Description
Foreliggende oppfinnelse angår nye derivater av [substituerte' (2,4-dioxo-1-pyrimidiny1)acetylamino]peniciIliner og cefalosporiner, fremgangsmåte ved fremstilling av disse og anvendelse av disse som antibakterielle midler.
Oppfinnelsen angår folgelig nye syntetiske forbindelser av penicillin og cefalosporinklassen som er nyttige som antibakterielle midler. Disse forbindelser utviser en hoy grad av aktivitet overfor et stort antall mikroorganismer, i særdeleshet penici llinase-produserende mikroorganismer. Som antibakterielle midler er forbindelsene ifolge oppfinnelsen terapeutisk aktive ved behandling av infeksjose sykdommer som skyldes gram-positive og gratn-nega tive bakterier i f jserf e .. og. dyr, innbefattet mennesket. 1 tillegg er forbindelsene ifolge oppfinnelsen nyttige som dyrefårtil-legg og som aktiv bestanddel i bakteriedrepende midler som anvendes som overflatedesinfeksjonsmidler.
Splitting av penicilliner til 6-aminopenicillansyre i 1959 og deh kjemiske splitting av cefalosporin til den tilsvarende 7-aminocefalosporansyre gjorde syntesen av nye syntetiske penicilliner og cefalosporiner som tidligere ikke var tilgjengelige via fermente-ringsprosedyrer mulig. Acylering av aminogruppen bar gitt derivater inneholdende en heterocyclisk ring i sidekjeden i 6-stillingen slik som tilfellet er i penicillanserien, eller i den tilsvarende 7-sidekjede som tilfellet er i cefalosporinserien. Slike heterocycliske deler innbefatter thiofenringen som beskrevet i US patentskrift 3.218.318, 3.449.338 og 3.498.979 (cefaloridin og cefalothin), pyridin, US patentskrift 3.422.100 (cefapirin), picolin, US patent 3.553.203, hydantoin, US patentskrift 3.227.712, og forskjellige andre nitrogenholdige heterocycliske deler innbefattet pyrrolidin og nicotinsyre, US patentskrift 3„308.120.
I hvert tilfelle er den heterocycliske halvdel bundet til en sidekjede, vanligvis til den av et acetyIradika1 via et av ring-carbonatomene'. Foreliggende oppfinnelse angår 2, 4-dioxo-1-pyrimid-inyIderivater som er bundet direkte til acetyIradikalet gjennom he-teroatomer. Kjente eksempler på denne type binding, og som i denne henseende ansees å representere det mest nærliggende innen teknik-kens stilling, er den tetrasolring som er beskrevet i US patentskrift 3.516.997 (cefazolin) og visse kinazolinyIderivater av peni-cillansyré, US patentskrift 3.652.547.
Foreliggende oppfinnelse angår nye 2,4-dioxo-1-pyrimidinyl-penicillin og cefalosporinderivater. Nærmere bestemt angår oppfin-ne Isen [s ubs tit ue rte (2, 4-di ox o-l-pyrimid i ny 1) a cetylami no] penici Hin og cefalosporinderivater som er nyttige som antibakterielle midler og som kan representeres av den generelle formel:

hvor X er hydrogen, hydroxy, acetoxy, N-pyridinium, 5-methy1-1,3,4-thiadiazol-2-ylthio og 1-methyl-l,2,3,4-tetrazol-5-ylthio, og farmasoytisk akseptable salter derav.
Forbindelsene ifolge foreliggende oppfinnelse fremstilles ved kondensasjon av en 6-aminopenicillansyre eller en 7-aminocefalosporansyre med en 7-aminocefalosporansyre med en (substituert)-2,4-dioxo-1-pyrimidinyleddiksyre som vist i det efterfolgende reaksjonsskjema.
