Description
Oppfinnelsen angår fremgangsmåte til fremstilling av et modifisert Pseudomonas eksotoksin sammenføyd til en målsøkende bærer, ekspresjonsplasmid og vertscelle benyttet i fremgangsmåten .
Toksiner er meget kraftige celledrepende midler som forårsaker en rekke sykdommer hos mennesker. På grunn av deres høye aktivitet er disse midler blitt festet til monoklonale antistoffer for å danne cytotoksiske midler (immunotoksiner)
som spesifikt binder til målceller. Disse immunotoksiner er derfor meget nyttige i kreftbehandling.
Pseudomonas eksotoksin A (PE) er et meget aktivt monomert protein (molekylvekt 66 kD), som utskilles av Pseudomonas aeruainosa, som hindrer proteinsyntese i eukaryote celler gjennom inaktiveringen av forlengelsesfaktor-2 (EF-2) ved katalyse av dets ADP-ribosylering (katalyse av overføringen av ADP-ribosylmolekyldelen av oksidert NAD på EF-2).
Intoksikasjonsprosessen antas å foregå ved de følgende trinn: Først binder PE til celler gjennom en spesifikk reseptor på celleoverflaten. Deretter blir PE-reseptorkomplekset inter-nalisert inn i cellen. Til slutt blir PE overført til cytosolen hvor det enzymatisk hindrer proteinsyntese. Over-føringsprosessen antas å finne sted fra et surt område, da cellulær intoksikasjon hindres av svake baser såsom NH4+,
som øker pH-verdien i sure vesikler. Ved utsettelse for sure betingelser kommer det hydrofobe domene av PE inn i membranen, hvilket fører til dannelsen av en kanal gjennom hvilken enzymdomenet i forlenget form passerer inn i cytosolen.
US-PS 4 545 985 viser at Pseudomonas toksin kan kobles kjemisk til et antistoff eller vekstfaktor. Disse kjemisk koblede toksiner er imidlertid vist å ha uønskede giftighetsnivåer. Det ville derfor være nyttig å ha toksiner som er sterkt mål-rettede for spesifikke celletyper uten den generaliserte celledreping som normalt er forbundet med disse toksiner. PE-holdige immunotoksiner konstrueres ved at nativt PE først omsettes med iminotiolan. Denne reaksjon tjener både til å introdusere to nye sulfhydrylgrupper som benyttes til kobling av et antistoff til toksinet og til å inaktivere bindingen av PE til dens egen reseptor. Denne fremgangsmåte avhenger av den kjemiske inaktivering av PE-bindingsseter for å bringe til et minimum uønskede bivirkninger som skyldes bindingen av PE til celler med PE-reseptorer. Skjønt denne fremgangsmåte har vært forholdsvis vellykket når det gjelder å produsere en spesiell celledrepende reagens i vevskultur og i tumor-bærende mus, har det ikke vært mulig å gi mer enn 2 \ iq PE-immunotoksin til en 2 0 g's mus eller 1 mg til en 3 kg's ape eller 4 mg til et voksent menneske på grunn av de giftige bivirkninger av immunotoksinet. Det er derfor ønskelig å kunne gi større mengder immunotoksiner for oppnåelse av større dreping av tumorceller. Det er derfor hensikten med foreliggende oppfinnelse å tilveiebringe en fremgangsmåte til å fremstille immunotoksiner som ikke utviser ovennevnte ulemper.
Den foreliggende oppfinnelse oppfyller denne hensikt ved at den skaffer et immunotoksin med høy virkning og lav giftighet. Videre, for å overvinne den ovenfor angitte avhengighet av kjemisk inaktivering av PE-bindingssetene innlemmer den foreliggende oppfinnelse rekombinant-DNA-teknikker for å klone det fullstendige toksingen (eller segmenter av det)
for å uttrykke på høye nivåer den fulle lengde av toksin-molekylet (eller partier av det) som inneholder forskjellige funksjonelle domener, innbefattet et som mangler celle-bindingsdomene. Se eksempel 1 for en sammenligningsunder-søkelse av disse kloner.
