Description
Foreliggende oppfinnelse angår nye biologisk aktive forbindelser av peptidtypen, nemlig en ny cyklisk analog av et naturlig-forekommende fagocytosestimuler-
ende treonyl-cyklo-/<=>N-"-lysyl-propyl-arginyl7 med en fagocytosestimulerende virkning og som er brukbar i medisin som immunostimulerende middel som.har et.vidt virkningsområde som som er motstandsdyktig mot karboksypeptidaser.
Oppfinnelsens bakgrunn
Tuftsin med treonyl-cyklo-/-N -lysyl.-prolyl-arginyl-7 som sin analoge omfatter et fragment av en tung kjede av immunoglobuliner, klasse IgG, av human opprinnel-se- (Thr-Lys-Pro-Aig). Dette tetrapeptid viser en betyde-lig stimulerende virkning med henblikk på forskjellige immunologiske responser in yivo og in vitro; således oker det vesentlig en fagocytisk aktivitet av leukocytter og makrofager, aktiverer immunogenese av det sistnevnte, jfr. Feldman "Tuftsin (an Ig-associated tetrapeptide) trig-gers the immunogenic function of macrophages: Application for activation of programmed cells", sidene 3400 - 3404.

En mangel ved tuftsin som medikament ligger i

De cykliske analoge av det naturlig forekommende fagocytosestimulerende peptid, treonyl-cyklo-/-N^-lysyl-prolyl-arginyl-7., er ennu ikke beskrevet i litteraturen.
Beskrivelse av oppfinnelsen
Foreliggende oppfinnelse er rettet mot å frem-bringe en ny forbindelse som er motstandsdyktig overfor enzymatisk spalting på grunn av karboksypeptidaser.
Dette oppnås ved en ny cyklisk analog av et naturlig-forekommende f agocytosestimulerende. peptid, nemlig treonyl-cyklo-^-N-lysyl-prolyl-arginyl-/ med formelen:
og en fremgangsmåte for fremstilling av denne ved trinnvis oppbygning av peptidkjeden fra C-enden ved bruk av aktiverte etere av benzyloksykarbonylprolin, tert.-butyloksykarbonyl-N -benzyloksykarbonyl-lysin og tert.-butyloksykarbonyl-treonin, fulgt av cyklisering av det resulterende delvis
blokkerte tetrapeptid ved hjelp av Woodwards reagens og isolering av det onskede produkt.

Ifblge oppfinnelsen har den nye forbindelse, dvs. treonyl-cyklo-/_-N' -lysyl-prolyl-arginyl-/ den folgende formel:
Cyklo-/-N -lysyl-prolyl-arginyl=7 er et amorft hvitt pulver som dekomponerer over 100°C, spesifikk rota-sjon /Z7q<2>- 63° (c 0,2; 5°/o CH^COOH) .

elektroforetisk mobilitet bestemmes ved forhol-det til histidin i IN eddiksyre på papir FN-15 (GDR) ved en spenningsforskjell på 900 volt-; For den hye forbindelse ifolge oppfinnelsen er E, . = 0,9 (pH2,4).
Kromatografisk analyse:
kromatografisk mobilitet R f = 0,07 (n-butanol-etanol-vartn-eddiksyre 80:10:30:5), Rf = 0,22 (n-butanol-pyridin-vann-eddiksyre 30:20:24:6) ble bestemt på "Merck"-plater.
Den biologiske aktivitet av treonyl-cyklo-^-N<c>'-lysyl-p.rolyl-arginyl-7 ble studert ved in vitro-forsbk. Virkningen av denne forbindelse på den fagocytiske evne for rotters blodsegmenterte noytrofiler studeres i sammen-ligning med virkningen av et kjent stimulerende middel for fygocytose, tuftsin (Thr. Lys. Pro. Arg. 2CH^C00H<*>3H20) fremstilt ved instituttet for organisk syntese i Latvia, SSR vitenskapsakademi. Den fagocytiske evne av segmenterte noytrofiler bestemmes i henhold til Gostev's prosedyre (jvf. Proe. USSR Acad. Med. Sei.,- Nutrition Problems, "Role of Protein in Nutrition", bind 13, utgave 2, 1950, Moskva; V.S. Gostev, M.N. Petriashina, S.A. Popovkina,-A.K. Saakov "Phagocytosis, complement and blood coagula-tion with protein deficiency in food", sidene 110 - 116).
