Description
Deri foreliggende oppfinnelse angår nye alkyleringsreagenser som er nyttige som startmaterialer i organisk syntese.
Beskrivelse av tidligere kjent teknikk
Flerfunksjonene forbindelser har stor verdi som startmaterialer i organisk syntese. De fleste enkle forbindelser med lav molekylvekt i denne kategori er velkjent i litteraturen, således er en rekke mulige isomerer av alkoholer med molekylformelen C4HsBr20 allerede blitt beskrevet (S.A. Pogorshelski, Chem. Zentralbl., 1, (1905), p. 668).
Der er imidlertid et behov for nye lavmolekylære flerfunksjonene forbindelser som enten kan lette syntesen av kjente forbindelser ved å skaffe nye syntetiske veier, eller gjøre det mulig å fremstille nye strukturer.
Sammendrag av oppfinnelsen
En hensikt med den foreliggende oppfinnelse er å skaffe nye propanolderivater som er nyttige som startmaterialer i organisk syntese.
En annen hensikt med oppfinnelsen er å skaffe en fremgangsmåte til fremstilling av propanolderivatene ifølge oppfinnelsen.
Oppfinnelsen angår propanolderivater med formel I
hvor X og Y er avgangsgrupper valgt hver for seg fra halogen, p-toluensulfonyloksy, metansulfonyloksy, Ci-g-alkylkarbonyloksy, fenylkarbonyloksy eller fenylkarbonyloksy hvor fenyldelen er substituert med elektrontiltrekkende grupper, og Rj_ er H eller en beskyttende gruppe valgt fra Ci_g-alkylkarbonyl, fenylkarbonyl, fenylkarbonyl hvor fenyldelen er substituert med elektrontiltrekkende grupper, silyl eller tetrahydropyranyl beskyttende grupper.
I den foreliggende sammenheng betyr uttrykket "avgangsgruppe"
("leaving group")' for X eller Y hvilke som helst av de grupper som brukes i faget og som lett spaltes av når karbonatomet, til hvilket de er festet, underkastes nukleofilt angrep. Typiske eksempler på avgangsgrupper er halogener, såsom klor, brom og jod, særlig brem; p-toluensulfonyloksy, metansylfonyloksy, esterfunksjo-ner såsom C-\ .g-alkylkarbonyloksy, f.eks. metylkarbonyloksy, etylkarbonyloksy,, propylkarbonyloksy etc, og arylesterfunksjo-ner, såsom fenylkarbonyloksy, idet fenylgruppene valgtfritt kan være substituert med elektrontiltrekkende grupper, såsom nitro eller fluoro.
Uttrykket "beskyttende gruppe" for Ri angir en hvilken som helst gruppe som på den ene side kan hindre oksygenatomet, til hvilket det er festet, r å ta del i substitusjons- eller eliminasjons-reaksjonene, og på den annen side lett kan fjernes. Eksempler på slike beskyttende grupper er enoletre (dannet med ketoner som har ct-protoner); acyl såsom C-j _8~karbonyl, f.eks. acetyl, propionyl, buturyl etc., og arylkarbonyl såsom fenylkarbonyl hvor fenylgruppene kan valgfritt være substituert med elektrontiltrekkende grupper såsom nitro eller fluoro, silylgrupper såsom timetylsilyl, tetrahydropyranyl eller en karbohydrat-gruppe.
Uttrykket "Ci _8-a<rkyl" slik det brukes' ovenfor i forbindélse med esterfunksjoner„ kan angi slike grupper som metyl, etyl, propyl, i-propyl, n-butyl',, i-butyl, tert.butyl, pentyl, heksyl og oktyl.
Når X og Y er forskjellige, kan forbindelsene ifølge formel I
opptre i etantiomeriske former. Avhengig av den nøyaktige egenskap av gruppene X, Y og Rj kan disse grupper også innføre stereoisomerisme- eller diastereoisomerisme i forbindelsene ifølge formel I. Det skal forstås at formel I omfatter alle slike stereoisomece og diastereomere former.
I forbindelsene med formel I foretrekkes det at X og Y er identiske, særlig?valgt fra avgangsgruppene beskrevet ovenfor, særlig halogener, såsom klor, brom eller jod, idet brom er særlig foretrukket.
Det foretrekkes av substituenten R^ er H, da slike forbindelser kan virke som nukleofile materialer.
Eksempler på foretrukne forbindelser er: 3-brom-2-brommetylpropan-l-ol
3-brom-2-brommetylprop-l-yl-acetat
3-brom-2-brommetylprop-l-yl-tetrahydropyranyleter.
En særlig foretrukket forbindelse er 3-brom-2-brommetyl-propan-l-ol.
Forbindelsene ifølge formel I har gode egenskaper som flerfunksjonene reagenser. Således, dersom R^ er H, kan forbindelsene ifølge formel I fungere som gode nukleofile materialer under alkaliske eller sure betingelser og på samme tid bære potensielle seter for nukleofile angrep. Forbindelsene kan omsettes med en rekke forskjellge nukleofile reagenser avhengig av egenskapene av avgangsgruppene X og Y. I denne sammenheng er merkaptoioner særlig interessante nukleofile reagenser.