Den foreliggende oppfinnelse angår fremgangsmåte til fremstilling av (1) et modifisert Pseudomonas eksotoksin som omfatter minst ADP-ribosyleringsaktivitet og eventuelt translokasjonsaktiviteten er definert som evnen til å translokere gjennom en cellemembran og hvor det modifiserte endotoksin mangler et funksjonelt domen Ia og eventuelt domene II i det native eksotoksin, (2) et målsøkende bærer-toksinkonjugat som består av det nevnte eksotoksin sammenføyd til en målsøkende bærer, særlig immunotoksin som omfatter eksotoksinet sammenføyd med et antistoff, kjennetegnet ved å: (a) transfektere en egnet vertscelle med et ekspresjonsplasmid som inneholder en DNA-sekvens som koder for nevnte eksotoksin eller nevnte målsøkende bærertoksinkonjugat og dyrking av vertscellen under betingelser slik at det modifiserte Pseudomonas eksotoksin eller det målsøkende bærer-toksinkonjugat blir fremstilt; eller (b) å konjugere nevnte modifiserte eksotoksin med målsøkende bærer for å fremstille nevnte målsøkende bærer-toksinkonjugat, særlig immunotoksin.
Den tredimensjonale struktur av PE er blitt bestemt ved røntgenkrystallografi. Som vist på fig. 2, inneholder PE-molekylet tre strukturelt forskjellige domener: Domene I inneholder aminosyrerester 1-252 (Domene Ia) og 365-404 (Domene Ib); domene II inneholder aminosyrerester 253-364
og domene III inneholder aminosyrerester 405-613.
Plasmider er blitt konstruert som uttrykker forskjellige partier av PE-molekylet, hvilket gir evnen til å korrelere forskjellige strukturelle domener med forskjellige funksjonelle aktiviteter og å bestemme (1) hvilke partier av molekylet er ansvarlige for cellegjenkjennelse (binding), (strukturelt domene I, aminosyre 1-252) ; (2) hvilket parti er nødvendig for enzymatisk aktivitet (ADP-ribosyleringsaktivitet, domene III pluss et parti av domene Ib) (aminosyre 385-613); (3) hvilket parti er ansvarlig for translokasjon gjennom cellemembranen (domene II). Beviset for at strukturelt domene Ia er involvert i cellegjenkjennelse er at proteiner fremstilt ved plasmider uten domene Ia, men inneholdende domene II, Ib og III, ikke i seg selv er cytotoksiske og ikke oppviser konkurrerende hindring av cytotoksisitet i intakte toksiner, mens et plasmid som koder for domene Ia, men mangler andre domener, blokkerte PE-cytotoksisitet i følsomme celler. PE fra plasmider med en delesjon av den første halvdel av strukturelt domene II oppviser både PE-blokkerende aktivitet og ADP-ribosyleringsaktivitet, men disse molekyler tapte all celledrepende aktivitet. Plasmider som koder for bare strukturelt domene III, produserer store mengder av proteinet, men mangler påvisbar enzymatisk aktivitet (ADP-ribosylering). Plasmider som koder for hele det strukturelle domene III pluss tilgrensende aminosyrer fra strukturelt domene Ib, uttrykker imidlertid store mengder av proteinet, og deres ADP-ribosyleringsaktivitet er høy.
Basert på den tredimensjonale struktur av PE er plasmider blitt konstruert som uttrykker forskjellige partier av PE. Proteinmønsteret av cellene som uttrykker de forskjellige konstruksjoner, ble analysert ved SDS-gelelektroforese, og ADP-ribosyleringsaktiviteten av de rekombinante toksiner
ble målt. Av disse plasmider (beskrevet i eksempel 1) er plasmid pJH8 som inneholder aminosyrer 253-613 (Domener Ib, II og III) og plasmid pJH17 som inneholder aminosyrer (fra Domene Ib), i stand til å kode store mengder modifisert PE som oppviser lav giftighet for menneskelige celler, men forblir enzymatisk meget aktive.
Sett under ett antyder plasmidmønstrene at strukturelt domene Ia er et reseptorbindende domene, at den første halvdel av strukturelt domene II er nødvendig for translokasjon av toksinet fra en vertscelles endocytiske vesikel til cytosolen, og at strukturelt domene III alene ikke er tilstrek-kelig til å uttrykke full ADP-ribosyleringsaktivitet.
