Description
Fremgangsmåte ved fremstilling av 4-oxc-l-pyridinylpenicillin og cefalosporinderivater
Foreliggende oppfinnelse angår nye syntetiske forbindelser av penicillin<p>g cefalosporinklassen som er anvendbare som antibakterielle midler. Disse forbindelser utviser en høy grad av aktivitet overfor et stort antall mikroorganismer. Cefalosporinderivatene ifølge oppfinnelsen er særlig anvendbare overfor peni-cillinase-produserende mikroorganismer. Som antibakterielle midler er forbindelsene ifølge oppfinnelsen terapeutisk effektive ved behandling av infektiøse sykdommer som skyldes gram-positive og gram-negative bakterier i fjærfe og dyr, innbefattet mennesket.
I tillegg er forbindelsene ifølge oppfinnelsen anvendbare som dyre-fortillegg og som aktiv bestanddel i bakteriedrepende preparater som anvendes som overflatedesinfiserende midler.
Splittingen av penicilliner til 6-aminopenicillan.syré i 1959 og den kjemiske splitting av cefalosporin under dannelse av den tilsvarende 7~aminocefalosporansyre gjorde syntesen av nye. syntetiske penicilliner og cefalosporiner som ikke tidligere var tilgjengelige via fermenteringsprosedyrer mulig. Acylering av. aminogruppen har gitt derivater inneholdende en heterocyklisk ring i 6-sidekjeden slik som tilfellet er i penicillinserien, eller i den tilsvarende 7-sidekjede, si±k som tilfellet er i cefalosporinserien. Slike heterocykliske deler innbefatter thiofenringen som beskrevet i US patentskrift 3 218 318, 3 449 338 og 3 498 979 (cefaloridin og cefalothin), picolin, US patentskrift 3 553 203, hydantoin, US patentskrift 3 227 712 og forskjellige andre nitrogén-holdige heterocykliske deler innbefattet pyrrolidin og nicotinsyre, US patentskrift 3 308 120.
I hvert tilfelle er den heterocykliske del bundet til en sidekjede, generelt til det av et acetylradikal, gjennom en. av ringcarbonatomene. Foreliggende oppfinnelse angår 4-oxo-l-pyridi-nylderivater som er bundet direkte til acetylradikalet gjennom heteroatomet. Kjente eksempler inneholdende denne type binding, og som i denne forbindelse representerer den nærmeste kjente lære, er den tetrazolring som er beskrevet i US patentskrift 3 516 997 (cefazolin) og visse kinazolinylderivater av penicillansyre, slik som beskrevet i US patentskrift 3 652 547.
Foreliggende oppfinnelse angår nye 4-oxo-l-pyridinylpeni-cillin og cefalosporinderivater. Nærmere bestemt angår oppfinnelsen substituerte(4-oxo-l-pyridinyl)-acetylamino-penicillin og cefalosporinderivater som er anvendbare som antibakterielle midler og som kan representeres av den generelle formel:

Forbindelsene ifølge foreliggende oppfinnelse fremstilles ved kondensasjon av en 6-aminopenicillansyre eller en 7-aminocefalosporansyre med en (substituert) 4-oxo-l-pyridinyleddiksyre som vist i følgende reaksjonsskjema:
Alle forbindelser ifølge oppfinnelsen inneholder et 4-oxo-l-pyridinylradikal eller en 4-pyridondel i endestilling av acetylaminosidekjeden, som angitt i generell formel I. Når det gjelder penicillinserien, er acetylaminosidekjeden numerert som 6-stillingen, mens forbindelsene i cefalosporinserien er numerert i. 7-stilling. Numereringssystemet for disse to serier av forbindelser er vist for utgangsforbindelsene 6-aminopenicillansyre (IV) og 7-aminocefalosporansyre (V) nedenfor:
4-pyridondelen bundet til acetylaminosidekjeden kan.være substituert eller usubstituert. Disse substituenter foreligger i enten 2-, 3-, 5- eller 6-stillinger i pyridinkjernen og innbefatter følgende radikaler<1>halogen, hydroxyl, lavere alkyl, trifluor-
methyl, nitro, amino, cyano, carboxyi og methyl- og ethylestere av carboxylradikalet. Uttrykket halogen innbefatter fluor, klor, brom og jodradikaler. Uttrykket lavere alkyl som anvendt heri omfatter både rettkjedet og forgrenet alifatiske hydrocarbonradi-kaler med fra 1 til 4 carbonatomer. Særlig innbefattet er slike som methyl, ethyl, propyl, isopropyl, butyl, isobutyl og t-butyl-radikaler.
I tillegg til de forskjellige substituenter som ovenfor er beskrevet, kan 4-pyridondelen betraktes som substituert med en tilstøtende mettet eller umettet 6-delt ring. Således kan symbolet R-j^ når det sees i kombinasjon med det tilstøtende symbol R2betraktes som å danne et bundet 6-delt alicyklisk eller aromatisk derivat ved 2,3-stilling i pyridinringen. Disse derivater angis mer nøyak-tig som 2-(substituerte(4-oxo-l-tetrahydrokinolinyl)}-acetylamino og 2-(substituert(4-oxo-l-kinolinyl))-acetylamino-derivater av penicilliner og cefalosporiner. På grunn av symmetrien av pyridin-molekylet behøves' således bare et par tilstøtende symboler å defi-neres. Foreliggende oppfinnelse innbefatter ikke de tricyliske heterocykliske dibenzo-pyridin- eller acridinringsystemer.
I tillegg til den obligatoriske substitusjon av 2-methylgruppen av acetylamino- eller acetamidodelen av molekylet med 4-oxo-l-pyridinylradikal, kan 2-methylgruppen inneholde ytterligere substitusjon i form av et methylradikal eller et carboxylradikal som representert ved symbolet R5. Når R5er methyl, er forbindelsene mer nøyaktig angitt som propionylderivater av 6-aminopenicillansyre eller av 7-aminocefalosporansyre. For å få ensartethet i nomenklaturen er de imidlertid her angitt som 2-(substituerte)-acetylaminoderivater.
Når det gjelder et cefalosporansyrederivat hvori. R^ er
.methyl og 5-pyridonet forblir usubstituert, er således forbindelsen angitt som 7-(2-(4-oxo-l-pyridyl)-2-methylacetylamino) -cefalosporansyre. I tillegg til carboxylradikalet ved R5innbefattes

Oppfinnelsen angår hovedsakelig fremstilling og beskriv-else av 2-(4-oxo-l-pyridinyl)-acetylaminosidekjedederivater av lactamantibiotica. Disse derivater fremstilles ved kondensasjon med den lett tilgjengelige 6-aminopenicillansyre eller en hvilken som helst av de tilgjengelige 7-aminocefalosporinmellomprodukter.
Når således Rg er radikalet
betraktes derivater av penicillinserien, mens når Rg er radikalet
beskrives derivater av cefalosporinserien.
g<->lactamkjernen kan forbli.usubstituert eller den kan substitueres med en methoxysubstituent som angitt ved symbolet Rg. Slik substitusjon skjer ved 6-stilling i penicillinserien og ved 7-stilling i céfalosporinserien av forbindelsene.
Både 3-stillingen i penicillinet og 2-stillingen i cefalosporinserien av forbindelsene er substituert med enccarboxylsyre eller en carboxylsyreester som angitt ved delstrukturen -COORy.
Når Ry er hydrogen, erholdes de tilsvarende penicillansyrer eller cefalosporansyrer. I tillegg kan symbolet Ry betegne følgende radikaler: alkanoyloxymethyl, alkanoylaminomethyl, alkoxycarbonylaminomethyl og p-(alkanoyloxy)-benzyl. Disse estere gir molekylet glimrende absorpsjonsegenskaper og er samtidig fysiologisk stabile. Således absorberes disse estere fra gastro-intestinaltractus og hydrolyseres enzymatisk til de tilsvarende penicillan- eller cefalosporansyrer og tilveiebringer således en god oral aktivitet.
Visse spesifike variasjoner innen cefalosporinserien er ytterligere angitt ved symbolet X. Når X er hydrogen beskrives desacetoxycefalosporansyrer, og når symbolet X er hydroxyl, beskrives desacetylcefalosporansyrer. Når symbolet X betegner et acetoxy-radikal er $-lactamkjernen den tilsvarende til cefalosporansyren. Ytterligere substituenter ved 3-stillingen av decefalosporansyr som er innbefattet innen foreliggende oppfinnelse og som er representert ved syiobblet X er n-pyridinium, 5-methyl-l,3,4-thiadiazol-2-ylthio

De farmasøytisk akseptable salter av forbindelsen av formel I omfatter de ikke-toksiske, carboxylsyresalter som dannes med en hvilken som helst egnet organisk eller uorganisk base. Disse salter innbefatter salter av alkalimetaller slik som f.eks. natrium og kalium, jordalkalimetaller slik. som kalsium og magnesium, lette metaller fra gruppe HIA innbefattet aluminium, og organiske primære, sekundære og tertiære aminer, slik som f.eks. trialkylaminer, innbefattet triethylamin, procain, dibenzylamin, 1-ethenamin, N,N'-dibenzylethylendiamin, dihydroabiethylamin, N-(lavere)-alkylpiper-idin, og dessuten aminer som er funnet å kunne danne ikke-toksiske salter med benzylpenicillin. Disse salter kan fremstilles under anvendelse av vanlige metoder slik som å bringe carboxylsyren i et polart løsningsmiddel i kontakt med en støkiometrisk mengde av basen.