Alle forbindelsene ifolge foreliggende oppfinnelse innehol-der et 2,4-dioxo-1-pyriraidinylradika1 eller en 2,4-pyrimidindiondel ved endestiIlingen av acetylaminosidekjederi som angitt i den ovenfor generelle formel (I). Når det gjelder penicillinseriene nummereres acetylarainosidekjeden som 6-stilling, mens forbindelsene i cefalosporinseriene nummereres i 7-stilling. Nummereringssystemet for disse to serier av forbindelser er illustrert for mellomproduktene 6-aminopenicillansyre (IV) og 7-aminocefalosporansyre (V):
2,4-pyrimidindion bundet til acetylaminosidekjeden kan enten være substituert eller usubstituert. De forskjellige substituenter er lokalisert enten på 5-stillingen eller 3-stillingen av 2,4-dioxo-pyrimidinkjernen. Substituentene ved 5-stillingen, representert av symbolet R^, innbefatter folgende radikaler: halogen, hydroxyl, lavere alkyl, trifluormethy1, nitro, amino, cyano, carboxy 1 og methyl og ethylestrene av carboxylradikalet. Uttrykket halogen innbefatter fluor, klor, brom og jodradikaler. Uttrykket lavere alkyl som anvendt her innbefatter både rettkjedede og forgren-ede alifatiske hydrocarbonradikaler med 1-4 carbonatomer. Spesi-fikt innbefattet er slike som methyl, ethyl, propyl, isopropyl, but-yl, isobutyl og t-buty lradikalene .■ Substitusjon ved imidnitrogen-atomet lokalisert ved 3-stillingen i pyrimidinringen er meget mere begrenset og innbefatter hydrogen eller et methylradika1, som indi-kert ved symbolet R^.
Når det gjelder acetylamino eller acetamidodelen av molekylet, er substitusjonen av 2-methylgruppen selvsagt obligatorisk med 2,4-dioxo-1-pyrimidinylradikalet. I tillegg kan 2-methylgruppen in-neholde substitusjonen i form av et methylradikal, et fenylradikal eller et carboxylradika1 som representert ved symbolet R3. Når f.eks. R^er et methylradikal, angis forbindelsene mere noyaktig som propionylaminoderivater av 6-am.inopenici 1 lansyre eller av 7-amino-cef a losporansyre . For enkelhets skyld, og for å anvende én nomen- klatur vil imidlertid disse derivater her bli angitt som 2r(substituerte) acetylaminoderivater . Når det f.eks. gjelder et cefa]osporan-syrederivat hvor R^er methyl og 2,4-pyrimidindionet forblir usubstituert, angis forbindelsen som 7-[2-(2,4-dioxo-1-pyrimidinyl)2-methylacetylamino]cefalosporansyre. På samme måte, når R^represen-terer fenylradikalet, vil den samme forbindelse bli angitt som 7-[2-(2,4-dioxo-1-pyrimidinyl)-2-fenylacetylamino]cefalosporansyre. I tillegg til carboxyIradikalet ved R^ innbefattes methyl og ethyles-tere-eller carbomethoxy og carbethoxyradikaler innen oppfinnelsens ramme.
Oppfinnelsen angår særlig fremstilling og beskrivelse av 2-(2, 4-dioxo-1-pyrimidinyl)-acetylaminoderivater av (3-lactam antibiotika. Disse derivater fremstilles ved kondensasjon med den lett tilgjengelige 6-aminopenicillansyre eller en hvilken som helst av de tilgjengelige 7-aminocefalosporinmellomprodukter..Således, hvor R4er radikalet
omfattes derivater av penicillinseriene, mens når R^er radikalet
beskrives derivater av cefalosporinseriene.
Variasjoner innen cefalosporinserien indikeres ytterligere av symbolet X. Hvor således X er hydrogen, beskrives desacet-oxycefalosporansyrer, og hvor symbolet X er hydroxyl, beskrives desacetylcefalosporansyrer. Når symbolet X betegner et acetoxyra-dikal, er (3-lactamkjernen den til cef a losporansyren. Behandling av cefalosporin C med pyridin efterfulgt av en syrehydrolyse gir forbindelsen 7-amino-3-(pyridiniummethy1)decefa losporansyre av den nedenfor viste formel. Fremstillingen av denne forbindelse er kjent innen faget og beskrevet f.eks. i US patentskrift 3.117.126 og britiske patentskrifter 932.644, 957.570 og 959.054.

Når forbindelsen 1-methy1-1,2,3,4-tetrazol-5-thiol anvendes, erholdes forbindelsen 7-amino-3-[(1-methy1-1,2,3,4-tetrazol-5-ylthio)methyl]decefalosporansyre av formelen 2,4-dioxo-1-pyridinylearboxylsyrene (II) som anvendes som utgangsmateria ler er flesteparten kjente forbindelser som synteti-seres i et eller to trinn via kondensasjon av et alkalimetallsa.lt av et 2,4-dioxo-pyrimidin (XI) med ethylbromacetat eller et substituert ethylbromacetat (XII). De substituerte ethoxyacrylamidderivater fremstilles på sin side ved kondensasjon av et substituert N-acetoacetylurethan (XIV) med triethylorthoformiat (XV) ved kokning med eddiksyreanhyd-rid under tilbakelopskjoling. Denne serie av reaksjoner er illustrert i det efterfolgende reaksjcnsskjema: hvor ' betegner acetyl, cyano, carbomethoxy og carbethoxy, en un-dergruppe av gruppen representert av R^jog R^og R^har de tidligere angitte betydninger.
