Description
The invention relates to the production of food products, namely the production of encapsulated fat-soluble food products, in particular fish fats with additives of biologically active substances.
Known gelatin base for encapsulation of fat-soluble drugs (castor oil, extract of male fern, the drug "Olimetin") by immersion method, consisting of the following ingredients, wt.%:
Edible gelatin - 25
Glycerin - 8
Antiseptic - 0.17
Distilled Water - Else
(see Nikolayenko N.S., Chzhen F.X., Nesmelova Z. // Status and prospects of the development, production and use of excipients for medicines: Collection. - Kharkov, 1982. - S.141-142) . However, the gelatin base of this composition does not provide a satisfactory rate of structuring and the required strength characteristics of the capsule shell when encapsulating fish oils with additives of biologically active substances.
The reasons that impede the achievement of the technical result indicated below when using a known substance include the fact that the additives of biologically active substances contained in encapsulated fish oil interact with the material of a known gelatin base (primarily with gelatin), slowing down the process of structuring and having a destabilizing effect on the emerging capsule shell structure. In addition, the gelatin base of this composition has no biological value, since its main component, gelatin, is a protein that is not balanced in amino acid speed, and is not a biologically active substance.
The composition of the same purpose closest to the claimed invention for the totality of features is the composition of the shell of a hard capsule containing gelatin and a hydrophilic compound, which is a cross-linked sodium salt of carboxymethyl cellulose with a fiber length of 50-300 μm in an amount of 1-2% (see USSR Patent 1743333 (USSR), MKI A 61 K 9/48. Publ. 23.06.92. BI 23). Adopted for the prototype.
The reasons that impede the achievement of the technical result indicated below when using a known substance adopted as a prototype include the fact that in the known substance a synthetic polysaccharide having no biological value is used as a modifying additive to the gelatin base.
The invention is expressed as follows. A natural sodium alginate polysaccharide that meets the requirements of TU 15-544-83 "Food sodium alginate. Technical conditions" is used as a modifying additive to the gelatin base. The following optimal ratio of gelatin base for encapsulation, wt.%:
Edible gelatin - 25 - 29
Glycerin - 8 - 10
Antiseptic - 0.1 - 0.2
Food sodium alginate - 0.1 - 0.2
Distilled Water - Else
As an antiseptic, it is recommended to use: citric acid, nipagin or nipazole.
The technical result of the invention is expressed in accelerating and increasing the uniformity of the structuring of the gelatin base, as well as in increasing the strength characteristics of the capsule shell. This technical result leads to an increase in the yield of the finished product, an improvement in its organoleptic quality indicators and enhances the resistance of the product to destructive loads during storage and transportation.
The specified technical result during the implementation of the invention is achieved by the fact that the gelatin base of the known composition additionally contains food sodium alginate in an amount of 0.1-0.2 wt.%. This substance is an additional structurant and has biological activity.
The required amount of antiseptic is dissolved in distilled water at a temperature of (60 ± 2) o C. The antiseptic solution is cooled to a temperature of (20 ± 5) o C. The required amount of sodium alginate is dissolved in distilled water at a temperature of (60 ± 2) o С with constant stirring . A hot solution of sodium alginate is filtered through a vacuum filter to remove insoluble substances, the filtrate is cooled to a temperature of (20 ± 5) o C.
To cook the gelatin base, it is necessary to use a steam-heated reactor (dead steam), an anchor stirrer and a lower drain cock. The required amount of distilled water, solutions of antiseptic and sodium alginate are added to the reactor. Then make the necessary amount of gelatin and carry out the process of swelling at a temperature of (20 ± 5) o C for a period of time not less than 1 hour
The swollen gelatin is heated in the reactor with dead steam to a temperature of (61 ± 1) o C without stirring. After that, the heating process is carried out with constant stirring with an anchor stirrer and a constant temperature for 1-1.5 hours. Then, without stopping mixing and maintaining the temperature (61 ± 1) o С, the required amount of glycerol is added to the gelatin base. The stirring process is continued at this temperature for 10 minutes.
The reactor stirrer is turned off, the heating process is stopped, a gelatin base sample is taken to determine the kinematic viscosity. The kinematic viscosity of the gelatin base at a temperature of 60 o With should be in the range from 750 to 900 mm 2 / s
After determining the viscosity of the gelatin base for 1 min, the stirrer is turned on, then the gelatin base is poured through a bottom drain valve into a clean container to prevent splashing. The collected gelatin base is sent for encapsulation in the capsule shell tank. The temperature in the tank tank is maintained at a level of (61 ± 1) o C.
Studies of the physicochemical properties of the gelatin base through mathematical modeling showed that the addition of sodium alginate at the specified ratio of the ingredients of the gelatin base provides accelerated and uniform structuring of the gelatin base, as well as hardening of the capsule shell of the fat-soluble food product. This is because the addition of sodium alginate to the gelatin base with the specified ratio of its ingredients causes the formation of complex gels with maximum structuring speed, strength and ductility. Sodium alginate macromolecules create additional nodes in the spatial network of the gelatin gel. In this case, the viscosity of the gelatin base will remain low enough to ensure its flow through the tubes of the capsulator. In addition, sodium alginate, which has the ability to remove heavy metals and radionuclides from the body, increases the biological value of the encapsulated product.
The effectiveness of the proposed composition of the gelatin base, including the addition of sodium alginate, was identified in the production of an experimental industrial batch of the encapsulated drug "Prlien" (ω-3 polyunsaturated fatty acid concentrate in fish oil) at the encapsulation site of ZAO MED-KAP (Murmansk).
According to the test results, the yield of the finished product amounted to 81.4% of the mass of the directed raw materials, whereas when using a gelatin base of known composition (without the addition of sodium alginate), the yield of the finished product is 67.2%. Thus, the finished product yield increased by 14.2%, which is explained by the increased rate of structuring of the gelatin base during capsule shell formation at the encapsulation stage. In terms of organoleptic quality indicators (appearance, color, transparency, smell and taste), as well as in the consistency and diameter of capsules, the encapsulated product meets the requirements established by TU 9281-001-00462663-97 "Food fish oil. Technical conditions".
The introduction of a modifying additive of sodium alginate into the gelatin base causes an increase in the elastic limit of capsule shells by 1.2 times.