Også innbefattet som farmasøytisk akseptable syreaddisjonssalter er de ikke-toksiske organiske eller uorganiske syreaddisjonssalter av baseforbindelsen av formel I. Illustrative uorganiske syrer som danner egnede salter innbefatter saltsyre, hydrobromsyre, svovelsyre og fosforsyre såvel som syremetallsalter slik som natriummonohydrogenorthofosfat og kaliumhydrogensulfat. Illustrative organiske syrer som danner egnede salter innbefatter mono-, di- og tricarboxylsyrer, slik som f.eks. eddiksyre, glycolsyre, melkesyre, pyrodruesyre, malonsyre, ravsyre, glutar-syre, fumarsyre, malinsyre, vinsyre, citronsyre, ascorbinsyre, maleinsyre, hydroxymaleinsyre, benzoesyre, p-hydroxybenzoesyre, fenyleddiksyre, cirmaminsyre, salicylsyre, 2-fenoxybenzoesyre og sulfonsyrer slik som methansulfonsyre og 2-hydroxyethansulfonsyre. Slike salter kan foreligge enten i form av et hydrat eller som en hovedsakelig vannfri form.
I tillegg til de ikke-toksiske carboxylsyresalter og de ikke-toksiske syreaddisjonssalter av basene, innbefatter uttrykket farmasøytisk akseptable salter indre salter eller zwitter-ioner av disse forbindelser av formel I som er amfotere i naturen. 6-lactamutgangsmaterialene (III) er alle kjente forbindelser. Forbindelsen 6-aminopenicillansyre av formelen
kan fremstilles under anvendelse av biologiske metoder og kan også fremstilles ved hydrolyse av forskjellige penicillener som beskrevet i US patent 3 499 909. Hydrolyse av antibioticumet cefalosporin C fører til dannelse av 7-aminocefalosporansyre, Loder et al., Biochem. J. 79, 408 - 417 (1961) av formelen Forbindelsen 7-aminodesacetoxycefalosporansyre med formelen
fremstilles ved katalytisk reduksjon av cefalosporin C, etterfulgt av hydrolytisk fjerning av 5-aminoadipoylsidekjeden som beskrevet i US patentskrift 3 129 224.
De forbindelser av 3-lactamutgangsmaterialet (III) hvor symbolet Rg betegner en methoxygruppe har tidligere vært beskrevet i US patentskrift 3 778 432.
Behandling av cefalosporin C med et acetylesterease fremstilt fra appelsinskall,Jeffery et al., Biochem. J., 81, 591 (1961) fører til dannelse av 3-hydroxymethyl-7-aminodecefalosporansyre eller 7-aminodesacetylcefalosporansyre med formelen
Behandling av cefalosporin C med pyridin etterfulgt av syrehydrolyse gir forbindelsen 7-amino-3-(pyridiniummethyl)-decefalosporansyre med den nedenfor viste formel. Fremstilling av denne forbindelse er kjent innen faget og beskrevet f.eks. i US patentskrift 3 117 126 og britisk patentskrifter 932 644, 957 570 og 959 054.
De 3-thiolaterte 7-aminocefalosporansyrer kan erholdes ved å omsette 7-aminocefalosporansyre med den passende thiol slik som beskrevet i US patentskrift 3 516 997. Når således 5-methyl-1,3,4-thiadiazol-2-thiol anvendes erholdes forbindelsen 7-amino-3-((5-methyl-l,3,4-thiadiazol-2-ylthio)-methyl)-decefalosporansyre som har formelen
Når forbindelsen 1-methyl-l,2,3,4-tetrazol-5-thiol anvendes erholdes forbindelsen 7-amino-3-/{ (1-methyl-l, 2 ,3 , 4-tetrazol-5*-ylthio) -methyl) -decefalosporansyre med formelen 4-oxo-l-pyridinyl-(substituert)-eddiksyrer (II) som anvendes som utgangsmaterialer er for det meste kjente forbindelser som kan syntetiseres i enten én eller to trinn via kondensasjonen av et alkalimetallsalt av et hydroxypyridin (XI) med ethylbromacetat eller et substituert ethylbromacetat (XII). Generelt fore-trekkes kaliumsaltet av hydroxypyridin for å bevirke kondensasjon, og den resulterende ester hydrolyseres til den ønskede 4-oxo-l-pyridinyl- (substituert) -eddiksyre (II) med en vandig base som vist i det følgende reaksjonsskjerna:

En foretrukket metode innbefatter en foregående silyli-ering av et (substituert)-4-hydroxypyridin (XI) i nærvær av en organisk base som slik triethylamin, med et tri-(lavere lakyl)-substituert halogensilan, slik som f.eks. klortrimethylsilan. Denne metode muliggjør rensing av det silulerte mellomprodukt som i sin tur resulterer i fremstilling av (substituert)e-(4-dxo-l-pyridinyleddiksyrer (II) med mindre forurensninger og i et høyere utbytte.
De (substituerte)-4-oxo-l-pyridinyleddiksyrer av formel (II) hvor Rcjer hydrogen kan også fremstilles ved behandling av pyron eller ved behandling av et substituert pyron med glycin i nærvær av en base. I denne nukleofile reaksjon åpnes pyronringen, vann elimineres og ringen lukkes igjen med aminonitrogenatomet nå inneholdt i pyridinringen. Denne reaksjon som vanligvis utføres ved forhøyede temperaturer i løpet av et lengre tidsrom, kan illu-streres som følger ved anvendelse av 2,6-dimethyl-4-pyron (XV) for fremstilling av 2,6-dimethyl-4-pyridon-l-eddiksyre (XVI).
Generelt kan 6-aminopenicillan eller 7-aminocefalosporan-syrederivatene ifølge oppfinnelsen fremstilles ved kondensasjon av en 4-oxo-l-pyridinyleddiksyre (II) og et amino-3-lactam (III) som tidligere angitt. Koblingsreaksjonen utføres vanligvis i løsning i. nærvær av et egnet løsningsmiddel. Egnede løsningsmidler innbefatter aceton, dioxan, acetonitril, kloroform, ethylenklorid, tetrahydrofuran eller andre inerte og lett tilgjengelige løsnings-midler. Koblingsreaksjonen utføres ennvidere i nærvær av en base, slik som et alkalimetallcarbonat.eller et alkalimetallsyrecarbonat, trialkylamin, i hvilket alkylgruppen har fra 1 til 5 carbpnatomer eller pyridin. Temperaturen ved koblingsreaksjonen kan variere fra -20° C til 100° C og hvor den foretrukne temperatur er romtemperatur eller svakt lavere enn romtemperatur. Reaksjonstiden kan variere fra 15 minutter til så lenge som 36 timer, avhengig selvsagt av temperaturen på reaksjonsblandingen og reaktiviteten av de anvendte reaktanter. Fortrinnsvis anvendes en periode av fra 1 til 8 timer. Etter kondensasjonsreaksjonen isoleres reaksjons-prdquktene og gjenvinnes under anvendelse av vanlige eksrraksjons-og krystallisasjonsmetoder som er vel kjent innen faget.
For å gi en egnet drivkraft for koblingsreaksjonen anvendes et koblingsmiddel. En type av koblingsmidler virker hovedsakelig som et dehydreringsmiddel, som aktiverer acyleringen og fjer-ner det vann som dannes under reaksjonen. Slike dehydrerende koblingsmidler eller dehydreringsmidler, som uttrykt her, innbefatter forbindelsene: dicyclohexylcarbo.diimid, N-cyclohexyl-N-morfolino-methylcarbodiimid, pen tame thylketon-N-cyclohexy'limin, N-ethyl-5-fenylisoxazolium-3-sulfonat og fosfortriklorid. Forbindelsen dicyclohexylcarbodiimid representerer et foretrukket dehydreringsmiddel særlig ved fremstilling av cefalosporinserien av forbindelsene .