Ifolge denne prosedyre blir rotteblod fortynnet med 2% natriumcitrat i et forhold på 1:1 og opplosninger av proveforbindelsene benyttes. En 1-dag gammel kultur av den koagulasopositive patogene stamme Staphylococcus aureus i en konsentrasjon på 1 bin i 1 ml tilsettes til denne.'Blandingen inkuberes i en termostat ved en temperatur på 37°C i 30 minutter. Deretter prepareres utsmbr-inger, disse fikseres med metanol og tijrkes i henhold til Romanovsky-Gymza. 100 segmenterte noytrofiler telles, an-tall bakterieabsorberende beregnes blant disse (% av aktive fago.cytter) samt et gjennomsnittstall for bakterier i én segmentert nbytrofil (fagocyttallet). De fagocytiske para-metre er vist i tabellen nedenfor.
Slik det fremgår av tabellen, viser treonyl-cyklo-/-N -l<y>s<y>l-<p>rol<y>l-ar<g>in<y>l-7i konsentrasjoner fra 3*10~ og 3"10 M en stimulerende virkning på den fagocytiske aktivitet' av leukocytter og viser ingen inhiberende virkning i forhold til virkningen av tuftsin.
Fremstilling av treonyl-cyklo-/_ -N g.-lysyl-prolyl-arginyl=7gjennomfbres på fblgende måte.
p-nitrofenyleteren av benzyloksykarbonylprolin omsettes med arginin hvorved man oppnår benzyloksykarbonyl-prolyl-arginin.
Den således fremstilte forbindelse blir deretter opplost i iseddik, behandlet med en opplbsning av hydrogenbromid i iseddik og man oppnår prolyl-argihin.
Den således oppnådde prolyl-arginin•omsettes deretter med pentafluorfenyleteren av tert-butyloksykarbonyl-benzyloksykarbonyl-lysin under dannelse av tert-butyloksykarbonyl-N^-benzyloksykarbonyl-lysyl-prolyl-arginin som behandles med 70% vandig opplbsning av trifluoreddiksyre hvorved man oppnår N^-benzyloksykarbonyl-lysyl-prolyl-arginin. Deretter utfores det en reaksjon for gjensidig på-virkning mellom pentafluorfenyleteren av tert-butyloksykarbonyl-treonin og N^-benzyloksykarbonyl-lysyl-prolyl-arginin under dannelse av tert-butyloksykarbonyl-treonyl-N^-benzyloksykarbonyl-lysyl-prolyl-arginin. Det således oppnådde produkt hydrogeneres hvorved man oppnår tert-butyloksykarbonyl-treonyl-lysyl-prolyl-arginin.
Tert-butyloksykarbonyl-treonyl-lysyl-prolyl-arginin cykliseres for å oppnå tert-butyloksykarbonyl-treonyl-cyklo-^N^-lysyl-prolyl-arginyl^• Det resulterende produkt behandles med en opplbsning av hydrogenklorid i iseddik for fremstilling' av treonyl-cyklo-^N^-lysyl-prolyl-arginyl-;7.

Den nye analoge av tuftsin, treonyl-cyklo-/=N - lysyl-prolyl-arginyl-7, bibeholder et hoyt nivå av fagocytosestimulerende virkning i forhold til segmenterte noytrofiler fra rotteblod in vitro karakteristisk for tuftsin, men til forskjell fra den sistnevnte er den motstandsdyktig overfor enzymatisk hydrolyse på grunn av karboksypeptidaser.
For en bedre forståelse av oppfinnelsen gis det nedenfor et eksempel som illustrerer denne.
