Abstract

Fremstilling av et modent benmorfogenetisk protein ved at et prosesseringsenzym virker på en forløper til benmorfogenetisk protein, som omfatter enten innføring av en ekspresjonsvektor for et benmorfogenetisk forløperprotein og en ekspresjonsvektor for et prosesseringsenzym i en dyrecellelinje, dyrking av den etablerte cellelinje for å gi et modent benmorfogenetisk protein, separering av proteinet fra kulturen eller alternativt tilsette en løsning av et prosesseringsenzym til en løsning av benmorfogenetisk forløperprotein og inkubering av blandingen. En egnet fremgangsmåte omfatter innføring av en ekspresjonsvektor for en human MP52-forløper og en ekspresjonsvektor for en sekretorisk furinvariant i en etablert dyrecellelinje, dyrking av cellelinjen for å gi et modent benmorfogenetisk protein og separering av denne fra kulturen.