Det substituerte N-acetoacetylurethan (XIV) som anvendes som utgangsmateriale i den ovenfor angitte reaksjonsrekke kan, når det er substituert med et acetylradika1 fremstilles ifolge den fremgangsmåte som er beskrevet av Dewar og Shaw, J. Chem. Soc., 3254 (1961). Når det er substituert med et cyanoradika1, kan ut- gangsmaterialet fremstilles som beskrevet av Atkinson, Shaw og Warrener, J. Chem. Soc., 4118 (1956). Carbomethoxy og carbethpxy-derivatene av forbindelsen av XIV fremstilles efter den fremgangsmåte som er beskrevet av Holy, Collect. Czech. Chem. Commun, 37, 1555 .( 1972) .
6-aminopenicillansyren eller en 7-aminocefalosporansyre (III) kan kobles direkte med en 2, 4--dioxy- 1-pyrimidiny 1- (substitu .
ert)-eddiksyre (II). Alternativt kan G-lactamsyren kobles som sitt noyfrale salt (fortrinnsvis som natrium eller triethylaminsaIt) eller kobles som dets S-lactamester. Estere representert av formelen (III) er de i hvilke den frie carboxyIgruppe av G-lactamforbindelsen er blitt forestret. Siden estergruppen må fjernes efter koblingsreaksjonen, anvendes fortrinnsvis de éstergrupper som lett kan fjernes for igjen å gi den frie carboxyIsyre, slik som f.eks. ved solvo-lyse, hydrogenolyse eller via en nucleofil ombytning,uten påvirkning eller forandring av den gjenværende del av molekylet. Forestrende
grupper som lett omdannes til den frie carboxyIsyre under milde be-tingelser er de'silylerte og og stannylerte carboxyIgrupper. Disse grupper dannes ved å behandle forbindelser som har en fri carboxyl-gruppe med et egnet silyleringsmiddel, slik som f.eks. et alkyldi-silazan, slik som hexamethyldisilazan. Egnede stannyleringsmidler innbefatter f.eks. et bis-(tri-lavere alkyltinn)oxyd s>ik som bis-(tri-n-butyltinn)-oxyd, et tri-lavere alkyltinn-hydroxyd slim som triethyltinnhydroxyd, en tri-lavere alkoxy-tinnforbindelse slik som triethoxytinnhydroxyd, og et tri-lavere alky1-tinnhalogenid slik som tri-n-buty1-tinnklorid. Den resulterende si lylerte. eIler stannylerte carboxy Igruppe kan. regenereres til den onskede frie carbox-ylsyre ved behandling med et ndytralt, hydrogendonerende middel. Vann eller en lavere alkanol, slik som f-eks. ethanol, anvendes fortrinnsvis som det hydrogendonerende middel.
Med hensyn til de 2,4-dioxo-1-pyrimidiny1-(substituerte)-eddiksyrer (II) som anvendes for å kobles til 6-aminopeniciHan eller 7-aminocefalosporansyrene, kan funksjonelle ekvivalenter for-skjellig fra den frie (substituerte)-eddiksyre også anvendes. Eksempler på slike reaktive ekvivalenter innbefatter de tilsvarende syrehalogenider, syreazider, blandede syreanhydrider med alkylfos-forsyre eller alky 1-carbonsyre, syreamider med imidazol eller -et 4-substituert imidazol, syrecyanomethylestere og syre p-nitrofeny1-estere.

Fortrinnsvis utfores koblingsreaks j onen i nærvær <• a v et kondensas jonsraiddel slik som dicyclohexylcarbodiimid , N-cyclohexj'!-N'-morfolinethylcarboiiraid, pentamethylenketon-N-cyclohexylimin, N-ethylfenylisoxazolium-3<1->sulfonat og fosfortriklorid. Under slike omstendigheter antas reaksjonen å forlope gjennom en aktiv form av carboxylradikalet i 2,4-dioxo-1-pyrimidinyl-(substituert)-eddiksyre eller via aminoradikalet av 6-aminopenicillansyren eller 7 -amino-cefalosporansyren.