For syntese av treonyl-cyklo-/-N -lysyl-prolyl-arginyl-7 bruker man aminosyrer og derivater derav som er tilgjengelige fra selskapet "Reakal" i Ungarn. Alle amino-syrene hadde L-konfigurasjon..Smeltepunktene bestemt i åpne kapillarer gis uten korreksjon. Homogeniteten av de resulterende forbindelser sjekkes på plater "Silufol W-254", "Eastman", og "Merck". Også vises de kromatografiske mo-bilitetsverdier R~i de folgende systemer: A -r n-butanol-etanol-vann-eddiksyre (80:10:30:5); B - n-butanol-pyridin-vann-eddiksyre (30:20:24:6), såvel som elektroforetisk mobilitet i forhold til histidin ) på papir"FN-15"
(GDR) i IN (pH 2,4) og 5N (pH 1,4) eddiksyre. Rotasjons-vinklene bestemmes i polarimeter "Perkin Eimer 140". For-dampingen gjennomføres i en vakuumrotasjonsfordamper under et gjenværende trykk på 1 - 10 mm Hg ved en temperatur på 30°C.
BgEyloksykarbonyl- prolyl- arginin ( I).
Til en opplbsning av 37 g eller 0,1 mol p-nitro-fenyleter av benzyloksykarbonylprolin i 360 ml dioksan tilsettes en opplbsning av 16 g eller 0,09 mol arginin i 160'ml vann under heftig omrbring. Etter 2 timer fordampes opplbsningsmidlet og resten opplbses i 100 ml dimetylformamid. Opplbsningen omrbres ved romtemperatur (25°C) i 72 timer og helles deretter i 1 liter etylacetat. Etter en oppholdstid for reåksjonsblandingen i 12 timer i en kjoler.blir det resulterende bunnfall filtrert av, vasket på filteret med etylacetat og dietyleter.. Etter omkrystallisering av produktet fra en blanding av etahol og dietyleter (1:5) oppnås 35 g eller 96% benzyloksykarbonyl-prolyl-arginin med de folgende egenskaper: smeltepunkt 137 - 140°C, /L7j5° - 46,6° (c 1,0; H20), Rf 0,53 (A, "Eastman");.0,66 (B, "Eastman") ,; Ehis 0,56 (pH 2,4).
Funnet, vekt-»/:' C 53,17, H 6,08, N 15,91. Beregnet for C]_9H27N5°5' H2°» vekt-%: C 56,28, H 6,71, N 17,27.
Prolyl- arginin ( II)
Dette fremstilles ved å behandle forbindelse (I) med IN hydrogenbromid i iseddik. Opplbsningen av hydrogen-bromidsalt av prolyl-ariginin (II) i 500 ml vann behandles med en anionitt til negativ reaksjon på Br -ioner. Utbyttet av prolyl-arginin er 23 g (kvantitativt). ' aT rer gt in- lbu nt-y(l Io Ik Is)ykarbonyl-N -benzyloksykarbonyl-lysyl-prolyl-Til en opplbsning av 53 g eller 0,1 mol penta-fluorf enyleter av tert-butyloksykarbonyl-N&-benzyloksykarbonyl-lysin i 300 ml dioksan tilsettes under heftig omrbring en opplbsning av 23 g eller 0,08 mol prolyl-arginin (II) i 100 ml vann. Etter 2 timer fordampes opplbsningsmidlet og resten opplbses i 150 ml dimetylformamid. Opplbsningen omrbres i 48 timer ved romtemperatur (25°C), dimetylformamid destilleres av til en tredjedels volum,. hvoretter reaksjonsblandingen helles i 1 liter dietyleter. En ytterligere behandling gjennomfbr.es som beskrevet i forbindelse med for bindelse (I). Etter omkrystallisering av produktet fra en blanding av etylacetat-etanol-dietyleter (3:1:30) oppnås det 47 g eller 75% tert-butyloksykarbonyl-N -benzyloksykarbonyl-lysyl-prolyl-arginin med de folgende egenskaper: Smeltepunkt 90°C, [ 17^ ° - 39° (c 1,0; 10% CH^COOH); Rf = 0,51 (A,. "Eastman"), 0,82 (B, "Eastman"), Ehis= 0,34 (pH 2,4).
