Description
Foreliggende oppfinnelse vedrører fremstillingen av forbindelser med anti-arrytmivirkning av formel:
og farmasøytisk akseptable syreaddisjonssalter derav, hvor X står for en halogen-, laverealkyl-, hydrogen-, trifluormetyl-, fenyl- eller laverealkoksysubstituent, Y står for -CN, -CONH2, C0N(R1)2 eller C02Ri hvor Ri er en laverealkyl; m er 1 eller 2 og n er et helt tall fra og med 1 til og med 3; R2 står for laverealkyl; R3 står for laverealkyl eller laverealkanoyl, benzoyl eller trifluoracetyl; A, B og D er karbonyl eller metylen, slik at når en av A, B eller D er karbonyl, er de andre metylen og R3 er laverealkyl, mens når R3 er laverealkanoyl, benzoyl eller trifluoracetyl, er A, B og D metylen.
Forbindelser med formelen

er beskrevet i U.S. patent nr. 4.107.205. Forbindelsene fremstilt ifølge foreliggende oppfinnelse adskiller seg fra forbindelsene ifølge tidligere kjent teknikk ved fraværet av et andre basisk senter (tertiært amin) i molekylet.
Forbindelsene som fremstilles ifølge oppfinnelsen adskiller seg fra forbindelsene ifølge tidligere kjent teknikk ved fraværet av et andre basisk senter (tertiært amin) i molekylet og denne endringen i strukturen resulterer i forbindelser med en mere fordelaktig hemodynamisk profil.
En foretrukket utførelse av oppfinnelsen omfatter fremstilling av en forbindelse som har en karbonyl ved A, og B og D er metylen og R3 er laverealkyl og Y er C02Ri, CN eller C0N(Ri)2. Nok en foretrukket utførelse av oppfinnelsen har en karbonyl ved B, og A og D er metylen og R3 er laverealkyl og Y er C02Ri, CN eller C0N(R1)2. Nok en foretrukket utførelse av oppfinnelsen har en karbonylgruppe ved D, A og B er metylen og R3 er laverealkyl og Y er C02Ri, CN eller CON-(<R>l)2.
En annen foretrukket utførelse omfatter fremstilling av en forbindelse som har metylen ved A, B og D og Y er C0NH2, CN, C02Ri eller C0N(Ri )2 hvor Rj er laverealkyl og R3 er laverealkanoyl, benzoyl eller trifluoracetyl.
Oppfinnelsen vedrører fremgangsmåter for fremstilling av forbindelsene med formel I som innbefatter at forbindelser med formelen:

Omsetning av nitrilet (II) med l-(kloracetyl)piperidin i nærvær av natriumhydrid gir det tilsvarende amidet (III)
(reaksjonsskjerna I). NItrilfunksjonaliteten kan erstattes av en ester- eller en dimetylamidfunksjonalitet ved å benytte esteren eller dimetylamldet analogt (II) som en reaktant. Mellomproduktene (II) og (V) i reaksjonsskjerna II er beskrevet i J. Med. Chem. 23, 1102-1108 (1980).
Omsetning av nitrilet (IV) med N-benzyl,N-isopropylamino-etylklorid i nærvær av natriumamid i toluen ved 85°C i 1 time gir det tilsvarende nitrilet (V). Hydrering av nitrilet (V) med svovelsyre etterfulgt av katalytisk hydrogenering gir det tilsvarende amidet (VI). Acylering av (VI) med acetylklorid eller andre acylklorider som f.eks. trifluoracetyl, aroyl eller fenacetyl gir det ønskede acylerte amidet (VII) i reaksjonsskjema II.
Behandling av nitrilet (IV) med natriumhydrid i N,N-dimetyl-formamid etterfulgt av 1-klor-N,N-bis-(1-metyletyl)acetamid gir nitrilamidet VIII som vist i reaksjonsskjerna III.
Omsetning av nitrilet II med litiumdiisopropylamid i THF etterfulgt av behandling med tris (isopropoksy) titanklorid Heiv. Chim. Acta., 64» 357 (1981) og etterfølgende konden-sasjon med N-(2-kloretyl)pyrrolidin-2-on gir nitrilpyrrolidon IX som vist i reaksjonsskjerna IV.
Fremstillingen av forbindelsene med formel I kan sammenfattes ved de følgende reaksjonsskjemaene. I forbindelser av formel I refererer betegnelsen laverealkyl til rette eller forgrenede alkylkjeder som inneholder 1 til 6 karbonatomer, som f.eks. metyl, etyl, propyl, isopropyl, butyl, pentyl, heksyl og lignende. Betegnelsen laverealkoksy refererer til alkoksygrupper som inneholder 1 til 6 karbonatomer fra rette eller forgrenede laverealkyler som beskrevet ovenfor. Følgelig er metoksy, etoksy, propoksy, butoksy og lignende egnede laverealkoksygrupper. Halogen refererer til halogener f.eks. klor, brom, fluor og jod.
Plassering av substituenten X relativt til tilknytnings-punktet for fenylen er ikke kritisk. Følgelig er orto, meta eller para monosubstituerte fenyler hvor X står for en halogen-, trifluormetyl-, fenyl-, laverealkyl- eller laverealkoksysubstituent innbefattet. En foretrukket utførelse er når X er klor, en 2-klor-substituert fenyl.
Farmasøytisk akseptable syreaddisjonssalter avledes fra syrer som f.eks. saltsyre, hydrojodsyre, hydrobromsyre, fosforsyre, svovelsyre, toluensulfonsyre, eddiksyre, maleinsyre, benzo-syre og lignende.
Forbindelser med formel I er nyttige på grunn av sin evne til å inhibere ventrikulær arrytmi som det fremgår av følgende forsøk.
Ventrikulær arrytmi induseres ved en 2-trinns underbinding av den foranliggende nedadgående grenen av den venstre krans-arterien i hver av to eller flere ubedøvede hunder. Forsøks-forbindelsen administreres intravenøst ved doser på 5 mpk til en maksimaldose på 20 mpk. En forbindelse vurderes som aktiv dersom den gir en reduksjon på minst 25$ i ektopiske slag I løpet av et tidsrom på minst 10 minutter i halvparten eller flere av hundene. Den gjennomsnittlige minimale effektive dosen bestemmes for hver aktiv forbindelse.
Anti-arrytmi midler som er effektive hos mennesker, som f.eks. kinidin, prokainamid og "Norpace" er aktive i dette forsøket.
De forbindelsene som er funnet aktive ved å gi en 75-1005É reduksjon i ventrikulær arrytmi, eller toksiske ved forsøket ovenfor ved 5 mpk undersøkes ved administrering av en 1 mpk dose med fem minutters intervaller. En forbindelse vurderes som aktiv dersom den gir en reduksjon på minst 25# i ektopiske slag i løpet av et tidsrom på minst 10 minutter i halvparten eller flere av de undersøkte hundene.
Tabell I illustrerer aktiviteten av foretrukne forbindelser med formel I.
Forbindelsene fremstilt ifølge foreliggende oppfinnelse adskiller seg fra forbindelsene ifølge kjent teknikk ved det faktum at de har en mer fordelaktig hemodynamisk profil. Mens forbindelsene med formel I undertrykker ventrikulær arrytmi i ubedøvede hunder i samme doseområde som de forbindelsene Ifølge tidligere kjent teknikk som er beskrevet i U.S. patent nr. 4.107.205, har de redusert myokardisk undertrykkings-aktivitet som vist i tabell II. Denne reduksjonen av negativ ionotrop virkning er spesielt gunstig i det disrytmiske hjerte hvor undertrykkelse av kontraktiviteten kan være spesielt livstruende.
Forbindelsene fremstilt ifølge oppfinnelsen formuleres i konvensjonelle doseringsformer som f.eks. tabletter, kapsler, og injiserbare oppløsninger.
De følgende eksemplene skal illustrere oppfinnelsen. Referanseeksemplene beskriver fremstillingen av mellom-produkter og eksemplene beskriver fremstillingen av slutt-produkter .
Referanseeksempel 1
Fremstilling av metyl a-2-bis(1-metyletyl)-amino)etyl-2-klorbenzenacetat
Til en omrørt oppløsning av 100 g a-2-bis(1-metyletyl)-aminoetyl-2-klorfenylacetonitril I 200 ml metanol tilsettes det 100 ml konsentrert svovelsyre. Reaksjonsblandingen oppvarmes under tilbakestrømning i 6 timer. Etter avkjøling til romtemperatur helles oppløsningen i isvann og denne oppløsningen nøytraliseres omhyggelig med 50% natrium-hydroksydoppløsning. Den basiske oppløsningen ekstraheres med 2 x 300 ml porsjoner av eter og ekstraktene vaskes med vann og tørkes over vannfritt magnesiumsulfat. Oppløsningsmidlet fjernes ved inndampning under redusert trykk slik at man får råproduktet som destilleres ved 142-148°C (0,3 mm) slik at man får 85,1 g metyl a-2-bis-(1-metyletyl)aminoetyl-2-klorbenzenacetat som har formelen