En annen klasse av koblingsmidler kan anses som å gjen-sidig reagere med de forskjellige 4-oxo-l-pyridinyleddiksyrer som anvendes på en slik måte at carbonylradikalet av eddiksyredelen av molekylet aktiveres under dannelse av et reaktivt mellomprodukt. Dette reaktive mellomprodukt acylerer i sin tur amino-(3-lactamet. Således er de tilsvarende syrehalogenider, syreazider, blandede syreanhydrider med alkylfosforsyre eller alkylcarbonsyre, syreami-der med imidazol eller et 4-subsittuert imidazol, syrecyanomethyl-estere og syre-p-nitrofenylestere alle egnecle reaktive forbindelser som kan anvendes med hell.
Fremstilling av et reaktivt mellomprodukt representerer
. en foretrukket fremgangsmåte for fremstilling av forbindelsene ifølge oppfinnelsen, og i særdeleshet for fremstilling av cefalosporinderivatene som her beskrevet. Egnede koblingsmidler innbefatter carbonyldiimidazol, alkylklorformiat i hvilken alkylgruppen har fra 1 til 5 carbonatomer, thionylklorid, kloracetonitril,'og bis-p-nitrofenylcarbonat. Det foretrukne koblingsmiddel er carbonyldiimidazol som, generelt, kan tilsettes til en løsning av 4-oxo-
1-pyridinyleddiksy.re ved en temperatur lavere enn romtemperatur; Reaksjonsblandingen tillates å nå romtemperatur og reaksjonsblandingen underkastes redusert trykk for å fjerne det carbondioxyd som utvikles under dannelse av imidazolidet. Løsningen inneholdene det reaktive imidazolidmellomprodukt avkjøles igjen og kobles nå rseci éiet" egnede 3~lactam. Koblingen utføres generelt ved en temperatur av fra 0 til 150° C i løpet av et tidsrom på 1 - 12 timer, hvorpå det ønskede produkt gjenvinnes under anvendelse av isoler-ingsteknikker som er vel kjent innen faget.
Som et alternativ til den direkte kobling av amino-B-lactamsyren, kan den passende 6-aminopenicillansyre eller 7-amino-cef alosporansyre kobles som et nøytralt salt eller i form av en ester.• Egnede salter innbefatter trialkylammoniumsaltene hvori alkylgruppen har fra 1 til 5 carbonatomer. Eksempler på slike salter er de som dannes ved trimethylamin eller triethylamin. Estere representert av formel (III) er de i hvilke den fri carboxy1-gruppe av amino-$-lactamet e.r blitt egnet forestret. I de tilfeller i hvilke estergruppen deretter fjernes for å gi den fri syre, anvendes fortrinnsvis de estergrupper som lett kan fjernes.
Både silyl og stannylestrene er blandt de estere som lett omdannes til den tilsvarende fri syre under relativt milde betingelser. Således kan f.eks. estrene underkastes hydrolyse, solvolyse eller en. nucleofil utveksling, uten påvirkning av den gjenværende del av molekylet. Egnede silyleringsmidler innbefatter alkyldisilazanene, som f.eks. tetramethyldisilazan og hexamethyldisilazan, eller bis-trimethylsilylacetamid. Egnede stannylerings-midler innbefatter f.eks. et bis-(tri-lavere alkyl-tinn)-oxyd slik som bis-(tri-n-butyl-tinn)-oxyd, et tri-lavere alkyl-tinn-hydroxyd slik som triethyl-tinn-hydroxyd,, en tri-lavere alkoxy-tinnforbin-delse som triethoxy-tinn-hydroxyd, og et tri-lavere-alkyl-tinn-halogenid slik som tri-n-butyl-tinn-klorid. Den resulterende silylerte eller stannylerte carboxylgruppe kan regenereres til den ønskede fri carboxylsyre ved behandling med et nøytralt hydrogen-donerende middel. Vann eller en lavere alkanol, slik som f.eks. ethanol, anvendes fortrinnsvis som det hydrogen-doneren.de middel.
En alternativ metode for fremstilling av forbindelsene ifølge oppfinnelsen innbefatter behandling av et 6a-halogenaceta-midopenicillin eller et 7a-halogenacetamido-cefalosporinderivat med et silylert 4-hydroxypyridin som vist i det følgende reaksjonsskjerna:
hvor-- R^/ R2 / R4 / R5 / Rg og Rg har de tidligere angitte betydninger, Rg er lavere alkyl med fra 1 til 5 carbonatomer og Y er brom eller klor.
a-halogenacetamido-ft-lactamutgangsmaterialet er kjente forbindelser som tidligere har vært beskrevet i J.Med. Chem. 16, 1413 (1973) , belgisk patent 758 587 og US patenter 2 941 995 og
3 516 997. Fremstilling av de silulerte 4-hydroxypyridiner finner sted med et egnet siluleringsmiddel som tidligere beskrevet. Egnede siluleringsmidler innbefatter: di-lavere alkylklorsilan, tri-lavere alkylklorsilan, di-lavere alkylbromsilan og tri-lavere
alkylbromsilan i hvilke den lavere alkylgruppe inneholder fra 1 til 5 carbonatomer, tetramethyldisilazan, hexamethyldisilazan og bis-trimethylsilyiacetamid.
Kondensasjonsreaksjonen utføres vanligvis i et inert løsningsmiddel slik som kloroform, aceton, methylenklorid, dimethylformamid, dioxan eller acetonitril. Temperaturen på reaksjonen kan variere fra -20 til 100° C, med en temperatur på 20° C som, foretrukket. Vanligvis finner reaksjonen sted i en inert atmosfære slik som nitrogen, argon eller helijm. Reaksjonen utføres vanligvis i et tidsrom av fra 15 minutter til 36 timer, avhengig av arten av reaktantene og temperaturen ved hvilken reaksjonen utføres. Reaksjonen fullføres vanligvis i løpet av fra 1 til 24 timer ved det foretrukne temperaturområde.
Generelt omsettes en ekvivalent av det silulerte 4-hydroxypyridinderivat med en ekvivalent av a-halogenacetamido-3-lactamderivatet. 3-lactamderivatet kan også anvendes i form av et salt, slik som natrium, triethylamin, N,N-diethylanilin eller diisopropylethylamin. Alternativt kan 3-lactamet foreligge i form av en ester. Estere innbefatter de som kan lett fjernes for å gi den fri syre under milde betingelser og som ikke påvirker den gjenværende deX av molekylet. Spesifikt innbefatter slike estere: t-butyl, trialkylsilyl og trialkylstannyl hvor alkylgruppen har fra 1 til 5 carbonatomer eller benzylestere, hvor de foretrukne estere er av trialkylsilyltypen. Slike estere kan lett hydrolyseres til den fri syre ved behandling med et nøytralt hydrogen-donerende middel slik som en alkohol.
I alle de ovenfor angitte koblingsreaksjoner beskyttes alle forbindelser ifølge oppfinnelsen som har reaktive funksjonelle grupper som kan innvirke i koblingsreaksjonen under anvendelse av egnede blokkerende grupper. Således kan carboxylgrupper lokalisert på 4-pyridondelen av molekylet eller på eddiksyredelen av molekylet silyleres eller forethres med andre labile estere som tidligere beskrevet. På samme måte kan amino- og hydroxylgrupper lokalisert på 4-pyrididondelen av molekylet hensiktsmessig beskyttes som labile derivater. Slike derivater innbefatter silylethere, benzyletbere og carbonatestere for hydroxylgrupper, carbobenzyloxy, carbo-t-butyloxy og trifenylmethylderivater for aminogrupper.
En alternativ reaksjonvei til cefalosporinderivatene av formel (I) hvori symbolet X betegner 5-methyl-l,3,4-thiadiazol-2-ylthio eller 1-methyl-l, 2 ,3,4-tet.razol-5-ylthio, består i fjerning (displacement) av acetoxygruppen fra methylgruppen ved 3-stillingen av de substituerte (4-oxo-l-pyridinylacetyl)-aminocefalosporansyrer (XIX). Dette er vist i de etterfølgende to reaksjonsskjemaer: hvor symbolene R-^, R2, Rg, R^, Rg og Rg har de tidligere angitte, betydninger.
Cefalosporansyrene av formel (XIX) løses med 2-mercapto-5-methyl-l,3,4-thiadiazol (XX) eller et metallsalt derav i et inert løsningsmiddel. Fortrinnsvis anvendes et alkalimetallsalt av cefalosporansyren (XIX). Disse salter kan fremstilles for eksempel ved behandling av cefalosporansyren med et alkalimetall-bicarbonat. Reaksjonen utføres i vann eller et vandig organisk løsningsmiddel slik som vandig aceton, vandig tetrahydrofuran eller vandig dimethylformamid. Om ønsket kan pH for reaksjonsblandingen reguleres ved tilsetning av vandige buffere. Hvis de fri cefalosporansyrer anvendes som utgangsmaterialer, kan reaksjonen- utføres i nærvær av en base, slik som natriumbicarbonat, triethylamin 1 eller kaliumbicarbonat.