Koblingsreaksjonen utfores vanligvis i nærvær av et egnet opplosningsmidde1. Egnede opplosningsmidler innbefatter aceton, dioxan, acetonitril, kloroform, ethylenklorid, tetrahydrofuran eller andre inerte, vanlig anvendte opplosningsmidler. Reaksjonen kan ytterligere utfores i nærvær av en base slik som et alkalime-tallcarbonat, trialkylamin eller pyridin. Vanligvis utfores reaksjonen ved romtemperatur eller lavere.
Efter at koblingsreaksjonen er fullfort, hvilket vanligvis skjer efter fra 30 minutter til 4 timer, isoleres reaksjonsproduk-tet under anvendelse av kjente metoder innen faget.
De nye forbindelser ifolge oppfinnelsen er biologisk aktive og er funnet å utvise en god antibakterie11 aktivitet. Således er de anvendbare antimikrobielle midler som har en bredspektret in vitro antimikrobiell aktivitet overfor de standard laboratoriemik-roorganismer som anvendes for å undersoke aktiviteten overfor pato-gene bakterier. Det antibakterielle spektrum til typiske forbindelser bestemmes ved en standardmetode ved den agar-fortynnings-plateteknikk som vanligvis anvendes for å teste nye antibiotika. Den hoye in vitro antibakterielle aktivitet til de nye forbindelser ifolge oppfinnelsen gjor dem ikke .bare anvendbare som farmakologiske midler per se, men gjor dem også anvendbare som additiver for dyre-for, såvel som additiver for materialer som underkastes mikrobiell nedbrytning, slik som boreoljer og brenselsoljer. Disse forbindelser er også anvendbare på grunn av deres antibakterielle effekt i såper, shampoer og i topiske komposisjoner for behandling av sår og brannskader.

Eksempel 1 2-( 2, 4-diox o- 1- pyrimidiny1) eddiksyre
En opplosning av 22 g (0,2 mol) i en 1 N opplosning av ka-liumhydroxyd ble oppvarmet til tilbakelopstemperatur, og 18 g (0,2 mol) kloreddiksyre ble tilsatt porsjonsvis. Efter 18 timers kokning ved tilbakelopstemperatur ble reaksjonsblandingen avkjblet, surgjort og filtrert under dannelse av 20 g hvit2,4-dioxo-1-pyrimidinyled-diksyre med et smeltepunkt på 285°C.

11,4 g (0,05 mol) av forbindelsen a-acety1-S-ethoxy-N-eth oxycarbonylacrylamid, fremstillet ifolge J.H. Dewar og G. Shaw,
J. Chem. Soc, 3254 (1961) ble tilsatt til en opplosning av 7,6 g (0,05 mol) a-fenylglycin i 20 ml 2 N natriumhydroxyd. Reaksjonsblandingen ble oppvarmet på dampbad i 5 minutter, avkjolet, surgjort og::filtrert under dannelse av 4 g 2-(5-acety 1-2, 4-dioxo-1-pyrimidinyl) f eny leddiksy re med et smeltepunkt på 221 - 223°C, (spaltning).
Eksempel 4 7-[2-(2,4-dioxo-1-pyrimidinyl)acetylamino]-cefalosporansyre, natriumsalt
3,4 g (0,02 mol) av forbindelsen 2,4-dioxo-1-pyrimidiny1-eddiksyre ble opplost i 50 ml dimethylformamid, og opplosningen ble avkjolet til 0°C. 3,2 g (0,02 mol) carbonyIdiimidazol ble tilsatt, og blandingen ble omrort i ni trogenatmosfære ved 0°C i 30 minutter og ble derefter oppvarmet til romtemperatur. Reaksjonskolben ble evakuert 30 minutter og avkjblet til -20°C. Opplosningen ble behandlet med 2 ml metbanol, og den utfel-te 7-aminoce.fa losporansyre ble f jernet, ved filtrering. En opples-ning av natrium-2-ethyIhexanoat i n-butanol (10 ml av en 2 N opplosning) ble tilsatt, og blandingen ble fortynnet med ether til et volum på 1 liter for å utfelle produktet. Efter utfelling pånytt fra methanol med ether ble der oppnådd et utbytte på 2,0 g hvit 7-[2-(2,4-dioxo-1-pyrimidiny1)acetylamino]cefalosporansyre med smeltepunkt 240°C (spaltning). Jodtitrering indikerte en renhet på 98,5 %.