Funnet, vekt-%: C 54,66, H 7,98, N 15,89, Beregnet for C^øH^yNyOg, vektprosent: C 56,86, H 7,48, N 15,47. N^- benzyloksykarbonyl- lysyl- prolyl- arginin ( IV)
Denne forbindelse fremstilles ved å behandle forbindelse (III) med en 70%-ig vandig opplbsning av trifluoreddiksyre og en anionitt for å fjerne trifluoracetat. Utbyttet av det bnskede produkt er 11 g (kvantitativt)."Tert-butyloksykarbonyl-treonyl-N c-benzyloksykarbonyl-lysyl-^ prolyl- arginin ( V)
2,18 g eller 4,1 mmol N -benzyloksykarbonyl-lysyl-prolyl-arginin (IV) i 30 ml vann tilsettes under omrbring til en opplbsning av 2,10 g eller 5,3 mmol pentafluorfenyl-eter av tert-butyloksykarbonyl-treonin i 70 ml dioksan. Deretter blir reaksjonsblandingen behandlet ved å folge
den prosedyre som er beskrevet for syntesen av forbindelse (III). Utbyttet av det bnskede produkt, tert-butyloksykarbonyl-treonyl-N c-benzyloksykarbonyl-lysyl-prolyl-arginin, . er 2,65 g (188%).
Den resulterende forbindelse (V) renses på en kolonne (3 x 100) med silikagel ("Chemapol" CSSR, L 40/100^u) i system A. De samlede fraksjoner konstrolleres ved TLC-teknikker. Fraksjonene med Rf = 0,45.(A, "Silufol") kombineres. Utbyttet fra produkt (V) er 180 g eller 60%. Den således fremstilte forbindelse har folgende egenskaper: Smeltepunkt 175°C; [ TJ^ - 64,6° (c 1,0; H2), Rf = 0,65 (B, "Merck"); EMs 0,41 (pH 1,4).
Funnet, vekt-%: C 52,44, H 7,26, N 14,42; beregnet for C34<H>55<N>8°io'2H °»vekt-%: C 52,98, H 7,59, N 14,54.
Tert- butyloksykarbonyl- treonyl- lysyl- prolyl- arginin ( VI) 1.23 g eller 1,67 mol tert-butyloksykarbonyl-treonyl-N^-benzyloksykarbonyl-l-lysyl-prolyl-arinin (V) hydrogeneres under atmosfærisk trykk i nærvær av palla-diumsort i en opplbsning av 20 ml metanol i 10 timer. Der-^etter blir opplbsningen filtrert,,fordampet til torr til-stand og tbrket under torr dietyleter. Den resulterende substans tbrkes over fosforpentoksyd i vakuum under et gjenværende trykk på 1 mm Hg.
Utbyttet av tert-butyloksykarbonyl-treonyl-lysyl-prolyl-arginin er l,05g eller 99%; produktet har folgende- egenskaper: Smeltepunkt 170°C; { Tj- 66° (c 1,0;H20); Rf = 0,05 (A, "Silufol"), 0,16 (A, "Merck"); EMs = 0,84 (pH 1,4).