Reaksjonen kan utføres over et temperaturområde fra
25° C til 110° C. Fortrinnsvis anvendes et temperaturområde fra 50 til 100° C. Om ønsket kan reaksjonen utføres i,nærvær av en
inert gass slik som nitrogen eller argon. Reaksjonstiden kan variere fra 15 minutter til 2 4 timer, hvor det anvendes fortrinnsvis en reaksjonstid fra 15 minutter til 6 timer.
I visse tilfeller fører fjerningen av acetoxygruppen fra methylgruppen til 3-stilling til vånding av dobbeltbindingen til 2-stillingen i g<->lactamkjernen. Under disse omstendigheter kan stillingen til dobbeltbindingen lett gjenopprettes ved oxydasjon av ringsvovelet til sulfoxydet med slike oxyderende midler som hydrogenperoxyd, natriummetaperjodat eller en organisk persyre. Etterfølgende reaksjon av sulfoxydet ved hjelp av katalytisk hydro-lysering eller natriumdithionit gir det ønskede cefalosporinderivat som er umettet i 3-stillingen i 3-lactamkjernen.
De nye forbindelser ifølge oppfinnelsen er biologisk aktive og er funnet å utvise en god antibacteriell aktivitet. Således er de anvendbare som anti-mikrobielle midler med en bred-spektret anti-mikrobiell aktivitet in vitro overfor standard laboratorie-mikroorganismer som anvendes for å teste aktiviteten overfor patogene bakterier. De anti-bakterielle spektrum for typiske forbindelser ifølge oppfinnelsen bestemmes ved en standard metode ved den agar-fortynningsstrekkplateteknikk som vanligvis anvendes for testing av nye antibiotica.
Nærvær av 7-methoxy-substituenten i cefalosporinserien medfører den fordelaktige effekt at den forbedrer eller forsterker spekteret av anti-mikrobiell aktivet overfor visse gram-negative mikroorganismer. Nærmere bestemt er forbindelser inneholdende 7-methoxysubstituenten aktive overfor visse gram-negative mikroorganismer som er resistente overfor forbindelser som ikke inneholder 7-methoxysubstituenten, som f.eks. Enterobacter aerogenes, Enterobacter cloacae, Serratia marcescens og de indol-positive arter av Proteus.
Den høye in vitro antibakterielle aktivitet til de nye forbindelser ifølge oppfinnelsen gjør dem ikke bare anvendbare som farmakolotiske midler per se, men gjør dem anvendbare som additiver for dyrefor, såvel som additiver for materialer som underkastes miktobiell nedbrytning, slik som boreoljer og brenseloljer. Disse ,forbindelser er også nyttige på grunn av deres antibakterielle effekt i såper, shampooer og topiske komposisjoner for behandling av sår og brannskader.
Oppfinnelsen beskrives nærmere i de etterfølgende eksempler.
Eksempel 1
4- pyridon- T- eddiksyre
En suspensjon av 19,0 g (0,2 mol) 4-hydroxypyridin,
22,2 g, 0,22 mol, triethylamin og 300 ml toluen ble oppvarmet til tilbakeløpstemperaturen og 23 g, 0,2 mol, klortrimethylsilan ble dråpevis tilsatt. Blandingen ble opparmet under omrøring ved til-bakeløpstemperatur i 18 timer og filtrert. Filtratet ble fordampet og residuet ble destillert ved redusert trykk under dannelse av 10,4 g silulert 4-hydroxypyridin, k.pkt. 102 - 104° C. 9 g, 0,054 mol, av det•silulerte 4-hydroxypyridin ble blandet med 25 ml ethylbromacetat og blandingen ble omrørt inntil den eksoterme reaksjon avtok. Den stivnede blanding ble triturert med ether og filtrert. Det faste materiale ble omkrystallisert fra isopropylalkohol hvorved det ble erholdt 8 g 4-pyridon-l-eddiksyre-ethylester-hydrobromidsalt med sm.p. på 195° C. 8 g, 0,03 mol, av esterhydrobromidsaltet fremstilt på denne måte, ble tilsatt til 80 ml av en IN natriumhydroxydløsning og blandingen ble omrørt i 5 timer, surgjort og fordampet til 20 ml. Løsningen ble avkjølt og filtrert under dannelse av 3,7 g 4-pyridon-
1- eddiksyre med sm.p. 265 - 266° C.
Ved å følge den samme prosedyre men anvende 3-methyl-4-pyridinol, 2,5-dimethyl-4-pyridinol/3-nitro-4-pyridinol i stedet for 4-hydroxypyridin ble det., erholdt 3-methyl-4-pyridon-l-eddiksyre, 2.5- dimethyl-4-pyridon-l-eddiksyre og 3-nitro-4-pyridon-l-eddiksyre.
Eksempel 2
4- kinolon- l- eddiksyre
20 g, 0,2 mol, 4-hydroxykinolintrihydrat ble løst i en
50 %-ig vandig kaliumhydroxydløsning og 20 g, 0,2 mol, kloreddiksyre ble gradvis tilsatt. Den resulterende løsning ble oppvarmet til tilbakeløpstemperaturen i 18 timer, avkjølt, surgjort og filtrert under dannelse av 7 g 4-kinolon-l-eddiksyre med sm.p. 278 - 279° C.
Ved å følge den samme prosedyre men å anvende 3-cyano-2.6- dimethyl-4-pyridinol, 5-klor-2-ethoxy-4-pyridinol eller 3-trifluormethyl-4-pyridinol i stedet for 4-hydroxykinolintrihydrat, ble det erholdt 3-cyano-2,6-dimethyl-4-pyridon-l-eddiksyre, 5-klor-2-ethoxy-4-pyridori-1-eddiksyre og 3-trifluormethy1-4-pyridon-1-eddiksyre.
Eksempel 3
2, 6- dimethyl- 4- pyridon- l- eddiksyre
12,4 g, 0,1 mol, 2,6-dimethy1-4-pyron bis tilsatt til en løsning av 20 g, 0,2 mol, triethylamin og 7,5 g, 9,1 mol, glycin i 100 ml ethanol og 10 ml vann. Blandingen ble oppvarmet til til-bakeløpstemperaturen i 4 dager, ble avkjølt, surgjort og filtrert under dannelse av 4 g 2,6-dimethyl-4-pyridon-l-eddiksyre med sm.p.
på 243o C.
Ved å følge den samme prosedyre men å anvende 3-methoxy-2- methyl-4-pyron, 3-hydroxy-4-pyron og 2,6-dicarbethoxy-4-pyron i stedet for 2,6-dimethyl-4-pyron, ble det erholdt 3-methoxy-2-methyl-4-pyridon-l-eddiksyre, 3-hydroxy-4-pyridon-l-eddiksyre og 2,6-dicarbethoxy-4-pyridon-l-eddiksyre.
Eksempel 4
7- f2-( 4- oxo- l- pyridinyl)- acetylamino)- cefalosporansyre- natriumsalt
3,06 g,- (0,02 mol) av forbindelsen 4-pyridon-l-eddiksyre, ble løst i 50 ml dimethylformamid og løsningen ble avkjølt til 0° C. 3,2 g (0,02 mol) carbonyldiimidazol ble tilsatt og blandingen ble omrørt under nitrogenatmosfære ved 0° C i 30 minutter og ble der-
etter oppvarmet til romtemperatur. Reaksjonskolben ble evakuert i 30 minutter for å fjerne carbondioxydet og ble avkjølt til -20° C. I en separat kolbe ble 7-aminocefalosporansyre silulert ved oppvar-ming av en suspensjon av 5,4 g (0,02 mol) 7-aminocefalosporansyre og 8 ml hexamethyldisilazan i 50 ml kloroform ved tilbakeløpskjø-ling i 30 minutter. Denne løsning ble fordampet til tørrhet for å fjerne dannet ammoniakk. En løsning av residuet i 50 ml kloroform ble avkjølt til -20° C og tilsatt imidazolid. Reaksjonsblandingen ble oppvarmet til 0° C i 1 time, ble deretter oppvarmet til romtemperatur og omrørt over natten.
Løsningen ble behandlet med 2 ml methanol og den utfelte 7-aminocefalosporansyre ble fjernet ved filtrering. En løsning av natrium-2-ethylhexanoat i n-butanol (10 ml av en 2N løsning) ble tilsatt, og blandingen ble fortynnet med ether til et volum på ca. 1 liter for å utfelle produktet. Etter utfelling påny fra methanol med ether, ble det erholdt 2,2 g 7-(2-(4-oxo-l-pyridinyl)-acetyl
amino) -cefalosporansyre som et hvitt fast materiale med smeltepunkt på 180° C (spaltning). Jodtitrering anga en renhet på 72,7 %.