Når det gis til dyr, bevirker PE karakteristisk død på grunn av leversvikt. Immunotoksiner fremstilt med PE angriper også leveren, og når det gis i store mengder, bevirker de død på grunn av leverforgiftning. Eksperimentene vist i tabell III antyder at Domene Ia er ansvarlig for cellebinding og antyder at et PE-molekyl hvor domene Ia er deletert, er mindre giftig for mus enn nativt PE. Dataene vist i eksempel 4 understøtter denne konklusjon, hvilket tyder på at modifisert PE er minst 200-ganger mindre giftig for mus enn nativt PE. Eksempel 5 viser at når proteinet fremstilt ved pJH8 renses og kobles til et antistoff til den humane transferrin-reseptor, blir der dannet et immunotoksin som er tilnærmet like aktivt som et immunotoksin som inneholder nativt PE,
men som er 100-ganger mindre giftig på ikke-målceller (museceller).
En side ved den foreliggende oppfinnelse er derfor fremstilling av PE-molekyler med en delesjon av Domene Ia som er virksomme immunotoksiner med nedsatte bivirkninger innbefattet forminsket levergiftighet.
BESKRIVELSE AV FIGURENE
Fig. 1 viser konstruksjonen av plasmidene ifølge oppfinnelsen, som benyttes til ekspresjon av forskjellige domener av PE. Fig. 2 er et forenklet kart over PE-molekyler med forskjellig størrelse, fremstilt som en del av den foreliggende oppfinnelse. Fig. 3 viser virkningen av PE45~HB21 og PE-HB21 på proteinsyntese i humane KB-celler. PE45er det toksinprotein som mangler Domene I kodet for ved plasmid pJH8. Celler ble inkubert i 24 h med det riktige immunotoksin og deretter

inkubert med H-leucin i 1 h. Innlemmelse av ( H)-leucin i protein ble målt. I panel 3B ble Swiss 3T3-celler inkubert i 24 h med enten nativt Pseudomonas
toksin (PE), nativt Pseudomonas toksin koblet til HB21,
PE„C alene eller PE,,. koblet til HB21. Celler ble utsatt

for disse midler i 24 h og deretter ble proteinsyntesen målt i 1 h som angitt ovenfor.
NÆRMERE BESKRIVELSE AV OPPFINNELSEN
I den foretrukne utførelsesform angår oppfinnelsen fremstil-lingen av en modifisert form av pseudomonas eksotoksin (PE) hvor modifikasjonen fører til et aktivt immunotoksin. Det modifiserte toksin og immunotoksinet fremstilt fra det har sterkt redusert ikke-spesifikk toksisitet overfor menneske-og museceller i vevskultur og med sterkt nedsatt giftighet for mus in vivo.
Genomisk DNA fås ved fullstendig fordøyelse av en stamme av Pseudomonas aeru<g>inosa med EcoRI og Pstl. Skjønt en hvilken som helst stamme av P. aeruginosa er egnet til bruk i oppfinnelsen, er den foretrukne stamme PA103, en stamme som produserer en stor mengde aktivt toksin. DNA-fragmenter i størrelsesorden 2,6-2,9 kb isoleres fra det genomiske DNA-bibliotek og ligeres med et 2,7 kb DNA-fragment fra plasmid pUC13 kuttet med EcoRI og Pstl. Plasmid pUC13 er tilgjengelig i handelen fra Boehringer Mannheim. En egnet vert, fortrinnsvis en E. coli-stamme, blir deretter transformert med de ligerte produkter (den foretrukne stamme av E. coli er HB101, tilgjengelig i handelen fra Bethesda Research Laboratories). Det 2,7 kb DNA-fragment som fås fra ptoxETA, et plasmid som inneholder det PE-strukturelle gen, benyttes som en probe, og plasmid-DNA fra forskjellige positive kolonier ble fremstilt og karakterisert ved Southern blotting. De forskjellige størrelser av rekombinant-PE blir deretter uttrykt i en egnet vert. En vert som bærer det bakteriofage T7 RNA-poly-merasegen i lysogenisk og induserbar form [BL21(DE3)], foretrekkes og er tilgjengelig fra Dr. F. W. Studier ved Brookhaven National Laboratories.
Også foretrukket er plasmider som bærer den bakteriofage sene T7-promotor og AMP<r> og Shine-Dalgarno-boks - pAR2156, også tilgjengelig fra Dr. Studier.
