Claims (3)
1. Лидерная последовательность, содержащая нуклеотидную последовательность poly(А) от 5 до 35 А или ее делецию или лидерная последовательность, содержащая дополнительную последовательность слитую с полным или делегированным вариантом последовательности poly(А), или лидерная последовательность, содержащая, по крайней мере, одну нуклеотидную замену, в полном или делетированом варианте последовательности poly(А) и обеспечивающая усиление трансляции мРНК в бесклеточной системе.
2. Лидерная последовательность по п.1, содержащая последовательность SEQ ID No.1.
3. Способ синтеза полипептидов в бесклеточной системе, в соответствии с которым на первом этапе получают генетическую конструкцию, содержащую лидерную последовательность в 5'UTR области, в состав которой входит poly(А), длину и состав лидерной последовательности, выбирают по результатам трансляции мРНК или совмещенной транскрипции/трансляции ДНК в эукариотических бесклеточных системах в диапазоне концентраций мРНК от 50 до 2000 pmol/ml.
