Изобретение относится к медицинской вирусологии и может быть использовано в здравоохранении для профилактики эпидемического гриппа среди взрослых и детей живой интраназальной гриппозной вакциной из штамма B/60/Петербург/95/20. Известный вакцинный штамм B/60/32/P вируса гриппа B - прототип [1] - утратил антигенную актуальность и вследствие этого не может вызывать защитную реакцию во время эпидемии, вызванной современными штаммами вируса гриппа B. The invention relates to medical virology and can be used in healthcare for the prevention of epidemic influenza among adults and children with a live intranasal influenza vaccine from strain B / 60 / Petersburg / 95/20. Вакцинный штамм B/60/Петербург/95/20 получен методом генетической реассортации эпидемического вируса B/Петербург/92/95 с холодоадаптированным штаммом B/СССР/60/69 - донором аттенуации - с последующей селекцией в присутствии антисыворотки к вирусу B/СССР/60/69. Донор аттенуации В/СССР/60/69 - холодоадаптированный температурочувствительный штамм вируса гриппа, разрешенный для получения безвредных живых интраназальных вакцин для взрослых и детей [2]. Методом сравнительного анализа электрофоретической подвижности фрагментов РНК родительских вирусов и реассортантных клонов в полиакриламидных гелях установлено, что вакцинный штамм B/60/Петербург/95/20 унаследовал 6 генов, кодирующих внутренние белки (PB1, PB2, PA, NP, М, NS) от донора аттенуации, и 2 гена, кодирующих поверхностные белки - HA (гемагглютинин) и NA (нейраминидазу) - от эпидемического вируса. Штамм является температурочувствительным (разность титров при 33oC и 39oC - 6.0 lg ЭИД50/0,2 мл) и холодоадаптированным (разность титров при 33oC и 25oC - 4,0 lg ЭИД50/0,2 мл).Using a comparative analysis of the electrophoretic mobility of RNA fragments of parent viruses and reassortant clones in polyacrylamide gels, it was found that vaccine strain B / 60 / Petersburg / 95/20 inherited 6 genes encoding internal proteins (PB1, PB2, PA, NP, M, NS) from attenuation donor, and 2 genes encoding surface proteins - HA (hemagglutinin) and NA (neuraminidase) - from the epidemic virus.
