Abstract
Description
A(H1N1). The invention relates to medical virology and can be used in healthcare for the prevention of epidemic influenza among adults with a live intranasal influenza vaccine from strain A / 17 / Texas / 91/1/3 (H1N1). The well-known strain A / Kharkov / 17/91 (H1N1) prototype has lost antigenic relevance and therefore cannot cause a protective reaction during the epidemic of influenza A (H1N1).
The problem to which the invention is directed, is to obtain a vaccine strain of the actual antigenic variety based on the epidemic virus A / Texas / 35/91 (H1N1).
The vaccine strain A / 17 / Texas / 91/1/3 (H1N1) was obtained by genetic reassortment of the epidemic virus A / Texas / 36/91 (H1N1) with a cold-adapted strain A / Leningrad / 134/17 (H2N2) by an attenuation donor, followed by selection in the presence of antisera to the virus A / Leningrad / 134/17/57 (H2N2).
Using PHK: PHK hybridization followed by polyacrylamide gel electrophoresis, it was found that vaccine strain A / 17 / Texas / 91/1/3 (H1N1) inherited 6 genes encoding the internal proteins PB1, PB2, PA, NP, M, NS from a donor attenuation and 2 genes encoding the surface proteins HA (hemagglutinin) and NA (neuraminidase) from the epidemic virus. The strain is temperature-sensitive (titer difference at 32 o and 40 o C 8.0 Ig EID 50 / 0.2 ml) and cold-adapted (titer difference at 32 o and 25 o C - 3.5 Ig EID 50 / 0.2 ml) .
Thus, the presented vaccine strain A / 17 / Texas / 91/1/3 (H1N1) is characterized by a combination of useful features necessary for the vaccine strain: antigenic specificity of the surface proteins of the epidemic virus A / Texas / 36/91 (H1N1), the structural genome is optimal for reassortant vaccine strains, temperature sensitivity and cold adaptation, which correlates with human attenuation characteristic of the attenuation donor.
The morphology of the strain is polymorphic, typical of the influenza virus.
Infectious activity during reproduction in developing chicken embryos at 32 o C for 48 hours 7.0 Ig EID 50 / 0.2 ml
1:256. Hemagglutinating activity 1: 256.
The strain exhibits genetic stability of biological traits after 5 passages on chicken embryos (when using large infectious doses).
The strain isreactogenic with intranasal administration to adults over 15 years of age.
Example. A group of 101 people was vaccinated intranasally in a volume of 0.5 ml with a reassortant strain A / 17 / Texas / 19/1/3 (H1N1) with an infectious activity of 7.0 Ig EID 50 / 0.2 ml. A group of people of the same age out of 102 received a placebo.
An average reaction with a short-term temperature increase above 37.5 o C was not observed, weak temperature reactions to 37.5 o C in 5 cases. In the group of people who received a placebo, average temperature reactions were not observed, and weak were observed in 5 cases. The reactogenicity index (the difference in the percentage of average reactions in vaccinated vaccines and received placebo) was 0%
Thus, the vaccine strain A / 17 / Texas / 91/1/3 (H1N1) in terms of reactogenicity meets the requirements for vaccine strains with pharmacopeia article FS 42-299 BC-89 for live influenza vaccine for intranasal use in adults.
