Description
Foreliggende oppfinnelse vedrorer diagnostiske anordninger for immuno-kjemiske målinger, spesielt utformet som test-strimler, omfattende kapillarholdig bæremateriale såvel som fremgangs-måten for sådanne bestemmelser.
Særlig omfatter de diagnostiske anordningene antistoff-bindende, kapillarholdig, porost bæremateriale. Bestemmelsesmetoden omfatter immuno-kjemisk målingkarakterisert vedbruken av kapillarkraften i porose bærematerialer hvortil er bundet antistoffer på en faglig kjent måte som f.eks. ved absorpsjon eller kovalent binding slik som cyanogen bromid eller glutarsyrealdehyd.
Prover som inneholder antigenet som skal måles settes til den kapillarholdige diagnostiske anordningen. En kapillarbevegelse får finne sted hvoretter de antigeriholdige områdene av den diagnostiske anordningen påvises ved tilsetning av antistoffer som er bundet til en egnet fargeindikator, slik som en fluoriscerende forbindelse eller et enzym som katalyserer en fargereaksjon.
De hittil brukte kvantitative immuno-kjemiske metodene er basert på immunodiffusjonen som, f.eks. Mancini<1>s eller Ouchterlony<1>s metoder, som er oppkalt etter deres respektive oppfinnere, eller på immuno-elektroforese som Laurell's elektroimmuno-prove. Disse metodene, såvel som deres varianter, frembringer den nodvendige påvirkning mellom antigen og antistoff ved en diffusjonsprosess og/eller en elektroforetisk prosess, mens metoden som heri er beskrevet er basert på bruken av kapillar-kraf t i et porost bæremateriale hvortil antistoffer er bundet.

Kapillarholdig materiale som er egnet for gjennomføringen av oppfinnelsen er forskjellige typer av cellulosefiberholdig materiale, som filterpapir, kromatografipapir, ionebytterpapir, celluloseacetat-membran, celluloseacetat-skiver, cellulose tynnsjikt-kromatografi-skiver, men også slike materialer som stivelse, Sephadex (Pharmacia Fine Chemicals, Uppsala), plastikkmateriale som polyvinylklorid, keramisk materiale og kombinasjoner som polyvinylklorid-kiselgel. Egnede antistoffer frembringes ved immunisering på fagmessig kjent måte.
Oppfinnelsen illustreres ved de folgende eksempler:
Fremstilling av diagnostiske anordninger
EKSEMPEL 1. Uopploseliggjorinq av antistoffer på kapillarholdig materiale med CNBr: lo g celluloseholdig materiale plasseres i destillert vann 5 minutter. Deretter tilsettes en losning av 8 g CNBr i
3oo ml destillert vann og pH-verdien justeres til lo,5 med
1 M NaOH. Under opprettholdelse av denne pH-verdi ved pH-justeringer, utfores reaksjonen gjennom en 2o minutters periode og losningen dekanteres så. Cellulosematerialet
vaskes med 2 1 o,oo5 M NaHCO^. Etter fjerning av vaske-losningen inkuberes cellulosematerialet natten over ved +4°C med en losning av 5oo mg kanin-gammaglobulin mot humant gammaglobulin (IgGFc fragment) i lo ml o,1 M NaHC03og inkuberes så igjen 3 timer med loo ml o,oo5 M etanolamin-losning, fulgt av en annen vask med 5oo ml o,5 M NaHCO^, og så 2oo ml o,l M acetatbuffer med pH 4,o og 5oo ml o,o75 M natriumbarbituratbuffer med o,3% albumin tilsatt ved +4°C.
EKSEMPEL 2. Uloseliggioring av antistoffer på kapillarholdig materiale med glutarsyrealdehyd: o,6 g celluloseacetat-membran inkuberes med en losning av loo mg kanin-gammaglobulin mot humant gammaglobulin (IgGFc fragment) i lo ml o,1 M fosfatbuffer med en pH på 6,8 i 3o minutter. Når losningen har blitt dekantert, inkuberes membran i lo ml 1% glutarsyrealdehyd-losning i 3o minutter, fulgt av inkubasjon i lo ml 1,o M metylamin i 15 minutter. I natten over ved +4°C med en losning av 5oo mg kanin-gamma globulin mot humant gammaglobulin (IgGFc fragment) i loo ml o,l M fosfatbuffer ved pH 6,8. Etter avsluttet inkubasjon vaskes materialet med 1 1 fosfatbufret fysiologisk NaCl ved pH 7,o. Materialet torkes mellom filterpapir.