Ved å gjenta den samme prosedyre, men anvende 7-amino-des-acetylcefalosporansyre, 7-amino-3-[(1-methyl-l,2,3,4-tetrazol-S-yl-thio) methy 1 ]decef a losporansyre og 7-a mi no-3- [ (5-me.thy 1- 1,3,4- thia-diazol-2-ylthio)methyl]cefalosporansyre i stedet for 7-aminocefalosporansyre ble de.r erholdt natriumsalter av 7-[2-(2, 4-dioxo-1-pyrimidiny 1) ace ty lamino ] des ace ty lcefalosporansyre, 7-[2-(2,4-dioxo-1-pyrimidiny1)acetylamino]-3-[(1-methy1-1,2,3,4-tetrazol-5-ylthio)-methyl]decefalosporansyre og 7-[2-(2,4-dioxo-1-pyrimidiny1)acety1-amino]-3-[(5-methy1-1,3,4-thiadiazol-2-ylthio)methyl]cefalosporan-syre.
Eksempel 5 7-[2-(5-cyano-2,4-dioxo-1-pyrimidinyl)acetylamino]-cef a losporansyre, natriumsa. lt
3,9 g (0,02 mol) 5-cyano-2,4-dioxo-1-pyrimidiny1)eddiksyre ble opplost i 50 ml dimethy lf orrnamid, og opplosningen ble avkjolet til 0°C. 3,2 g (0,02 mol) carbonyIdiimidazol ble tilsatt, og blandingen ble omrort under nitrogenatmosfære ved 0°C i 30 minutter, og ble derefter oppvarmet til romtemperatur. I en separat kolbe ble 7-aminocefalosporansyre silylert ved å oppvarme en suspensjon av 5,4 g (0,02 mol) 7-aminocefalosporansyre og 8 ml hexamethyIdisila-zan i 50 ml kloroform ved tilbakelbpskjbling i 30 minutter. Det faste materiale ble opplost i 100 ml vann, og opplosningen ble skylt med 100 ml ethylacetat. Opplosningen ble kraftig omrort og surgjort til pH 2 med 6 N saltsyreopplosning. Blandingen ble filtrert, og det organiske lag ble fraskilt fra filtratet. Det vandige lag ble ekstrahert med ytterligere 100 ml ethylacetat. De kombinerte ethylacetatlag ble torket over magnesiumsulfat, og en opplosning av natrium 2-ethyIhexanoatopplosning (10 ml av en 2 N opplosning i n-butanol) ble tilsatt.
Det resulterende bunnfall, ble frafiltrer og vakuumtorket under dannelse av et utbytte på 0,7 g 7-[2-(5-cyano-2,4-dioxo-1-pyrimidinyl)acetylaminojcefalosporansyre med et smeltepunkt på 272°C, (spa l.tning) .
Ved å folge den samme prosedyre, men anvende 2-(2,4-dioxo-5-nitro-1-pyrimidimy1)eddiksyre,
2- (2, 4-dioxo-5-methy 1- 1-py rimidi ny 1.) f eny leddiksy re,
2-(5-brom-2,4-dioxo-1-pyrimidinyl)eddiksyre,
2-(5-carbethoxy-2,4-dioxo-1-pyrimidiny1)-2-methyleddiksy.re og 2-(2,4-dioxo-3-methyl-1-pyrimidinyl)eddiksyre i stedet for 2-(5-cyano-2,4-dioxo-1-pyrimidinyl)eddiksyre,
ble der erholdt natriumsalter av
7-[2-(2,4-dioxo-5-nitro-1-pyrimidiny1)acetylaminojcefalosporansyre, 7-[2-(2,4-dioxo-5-methy 1-1-pyrimidinyl)-2-fenylacetylaminojcefalo-sporansyre,
7-[2-(5-brom-2,4-dioxo-1-pyrimidinyl)acetylamino]cefalosporansyre, 7-[2-(5-carbethoxy-2,4-dioxo-1-pyrimidiny1)-2-methylacetylamino]-cefalosporansyre,
7-[2-(2,4-dioxo-3-methyl-l-pyrimidinyl)acetylamino]cefalosporansyre.
Eksempel 6 6-[2-(5-acety1-2,4-dioxo-1-pyrimidiny1)acetylamino]-peni cillansyre, natriumsaIt
En opplosning av 1,9 g (0,0082 mol) 2-(5-acety1-2,4-dioxo-1-pyrimidiny 1.) -eddiksyre i 25 ml dimethylf ormamid ble . avkjblet til -5°C, og 1,3 g (0,0082 mol) carbonyIdiimidazol ble tilsatt i én porsjon. Blandingen ble omrort ved -5°C til 0°C under nitrogénat-mosfære i 1 time og evakuert i 15 minutter for å fjerne det utvik-lede carbondioxyd i imidazoliddanneIsen.