Som vist i eksemplene er de foretrukne plasmider pJH8 og pJH17. pJH8 inneholder et DNA-fragment på 5,1 kb og kan uttrykke Domener II, Ib og III av PE. Dette plasmid frem-stilles ved delvis kapping av plasmid pJH4 med Aval. Det lineariserte DNA-fragment blir deretter fullstendig kappet med Hindlll for oppnåelse av et 5,1 kb DNA-fragment som har Aval- og Hindlll-seter på sine ender. Dette fragment blir deretter inkubert med Sl-nuklease for å fjerne dets kohesive ender, fulgt av ligatisering med T4-ligase for dannelse av plasmid pJH8.
Plasmid pJH17 inneholder et DNA-fragment på 4,8 kb og kan uttrykke Domene III med de tilgrensende 20 aminosyrer av PE. Dette plasmid produseres ved fullstendig kapping av plasmid pJH4 med Hindlll og Apal. Det 4,8 kb DNA-fragment blir deretter isolert og inkubert med Klenow DNA-polymerase I og dNTP for å fylle opp de kohesive ender, fulgt av ligasjon med T4-ligase.
Ekspresjon av rekombinante toksiner i BL21 (DE3)
BL21(DE3) inneholdende plasmider for ekspresjon av forskjellige størrelser av PE dyrkes i LB-næringsvæske med 50 /xg ampicillin/ml ved 37°C. Når absorbans ved 650 nm når 0,3, blir IPTG (isopropyl-beta-D-tiogalatopyranosid) satt til kulturen ved en endelig konsentrasjon på 1 mM. Celler høstes 90 min senere og analyseres for mengden av rekombinante toksiner ved SDS-PAGE, immunoblotting, ADP-ribosylering og celledrepende forsøk.
SDS- PAGE og immunoblottincr
Cellepellets løses opp i Laemmli-buffer. Prøver kokes i
5 min før påføring på en 0,1% SDS, 10% akrylamid-gelplate. For immunoblotting blir prøver etter elektroforese overført til et nitrocellulosepapir, fulgt av reaksjon med antistoff til PE, deretter et nytt antistoff (geit-antikanin) og farging. Antistoffet for PE fås ved hyperimmunisering av kaniner med glutaraldehyd (0,2%) omsatt PE (250 \ iq PE pr. injeksjon). En IgG-fraksjon ble fremstilt for immunoblotting.
Undersøkelse av ADP- ribosyleringsaktivitet
Kjente fremgangsmåter benyttes til undersøkelse av ADP-ribosyleringsaktivitet. Kort fortalt blir retikulocytt-preparater fra kanin eller hvetekim-ekstrakter anriket med forlengelsesfaktor-2 (EF-2) benyttet som en kilde til EF-2. Analyseprøver (500 ul samlet volum) inneholder ca. 10 pmol av EF-2, 37 pmol av <14>C-NAD (0,06 ^Ci), 0,25-1,25 fig av pE og buffer (40 mM
DTT, 1 mM EDTA og 50 mM Tris, pH 8,1). Aktiviteten måles
som pmol av NAD overført til EF-2 i løpet av 3 0 min. En standardkurve for kjente konsentrasjoner av PE opprettes og benyttes til bestemmelse av aktiviteten av PE i ekstrakter fra E. coli. Etter inkubasjon i 30 min ved 37°C blir 0,5 ml 12% TCA satt til hver prøveblanding. Prøveblandingene blir deretter satt i et isbad i 15 min, fulgt av sentrifugering ved 4°C, 3000 x g i 10 min. Pelleten vaskes med 1 ml 6% TCA og sentrifugeres som angitt ovenfor. Pelleten måles deretter for <14>C-radioaktivitet i en væske-scintillasjonsteller som indeksen for ADP-ribosyleringsaktivitet.
Cellecvtotoksisitets- forsok
Forsøk for cytotoksisk aktivitet for det modifiserte PE utføres i NIH 3T3-cellekulturer og humane KB-celler. NIH 3T3-celler eller KB-celler podes 24 h før cytotoksisitets-forsøket i en 24-brønns vevskulturplate ved en tetthet på 2 x 10 celler pr. brønn. Etter inkubasjon i 48 timer med forskjellige konsentrasjoner av PE eller proteinekstrakter isolert fra BL21(DE3) med plasmider som uttrykker forskjellige størrelser av PE, blir monolagene farget med metylenblått for påvisning av de overlevende celler. Resultatene er vist i tabell 1.