En særlig interessant bruk av forbindelsene ifølge formel I angår fremstillingen av syntetiske karbohydratreseptorer, såsom de som er beskrevet i søkerens samtidig innleverte søknad med tittelen "Glycosidic Derivatives". I forbindelsene er gruppe Ri i formel I ovenfor en reseptoraktiv karbohydratmolekyldel, og gruppen X og, valgfritt, Y er erstattet med forskjellige funksjonelle grupper, såsom lipider, bærere etc. Synteser av denne type er eksemplifisert i eksempel 4 og 5.
Den reseptoraktive karbohydratmolekyldel kan innføres i forbindelsene ifølge formel I ved at en forbindelse ifølge formel I hvor R-| er H, omsettes med et derivat av det passende reseptoraktive karbohydrat som har en avgangsgruppe ved den reduserende ende av karbohydratet. På denne måte blir den reseptoraktive karbohydratmolekyldel innført isteden for gruppen R-] i formel I ovenfor. Etter iranføringen av den reseptoraktive karbohydratmolekyldel kan forskjellige grupper, såsom lipidgrupper, bærere etc. innføres istedenfor gruppen X i formel I ovenfor gjennom reaksjon med passende nukleofile derivater av de grupper som skal innføres. så'ledes kan tioler omsettes med forbindelser inneholdende karbohydratmolekyldelen, hvilket tillater dannelsen av bis-sulfider. Sulfidene kan valgfritt være oksidert til sul.foksider eller sulfoner. Forbindelsene ifølge formel I som inneholder den reseptoraktive karbohydratmolekyldel, kan også omsettes med aminer eller alkoholer som nukleofile midler for dannelse av aminer eller etere.
Oppfinnelsen angår også en fremgangsmåte til fremstilling av propanolderivater ifølge formel I som angitt ovenfor.
En fremgangsmåte a) for fremstilling av forbindelser ifølge formel I omfatter å redusere den tilsvarende syre ifølge formel II.
fulgt av valgfri beskyttelse av hydroksygruppen som dannes.
Reduksjonen kan utføres ved en rekke forskjellige reduksjonsmid-ler, såsom NaBH:4 i vann eller protiske eller aprotiske, polare eller ikke-polare organiske oppløsningsmidler, såsom metanol, etanol, isopropanol, diglyme, benzen, toluen, eter, tetrahydrofuran, eller 1.,,2:-dimetyoksyetan, eller ved hjelp av reduksjons-midler såsom dib.oran eller LiAlH4 i aprotiske, polare eller ikke-polare organiske oppløsningsmidler såsom benzen, toluen, eter, tetrahydroÆuran eller 1,2-dimetoksyetan. Reduksjonen kan også utføres vedi reduksjon av det tilsvarende syreklorid eller en ester ved behandling av et alkalimetall såsom kalium, litium eller natrium i flytende ammoniakk, ved behandling med et hydrid som angitt ovenfor, eller ved behandling med et reduserende kompleks såsom Red-Al (Na-bis(2-metoksyetoksy)AI2), idet oppløsningsmiddelet er et av de tidligere nevnte oppløsningsmid-ler. Syrekloridet eller esteren kan også underkastes en første reduksjon til aldehydet Ved bruk av hydrogen katalysert ved Pd/BaSC>4, fulgt av en andre reduksjon til alkoholen, ved bruk av NaBH4. Reaksjonen kan utføres ved temperaturer i området fra -78°c til +150°c, normalt fra 0°C til 50°C, f.eks. værelsestemperatur, i en periode på 0,1-48 timer, normalt 8-24 timer, f.eks. 16 timer. I en ytterligere fremgangsmåte b) blir propanolderivater ifølge formel I, hvor X og Y er halogen, fremstilt ved omsetting av en diol med formel III
hvor R-| er som angitt ovenfor med et halogeneringsmiddel. Halogeneringsmiddelet som anvendes kan være et hvilket som helst av de midler som vanligvis anvendes i faget, f.eks. tionyl-klorid, fosfortribromid, eller fosforpentabromid, i f.eks. pyridin eller trifenylfosfin i CCI4 eller trifenylfosfin i CBr4. Reaksjonen kan utføres ved temperaturer i området fra -78°C til +200°C, normalt fra 0°C til 100°C, f.eks. ved tilbakeløpstempe-raturen av det oppløsningsmiddel som anvendes i 0,1-100 timer, normalt 8-24 timer, f.eks. 16 timer. Fremstillingen av iodider kan utføres ved omsetting av kloridene eller bromidene som fås ovenfor, eller f.eks. tosylatet eller metansulfonåtet med natriumiodid i aceton under tilbakeløpsstrømning.
