Abstract
Fremgangsmåte for tilsetning av ekstracellulære matrlkskomponenter til stasjonær fase av matriks for bipolar adhesjon av hepatocytter muliggjør at disse substansene blir konsentrert og bundet til en definert basismatriks bestående av en enkel baslsmatriks (f.eks. type I kollagen). En fordel, for eksempel når flbro- nektin blir tilsatt, er å skaffe til veie en komplett serumfri kultur av leverceller mens man opprettholder normal morfologi og funksjon; som et resultat er konvensjonelle dyrkningsmedier som tidligere er uegnet for serumfri dyrking av leverceller nå også velegnet for realtlvt langvarig serumfri dyrking. Reproduser- barhet av prøveresultater og mekanisk stabilitet er vesentlig forbedret l forhold til konvensjonell form av matrlkslagdeling. Fremgangsmåten blir anvendt for å anvende et spesielt tilpasset sett. Andre matrikskompo- nenter kan også bli tilsatt matriks ved denne frem- gangsmåten, og konsentrasjon og binding kan bli gjennomført i det fibrillholdige matriksskjelettet etter at basismatriks er geldannet.