Inhibision av proteinsyntese
Målinger av inhibisjonen av proteinsyntese ved PE eller PE med ekstrakter fra B121(DE3)/pJH8 utføres i Swiss 3T3-celle-kulturer. Swiss 3T3-celler podes en dag før undersøkelse i 24-brønns vevskulturplater med en tetthet på IO<5> celler pr. brønn. Cellene vaskes en gang ved at mediet erstattes med DMEM og 0,2% BSA før tilsetning av PE (100<*> ng/ml) eller PE (100 ng/ml) med ekstrakter som inneholdt et overskudd av forskjellige størrelser av PE (tabell 2). Etter 15 min ved 37°C blir mediet så fjernet og erstattet med nytt DMEM og 0,2% BSA. Fire timer senere blir hastigheten for proteinsyntese målt ved tilsetning av [ H]-leucin til mediet (til en endelig konsentrasjon på 2-4 /xCi/ml) i 1 h.
I en foretrukket utførelsesform av oppfinnelsen blir det strukturelle domene til Pseudomonas eksotoksin-genet innsatt i en T7-ekspresjonsvektor nedstrøms for ribosom-bindingssetet og dets ledsagende ATG-initieringskodon (som vist på fig. 1), som beskrevet ovenfor. For å produsere rekombinante proteiner blir celler dyrket ved 37°C til Ag5() = 0,3. IPTG blir deretter tilsatt ved 1 mM for å indusere T7 RNA-polymerase og inkuberes i 2 h. En serie plasmider produseres som vist i eksempel 1. Deretter blir en klon konstruert som koder for et PE-molekyl uten en ledersekvens (pJH4) og i hvilken et metionin anbringes grensende opp til alaninet ved aminoterminusen av den prosesserte form av nativt PE (tabell 1). Det protein som produseres med pJH4, betegnes Met-PE. Store mengder Met-PE produseres ved induksjon med IPTG, hvilket representerer 20% av det samlede celleprotein. ADP-ribosyleringsaktivitet svarende til 0,1 mg nativt PE blir funnet i supernatanten,
og 0,2 mg i pelleten pr. mg samlet celleprotein. PE-molekylet som produseres ved pJH4, avviker fra native PE-molekyler
ved nærværet av én ekstra metioninrest ved amino-terminusen.
pJH8 fremstilt fra pJH4, som angitt ovenfor, uttrykker et protein hvor mesteparten av Domene Ia er slettet (bibeholder kun det tilsatte metionin og tre aminosyrer ved amino-terminusen) . Immunoblotting viser at dette protein er 45 kD, og det uttrykkes ved en konsentrasjon på tilnærmet 0,04 mg/mg celleprotein. Høy ADP-ribosyleringsaktivitet oppvises når urea og DTT ekskluderes fra reaksjonsblandingen. I motsetning til nativt PE (hvor tilsetningen av urea og DTT aktiverer ADP-ribosyleringsaktivitet) gir tilsetningen av urea og DTT til pJH8 redusert ADP-ribosyleringsaktivitet (med ca. 3 0%).
Fordi ADP-ribosyleringsaktiviteten av PE ligger i karboksyl-enden av molekylet, ble plasmid pJH17, konstruert fra pJH4 (som angitt ovenfor), produsert. Dette plasmid inneholder Domene III og 20 aminosyrer fra Domene Ib. Ekstrakter fra dette plasmid inneholder høye nivåer av ADP-ribosyleringsaktivitet - ekvivalent med 0,06 mg PE (tabell 1). Ved immunoblotting ble et 31 kD protein påvist, som foreligger i en konsentrasjon på 0,03 mg/mg celleprotein.
Ingen av de plasmider som produseres ved den ovenfor angitte fremgangsmåte (og som er beskrevet i eksemplene), med unn-tagelse av pJHl, pJH2 og pJH4, oppviser betydelig celledrepende evne, skjønt mange av disse oppviser høy enzymatisk aktivitet (tabell 1). Disse plasmider er vist å hindre inhi-bisjon av proteinsyntese eller celledreping ved nativt PE ved at cellene utsettes i 15 min for nativt PE ved 0,1 ixg/ml i nærvær av 3-5 ixg/ml forskjellige modifiserte toksiner. Dataene i tabell III viser at plasmider som uttrykker enten Domene Ia alene eller Domene I, halvparten av Domene II og Domene III hindrer PE i å inhibere proteinsyntese.
