Abstract
Det er oppdaget at TIR-reseptorer går sammen om å danne funksjonelle smaksreseptorer.Særlig er det oppdaget at koekspresjon av T1R1 og T1R3 resulterer i en smaksreseptor som responderer på umamismaksstimuli, inklusivt mononatriumglutamat. Videre er det oppdaget at koekspresjon av T1R2- og T1R3 reseptorer resulterer i en smaksreseptor som responderer på søtsmaksstimuli,. inklusivt naturlig forekommende og kunstige søtningsmidler. Dessuten beskrives anvendelsen av heterooligomere smaksreseptorer omfattende T1R1/T1R3 og T1R2/T1R3 i tester for å identifisere forbindelser som responderer henholdsvis på umamismaksstimuli og søtsmaksstimuli.Videre beskrives konstitutive cellelinjer som stabilt eller transient koeksprimerer en kombinasjon av T1R1 og T1R3, eller T1R2 og T1R3, under konstitutive eller induserbare betingelser. Anvendelsen av disse cellelinjene i cellebaserte tester for å identifisere umamismaks- og søtsmaksmodulerende forbindelser, beskrives også, særlig massescreeningstester som påviser reseptoraktivitet ved bruk av fiuorometrisk avbildning. Endelig beskrives oppdagelsen av at enkelte forbindelser, feks. lactisol, inhiberer både aktivitetene av humane T1R2/ T1R3- og TlRl/TlR3-reseptorer, og følgelig søt- og umami-smaken, hvilket tyder på at disse reseptorene kan være de eneste søt- og umami-reseptorer.