Ved å følge den samme prosedyre men å anvende 6-aminopenicillansyre, 7-aminodesacetylcefalosporansyre, 7-amino-3-((5-methyl-1, 3 , 4-thiadiazol-2-ylthio) -methyl) -decef alosporansyre , 7-amino-3-X(1-methyl-l,2,3,4-tetrazol-5-ylthio)-methyl)-decefalosporansyre og 7-amino-7-methoxycefalosporansyre i stedet for 7-aminocefalosporansyre ble det erholdt natriumsaltet av 6-(2-(4-oxo-l-pyridinyl) -acetylamino) -penicillansyre, 7-(2-(.4-oxo-l-pyridinyl) - acetylamino).—desacetylcef alosporansyre, 7- (2- (4-oxo-l-pyridinyl) - acetylamino)—3-'((5-methyl-l,3,4-thiadiazol-2-ylthio)-methyl)-decefalosporansyre, 7-(2-(4-oxo-l-pyridinyl)-acetylamino)-3-((1-methyl-1,2,3,4-tetrazol-5-ylthio)-methyl)-decefalosporansyre og 7-methoxy-7-(2-(4-oxo-l-pyridinyl)-acetylamino)-cefalosporansyre.
Eksempel 5
6- f2-( 4- oxo- l- kinolinyl)- acetylamino)- penicillansyre, natriumsalt En løsning av 4,1 g (0,02 mol) 4-kinolon-l-eddiksyre i 50 ml dimethylformamid ble plassert under nitrogenatmosfære, ble
avkjølt til 10° C og 3,2 g (0,02 mol) carbonyldiimidazol ble tilsatt i én porsjon. Etter at blandingen var oppvarmet til romtemperatur ble kolben evakuert i løpet av 15 minutter for å fjerne det carbondioxyd som var utviklet ved dannelsen av imidazolidet. Løs-
ningen ble avkjølt til 10° C og en løsning av 4,4 g (0,02 mol) 6-aminopenicillansyre og 5 g (20 % overskudd) triethylamin i --50 ml kloroform ble tilsatt. Reaksjonsblandingen ble omrørt ved 10° C i 1 time, ble oppvarmet til romtemperatur og omrøringen ble fortsatt i ytterligere 3 timer. 10 ml av en 2N løsning av natrium-2-ethylhexanoat i butanol ble tilsatt og produktet ble utfelt ved tilsetning av 700 ml ether. 6-(2-(4-oxo-l-kinolinyl)-acetylamino)-penicillansyre ble filtrert, utfelt på nytt fra methanol med ether og vakuumtørket under dannelse av et utbytte på 4,8 g hvitt fast materiale med smeltepunkt 204° C (spaltning). Jodtitrering indikerte en renhet på 86,4 %.
Ved å følge den samme prosedyre men å anvende 2,5-dimethyl-4-pyridon-l-eddiksyre, 3-nitro-4-pyridon-l-eddiksyre, 3-hydroxy-4-pyridon-l-eddiksyre og 3,5-dijod-4-pyridon-l-eddiksyre i stedet for 4-kinolon-l-eddiksyre, ble det erholdt natriurnsaltet av 6-(2-(2,5-dimethyl-4-oxo-l-pyridinyl)-acetylamino)-penicillansyre, 6- (2- (3-nitro-4-oxo-l-pyridinyl)-acetylamino)-penicillansyre, 6-(2-(3-hydroxy-4-oxo-l-pyridinyl)-acetylamino)-penicillansyre og 6-(2-(3,5-dijod-4-oxo-l-pyridinyl)-acetylamino)-penicillansyre.
Ved å anvende 7-amino-7-methoxycefalosporansyre i stedet for 6-aminopenicillansyre erholdes natriumsaltene av 6-(2-(4-oxo-l-kinolinyl)-acetylamino)-7-methoxycefalosporansyre, 6-(2-(2,5-dimethyl-4-oxo-l-pyridinyl)-acetylamino)-7-methoxy-cef alosporansyre, 6-(2-(3-nitro-4-oxo-l-pyridinyl)-acetylamino)-7-methoxycefalo-spraransyre,

6- (2-(3,5-dijod-4-oxo-l-pyridinyl)-acetylamino)-7-methoxycefalo-sporansyre.
Eksempel 6
7- f 2- ( 2 , 6- dimethyl- 4- oxo- l- pyridinyl) - acetylaminoi[)- cef alosporansyre, natriumsalt'
3,6 h (0,02 mol) 2,6-dimethyl~4-pyridon-1-eddiksyre ble løst i 50 ml dimethylformamid og løsningen ble avkjølt til 0° C. 3,2 g (0,0 mol) carbonyldiimidazol ble tilsatt og blandingen ble omrørt under nitrogenatmosfære ved 0° C i 30 minutter og fikk oppvarmes til romtemperatur. Reaksjonskolben ble evakuert i 30 minut-
ter for å fjerne utviklet carbondioxyd. Den resulterende løsning ble avkjølt til -20° C og 50 ml av en kloroformløsning av etri-methylsilyl-7-aminocefalosporansyre (0,02 mol) ble' fremstilt som beskrevet i eksempel 4, ble tilsatt til denne. Reaksjonsblandingen ble omrørt ved 0° C i 1 time, oppvarmet til romtemperatur og omrørt over natten.
Reaksjonsblandingen ble behandlet med 2 ml methanol og den utfelte 7-aminocefalosporansyre ble fjernet ved filtrering. En 2N-løsning av natrium-2-ethylhexanoat i n-butanol (10 ml) ble tilsatt til reaksjonsblandingen, som ble fortynnet til 1 liter med ether og filtrert. 7-(2-(2,6-dimethyl-4-oxo-l-pyridinyl)-acetylamino )--cef alosporansyre ble utfelt på nytt fra methanol med ether og ble vakuumtørket under dannelse av 5,0 g av et hvitt pulver med smeltepunkt på 2 40° C.
Ved å gjenta den samme prosedyre men å anvende 3-methyl-4- pyridon-l-eddiksyre, 5,6,7,8-tetrahydro-4-kinolon-l-eddiksyre, 5- klor-2-ethoxy-4-pyridon-l-eddiksyre, 3-trifluormethyl-4-pyridon-1-eddiksyre og 3-hydroxy-4-pyridon-l-eddiksyre i stedet for 2,6-dimethyl-4-pyridon-l-eddiksyre fører dette til fremstillingen av natriumsaltene av 7-(2-(3-methyl-4-oxo-l-pyridinyl)-acetylamino}-cefalosporansyre, 7-(2-(5,6,7,8-tetrahydro-4-oxo-l-kinolinyl)-acetylamino}-cefalosporansyre, 7- (2- (6-klor-2-ethoxy-4-oxo-l-pyridinyl) -acetylamino)-cefalosporansyre, 7-(2-(3-trifluormethyl-4-oxo-l-pyridinyl) -acetylamino)-cefalosporansyre og 7-(2-(3-hydroxy-4-oxo-1-pyridinyl) -acetylamino)--cef alosporansyre.
Ved å anvende trimethylsilyl-6-amino-6-methoxypenicillan-syre i stedet for trimethylsilyl-7-aminocefalosporansyre erholdes natriumsaltene av
6- (2-(2,6-dimethyl-4-oxo-l-pyridinyl)-acetylamino)-6-methoxy-penicillansyre,
6-(2-(3-methyl-4-oxo-l-pyridinyl)-acetylamino)-6-methoxypenicillan-syre,
6-(2-(5,6,7,8-tetrahydro-4-oxo-l-kinolinyl)-acetylamino}-6-methoxy-penicillansyre,
6-(2-(5-klor-2-ethoxy-4-oxo-l-pyridinyl)-acetylamino)-6-methoxy-penicillansyre,
6-(2-(3-trifluormethyl-2-oxo-l-pyridinyl)-acetylamino) -6-methoxy-penicillansyre og
6-(2-(3-hydroxy-4-oxo-l-pyridinyl)-acetylamino)-6-methoxy-penicillansyre.
Eksempel 7
7-(2-(4-oxo-l-pyridinyl)-acetylamino)-3-(pyridiniummethyl)-decef alosporansyre
7-(2-(4-oxo-l-pyridinyl)-acetylamino)-cefalosporansyre-natriumsalt ble løst i vann og omsatt med pyridin i nærvær av kaliumthiocyanat ved 60° C i 6 timer. Opparbeidelsen ble utført ifølge J.L. Spencer et al., J. Org, Chem. 32, 500 (1967) og førte til fremstilling av 7-(2-(4-oxo-l-pyridinyl)-acetylamino)-3-(pyridiniummethyl) -decefalosporansyre som zwitterione.