Giftighet for dvr
Mus som mottar nativt PE, bukker under på grunn av lever-ødeleggelse. Den dødelige dose for en 20 gs mus er 0,1-0,2 jug. Dataene i tabell IV viser at PE med en delesjon av Domene
Ia oppviser betydelig forminsket giftighet i mus. Mus ble injisert I.P. med enten nativt PE eller manglende Domene Ia. Alle mus som mottok 1,0 /xg nativt PE og halvparten av
de som mottok 0,2 ug PE, var døde etter 40 h. To av tre mus som mottok 50 /xg av PE Ia, døde, og alle som mottok 20 ug av PE Ia, overlevde, og alle mus som mottok 5 /xg PE Ia, overlevde. Således er PE Ia mer enn 100 ganger mindre giftig på vektbasis enn nativt PE.
Genfusioner ved bruk av rekombinant modifisert Pseudomonas eksotoksin
Genet for modifisert PE ifølge oppfinnelsen som nærmere bestemt inneholdes i pJH8, ble sammensmeltet med DNA-sekvenser som koder for den humane alfa-transformerende vekstfaktor (a-TGF) eller humant interleukin-2 (IL-2). Se eksempel 7 og 8 for detaljer for disse genfusjoner. Modifisert PE er egnet til bruk i fusjoner med et hvilket som helst peptidhormon, vekstfaktor eller annet polypeptid-cellegjenkjennelsesprotein for hvilket en spesifikk reseptor foreligger på celler.
Noen eksempler innbefatter: insulin, glukagon, endorfiner, veksthormon, melanocytt-stimulerende hormon, transferrin, bombesin, lipoprotein med lav tetthet, luteiniserende hormon og asialoglykoproteiner.

Eksempel 1
Som vist i tabell l ble fremgangsmåten ifølge oppfinnelsen brukt til å konstruere en rekke plasmider inneholdende fusjoner av de forskjellige størrelser av PE-strukturelt gen og den T7 sene promotor. pJH2 inneholder det intakte PE-strukturelle gen med et segment som koder for en modifisert ledersekvens foran. pJH4 inneholder det intakte PE-strukturelle gen med tilføyelsen av et metioninkodon ved amino-terminusen. pJH7 inneholder det gen som koder for strukturelt Domene III av PE (aminosyre 405-613). pJH8 inneholder det
gen som koder for strukturelt Domene II, Ib og III av PE (aminosyre 253-613). pJH13 koder for PE med en utslettelse
av den første halvpart av strukturelt domene II som omfatter aminosyrer 253-307. pJH14 koder for strukturelt domene Ia av PE (aminosyrer 1-252). pJH17 koder for strukturelt domene III med en ytterligere tilgrensende sekvens på 2 0 aminosyrer fra Domene Ib (aminosyrer 385-613) .
pJHl ble konstruert ved delvis kapping av det native PE
(pE 0) med BamHI, og den lineære form av DNA ble eluert og fullstendig kappet med EcoRI. Det 2,0 kb DNA-fragment som ble oppnådd fra pE 0 som har BamHI og EcoRI ved de to ender, ble deretter innsatt i pAR 2156 som var blitt fullstendig kappet med BamHI og EcoRI.
pJH2 ble konstruert ved delvis kapping av pJHl med BamHI.
Den lineære form av DNA ble isolert og fullstendig kappet
med Ndel. Det 610 kb DNA-fragment ble spart, for å konstruere pJH2 ved at det ble ligert med syntetisk oligonukleotid-dupleks.
pJH4 ble konstruert ved delvis kapping av pJH2 med Taql. Det lineariserte DNA (610 kb) ble isolert og fullstendig
kappet med Ndel. Det storste DNA-fragment (5,9 kb) ble sepa-rert og ligert med syntetisk oligonukleotid-dupleks.