Referanseeksempel 2
Fremstilling av N,N-dimetyl-a-2-bis(1-metyletyl)-amino-etvlfenvlacetamid
Til 6,0 g kaliumhydrid (35* i olje) i 20 ml nydestillert THF under argonatmosfære tilsettes det 4,0 g N,N-dimetylfenyl-acetamid i 20 ml THF i løpet av 33 minutter ved hjelp av en motordrevet sprøyte, og blandingen omrøres ved romtemperatur i 10 minutter. 4,32 g N,N-diIsopropylamino-etylklorid i 6 ml THF tilsettes i løpet av et tidsrom på 33 minutter, og blandingen omrøres ved romtemperatur i 6 timer. Reaksjonsblandingen helles så på isvann og ekstraheres med 3 50 ml's porsjoner av eter. Eterekstraktene vaskes med vann, etterfulgt av mettet natriumkloridoppløsning og tørkes over magnesiumsulfat. Oppløsningsmidlet fjernes ved inndampning under redusert trykk, slik at man får råproduktet, som tas opp i IN saltsyreoppløsning. Den sure oppløsningen vaskes med pentah før den gjøres basisk med 50* natriumhydroksyd-oppløsning og den vandige oppløsningen ekstraheres med 3 50 ml's porsjoner av eter. Eterekstraktene tørkes over magnesiumsulfat og oppløsningsmidlet fjernes under redusert trykk slik at man får 3,77 g N,N-dimetyl-a-2-bis( 1-metyl-etyl )-aminoetyl-fenylacetamid som har formelen

Fremstilling av a-2-(1-metyletyl)aminoetyl-a-(2-klorfenyl)-1- piperidinbutanamid
Syntesen av det sekundære aminet (IV, Reaksjonsskjema II) ble oppnådd ved den katalyttlske debenzyleringen av det tilsvarende tertiære benzylaminet. Tyve gram a-2-l-metyletyl-(fenylmetyl )amino-a-(2-klorfenyl )-l-plperldinbutanamld ble hydrogenert i 200 ml etanol som inneholdt 7,54 ml konsentrert saltsyre over 2,0 g Pd/C ved 34,5 kPa ved romtemperatur i 3 timer. Katalysatoren ble fjernet ved filtrering under vakuum. Oppløsningsmidlet ble fjernet ved inndampning under redusert trykk slik at man fikk et råsalt som ble tatt opp 1 vann. Den sure oppløsningen ble vasket med pentan før den ble gjort basisk med 50* natriumhydroksydoppløsning, og den vandige oppløsningen ble ekstrahert med 3 200 ml's porsjoner av metylenklorid. De organiske ekstraktene ble tørket over natriumsulfat og oppløsningsmidlet ble fjernet under redusert trykk slik at man fikk råproduktet som ved triturering med en blanding av eter og heksan ga 14,8 g a-2-( 1-metyletyl )-aminoetyl-a-(2-klorfenyl)-l-plperidinbutamid som har formelen

