Abstract
Basalmembranproteiner isoleres fra menneskelig eller dyrisk vev i vandig oppløsning i nærvær av en chelat-danner som funksjonsegnede proteiner i relativt store mengder. Ved hjelp av disse proteiner kan det fremstilles høyspesifike antistoffer som anvendes til immunologisk bestemmelse av disse proteiner.
Description
Foreliggende oppfinnelse angår en fremgangsmåte for isolering av basalmembranproteiner fra human- eller animalvev.
Oppfinnelsen angår også anvendelsen av de oppnådde proteiner.
Laminin, nidogen og BM40 er glykoproteiner, som tilskrives en nøkkelrolle i organisasjonen av basalmembraner og ved vekselvirkning av celler med basalmembraner [Paulsson, M. et al. "Eur. J. Biochem." 161, 455-464 (1986), R. Timpl. M. Dziadek, "Int. Rev. Exp. Pathol." 29, 1-112 (1986)]. Således understøtter laminin utvekst av neuritter fra neuronene av perifere nerver [Edgar D. et al., "EMBO" J. 3, 1463-1467
(1984)] og deres regenerasjon in situ [Madison et al. "Exp. Neurol." 88, 767-772 (1985)].
En bred anvendelse av disse proteiner hindres hittil frem for alt ved at det ikke står noen egnede fremgangsmåter til disposisjon, hvormed disse proteiner kan isoleres på skånende måte fra humant vev. Den av Risteli og Timpl ["Biochem. J." 193, 749-755 (1981)] omtalte fremgangsmåte behandler isoler-ingen av det proteaseresistente lamininfragment Pl fra humanplasenta. Ifølge von Ohno et al. ["Biochem. Biophys. Res. Com." 112, 1091-1098 (1983)] og Wewer et al. ["J. Biol. Chem." 258, 12654-12660 (1983)] omtalte fremgangsmåte krever man enten denaturerte betingelser eller proteolyse for å ekstrahere signifikante mengder av laminin eller laminin-bruddstykker fra vevet. Også nidogen eller BM40 kan hittil bare isoleres under denaturerende betingelser [Dziadek et al., "Eur. J. Biochem." 161, 455-464 (1985), Paulsson et al., "Eur. J. Biochem." 156, 467-478 (1986)].
Med fremgangsmåten ifølge oppfinnelsen er nå mulig å over-vinne disse ulemper og på skånende måte å isolere biologisk aktivt material. Overraskende kan man fra vandig oppløsning i nærvær av en gelatdanner ekstrahere intakte basalmembranproteiner og også i større mengder enn tidligere mulig.
Foreliggende oppfinnels angår således en fremgangsmåte for isolering av basalmembranproteiner fra human- eller animalvev, og denne fremgangsmåte karakteriseres ved at
a) det gjennomføres en forekstrasjon av det basalmembran-holdige vev med en buffer som bibeholder den naturlige
konformasjon til basalmembranproteiner samt proteaseinhibitorer; og b) det gjennomføres en ekstrahering, under tilsetning av 1 til 500 mM chelatdannende polysyrer, av basalmembranproteinene fra human- eller animalvev ved hjelp av en buffer som bibeholder den naturlige konformasjon av basalmembranproteiner samt proteaseinhibitorer og chelatdannere.
Oppfinnelsen angår også anvendelsen av de ved denne fremgangsmåte oppnådde basalmembranproteiner til fremstilling av høyspesifikke antisera eller antistoffer og dermed til bestemmelse av basalmembranproteiner i kroppsvæsker og vevekstrakter.
I det følgende forklares oppfinnelsen detaljert, spesielt i de fortrukkede utførelsesformer. Videre defineres oppfinnelsen i patentkravene.