Eksempel 8
7- f2-( 4- oxo- l- pyridinyl)- acetylamino)- desacetylcefalosporansyre
7-(2-(4-oxo-l-pyridinyl)-acetylamino)-cefalosporansyre-natriumsalt ble behandlet med et acetylesterase isolert fra appelsinskall ifølge J. D'A, Jeffery, et al., Biochem. J., 81, 591
(1961) under dannelse av 7-(2-(4-oxo-l-pyridinyl)-acetylamino)-des-acetylcef alosporansyre-natriumsalt .
Eksempel 9
4- trimethylsilylaxypyridin
Til 500 g urent 4-hydroxypyridin ble tilsatt 5,5 liter toluen. Blandingen ble oppvarmet under omrøring og ca. 700 ml toluen ble destillert fra for å fjerne eventuelt tilstedeværende vann. Reaksjonsblandingen ble opprettholdt ved tilbakeløpstempera-turen og 543 g trimethylsilylklorid ble sakte tilsatt. Reaksjonsblandingen ble kokt under tilbakeløp i ca. 3 timer, ble avkjølt og filtrert. Filtratet ble fordampet under redusert trykk for å fjerne toluenet og residuet ble destillert ved 18 - 20 mm under dannelse av et utbytte på 620 g 4-trimethylsilyloxypyridin med et kokepunkt på 95 - 96° C.
Eksempel 10
7- f2-( 4- oxQ- l- pyridinyl)- acetylamino)- cefalosporansyre- natriumsalt
Til 20,0 g 7-(2-bromacetamido)-cefalosporansyre fremstilt ifølge US patentskrift 3 647 789, ble tilsatt 135 ml kloroform og 20 ml N,0-bis-(trimethylsilyl)-acetamid. Blandingen ble omrørt i
ca. 1 time under en atmosfære av tørr nitrogeriVog den resulterende blanding ble omrørt ved romtemperatur i ca. 15 timer under en atmosfære av tørr nitrogengass. 80 ml methylalkohol ble tilsatt til reaksjonsblandingen under omrøring inntil alt bunnfall som umiddel-bart ble dannet var løst. Blandingen ble heldt over i ca. 1 liter
vandig ether under omrøring, løsningsmidlet ble fjernet ved dekan-tering og det klebrige bunnfall som var tilbake ble løst i ytterligere 400 ml methylalkohol. En løsning av 65 ml 2N~natrium-2-ethylhexanoat i n-butanol ble tilsatt til løsningen inneholdende det løste bunnfall, og den resxilterende blanding ble behandlet med benkull og filtrert gjennom et sjikt av diatoméjord. 900 ml isopropylalkohol ble sakte tilsatt til filtratet. Det bunnfall som ble dannet ble fjernet ved filtrering, vasket med ether og tørket i vakuum under dannelse av 17,2 g 7-(2-(4-oxo-l-pyridinyl)-acetylamino)-cefalosporansyre-natriumsalt.
Ved å følge den samme prosedyre og anvende(7- 2-klor-acetamido) -7-methoxycefalosporansyre, 7-(2-klor-2-methylacetamido) - desacetylcefalosporansyre, 7-(2-brom-2-carbethoxyacetamido) -des-acetoxycefalosporansyre, 6-(2-klor-2-methylacetamido) -penicillansyre, 6-(2-bromacetamido)-6-methoxypenicillansyre og 6-(2-brom-2-carbethoxyacetamido)-penicillansyre i stedet for 7-(2-bromacetamido)-cefalosporansyre ble det erholdt 7-(2-(4-oxo-l-pyridinyl)-acetylamino) -7-methoxycefalosporansyre, 7 -(2-(4-oxo-pyridinyl)-2-methyl-acetylamino}-desacetylcefalosporansyre, 7-(2-(4-oxo-l-pyridinyl)-2-carbethoxyacetylamino)-desacetoxycefalosporansyre, 6-(2-(4-oxo-l-pyridinyl) -2-methylacetylamino)-penicillansyre, 6-(2-(4-oxo-l-pyridinyl) -acetylamino) -6-methoxypenicillansyre og 6-(2-(4-oxo-l-pyridinyl) -2-carbethoxyacetylamino) -penicillansyre som deres natriumsalter.
Ved å anvende 2, 6-dimet.hyl-4-trimethylsilyloxypyridin, 2,6-bis-(carbotrimethylsilyloxy)-4-trimethylsilyloxypyridinvble de tilsvarende 2-(2,6-dimethyl-4-oxo-l-pyridinyl), 2-(2,6-dicarboxy-4-oxo-l-pyridinyl) og 2-(3-klor-4-oxo-l-pyridinyl)-derivater av de forskjellige cefalosporan- og penicillansyre erholdt som deres natriumsalter.
Eksempel 11
7- (tx-bromacetylamino) -3- ((5-methyl-l, 3 , 4-thiadiazol-2-ylthio) - methyl)-- decefalosporansyre ' \
Til 1,4 g 7-amino-3-((5-methyl-l,3,4-thiadiazol-2-ylthio)-methyl}-decefalosporansyre, fremstilt som beskrevet i US patentskrift 3 516 997, og inneholdt i en omrørt blanding av 25 ml vann og 25 ml aceton ble tilsatt 3 g natriumbicarbonat. Blandingen ble avkjølt til -10° C og 2,2 g bromacetylbromid i 1 ml aceton ble sakte tilsatt til den omrørte blanding i løpet av 10 minutter. Blandin gen ble omrørt i ytterligere 1 time ved -10° C og fikk oppvarmes til romtemperatur. Reaksjonsblandingen ble ekstrahert med ethylacetat og den organiske fase ble fraskilt. Den vandige fase ble skilt med 100 ml ethylacetat og den vandige fase ble justert til pH 2 med en 40 %-ig løsning av fosforsyre. Den organiske fase ble fraskilt, tørket over natriumsulfat og avfarget med benkull. Løs-ningen ble filtrert, fordampet og residuet ble triturert med ether. Residuet ble tørket i vakuum under dannelse av 0,7 g 7-(a-bromacetylamino)-3-((5-methyl-l,3,4-thiadiazol-2-ylthio)-methyl}-decefalosporansyre.
Ved å følge den samme prosedyre og anvende 7-amino-7-methoxy-3-((5-methyl-l,3,4-thiadiazol-2-ylthio)-methyl)-decefalosporansyre, 7-amino-3-((1-methyl-l,2,3,4-tetrazol-5-ylthio)-methyl)-decefalosporansyre og 7-amino-7-methoxy-3-((1-methyl-l,2,3,4-tetra-zol-5-ylthio)-methyl)—decefalosporansyre i stedet.for 7-amino-3-((5-methyl-l,3,4-thiadiazol-2-ylthio)-methyl)-decefalosporansyre ble det erholdt 7- (a-bromacetylamino)-7-methoxy-3-((5-methyl-1,3 , 4-thiadiazol-2-ylthio) -methyl}-decef alosporansyre, . 7-=- (a-bromacetylamino) -3-((1-methyl-l,2,3,4-tetrazol-5-ylthio)-methyl)-decefalosporansyre og 7-(a-bromacetylamino)-7-methoxy-3-((1-methyl-1,2,3,4-tetrazol-5-ylthio)-methyl)-decefalosporansyre.
Eksempel 12
.7-(2-(4-oxo-l-pyridinyl)-acetylamino}-3-((5-methyl-l,3,4-thia-diazol- 2- ylthio) - methyl)— decefalosporansyre- natriumsalt
Til 4,65 g . 7-(a-bromacetylamino)-3-((5-methyl-l,3,4-thiadiazol-2-ylthio)-methyl)—decefalosporansyre fremstilt som beskrevet i eksempel 11, ble tilsatt 50 ml kloroform og 5 ml N,0— bis-(trimethylsilyl)-acetamid. Blandingen ble omrørt under en nitrogenatmosfære inntil en klar løsning ble erholdt, 1,6 g 4-trimethylsilyloxypyridin ble tilsatt og blandingen ble omrørt i 16 timer under nitrogen. 5 ml methylalkohol ble tilsatt og det faste bunnfall ble oppsamlet ved filtrering. Bunnfallet ble løst i 100 ml methylalkohol, 5 ml av en 2N-natrium-2-ethylhexanoatløsning i n^. butanol ble tilsatt, etterfulgt av 200 ml ether. Det dannede bunnfall ble oppsamlet ved filtrering og tørket under vakuum under dannelse av natriumsaltet av 7-(2-(4-oxo-l-pyridinyl)-acetylamino)-3-((5-methyl-l,3,4-thiadiazol-2-ylthio)-methyl)-decefalosporansyre.