Jiolen ifølge formel III kan fremstilles ved at 2-hydroksymetyl-1,3-propandiol beskyttes med en acetalgruppe. En slik acetalgruppe kan være en benzylidenacetalgruppe eller en acetalgruppe som fås fra et keton, såsom cykloheksanon. En benzylidenacetalgruppe kan opprettes ved omsetting av triolen med benzaldehyd og syre, eller med a,a-dimetoksytoluen og syre. Den acetalbeskyt-tede triol beskyttes deretter med R-| , hvoretter acetalfunksj onen fjernes ved omsetting med en syre, såsom saltsyre. Forbindelsene ifølge formel I hvor X og Y er p-toluensulfonyloksy, metansulfonyloksy, C^-g-alkylkarbonyloksy, eller fenylkarbonyloksy kan fremstilles ved en fremgangsmåte c) som omfatter å omsette diolen ifølge formel III som angitt ovenfor med p-toluensulfonylklorid, metansulfonylklorid, et Ci-g-alkyl-karbonylklorid, eller et fenylkarbonylklorid i et polært oppløsningsmiddel, såsom pyridin, ved temperaturer i området fra -78°C til +150°C, normalt fra 0°C til 100°C, såsom tilbakeløps-temperaturen av det oppløsningsmiddel som anvendes, i en periode på 0,1-24 timer, normalt 8-24 timer, f.eks. 16 timer.
Oppfinnelsen er videre illustrert ved de følgende ikke-begren-sende eksempler.
EKSEMPEL 1
3-Brom-2-brommetylpropan-1-ol (DIBol)
3-Brom-2-brommetylpropansyre (15,3 g, 62 mmol) (se A.F. Ferris, J. Org, Chem., 20 (1955) p. 780) ble oppløst i tørt diklormetan (400 ml) og avkjølt (0°C). Reaksjonsblandingen ble holdt under nitrogen. En oppløsning av diboran i tetrahydrofuran (190 ml,
190 mmol); 1 M oppløsning av BH3 i THF) ble dråpevis tilsatt med omrøring. Etter 1 time ble kjølebadet fjernet, og blandingen ble etterlatt over natten ved værelsestemperatur. Saltsyre (210 ml; 1 M) ble tilsatt, den organiske fase ble separert, og den vandige fase ble ekstrahert med diklormetan (3 x 50 ml). De kombinerte organiske faser ble tørket (Na2S04) og konsentrert. Hurtigkromatografi (flash chromatography) av residuet ga rent DIBol (13,8 g; 96%). Kp. ca. 45°C (0,1 mm Hg); n<?<3> 1 ,5439;

EKSEMPEL 2
3-Brom-2-brommetylpropan-1-yl-acetat
3-Brom-2-brommetylpropan-1-ol (512 mg, 2,21 mmol), pyridin (10 ml) og eddiksyreanhydrid (10 ml) ble omrørt ved værelsestemperatur i 17 timer. Oppløsningsmidlene ble fjernet (ko-fordampning med toluen), etylacetat (20 ml) ble tilsatt, og oppløsningen ble vasket med vann (2 x 10 ml). Den vandige fase ble ekstrahert med etylacetat (10 ml), og de kombinerte organiske faser ble tørket (Na2S04) og konsentrert. Residuet ble filtrert gjennom silika med heptan/etylacetat 4:1 for å gi tittelacetatet (483 ml, 81%) som hadde IR: -ymaks 1752, 1230, 1050 cm-<1>.

EKSEMPEL 3
3-Brom-2-brommetylprop-1-yl-tetrahydropyranyleter
3-Brom-2-brommetylpropan-1-ol (1,0 g, 4,3 mmol) og dihydropyran (1,81 g, 21,6 mmol) ble oppløst i tørt diklormetan (20 ml) og avkjølt (0°C). Toluen-p-sulfonsyre (10 mg) ble oppløst i diklormetan (2 ml) og satt til blandingen. Etter 4 timer ble kjølebadet fjernet, og reaksjonsblandingen ble etterlatt ved værelsestemperatur i 5,5 timer. Blandingen ble avkjølt (0°C), og en ytterligere <p>orsjon toluen-p-sulfonsyreoppløsning ble tilsatt. Etter 7 timer ble toluen (30 ml) og eter (20 ml) tilsatt, og blandingen ble vasket med mettet natriumhydrogen-karbonatoppløsning (50 ml) og mettet natriumkloridoppløsning (50 ml). De vandige faser ble ekstrahert med toluen (50 ml), og de kombinerte organiske faser ble tørket (Na2S04) og konsentrert.
Residuet ble destillert for å gi tittel-tetrahydropyranyleteren (1,07 g, 79%) (kokepunkt 85-105°C, 0,08 mm Hg). Kromatografi ga den rene forbindelse (0,88 g, 65%) som hadde n<1>0<3> 1 ,5120; IR: Ymaks 1130' 1060 cm_1' MS (m/e) 85' 133' 135'

Ri er H eller en beskyttende gruppe valgt fra C^-g-alkylkar-bonyl, fenylkarbonyl, fenylkarbonyl hvor fenyldelen er substituert med elektrontiltrekkende grupper, silyl eller tetrahydropyranyl beskyttende grupper.