Ved hjelp av fremgangsmåten ifølge oppfinnelsen kan alle humane eller animalsk vevtyper, hvori det befinner seg basalmembranproteiner, for eksempel humanplasenta, humant eller animalsk tumorvev eller nyrene, ekstraheres. Det kan i første rekke gjennomføres en forekstrahering. Vev må induseres i en for basalmembranproteiner fysiologisk buffer i nærvær av proteaseinhibitorer og sentrifugeres. Under behandling fjernes oppløslige proteiner fortrinnsvis lett, for eksempel serumproteiner og cytoplasmatiske proteiner av vevet. Vevresten opptas så igjen i en fysiologisk buffer og homogeniseres og ekstraheres i nærvær av proteaseinhibitorer og chelatdannende stoffer. Egnede chelatdannere er organiske polysyrer som for eksempel EDTA, EGTA og citrat. Ekstraher-ingen gjennomføres under tilsetning av 1-500 mmol av chelatdanneren, fortrinnsvis 10-50 mmol, hensiktsmessig under avkjøling ved 0-15°C, fortrinnsvis 4-10°C. Ved høyere temperatur er en ekstrahering også ennå mulig, den følges imidlertid da av forstyrrende avbygningsreaksjoner. Ekstraktene inneholder de ønskede basalmembranproteiner. Spesielt foretrukket isoleres ifølge oppfinnelsen BM40, nidogen og laminin, delvis som ikke-kovalent kompleks.
De enkelte proteiner ekstraheres fra ekstraktet ved moleky-lar siktkromatograf i på egnede søyler som dekstran, spesielt allyldekstran, kryssbundet med N,N'-metylenbisacrylamid eller agarose. Finrensningen av basalmembranproteinet foregår, alt efter proteinet, på fortrinnsvis svak basiske ionebyttere med efterfølgende molekylsiktkromatografi, respektivt ytterligere rensning på agarose- eller sepharose-søyler.
På denne måte kan det isoleres mengder av intakte basalmembranproteiner, som ikke oppnås med vanlig isoleringsfrem-gangsmåter.
De oppdelte rensede basalmembranproteiner kan da anvendes som antigen til fremstilling av høyspesifik antisera eller som antistoffer. Dette foregår på vanlig måte ved subkutan eller intramuskulær injeksjon på forsøksdyr, fortrinnsvis på kaniner. Hensiktsmessig arbeides det derved i nærvær av komplett Freund's adjuvans. Det kan anvendes de i disse tilfeller vanlige antigendoser. Den foretrukkede dose på kaniner er 0,5 til 1 mg pr. dyr. Det dannede antiserum fremstilles derefter på den for fagfolk kjente måte og kan anvendes som sådan. Det er også mulig, på forhånd å rense de i serumet tilstedeværende antistoffer. En foretrukket metode hertil er affinitetskromatografi.
Det med hjelp av de rensede basalmembranproteiner fremstilte høyspesifike antiserum muliggjør bestemmelsen av basalmembranproteiner i kroppsvæske, som for eksempel serum, urin etcetra og vevekstrakter under anvendelse av de i og for seg kjente immunologiske påvisningsmetoder.
Det kan ved disse bestemmelser anvendes såvel de kjente radio-immun-analyse-(RIA)-varianter som også enzym-immun-analyse-varianter og analoge fremgangsmåter.
Den for den immunologiske bestemmelse nødvendige markering av antigenet kan foregå på for proteinmarkering kjente metoder. Det blir fortrinnsvis radioaktiv-, enzym- eller fluorescens-markert. I tilfelle den radioaktive markering anvendes som radionuklid fortrinnsvis jod 125. Markeringen kan deretter foregå etter den kjente kloramin T-metoden ("Int. Arch. Allergy" 429, 185, 1966).
Ved disse påvisningsreaksjoner konkurrerer det markerte basalmembranprotein på kjent måte om antistoffet, således at i det dannede antigen-antistoffkompleks er mengden av markert antigen desto mindre, jo mere markerte antigener som er inneholdt i prøven som skal bestemmes. Det kan enten anvendes markering av komplekser, for eksempel radioaktivi-teten eller enzymaktiviteten eller markering av det overstå-ende efter adskillelse av antigen-antistoffkomplekset, for ved hjelp av en vurderingskurve, som ble oppstilt ved hjelp av prøver er kjent innhold av basalmembranprotein, å fastslå den inneholdte mengde antigen i prøven som skal undersøkes. Fortrinnsvis bestemmes markering i kompleks.