Ved å følge den samme prosedyre, men anvende 7-(a-brom acetylamino) -7-metho:xy~3- ((5-methyl-l,3, 4-thiadiazol-2-ylthio) - - methyl)—decefalosporansyre, 7-(a-bromacetylamino)-3-((1-methyl-1,2,3,4-tetrazol-5-ylthio)-methyl)—decefalosporansyre eller 7-(a-bromacetylamino)-7-methoxy-3-((1-methyl-l,2,3,4-tetrazol-5-ylthio)-methyl)--decefalosporansyre i stedet for 7-(a-bromacetylamino)-3-((5-methyl-l.,.3,4-thiadiazol-2-ylthio) -methyl) -decef alosporansyre, ble natriumsaltene av 7-methoxy-7-(2-(4-oxo-l-pyridinyl) -acetyl^-amino)—3-((5-methyl-l,3,4-thiadiazol-2-ylthio)-methyl)-decefalosporansyre, 7-(2-(4-oxo-l-pyridinyl)-acetylamino)—3-((1-methyl-1,2;3,4-tetrazol-2-ylthio)-methyl)—decefalosporansyre og 7-methoxy-7-(2-(4-oxo-l-pyridinyl)-acetylamino)-3-((1-methyl-l,2,3,4-tetra-zol-2-ylthio)-methyl)—decefalosporansyre erholdt.
Eksempel 13
7-(2-(3-cyano-4-oxo-l-pyridinyl)-acetylamino) -3-((5-methyl-l,3,4-thiadiazol- 2- ylthio)- methyl}- decefalosporansyre- natriumsalt
Til en kloroformløsning av 7-(a-bromacetylamino)-3-((5-methyl-1,3,4-thiadiazol-2-ylthio)-methyl)—decefalosporansyre og N,N-diethylanilin ble tilsatt en ekvivalent av 3-cyano-4-trimethylsilyloxypyridin. Blandingen ble omrørt i ca. 15 timer ved romtemperatur under nitrogenatmosfære. Methylalkohol ble tilsatt etterfulgt av vannfri ether. Det dannede bunnfall ble fjernet ved filtrering, løst i methylalkohol og en ekvivlant mengde av;en løsning av 2-N-natrium-2-ethylhexanoat i butanol ble tilsatt. Et like stort volum med ether ble tilsatt og det dannede bunnfall hie fjernet ved filtrering og tørket under vakuum under'dannelse av 7-(2-(3-cyano-4-oxo-l-pyridinyl)-acetylamino) -3-((5-methyl-l,3,4-thia-diazol-2-ylthio)-methyl)-decefalosporansyre som natrxumsaltet.
Eksempel 14
7-methoxy-7-(2-(4-oxo-l-kinolinyl)-acetylamino)-3-((1-methyl-1, 2, 3, 4- tetrazol- 5- ylthio)- methyl)- decefalosporansyre- natriumsalt
En blanding av 3,5 g natrium-7-methoxy-7-(2-(4-oxo-l-kinolinyl) -acetylamino)—cef alosporan, 2,5 g natriumbicarbonat, 2,6 g l-methyl-5-mercapto-l,2,3,4-tetrazol og 60 ml vann ble oppvarmet til ca. 70° C under nitrogenatmosfære i 4 timer. Reaksjonsblandingen ble avkjølt og fordampet under redusert trykk. Residuet ble triturert med aceton, løst i methanol og filtrert. Isopropylalkohol ble tilsatt til filtratet for å danne et bunnfall som ble oppsamlet ved filtrering og vakuumtørket under dannelse av et bunnfall av 7-mei:hoxy-7- (2- (4-oxo-l-kinolinyl) -acetylamino) -3- ((1-methyl- 1,2, 3,4-tetrazol-5-ylthio) -methyl)~-decef alosporansyre som natriumsalt.
Ved å følge den samme prosedyre men anvende 7-(2-(4-oxo-l-pyridinyl) -acetylamino}-cefalosporansyre eller 7-(2-methyl-2-(2,6-dimethyl-4-oxo-l-pyridinyl)-acetylamino -cefalosporansyre førte til dannelse av 7-(2-(4-oxo-l-pyridinyl)-acetylamino)-3-((1-methyl-1,2,3,4-tetrazol-5-ylthio)-methyl)-decefalosporansyre og 7-(2-methyl-2-(2,6-dimethyl-4-oxo-l-pyridinyl)-acetylamino) -3-((1-methyl-1,2,3,4-tetrazol-5-ylthio)-methyl)-decefalosporansyre som deres natriumsalter.
Eksempel 15
7-(2-(4-oxo-l-pyridinyl)-acetylamino)-3-((5-methyl-l,3,4-thiadia-zol- 2- ylthio)- methyl)- decefalosporansyre- natriumsalt
Til en løsning av 5 g natrium-7-(2-(4-oxo-l-pyridinyl)-• acetylamino)-cefalosporan ig 500 ml vann ble tilsatt 0,95 g natriumbicarbonat og 2;96-g 2-mercapto-5-methyl-l,3,4-thiadiazol. Blandingen ble oppvarmet under nitrogenatmosfære til 70° C i 3 timer og fordampet under redusert trykk. Residuet ble løst i 50 ml methanol og behandlet med et overskudd av acetonitril. Det hvite bunnfall ble fjernet ved filtrering, vasket med acetonitril og tørket under vakuum under dannelse av 4,3 g 7-(2-(4-oxo-l-pyridinyl)-acetylamino)-3-((5-methyl-l,3,4-thiadiazol-2-ylthio)-methyl)-decefalosporansyre som natriumsaltet.
Ved å følge den samme prosedyre men anvende natriumsaltet av 7-methoxy-7-(2-(4-oxo-l-kinolinyl)-acetylamino)-cefalosporansyre, 7-methoxy-7-(2-(4-oxo-l-pyridinyl-acetylamino)-cefalosporansyre eller 7-(2-methyl-2-(2,6-dimethyl-4-oxo-l-pyridinyl)-acetylamino) - cefalosporansyre ble det erholdt 7-methoxy-7-(2-(4-oxo-l-kinolinyl)-acetylamino),— 3- ( (5-methyl-l,3,4-t,hiadiazol-2-ylthio) -methyl) - decefalosporansyre, 7-methoxy-7-(2-(4-oxo-l-pyridinyl)-acetylamino)-3-((5-methyl-l,3,4-thiadiazol-2-ylthio)-methyl)-decefalosporansyre eller 7-(2-methyl-2-(2,6-dimethyl-4-oxo-l-pyridinyl)-acetylamino)-3-((5-methyl-l,3,4-thiadiazol-2-ylthio)-methyl)-decefalosporansyre som deres natriumsalter.
Eksempel 16
givaloyloxyraethyl- 7- f 2- ( 4- oxo- l- pyridinyl) - acetylamino) - cef alosporan
Til 4,5 g natrium-7-(2-(4-oxo-l-pyridinyl)-acetylamino)-cefalosporanat ble løst i 40 ml dimethylformamid og avkjølt til 0°C ble tilsatt 2,6 g pivaloyloxymethyljodid og løsningen ble omrørt i ca. 25 minutter. Blandingen ble fortynnet med .1.70 ml ethylacetat, vasket godt med vann og ble deretter vasket med en fortynnet løs-ning av natriumbicarbonat. Den organiske fase ble tørket over natriumsulfat, filtrert og fordampet til tørrhet under dannelse av pivaloyloxymethyl-7-(2-(4-oxo-l-pyridinyl)-acetylamino)-cefalospo-ranat .
Ved å følge den samme prosedyre men å anvende acetoxy-methyljodid, N-klormethyl-N-methylurethan eller p-acetoxybenzyl-bromid i stedet for pivaloyloxymethyljodid, førte til dannelse av acetoxymethyl-7-(2-(4-oxo-l-pyridinyl)-acetylamino)-cefalosporanat, N-ethoxycarbonyl-N-methylaminomethyl-7-(2-(4-oxo-l-pyridinyl)-acetylamino)--cef alosporanat og p-acetyloxybenzyl-7- (2- (4-oxo-l-pyridinyl) -acetylamino)--cef alosporanat.
Anvendelse av natriumsaltene av 7-methoxy-7-(2-(4-oxo-l-pyridinyl) -acetylamino)-3-((1-methyl-l,2,3,4-tetrazol-5-ylthio)-methyl)--decef alosporansyre, 6-methoxy-6- (2 - (3-cyano-470xo-l-pyridinyl)-acetylamino)-penicillansyre eller 6-(2-(4-oxo-l-kinolinyl)-2- methylacetylamino)-penicillansyre i stedet for 7-(2-(4-oxo-l-pyridinyl) -acetylamino )--cef alosporansyre , førte til dannelse av pivaloyloxymethyl-7-methoxy-7-(2-(4-oxo-l-pyridinyl)-acetylamino) - 3- ((1-methyl-l,2,3,4-tetrazol-5-ylthio)-methyl) -decefalosporanat, pivaloyloxymethyl-6-methoxy-6-(2-(3-cyano-4-oxo-l-pyridinyl)-acetylamino}-penicillanat og pivaloyloxymethyl-6-(2-(4-oxo-l-kinolinyl) -2-methylacetylamino) -penicillanat .