Ved -10°C ble en opplosning av 1,75 g (0,0082 mol) 6-amino-penici llansyre og 2 g 0,02 mol) triethylamin i 25 ml kloroform tilsatt. Reaksjonsblandingen ble omrort ved -10°C i 1 time, oppvarmet til romtemperatur, og 5 ml av en 2 N opplosning av natrium 2ethylhexanoat i n-butanol ble tilsatt. Utfelling av produktet ble fullfort ved tilsetning av 500 ml ether. Det faste materiale ble isolert ved filtrering og vakuumtorket under dannelse av 2,7 g hvitt fast materiale som var en blanding av 6-aminopenicillansyre og produktet.
Den faste blanding ble opplost i 70 ml koldt vann, .'og den resulterende opplosning ble skilt med 70 ml ethylacetat. Mens blandingen ble omrort ble en opplosning av 6 N saltsyre tilsatt for å justere oppløsningens pH til 2. Lagene ble fraskilt, og det vandige lag ekstrahert med ytterligere 70 ml ethylacetat. De kombinerte ethylacetatekstrakter ble tbrket over magnesiumsulfat og filtrert. Filtratet ble behandlet med 5 ml natrium 2-ethyIhexanoatopplosning, og ethylacetatet ble dekantert fra bunnfallet. Efter utfelling pånytt fra methanol med ether ble der erholdt 1,2 g hvitt, fast materiale med smeltepunkt 227°C (spaltning).

2-(2,4-dioxo-5-methyl-1-pyrimidiny1)eddiksyre,
2-(5-cyano-2,4-dioxo-1-pyrimidinyl)eddiksyre,
2-(2,4-dioxo-5-methy1-1-pyrimidinyl)-2-methyleddiksyre, 2-(5-acetyl-2,4-dioxo-1-pyrimidinyl)-2-fenyleddiksyre - og 2-(2,4-dioxo-3-methy1-1-pyrimidiny1)eddiksyre i stedet for
2-(5-acety1-2,4-dioxo-1-pyrimidinyl)eddiksyre ble der erholdt natriumsalter av 6-[2-(2,4-dioxo-1-pyrimidinyl)-acetylamino]penicillansyre, 6 - [2 - (2, 4-dioxo-5-methyl- 1-pyrimidinyl) acety lamino] penici llansyre, 6-[2-(5-cyano-2,4-dioxo-1-pyrimidiny1)acetylamino]penicillansyre, 6-[2-(2,4-dioxo-5-methy1-1-pyrimidinyl)-2-methylacetylamino]-penicillansyre,
6-[2-(5-acety1-2,4-dioxo-1-pyrimidinyl)-2-fenylacetylamino]
penicillansyre og 6-[2-(2,4-dioxo-3-methyl-1-pyrimidiny1)acetylamino]penicillansyre.
Eksempel 7 7-[2-(2,4-dioxo-1-pyrimidinyl)acetylamino]-3-pyridiniumdecefalosp oransyre
En opplosning av 4,8 g (0,01 mol) 7-[2-(2,4-dioxo-1-pyrimidinyl) acety lamino]cef alosporansyre, natriumsalt, 0,02 mol kalium-thiocyanat og 0,03 mol pyridin i 15 ml vann ble behandlet med 85 %'s fosfo.rsyre til pH 6,5. Reaksjonsblandingen ble oppvarmet til 60°C
i 6 timer, og 7-[2-(2,4-dioxo-1-pyrimidiny1)acetylamino]-3-pyridin-iumdecefalosporansyre ble isolert efter den metode som er beskrevet av J'.L. Spencer, et. al., J. Org. Chem., 32, 500 (1967).

R3 er valgt fra gruppen bestående av hydrogen, methyl, fenyl,
carboxy, carbomethoxy og carbethoxy,R^ er valgt fra gruppen bestående av
hvor X er hydrogen, hydroxy, acetoxy, N-pyridinium, 5-methy1-1,3,4-
thiadiazol-2-y.lthio og 1-raethy1-1,2,3,4-tetrazol-5-ylthio og farmasbytisk akseptable salter derav, karakterisert v e d at.a) en 2,4-dioxo-1-pyrimidiny1-eddiksyre av formelen
