Варианты, полученные через 10, 15, 20 и т.д. пассажей, изучали по ряду лабораторных тестов (маркеров) для выявления отличий от исходного эпидемического вируса, а затем проверяли на людях в отношении безвредности и иммуногенности для людей. Вирусы, удовлетворяющие требованиям фармакопейных статей на живую гриппозную вакцину для взрослых или детей, использовали в производстве указанных вакцин. Эти штаммы, как правило, не отличались по биологическим свойствам от исходных эпидемических вирусов, что затрудняло отбор аттенуированных вариантов перед их испытанием на людях и обусловливало необходимость повторных проверок вариантов, полученных после проведения ряда дополнительных пассажей в развивающихся куриных эмбрионах.Given the variability of the influenza pathogen, vaccine strains have to be replaced during the emergence of new influenza pathogens. Получение аттенуированного штамма вируса гриппа В/Ленинград/14/17/55, который наряду с ареактогенностью имеет отличительные признаки: температурочувствительность (ts маркер) и холодоадаптированность (ха маркер), достигается проведением пассажей вируса В/Ленинград/14/17/55 не при оптимальной (32-33oС), а при температуре, пониженной до 25oС. ts маркер: (сниженная способность к репродукции в куриных эмбрионах при повышенной (38oС) температуре в lg ЭИД 50/0,2 мл) 4,0-5,0 lg, т.е. штамм является температурочувствительным (ts-).ts marker: (reduced ability to reproduce in chicken embryos at elevated (38 o C) temperature in log EID 50 / 0.2 ml) 4.0-5.0 log, i.e. the strain is temperature sensitive (ts-).
Ха маркер (маркер холодоадаптированности), т.е. повышенной способности к репродукции в куриных эмбрионах при 25oС в lg ЭИД 50/0,2 мл, 4,0-5,0 lg, т. е. штамм является холодоадаптированным (ха+).Xa marker (cold adaptation marker), i.e. increased ability to reproduce in chicken embryos at 25 o C in lg EID 50 / 0.2 ml, 4.0-5.0 lg, i.e. the strain is cold adapted (ha +).
Штамм проявляет генетическую стабильность биологических признаков, т.е. не меняет свойств (маркеров) после 5 пассажей в куриных эмбрионах при оптимальной температуре. Пример 2. Была организована интраназальная вакцинация 228 взрослых серонегативных добровольцев вакциной на основе вируса В/Ленинград/14/17/55.