Eksempel 1
Fremstilling av l-5-bis(1-metyletyl)amino-3-(2-klorfenyl)-3- cvano- l- oksopentvlpiperidin
2,0 g natriumhydrid (50* i olje) vasket med heksan for å fjerne oljen, tilsettes til en oppløsning av 11,2 g a-2-bis(1-metyletyl)aminoetyl-2-klorfenylacetonitril i 75 ml N,N-dimetylformamid. Blandingen omrøres ved romtemperatur under nitrogenatmosfære i 45 minutter, og deretter tilsettes 6,4 g l-(kloracetyl)piperidin til blandingen. Reaksjonsblandingen omrøres ved romtemperatur i 1 time, helles i vann og ekstraheres med eter. Eterekstraktet vaskes med vann, tørkes over magnesiumsulfat. 37,5 ml 1,6 M n-butyllitium tilsettes til 8,4 ml diiso-propylamin i 200 ml nydestlllert THF ved 0°C under en nltrogenatmosfære. Blandingen omrøres ved 0°C i 30 minutter, avkjøles til -50"C, og 18,6 g av materialet fremstilt i referanseeksempel 1 i 100 ml THF tilsettes. Reaksjonsblandingen omrøres ved -50°C i 30 minutter før 8,5 g l-(klor-acetyl)-piperidin tilsettes ved -50°C og oppvarmes deretter til romtemperatur hvor den så holdes i 2 timer. Blandingen helles i vann og ekstraheres med 3 200 ml's porsjoner av eter og ekstraktene vaskes med vann, tørkes over vannfritt magnesiumsulfat og deretter fjernes oppløsningsmidlet under redusert trykk. Det resulterende råproduktet krystalliseres fra heksan slik at man får metyl-a-2-bis-(1-metyletyl)-aminoetyl-a-(2-klorfenylJ-^-okso-l-piperidinbutanoat, som har formelen

Reaksjonsb land ingen omrøres ved 0°C i 1 time, får deretter oppvarmes til romtemperatur og helles i vann. Lagene separeres og det organiske ekstraktet vaskes med vann og tørkes over magnesiumsulfat. En oppløsning av a-2-bis(1-metyletyl)aminoetyl-2-klor-acetonitril i THF behandles med en ekvivalent nyfremstilt LDJ ved -78"C. Etter 10 minutter tilsettes en ekvivalent av 2B tris (isopropoksy)titanklorid i heksan og reaksjonsblandingei får oppvarmes til romtemperatur. Fjernelse av oppløsningsmidlel i vakuum gir a-(2-klorfenyl)-a-2-bis(1-metyletyl)amino-etyl-2-okso-l-pyrrolidinbutannitril som har formelen:

1.
Analogifremgangsmåte for fremstilling av terapeutisk aktiv forbindelse med formelen
og farmasøytisk akseptable syreaddisjonssalter derav, hvor X står for en halogen-, laverealkyl-, hydrogen-, trifluormetyl-, fenyl- eller laverealkoksysubstituent, Y står for —CN, -CONH2, C0N(R_)2 eller C02R_ hvor R_ er en laverealkyl;
m er 1 eller 2 og n er et helt tall fra og med 1 til og med 3; R2 står for laveralkyl; R3 står for laverealkyl eller laverealkanoyl, benzoyl eller trifluoracetyl; A, B og D er karbonyl eller metylen, slik at når en av A, B eller D er karbonyl, er de andre metylen og R3 er laverealkyl, mens når R3 er laverealkanoyl, benzoyl trifluoracetyl, er A, B og D metylen, karakterisert ved at den innbefatter at
1) forbindelser med formelen:
hvor X står for en halogen-, laverealkyl-, hydrogen^, trifluormetyl-, fenyl- eller laverealkoksysubstituent; Y står for —CN, -C0NH2, CON(R_)2 eller C02R_ hvor R_ er en laverealkyl; m er heltallet 1 eller 2 og n er et helt tall fra og med 1 til og med 3; R2 står for laverealkyl; A, B og D er metylen, omsettes med a) konsentrert svovelsyre, eller b) hydrogeneres katalytisk vedhjelp av Pd/C hvorved det
oppnås forbindelser med formelen
som deretter acyleres med acylklorider
hvor R er laverealkyl, fenyl eller trifluormetyl; eller 2) for fremstilling av en forbindelse med formel I hvor R3 er laverealkyl, A er karbonyl, B er metylen og m er 2 og resten av substituentene er som definert ovenfor,
omsetning av en forbindelse med formelen
hvor R3 er laverealkyl og de øvrige substituentene er som definert ovenfor, med en forbindelse av formelen hvor n er som definert ovenfor,
og eventuelt, overføres en oppnådd forbindelse med formel I til et farmasøytisk akseptabelt salt derav.

Analogifremgangsmåte ifølge krav 1 for fremstilling av a-2-acetyl (1-metyletyl )aminoetyl-a-(2-klorfenyl )-l-piperidin-butanamid og de farmasøytisk akseptable syreaddisjonssaltene derav, karakterisert ved at tilsvarende utgangsmaterialer anvendes.