Funnet vektprosent: C 49,26, H 8,30, N 16,82; Beregnet for<C>^H^<NqOq>, vekt-% C. 49,48, H.8,35, N 16,58. Tert-butyloksykarbonyl-treonyl-cyklo-^-N^-lysyl-prolyl-arginyl-/(VIT): 0,85 g eller 1,34 mmol tert-butyloksykarbonyl-treonyl-lysyl-prolyl-arginin (VI) opplbses under omrbring i 260 ml dimetylformamid hvoretter opplbsningen avkjbles til 0°C og en opplbsning av 1,86 ml eller 1,34 mmol trietyl-amin i 200 ml dimetylforamid tilsettes dråpevis il time. Deretter, mens temperaturen holdes ved 0°C, tilsettes en suspensjon av 0,33 g eller 1,34 mmol Woodward-reagens (N-etyl-5-fenylisooksazolium-3'-sulfonat) i 150 ml dimetylformamid i 2 timer og kjblingen fortsettes i ytterligere 30 minutter. Deretter omrbres reaksjonsblandingen ved romtemperatur i 170 timer. Opplbsningsmidlet fordampes, resten behandles med torr eter, filtreres fra og tbrkes under 1 mm Hg. Utbyttet er 1,18 g (blanding av utgangsproduktet, dannet substans og Woodward-reagens). Etter rensing i en kolonne 3 x 100 med silikagel .("Chemapol", CSSR, L 40/100 ^ u] i system A samles fraksjoner med R^= 0,34 (A, "Merck") og man oppnår 0,17 g eller 20% tert-butyloksykarbonyl-treonyl-cyklo-/-N. -lysyl-propyl-arginyl=7 me<i folgende egenskaper: IL7^ ° - 46,6° (c 0,03; MeOH); Rf - 0,73 (B, "Merck"), EMs = 0,38 (ph 2,4).
Funnet, vekt-%: C 46,93, H 7,03, N 13,70; beregnet for CggH^N<g>O<y>'<2>CH3c6oH* 2H20, vekt-%: C 48,77, H 7,91, N 15,16. Treonyl- cyklo- Z- N - lysyl- prolyl- arginyl- 7 ( VIII) 0,13 g eller 0,21 mmol tert-butyloksykarbonyl-cyklo-^-N^-lysyl-prolyl-arginyl-/ (VII) opplbses i 5 ml iseddik og behandles med 0,5 ml 9,2N hydrogenklorid i dioksan. 20 minutter deretter fordampes opplbsningsmidlet, resten behandles med torr dietyleter, tbrkes. under 1 mm Hg over kaustisk kalium. Produktet resnes i en kolon-, ne. med CM-cellulose (Whatman "CM-32") ved en gradient for ammoniumacetat fra 0,01 M (pH 4,5) til 0,25 M (pH 6,5). Utbyttet av rent treonyl-cyklo-/=N^'-lysyl-prolyl-arginyl=7er 0,03 g eller 23%, og forbindelsen har folgende egenskaper: /l7q° = -63° .(c 0,2; 5% OH^COOH); •Rf = 0,07 (A, "Merck"), 0,22 (B, "Merck"), Eh±s .=<0>,9(pH ' 2,4).
Funnet, vekt-%: C.40,18, H 7,47, N 14,68; beregnet for C21H38N8°5'2CH3C00H<*>4H2°>vekt-%: C '44,50, H 8-,06, N 16,60.
Hydrolyse av treonyl-cyklo-/-N^-lysyl-prolyl-arginyl-7 (VIII) med karboksypeptidase B under optimale betingelser har vist motstandsevne for C-avsluttet arginin mot spalting i motsetning- til tuftsin.
Industriell anvendelse
Treonyl-cyklo-Z-N^-lysyl-prolyl-arginyl-/ kan benyttes i medisinen som et immunostimulerende middel med et vidt virkningsområde og det er motstandsdyktig overfor enzymatisk spalting under in vivo-betingelser. Dette muliggjbr anvendelse som farmasbytisk middel per os.

Fremgangsmåte for fremstilling av treonyl-cyklo-/-N^-lysyl-propyl-arginyl-7 ifolge krav 1,karakterisert vedat syntesen av treonyl-cyklo-^N^-lysyl-prolyl-arginyl-7 gjennomføres ved hjelp av en trinnvis oppbygning av peptidkjeden fra C-enden ved bruk av aktiverte etere av benzyloksykarbonyl-prolin, tert-butyloksykarbonyl-N^-benzyloksykarbonyl-lysin og tert-butyloksykarbonyl-treonin, fulgt av cyklisering av det resulterende delvis blokkerte tetrapeptid ved hjelp av Woodward-reagens i et overskudd av dimetylformamid samt isolasjon av det onskede produkt.