pJH7 ble konstruert ved delvis kapping av pJH4 med Aatll. Det lineariserte DNA-fragment (5,9 kb) ble deretter fullstendig kappet med Hindlll. Det 4,7 kb DNA-fragment som har Aatll- og Hindlll-seter ved sine ender etter separasjon,
ble inkubert med Sl-nuklease for å fjerne de kohesive ender, fulgt av ligasjon med T4-ligase.
pJH8 ble konstruert som beskrevet i den nærmere beskrivelse.
pJH13 ble konstruert ved delvis kapping av pJH4 med Aval. Det lineariserte DNA-fragment ble deretter fullstendig kappet med EcoRI. Det 4,7 kb DNA som har Aval og EcoRI kappet ved begge ender, ble inkubert med Sl for å fjerne kohesive ender (DNA-fragment 1). pJH4 ble delvis kappet med Sali. Det lineariserte DNA-fragment ble deretter fullstendig kappet med EcoRI. Det 1,0 kb DNA-fragment som har Sali- og EcoRI-seter ved endene, ble inkubert med Klenow DNA-polymerase I og dNTP for å fylle de kohesive ender (DNA-fragment 2). DNA-fragment 1 (4,7 kb) og DNA-fragment 2 (1,0 kb) ble ligert ved 4°C over natten.
pJH14 ble konstruert ved delvis kapping av pJH4 med Aval. Det lineariserte DNA-fragment ble deretter fullstendig kappet med EcoRI. 4,7 kb DNA som har Aval- og EcoRI-seter ved sine ender, ble inkubert med Sl for å fjerne den kohesive ende, fulgt av ligasjon med T4-ligase.
pJH17 ble konstruert som beskrevet i den nærmere beskrivelse.
Eksempel 2
Mengden og aktiviteten av de rekombinante toksiner (produsert ved plasmidene beskrevet i eksempel 1) ble målt ved SDS-PAGE, ADP-ribosylering og celledrepende evne. Resultatene
er vist i tabell 1.
Eksempel 3
Eksperimentene ble utført for å undersøke egenskapene av
den rekombinante giftighet ifølge oppfinnelsen. Resultatene er vist i tabell 2.
Eksempel 4
Virkningen av forskjellige delesjoner på celleproteinsyntese ble bestemt, i nærvær og fravær av nativt PE. Resultatene er vist i tabell 3. Strukturelt Domene Ia (pJH14) og strukturelt Domene I, halvparten av II og III (pJH13) ble ekstra-hert fra pelleten av de lydbehandlede celler med 8M urea.
10 /il av hvert ekstrakt svarende til 3-5 /xg rekombinante proteiner ble brukt i hver undersøkelse. Strukturelt II, Ib og III (pJH8) forelå i supernatanten av de sonikerte BL21(DE3)/pJH8-celler. 10 /il av ekstrakt svarende til 2 /xg rekombinant toksin ble benyttet. Cellene ble behandlet som angitt i tabell 3 i 15 min med og uten nativt PE ved 0,1 /xg/ml fulgt av 1 ml DMEM-vasking, inkubert i 4 h i DMEM og 0,2%
BSA og deretter inkubert med <3>H-leucin i 1 h.
Eksempel 5
Som vist i tabell 4 ble dosen som forårsaker død hos mus bestemt ved injisering av Balb/c-mus I.P. med forskjellige mengder av PE eller PE Ia inneholdt i 1,0 ml sterilt saline og 10 mg/ml sterilt humanalbumin. Dyrene ble overvåket daglig i to uker. Alle dødsfall fant sted ved 48 h.
Eksempel 6
Som vist på fig. 3 dreper et immunotoksin bestående av det
45 kD-protein fremstilt ved pJH8 konjugert ved et disulfid bundet til et antistoff til den humane transferrin-reseptor (PE45~HB2l) humane celler som uttrykker transferrin-reseptoren (ID5Q 3 ng/ml). Det har liten eller ingen virkning på museceller som ikke uttrykker den humane transferrin-reseptor ved 1000 ng/ml, mens nativt PE konjugert ved et sulfid bundet til HB21 dreper ikke-spesifikt museceller ved en ID50 på 29 ng/ml. Dataene på fig. 3 viser at den ikke-spesifikke giftighet av PE45~HB21 er 100 ganger mindre enn PE-HB21.
Molekylvekten av PE45 ble senere bestemt på nytt. Molekylvekten ble da funnet å være 40 000.
