Description
Foreliggende oppfinnelse angår en fremgangsmåte for å danne nye radiofarmasøytiske avbildingsmidler.
Flere Ikke-invasive metoder for avbilding av skjelettstruk-turer og legemsorganer og vev har blitt utviklet i løpet av de siste tiår. Disse metodene er basert på tendens til det spesielle skjelett, organ eller vev å konsentrere visse kjemikalier som kan detekteres, slik som ved anvendelse av seintIfotografi eller bestrålingsdeteksjon. Anvendelsen av radiofarmasøytiske avbildingsmidler i avbilding av skjelett-strukturer, organer og vev, er velkjente innenfor biologiske og medisinske forskningsfelt så vel som diagnostiske evalueringsprosedyrer. Metallbaserte radiofarmasøytiske midler, slik som de som er basert på technetium, har vært funnet å skaffe tilveie særlig nyttige bilder av skjelett-strukturer, og legemsorganer og vev fra hvilke diagnostisk informasjon kan bli oppnådd. Mer spesifikt har radiofarmasøy-tiske midler basert på technetium 99m blitt anvendt med hell som diagnostiske avbildingsmidler.
I tillegg har metallbaserte radiofarmasøytiske midler, slik som de som er basert på rhenium, blitt funnet å være nyttige som terapeutiske midler i behandling av forskjellige sykdommer. Mer spesifikt har radiofarmasøytiske midler basert på rhenium 186 eller rhenium 188 blitt anvendt med hell som terapeutiske midler.
De radiofarmasøytiske midlene innbefatter generelt et metallradlonuklid, forskjellige ligander for binding av radionuklidet til den ønskede skjelettstruktur, organ eller vev, reduksjonsmidler, stabiliserende midler, bærere og avleveringsbærere som er velegnet for injeksjon i, eller ved aspirasjon av pasienten etc.
På grunn av de relativt korte halveringstidene til metall-radionukleidet som ble anvendt i det radiofarmasøytiske midlet, er det ønskelig å skaffe tilveie Ikke-radioaktlve forbindelser av midlet som et sett til hvilken en radionukleidinneholdende oppløsning kan bil tilsatt for å danne midlet. Et reduksjonsmiddel er en nødvendig komponent i mange radiofarmasøytiske sett, reduksjonsmidlet virker ved å redusere den radionukleidinneholdende oppløsning, slik som en pertechnetatoppløsning, for å oppnå det endelige radiofarma-søytiske midlet. Et reduksjonsmiddel må bli inkludert 1 settene for dannelse av technetium-radiofarmasøytiske midler. Tinn(II)-ionet er det mest anvendte reduksjonsmidlet i settene for å danne metallbaserte radiofarmasøytiske midler. Dette innbefatter kjente sett for dannelse av technetium 99m diagnostiske midler for avbilding av hjertet, nyre, lunger og hepatobiliære system, så vel som sett for avbilding og terapeutisk behandling av hjerne og skjelett. Anvendelse av tinn(II)-ion som et reduksjonsmiddel har flere ulemper som generelt har opprinnelse i de iboende problemer som er beslektet til komplisert faststoff og oppløsningskjemi til tinnforbindelser. Særlig er tinn(II) ofte et "ikke-uskyldig" reduksjonsmiddel som interfererer med eller er inkorporert i det endelige radiofarmasøytiske midlet.
Anvendelsen av tinn(II)-ion som en reduktant for fremstilling av radiofarmasøytiske midler er velkjent. Tinn(II)-ionet blir derfor vanligvis inkludert som en komponent i settene for dannelse av radiofarmasøytiske midler, slik som midler for avbilding av hjertet, nyrer, lunger og hepatobiliært system og midler for avbilding og terapeutisk behandling av hjernen og skjelett.
Anvendelse av tlnn(II)-ion som et reduksjonsmiddel i dannelsen av radiofarmasøytiske midler har flere ulemper. Spesielt har tinn(II)-forbindelser iboende ulemper forbundet med komplisert faststoff og oppløsningskjemi. Tinn(II)-ionet virker ofte som en "ikke-uskyldig" reduktant og kan interfere med dannelse av det endelige radiofarmasøytiske midlet, eller bli inkorporert i det endelige radiofarmasøytiske midlet.
Det har nå blitt funnet at det er antall farmasøytisk aksepterbare ikke-tinn-reduksjonsmidler som ikke innehar de samme ulempene ved anvendelse av tinn(II)-ion som reduksjonsmiddel ved dannelse av radiofarmasøytiske midler.
Særlig vil metalliske forbindelser som Cu(I), Cu(II), Co(II), Fe(II), Sn(0), Zr(0), Cr(II) og Zn(0), virke ved at de effektivt reduserer radionuklidinneholdende oppløsninger, slik som pertechnetatoppløsnlnger, for å oppnå det ønskede radiofarmasøytiske midlet.
Flere ikke-metalliske forbindelser, slik som syrer generelt, dltlonitt, formamidin, formamidinsulfinsyre, fosfitt, hypofosfitt, ditiotreitol, saltsyre og borsyre, kan også bli effektivt anvendt for å redusere radionuklidinneholdende oppløsninger.
Det har videre blitt funnet at flere midler slik som fosfiner, sulfhydrylforbindelser, fosfitter, tioler, tioetere, borater, borcyanogrupper, askorbater og gentisater, effektivt reduserer den radionuklidinneholdende oppløsningen og komplekserer med radlonuklidet samtidig. Dette er meget fordelaktig da man reduserer antall komponenter som må bli inkludert i settene for å danne de radiofarmasøytiske midlene, og ved forenkling av kjemien som er nødvendig for å fremstille det radiofarmasøytiske midlet.
Således angår foreliggende oppfinnelse en fremgangsmåte for å danne et radiofarmasøytisk avMldnlngsmiddel, kjennetegnet ved at fremgangsmåten Innbefatter å kombinere en ikke-redusert radionuklidløsning med en multidentatligand og et kjemisk middel, der det kjemiske midlet reduserer radionuklidet og komplekserer med radionuklidet på en måte som inkorporerer det kjemiske midlet inn i det radiofarmasøytiske avbildnlngsmldlet.
Ifølge en utførelsesform av oppfinnelsen omhandler et sett for dannelse av et technetium-myokardialt avbildlngsmiddel tris(3-metoksypropyl)fosfin (TMPP), som både et reduksjonsmiddel og som et komplekseringsmiddel. Ytterligere komponenter, slik som kobberaskorbat, kan også bli Inkludert i settet for å øke det radiofarmasøytiske utbyttet. HypofosfItt og fosfitt er totalt fargeløse, enkle å analysere, lyofiliserbare og inj iserbare.
Det er også blitt funnet at hydrogenfosfitt (HP03<2>~) kan bli anvendt som en antioksidant og reduktant i dannelse av Tc-99m radiofarmasøytiske midler. Reduksjonspotentialet for hydrogenfosfitt kan sammenlignes med askorbinsyre. Videre er oksidasjonsproduktet av hydrogenfosfitt fosfat, PO^-, som er totalt harmløst i Tc-99m radiofarmasøytiske preparater. Fosfitt og fosfat er fargeløse, enkle å analysere, lyofiliserbare og injiserbare.
Det foregående har vært en beskrivelse av visse foretrukne utførelsesformer av foreliggende oppfinnelse, men har ikke til hensikt å begrense oppfinnelsen på noen måte. Mange modifikasjoner, variasjoner og endringer i detaljer kan bli utført innenfor rekkevidden av foreliggende oppfinnelse.