En foretrukket utførelsesform av metoden ifølge oppfinnelsen består i, å gjennomføre separeringen med det spesifike antiserum dannede antigen-antistoff-komplekset ved hjelp av et annet antistoff. Fortrinnsvis anvendes dertil et mot Immunglobulin G av den for fremstilling av det spesifikke antiserum anvendte dyretyper rettet antistoff. Separeringen av antigen-antistoff-komplekset fra oppløsningen kan foregå ved vanlige metoder som felling, sentrifugering eller frafiltrering. Alternativt bindes antiserumet respektivt det andre antistoff på en fast bærer. Efter separering av antigen-antistoff-komplekset bestemmes da den i komplekset inneholdte respektivt den i oppløsning gjenblivende radioak-tivitet eller annen markering.
Enzym-immun-analyse-variantene til bestemmelse av basalmembranprotein kan også gjennomføres således at antistoffet mot immunglobulin G av den angjeldende dyretype, for eksempel av kaniner, er enzymmarkert. Ved denne type av enzym-immun-analyse bestemmes den efter dannelse av antigen-antistoff-komplekset gjenblivende del av anti-basalmembranproteinseru-met, idet man binder disse til bærerbundne basalmembranprotein og derefter bringer det hele til reaksjon med enzymmark-erte antistoffer mot immunglobulin G av kaniner. Mengden av bundet enzymmarkert antistoff bestemmes derefter ved måling av enzymreaksjonen og er indirekte proporsjonalt den ukjente mengde av basalmembranproteinet i prøven.
Oppfinnelsen skal forklares nærmere ved hjelp av noen eksempler.
Eksempel 1
Isolering av basalmembranproteinene laminin, nidogen og BM40.
EHS-tumorceller injiseres subkutant på C57Bl-mus [Orkin et al., "J. Exp. Med." 145, 204 (1977 )]. Efter 10-14 dager høstes tumoren og det derefter rensede vevmaterial lagres ved
-20° C. 75 g av den frosne tumor ble homogenisert i 1
minutter i 1,5 liter 0,15 M NaCl, 0,05 M tris-HCl (pH 7,4)
(TBS) med 0,05 mM f enylmetansulf onylf luorid (PMSF) og N-etylmaleimid (NEM). Efter sentrifugering ved 8 000 omdrei-ninger pr. minutt i et tidsrom på 20 minutter plasseres supernatanten og resten homogeniseres igjen i 375 ml av den ovennevnte buffer under tilsetning av 10 mM EDTA og omrøres i 1 time i kjølerom. Ekstraktet som ble utvunnet efter sentrifugeringen, ble enten anvendt videre med en gang eller lagret ved -20°C. Utbytte og renhet av det ønskede produkt kan økes ytterligere hvis ekstraheringstrinnene blir gj entatt.
En alikvot mengde av TBS-EDTA-ekstraktet has over en agarose-(Biogel A5m) eller en sepharose Cl-6B-søyle (3 x 135 cm), som er ekvilibrert i TBS med 2 mM EDTA, 0,5 mM PMSF og 0,5 mM NEM. Derved kom det til oppspaltning i 3 til 4 fraksjone-ringsområder. Det først med utlukkelsesvolumene eluerte material inneholder laminin-nidogen-kompleks og fritt laminin, i følgende fraksjoner inneholdt fritt nidogen i intakt (MG 150 KD) og delvis nedbygget form (MG 130, 100 KD). Deretter eluerer BM 40.
Rensingen av laminin foregikk i første rekke på en svakt basisk ionebytter (DEAE cellulose) med 2 M urinstoff i 0,05 M Tris/HCl (pH 8,6) under tilsetning av proteaseinhibitorer. Det elueres ved hjelp av en lineær NaCl-gradient (0 til
0,04 M). Ved dette trinn fjernes forstyrrende forurensninger fra proteoglykan og nukleinsyrer. Laminin befris til slutt ved kromatografi på sepharose C1-4B i 2 M guanidinklorid (pH 7,4) for resterende nidogen og mindre proteiner.
