проницаемость кожи для содержащихся в креме или мази биологически активных веществ. Экстракции подвергают лиофилизированную массу медицинских пиявок, причем отношение лиофилизат:экстрагент при экстракции как 1:(3÷10). Соотношение лиофилизат:экстрагент как 1:(3:10). Экстракцию проводят в течение 5-70 часов при постоянном перемешивании ультразвуковой мешалкой в режиме, позволяющем сохранять стабильность эмульсии. По окончании экстракции экстракт фильтруют и затем удаляют воду до получения необходимой консистенции целевого продукта: порошкообразное вещество или жидкость, представляющая собой водный или масляный раствор целевого продукта. Последняя представляет собой емкость, герметично разделенную исследуемым кожным лоскутом на две камеры. Камеры заполнялись физиологическим раствором, а затем в первую добавляли по 200 мМ СаСl2, KНСО3, NaH2PO4, а также лизин. Через 1 час после начала опыта определяли концентрацию ионов Са+2, K+, Na+ и лизина во второй камере в опыте (Соп) и в контроле(Сконтр). В качестве контроля исследовалась проницаемость кожного лоскута после предварительного 15 и 30-минутного воздействия физиологическим раствором. Увеличение проницаемости обработанной экстрактом кожи для исследованных ионов выражено как отношение концентрации ионов, прошедших через кожный лоскут в контроле, к аналогичной величине в опыте. Во всех случаях различия статистически достоверны. Сравнение изменения активности протеазы и гиалуронидазы исследовалось в двух растворах: опыт 1 - в полученном по прототипу водном экстракте измельченных лиофильно высушенных медицинских пиявок и опыт 2 - в водном растворе сухого экстракта медицинских пиявок, полученном по заявляемому методу. В обоих случаях использовалась дистиллированная вода. Во 2-м опыте концентрация сухого экстракта медицинской пиявки была подобрана таким образом, чтобы исходная активность ферментов была достаточно близка. Активность гиалуронидазы (в нмоль глюкуроновой кислоты/мг белка/мин) и протеазы (мкмоль лейцина/мг белка/мин) определяли сразу после приготовления (получения) раствора, а также к концу 1, 3, 7, 15, 30, 60, 90, 180-х суток с момента приготовления. Весь период эксперимента растворы хранились в термостате при Т=20oС. С целью предупреждения процессов брожения в оба раствора добавлялся стандартный консервант, используемый в косметической промышленности. По результатам испытаний, представленным в таблице 3, видно, что в водном растворе биологически активной добавки, полученной по заявленному способу, активность протеазы и гиалуронидазы сохраняется в течение шести месяцев в отличие от прототипа, где активность сохраняется только 15 суток.A comparison of changes in the activity of protease and hyaluronidase was studied in two solutions:
Таким образом, биологически активная добавка, полученная предложенным способом, увеличивает в 1,5-2,3 раза проницаемость кожи, что приведет к повышению активности косметических и лекарственных средств, содержащих эту добавку. Кроме того, предложенная БАД сохраняет активность протеазы и гиалуронидазы в жидкой среде в течение 6 месяцев, что более чем в 10 раз больше, чем у прототипа.
