Abstract

Det er beskrevet sammensetninger og fremgangsmåter for fremstilling av et heterologt protein av interesse ved innskudd av en kopi av et gen kodende for det heterologe proteinet av interesse inn i kromosomet til en vertcelle, så som E. coli. Et kromosomalt transfer DNA (et sirkulært, ikke-selv-replikerende DNA) blir anvendt for å integrere genet kodende for det heterologe proteinet av interesse inn i vertcellekromosomet. Kromosomal transfer DNA omfatter minst en selekterbar markør og kan eventuelt innbefatte gjentatte DNA sekvenser som flankerer den selekterbare markøren, som letter den kromosomale amplifikasjonen av integrert DNA. Genet kodende for proteinet av interesse kan bli uttrykt etter integrering inn i kromosomet til vertcellen og seleksjon for kromosomal amplifikasjon kan bli utført før ekspresjon av genet.