Eksempel 7
Konstruksjon av alfa- transformerende vekstfaktor- PE- fusionsgen
PJH8 ble behandlet med Tth 1111 og SphI for å konstruere et mindre plasmid pVC8 som har færre Avall-seter. pVC8 ble delvis kappet med Avall og ligatisert til et syntetisk oligonukleotid (30 bp) som inneholder et STuI-sete, et Tth 1111-sete og et stoppkodon for å danne pVC31. pVC31 ble kappet med Tth 1111 og fylt opp med et Klenow-fragment, et fragment av DNA-polymerase og ligatisert til en klon med butt ende som inneholdt alfa-TGF-genet (pVC 33). Alfa-TGF-DNA, p-hTGF-10-925 [Derynck et al. Cell. 38:287-297 (1984)] ble kappet med EcoRI og Bgll for å gi et 322 bp-fragment som ble isolert og kappet med Fnu 4HI og behandlet med T4-polymerase for å gi et 152 bp-fragment som i sin tur ble ligatisert til pVC31 for å danne PE-alfa-TGF-fusjonsgenet. Når det uttrykkes i E. coli B121, produserer dette plasmid et protein som reagerer med antistoffer for alfa-TGF og for PE og har en molekylvekt på 51 000.
Eksempel 8
Konstruksjon av IL- 2- PE- fus~ ionsqen
pVC8 ble kappet med Avall ved posisjon 1190, 3,6 kb-fragmentet ble isolert, dets enkelttrådede ender fylt med Klenow-enzym og defosforylert for å produsere fragment I. Klon PST-5 [Gallo et al., PNAS. 81:2543-2547 (1984)] ble kappet med Pstl for å gi et 1 kD-fragment av IL-2 som deretter ble kappet med Bsp 1286 ved posisjon 105 og 669 og behandlet med T4-polymerase for å fylle opp endene. 564 bp-fragmentet ble ligert til fragment I for å danne pHL-1 og transfor-
mert inn i BL21-ekspresjonsceller. Ved induksjon ble et 60 kD-protein produsert som reagerer med antistoffer for IL-2 og for PE og inneholder ADP-ribosyleringsaktivitet.
Deponeringserklæring
De følgende plasmider er blitt deponert i the American Type Culture Collection i Rockville, Maryland under de respektive ATCC-nummer før innlevering av søknaden, og ved utstedelse av søknaden som patent vil de opprettholdes i en periode av tretti (30) år fra deponeringsdatoen eller fem (5) år etter den siste forespørsel etter slike deponeringer eller for patentets effektive levetid, hva som måtte være lengst. Deponeringene skal erstattes dersom kulturene muterer eller blir ikke-levedyktige i løpet av deponeringsperioden.
Claims (19)
1. Fremgangsmåte til fremstilling av (1) et modifisert Pseudomonas eksotoksin som omfatter minst ADP-ribosyleringsaktivitet og eventuelt translokasjonsaktivitet, hvor translokasjonsaktiviteten er definert som evnen til å translokere gjennom en cellemembran og hvor det modifiserte endotoksin mangler et funksjonelt domene Ia og eventuelt domene II i det native eksotoksin, (2) et målsøkende bærertoksinkonjugat som består av det nevnte eksotoksin sammenføyd til en målsøkende bærer, særlig immuntoksin som omfatter eksotoksinet sammenføyd med et antistoff, karakterisert ved å: (a) transfektere en egnet vertscelle med et ekspresjonsplasmid som inneholder en DNA-sekvens som koder for nevnte eksotoksin eller nevnte målsøkende bærertoksinkonjugat og dyrking av vertscellen under betingelser slik at det modifiserte Pseudomonas eksotoksin eller det målsøkende bærertoksinkonjugat blir fremstilt; eller (b) å konjugere nevnte modifiserte eksotoksin med målsøkende bærer for å fremstille nevnte målsøkende bærer-toksinkonjugat, særlig immunotoksin.
2. Fremgangsmåte som angitt i krav 1, karakterisert ved at det modifiserte Pseudomonas eksotoksin mangler minst én del av domene II i det native eksotoksin.

karakterisert ved at det omfatter en DNA-sekvens som koder for ethvert av proteinene som angitt i krav 1-4, og som er i stand til å uttrykke nevnte DNA-sekvens.