Eksempel 17
N-ethoxycarbonyl-N-methylaminomethyl-7-(2-(4-oxo-l-pyridinyl)-acetylamino}-3-((5-methyl-l,3,4-thiadiazol-2-ylthio)-methyl)-decef alosporanat
Til en oppslemming av 0,01 mol natrium-7-(2-(4-oxo-l-pyridinyl) -acetylamino)-3-((5-methyl-l,3,4-thiadiazol-2-ylthio)-methyl -decefalosporanat inneholdt i 20 ml dimethylformamid og holdt ved en temperatur på 0 - 5° C, ble tilsatt en løsning av 0,01 mol N-klormethyl-N-methylurethan inneholdt i 5 ml dimethylformamid. Blandingen ble omrørt i 1 time og heldt over i vann.
Det dannede bunnfall ble løst i ethylacetat, vasket med vann etterfulgt av en fortynnet løsning av natriumbicarbonat og ble tørket
over magnesiumsulfat. Løsningen ble fordampet under redusert trykk under dannelse av det ønskede N-ethoxycarbonyl-N-methylaminomethyl-7-(2-(4-oxo-l-pyridinyl)-acetylamino)-3-((5-methyl-l,3,4-thiadiazol-
2-ylthi.o). -methyl}-decefalosporanat.
Eksempel 18
Spesifike næringsagarplater ble fullstendig innpodet med forskjellige testorganismer. Filterpapirskiver ble plassert på overflaten av agaren og fuktet med 0,1 ml av.en løsning inneholdende 10, 100 og 1000 mikrogram av testforbindelsen. Inhiberings-soner av mikrobiell vekst ble anvendt for å indikere den antibakterielle aktivitet av testforbindelsen overfor de forskjellige anvendte testorganismer.

1. Fremgangsmåte ved fremstilling av forbindelser av formelen

X er valgt fra gruppen bestående av hydrogen, hydroxy, acetoxy, N-pyridinium, 5-methyl-l,3,4-thiadiazol-2-ylthio og 1-methyl-l,2,3,4-tetrazol-3-ylthio,
R- er valgt fra gruppen bestående av hydrogen, alkanoyloxymethyl, alkanoylaminomethyl, alkoxycarbonylaminomethyl og p-(alkanoyloxy)-benzyl i hvilket alkanoyl- eller alkoxygruppen inneholder fra 1 til 5 carbonatomer,
Rg er hydrogen eller methoxy, og
farmasøytisk akseptable salter derav, karakterisert ved at(a) en 4-oxo-l-pyridinyl-eddiksyre av formelen
hvor R17 R2/ R3, R^ og R5 er som tidligere angitt og et amino-$-lactam av formelen
hvor Rg og Rg er som tidligere angitt, løses i et inert løsnings-middel,
(b) pyridinyleddiksyren og g-lactamet kobles dehydrativt i løsning ved en temperatur på fra -20 til 100° C i løpet av et tidsrom varierende fra 15 minutter til 36 timer i nærvær av et dehydreringsmiddel valgt fra gruppen bestående av dicyclohexylcarbodiimid, N-cyclohexyl-N * -morf olinomethylcarbodiimid, pentarnethylketon-N-cyclohexylimin, N-ethyl-5-fenylisoxazolium-3'-sulfonat og fosfortriklorid, og
(c) det ønskede produkt gjenvinnes fra blandingen.
2. Fremgangsmåte ifølge krav 1, karakterisert ved at Rg er gruppen
3. Fremgangsmåte ifølge krav 2, karakterisert ved at kondensasjonsmidlet er dicyclohexylcarbodiimid.
4. Fremgangsmåte for fremstilling av en forbindelse ifølge krav 1, karakterisert ved at(a) en 4-oxo-l-pyridinyleddiksyre av formelen
hvor R <-> j^ , <R>2 , <R>3 , R4 og R5 har de tidligere angitte betydninger, omsettes med en forbindelse valgt fra gruppen bestående av carbonyldiimidazol, alkylklorformiat hvori alkylgruppen har fra 1 til 5 carbonatomer, thionylklorid, kloracetonitril og bis-p-nitrofenylcarbonat under dannelse av et reaktivt 4-oxo-l-pyridinylacetyl-mellomprodukt,
(b) pyridinylacetyl-mellomproduktet kobles med et amino-g-lactam av formelen
hvor Rg og Rg har de tidligere angitte betydninger, i løsning ved en temperatur fra -20 til 100° C i løpet av fra 15 minutter til 36 timer, og
(c) det ønskede produkt gjenvinnes.
5. Fremgangsmåte ifølge krav 4, karakterisert ved at Rg er gruppen
6. Fremgangsmåte ifølge krav 5, karakterisert ved at 4-oxo-l-pyridinyleddiksyren omsettes med carbonyldiimidazol under dannelse av et 4-oxo-l-pyridinylacetylimidazolid-mellomprodukt.
7. Fremgangsmåte ved---fremstilling av en forbindelse ifølge krav 1, karakterisert ved at(a) et 4-hydroxypyridin av formelen
hvor R-^ , R2/ Rg og R^ har de tidligere angitte betydninger, silu-leres med et siluleringsmiddel valgt fra gruppen bestående av di-lavere alkylklorsilan, trilavere alkylklorsilan, dilavere alkylbromsilan, trilavere alkylbromsilan i hvilket den lavere alkylgruppe inneholder fra 1 til 5 carbonatomer, tetramethyldisilazan, hexamethyldisilazan og bis-trimethylsilylacetamid under dannelse av en 4-hydroxypyridinsilylether,
(b) silyletheren kondenseres med et a-halogenacetamido-3-lactam av formelen
hvor Rg, Rg og Rg har de tidligere angitte betydninger, og hvor Y
. ér valgt fra gruppen bestående av klor, brom og jod, idet silyletheren kondenseres i løsning ved en ..temperatur fra -20 til 100° C i.løpet av fra 15 minutter til 36 timer, og
(c) at det ønskede produkt utvinnes.
8. Fremgangsmåte ifølge krav 7, karakterisert ved at Rg er gruppen
9. Fremgangsmåte ifølge krav 8, karakterisert ved at siluleringsmidlet er et trimethylklorsilan og Y er brom.
10. Fremgangsmåte ved fremstilling av en forbindelse ifølge krav 1 hvorR g er gruppen
og X er gruppen 5-methyl-l,3,4-thiodiazol-2-ylthia, karakterisert ved at(a) et 4-oxo-l-pyridinyl-B-lactam av formelen
hvorR^ ,R2 , R3/ R^/ Rg og Rg er som tidligere angitt, og 2-mer-capto-5-methyl-l,3,4-thiadiazol løses i et vandig løsningsmiddel valgt fra gruppen bestående av vann eller en vandig løsning av aceton, tetrahydrofuran og dimethylformamid,
(b) løsningen omsettes v.ed en temperatur fra 25 til 150° C i løpet av fra 15 minutter til 2 4 timer, og
(c) det ønskede produkt utvinnes.
11. Fremgangsmåte ved fremstilling av en forbindelse ifølge krav 1 hvor Rg er gruppen
og X er gruppen 1-methyl-l,2,3,4-tetrazol-5-ylthio, karakterisert ved at(a) et 4-oxo-l-pyridinyl-B-lactaia av formelen
hvor R^ , R2/ Rg7 R^ , R_ og Rg har de tidligere angitte betydninger og 5-mercapto-l-methyl-l,2,3,4-tetrazol løses i et vandig løsnings-middel valgt fra gruppen bestående av vann eller en vandig løsning av aceton, tetrahydrofuran og dimethylformamid,
(b) løsningen omsettes ved en temperatur av fra 25 til 150°C i løpet av fra 15 minutter til 24 timer, og
(c) det ønskede produkt gjenvinnes.
12. Fremgangsmåte ifølge krav 1 - II ved fremstilling av 7-(2-(4-oxo-l-pyridinyl)-acetylamino}-cefalosporansyre og farmasøy-tisk akseptable salter derav.
13. Fremgangsmåte ifølge krav 1-11 ved fremstilling av 6-(2-(4-oxo-l-pyridinyl)-acetylamino}-penicillansyre og farmasøytisk akseptable salter derav.
14. Fremgangsmåte ifølge krav 1-11 ved fremstilling av 7-(2-(4-oxo-l-pyridinyl)-acetylamino)-3-((5-methyl-l,3,4-thiadiazol-2-ylthio)-methyl)--decef alosporansyre og farmasøytisk akseptable salter derav.
15. Fremgangsmåte ifølge krav 1-11 ved fremstilling av 7-(2- (4-oxo-l-pyridinyl) -acetylamino}-3- ((1-methyl-l, 2 , 3 ,4-tetrazol-~ 5-ylthio)-methyl)—decefalosporansyre og farmasøytisk akseptable salter derav.