EKSEMPEL . 4. Fre mstilling av fluoriscerende antistoff konju-qater: a) 72o mg kanin-gammaglobulin mot humant gammaglobulin (IgGFc fragment), o,57 g NaCl, o,259 g NaHC03og o,o49 g
Na2CC>3.lo H^O opploses i 72 ml destillert vann. Reagens-losningen avkjoles i et isbad og 36 mg fluorescinisotiocyanat tilsettes under roring. Losningen blir plassert kaldt under roring 18 timer. Deretter blir blandingen dialysert mot fosfatbufret fysiologisk NaCl ved en pH-verdi på 7,o inntil dialysevæsken opphorer å fluorescere. '• Det resulterende antistoff konjugatet fryses.
b) 72o mg kanin-gammaglobulin mot humant gammaglobulin (IgGFc fragment), o, 57 g NaCl, o, 259 g NaHCC>3og o,o49 g
Na2C03.lo H20 opploses i 72 ml destillert vann. Reagens-losningen avkjoles i et isbad og 9 mg rodamin opplost i 2 ml aceton tilsettes under roring. Losningen plasseres kaldt under roring 18 timer. Deretter blir blandingen dialysert mot fosfatbufret fysiologisk NaCl ved en pH-verdi på 7,o inntil dialysevæsken opphorer å fluorescere. Det resulterende antistoff konjugatet fryses.
c) 72o mg kanin-gammaglobulin mot humant gammaglobulin (IgGFc fragment), o,57 g NaCl, o,259 g NaHC03og o,o49 g

EKSEMPEL 5. Fremstilling av LDH isoenzym H^ antistoff konjuqat: En suspensjon av 4,5 mg LDH isoenzym H . i ammoniumsulfat dia-lyseres mot o, 1 M fosfatbuffer ved pH 6,8. Under roring tilsettes 2,25 mg kanin-gammaglobulin mot humant gammaglobulin (IgGFc fragment) og fosfatbuffer til et totalvolum på 1,35 ml. Når alt kanin-gammaglobulin er opplost, tilsettes 45[il 1% glutarsyrealdehyd-losning dråpevis, og reaksjonen for stå ved romtemperatur i 2 timer. Blandingen blir så dialysert mot
en natriumbarbituratbuffer . og konjugatet lagres ved +4°C. For bruk fortynnes det med lo ml barbituratbuffer hvortil
2% albumin er blitt tilsatt.
EKSEMPEL 6. Kontroll av uloseliggjbringen:
To materialstrimler som er behandlet med antistoffer ifolge
et foregående eksempel, og to ubehandlede strimler benyttes for testen. En strimmel av hver type inkuberes i en losning av 1 mg humant gammaglobulin (IgG) pr. ml natriumbarbiturat-buf f er. Alle strimlene blir så vasket med 2o ml natrium-barbituratbuf f er som inneholder o,15 M NaCl, lo ganger ialt, hvoretter de inkuberes i fluorescinisotiocyanat antistoff konjugat i lo minutter. Etter gjentatte vaskinger som i det foregående, torkes stripene og inspiseres i ultrafiolett lys ved en bolgelengde på 34o[im, for å fastslå at fluorescens bare foreligger hos strimlene som inneholder uloseliggjorte antistoffer og som har blitt inkubert i IgG-losningen. Under kontroll av strimler ifolge eksempel 3, benyttes strimler behandlet med bovin albumin som kontroller.
Immuno- kjemisk kvantifisering
EKSEMPEL A.
Diagnostiske anordninger som er fremstilt innen rammen av foreliggende oppfinnelse benyttes for. immuno-kjemisk kvantifisering for diagnostiske formål på folgende måte: i Materialet som skal testes plasseres - i en kjent mengde,
fortrinnsvis mellom 1 (il og 3 u-l - på det kapillarholdige bærematerialet på samme tid som tilsvarende mengder av en standard-losning med et kjent innhold av antigenet som skal testes og plasseres i forskjellige fortynninger ved siden av testlosningen.
Som en mobil fase for å lette kapillarbevegelsen, kunne f.eks. o,o75 M natriumbarbituratbuffer brukes med en tilsetning av o,3% albumin.