Rensningen av nidogen foregår i første rekke på DEAE-cellulose og sepharose CI4-B som beskrevet ved laminin. Ytterligere rensningstrinn, hvis nødvendig, omfatter kromatografi på CM-cellulose med 2 M urinstoff 0,02 M Na-acetat (pE 4,8) og kryssbundet allyldekstran (sepharcryl S200) med 0,2 M NH4HCO3.
Den ytterligere rensing av BM 40 foregår likeledes på DEAE-cellulose med 2 M urinstoff 0,05 M tris (pH 8,6) ved eluering ved hjelp av en lineær NaCl-gradient (0-0,4 M NaCl). BM 40 eluerer ved 0,2-0,25 M NaCl og renses endelig efter konsen-trering over sepharcryl S200 med 0,2 M NH4HCO3.
Tabellen viser for eksempel, at med den skånende ekstrahering ekstraheres det mengder av intakt basalmembranproteiner, for eksempel EHS-tumor, som ikke kan oppnås ved de vanlige fremgangsmåter.
Eksempel 2
Fremstilling av det markerte antigen.
25 >jg av det ifølge eksempel 1 isolerte basalmembranprotein markeres med 0,5 mCi jod 125 efter kloramin T-metoden og ubundet jod fjernes ved dialyse eller gelfiltrering på en polyacrylamidgel (for eksempel "Biogel P2", Pharmacia Fine Chemicals Inc. ).
Eksempel 3
Gjennomføring av den immunologiske bestemmelse.
Alle de for den immunologiske bestemmelse nødvendige trinn gjennomføres i nærvær av 0,04$ av et ikke-ionisk detergens, for eksempel "Tween" 20, et polyetoksylert sorbitmonolaurat. Bindingskurver bestemmes med 1 ng markert, ifølge eksempel 1 isolerte basalmembranprotein. Konsentrasjonen av basalmembranprotein i en ukjent prøve av serum eller andre kroppsvæsker bestemmes i følgende inhibisjonsprøver.
En bestemt mengde av det spesifikke antistoff eller antiserum forinkuberes med den ukjente prøve i 16 timer ved 4°C og efter tilsetning av 1 ng markert antigen inkuberes det hele ytterligere i 8 timer ved 4°C. Derefter tilsettes et overskudd av antistoff mot kanin-immunoglobulin G, og efter ytterligere 16 timer ved 4°C, separeres det i immunkomplekset bundne antigen ved sentrifugering. Inhibisjonsaktiviteten av den ukjente prøve sammenlignes med aktiviteten av en standardkonsentrasjon av ikke-markert antigen.
Eksempel 4
Fremstilling av antiserumet.
Kaniner immuniseres med 0,5 til 1 mg fra det ifølge eksempel 1 isolerte basalmembranprotein pr. dyr, idet antigenoppløs-ningen, blandet med et likt volum komplett Freund's Adjuvans, injiseres subkutant eller intramuskulært. 4 uker efter den første injeksjon injiseres en tilsvarende mengde antigenopp-løsning blandet med komplett Freund's Adjuvans, subkutant eller intramuskulært. 4 til 8 uker efter den andre injeksjon tas blod og fra dette fås antiserum efter koagulering.
Claims (4)
1.
Fremgangsmåte til isolering av basalmembranproteiner fra human- eller animalvev,
karakterisert ved at a) det gjennomføres en forekstrasjon av det basalmembran-holdige vev med en buffer som bibeholder den naturlige konformasjon til basalmembranproteiner samt proteaseinhibitorer; og b) det gjennomføres en ekstrahering, under tilsetning av 1 til 500 mM chelatdannende polysyrer, av basalmembranproteinene fra human- eller animalvev ved hjelp av en buffer som bibeholder den naturlige konformasjon av basalmembranproteiner samt proteaseinhibitorer og chelatdannere.