Kapillarbevegelsen kan utfores i både åpne og lukkede test-beholdere; videre kan den mobile fasen bevege seg oppover eller nedover. Fortrinnsvis bor kombinasjonen lukket beholder og stigende fase anvendes. På et fritt valgt tidspunkt etter påbegynnelsen av kapillarbevegelsen, overfores det våte bærematerialet til en antistofflosning som inneholder enten fluorescinisotiocyanat- eller laktatdehydrogenase (LDH)-bundne antistoffer. Den foretrukne inkubasjonstiden for hver av disse losningene er lo minutter. Etter inkubering vaskes bærematerialet under rennende vann ved +37°C i 5 minutter, fulgt av inkubering i fortrinnsvis 2x5 minutter i 2o ml natriumbarbituratbuffer som inneholder o,2% albumin og o,15'
M NaCl.
I den utstrekning fluorescerende antistoffer benyttes for identifikasjonen, måles antistoffets bevegelsesavstand i ultrafiolett lys ved 34o (im. Hvis LDH antistoff losning benyttes, behandles bærematerialet i et fargingsbad som har folgende sammensetning: 3o mg nitroblått tetrazolium og 4o mg NAD opploses i 56 ml o,o5 M TRISbuffer ved pH 7,4. Til blandingen settes 1,6o ml 3,6 M litiumacetat; 5 ml0,06 M kalsiumcyanid og noen få
korn metylfenazonium metasulfat opplost i 2 ml destillert vann. Det kapillarholdige materialet inkuberes i fargings-badet ved + 37°C inntil en tydelig farging kan observeres. Fargeutviklingen baserer seg på det faktum at LDH1 et katalyserer reaksjonen av laktat + NAD<—» pyruvat + NADH2 og at NADH2 kvantitativt reduserer nitroblått tetrazolium til et uloselig lillafarget formazan.
De indikerte antigen-dekkede områdene av den diagnostiske anordningen vil så oke i styrke med det tiltagende innholdet av antigen i de testede provene. De matrix-bundne antistoff-molekylene holder tilbake bevegelsen av antigen-molekylene ved utbytningsreaksjoner under kapillarbevegelsen. Jo hoyere konsentrasjon av antigener, jo mindre tilbakeholdende effekt på grunn av at gjennomsnittstiden for et antigen-molekyl
som er bundet til et motstoff vil avta ved en okende konsentrasjon av antigener.
EKSEMPEL B.
Strimler av antistoffholdig bæremateriale med kapillarer
og fremstilt innen foreliggende oppfinnelses område senkes
til en bestemt dybde av, f.eks., 3 mm i et lite volum av en losning av materialet som skal testes. Den således initierte kapillarbevegelsen får fortsette i 3 - 2o minutter og avbrytes så ved å fjerne strimlen fra losningen. Bevegelsesavstanden for antigenet blir så målt som nevnt i eksempel A og resultatet evalueres ifolge eksempel A.

Claims (7)
1. Diagnostiske anordninger for immuno-kjemiske bestemmelser,karakterisert vedat de fortrinnsvis er utformet som test-strimler bestående av kapillarholdig, porost bæremateriale hvortil antistoffer er bundet.
2. Diagnostiske anordninger ifolge det foregående krav,karakterisert vedat antistoffene er bundet til det kapillarholdige bærematerialet ved absorpsjon eller kovalent binding slik som cyanogen bromid eller glutarsyrealdehyd.
3. Diagnostiske anordninger ifolge kravene 1-2,karakterisert vedat det kapillarholdige bærematerialet består av cellulosefibre, stivelse, Sephadex (Pharmacia Fine Chemicals), plastikkmateriale slik som polyvinylklorid, keramisk materiale, kombinasjoner som polyvinylklorid- si lis iumoksyd.
4. Fremgangsmåte for immuno-kjemisk bestemmelse,karakterisert vedat tilsetningstrinnene av provene som inneholder antigenene som skal bestemmes til en kapillarholdig, diagnostisk anordning ifolge de foregående krav, initiering av en kapillarbevegelse fulgt av måling av de antigenholdige områdene i den diagnostiske anordningen ved tilsetninger av antistoffer som er knyttet til en egnet fargeindikator som en fluorescerende forbindelse eller et enzym som katalyserer en farge-utviklende reaksjon.
5. Fremgangsmåte ifolge krav 4,karakterisert vedat fargeindikatoren er fluorescinisotiocyanat, rodamin, dansylklorid.
6. Fremgangsmåte ifolge krav 4,karakterisert vedat den diagnostiske anordningen er en test-strimmel av polyvinylklorid-silisiumoksyd hvorpå er absorbert antistoffer mot humant gammaglobulin.
7. Fremgangsmåte ifolge krav 6,karakterisert vedat den antistoffbindende fargeindikator er fluorescinisotiocyanat.
