Description
Foreliggende oppfinnelse angår en fremgangsmåte for å
belegge en polymeroverflate med et hydrofilt belegg som har lav friksjonskoeffisient når det fuktes med vannbasert væske, samt medisinske artikler som er belagt med et slikt hydrofilt belegg.
Britisk patent nr. 1.600.963 (Biosearch Medical Products, Inc.) beskriver en fremgangsmåte for påføring av et hydrofilt belegg, og hvor dette består av en interpolymer av polyvinyl-pyrrolidon og polyuretan, og hvor nevnte belegg har langt lavere friksjonskoeffisient i våt tilstand enn i tørr tilstand. Slike belegg er blant annet meget fordelaktige for medisinske instrumenter som skal føres inn i kroppshulen,
fordi instrumentet gir godt håndgrep i tørr tilstand samtidig som det blir meget glatt når det kommer i kontakt med kropps-væsker eller våte slimhinner, og det kan således lett inn-settes i pasienten uten at denne skades. Den fremgangsmåte som er beskrevet i nevnte britiske patent, har vist seg å
kunne brukes for fremstilling av slike belegg, men den er på den annen side for komplisert til at den kan brukes ved produksjon i større skala. Den fremgangsmåte som f.eks.
er beskrevet i eksempel 1 i nevnte patent, innbefatter ikke mindre enn 10 trinn, hvorav ett av trinnene tar 6 timer å utføre. En videre ulempe ved denne kjente fremgangsmåten er at det ofte dannes sprekker i belegget.
Hensikten ved foreliggende oppfinnelser er å tilveiebringe en fremgangsmåte for fremstilling av et hydrofilt belegg som har langt lavere friksjonskoeffisient i våt tilstand enn i tørr tilstand, og hvor fremgangsmåten er langt lettere og raskere å gjennomføre enn den fremgangsmåte som er nevnt i ovennevnte britiske patent nr. 1.600.963. Ved hjelp av foreliggende fremgangsmåte er det også mulig å fremstille et belegg som er fritt for sprekker.
Ifølge foreliggende oppfinnelse er det således tilveiebragt
en fremgangsmåte for på en polymeroverflate å anbringe et hydrofilt belegg som har lav friksjonskoeffisient når det
fuktes med en vannbasert væske, og denne fremgangsmåten er kjennetegnet ved at man på polymeroverflaten påfører en oppløsning inneholdende mellom 0,05 og 40% (vekt/volum)
av en forbindelse som inneholder minst to uomsatte isocyanatgrupper pr. molekyl, fordamper oppløsningsmiddelet, påfører en oppløsning inneholdende mellom 0,5 og 50% (vekt/volum) av polyetylenoksyd på den således behandlede polymeroverflaten, hvoretter man fordamper oppløsningsmiddelet fra sistnevnte oppløsning og herder belegget ved forhøyet temperatur.
Videre er det ifølge oppfinnelsen tilveiebragt en medisinsk artikkel for innsetting i kroppen, og denne artikkel er kjennetegnet ved at den har et hydrofilt belegg fremstilt ifølge den ovenfor angitte fremgangsmåte.
Oppfinnelsen er detaljert beskrevet i det etterfølgende.
Foreliggende fremgangsmåte kan brukes for belegging av mange forskjellige typer polymeroverflater, såsom lateksgummi, andre typer gummi, polyvinylklorid, andre vinylpolymerer, polyuretaner, polyestere og polyakrylater. Fremgangsmåten har vist seg å være spesielt fordelaktig i forbindelse med lateksgummi, polyvinylklorid og polyuretaner.
Foreliggende fremgangsmåte innbefatter at man påfører en polymeroverflate en forbindelse som inneholder minst to uomsatte isocyanatgrupper pr. molekyl oppløst i et oppløsningsmiddel, noe som kan gjøres ved dypping, sprøyting eller lignende, hvoretter man fordamper oppløsningsmiddelet fortrinnsvis ved lufttørking. Dette trinn danner et belegg med uomsatte isocyanatgrupper på polymeroverflaten. Eksempler på isocyanat-holdige forbindelser som kan brukes, er polyisocyanater såsom polymetylen-, polyfenylisocyanat, 4,4'-difenylmetandiiso-cyanat og 2,4-toluendiisocyanat. Prepolymerer eller andre addisjonsprodukter av isocyanater og polyoler er spesielt brukbar, f.eks. prepolymerer av toluendiisocyanat eller heksametylendiisocyanat og trimetylolpropan.eller trimerisert heksametylendiisocyanatbiuret. Slike prepolymerer er kommersielt tilgjengelige under varebetegnelsen "Desmodur"
(Bayer AG).
Oppløsningsmiddelet for isocyanatforbindelsen er fortrinnsvis et som ikke reagerer med isocyanatet. Det foretrukne oppløsningsmiddelet er metylenklorid, men det er også
mulig å bruke f.eks. etylacetat, aceton, kloroform, metyl-etylketon og etylendiklorid.
Isocyanatoppløsningen kan med fordel inneholde mellom
0,5 og 10% (vekt/volum) av isocyanatforbindelsen, og med inn-holdet fortrinnsvis mellom 1 og 6 (vekt/volum) av isocyanatforbindelsen. Oppløsningen trenger vanligvis bare å være i kontakt med overflaten i et kort tidsrom, f.eks. fra 5 til 60 sekunder. Når man f.eks. belegger en gummilateks, så
kan det være ønskelig å bruke lengre perioder, f.eks. fra
1 til 100 minutter, for å oppnå en sterk tilfesting. En annen fremgangsmåte for å øke festeevnen er å svelle polymeroverflaten på forhånd ved hjelp av et egnet oppløsnings-middel. En annen fremgangsmåte er å velge et oppløsnings-middel for isocyanatet som i seg selv har evnen til å
svelle eller oppløse den polymeroverflate som skal belegges.
Etter at man har fordampet oppløsningsmiddelet for isocyanatet fra polymeroverflaten, så kan denne belegges med polyetylenoksyd oppløst i et oppløsningsmiddel, hvorved man oppnår en hydrofil overflate som ved endelig herding av isocyanatet vanligvis inneholder en polyetylenoksyd-polyureainterpolymer. Polyetylenoksydet bør ha en midlere molekylvekt på mellom
10 4 og 10 7, og den foretrukne midlere molekylvekten er ca. 10^. Polyetylenoksyd med en slik molekylvekt er kommersielt tilgjengelig, f.eks. under varebetegnelsen "Polyox" (Union Carbide Corporation, USA). Eksempler på egnede oppløsnings-midler for polyetylenoksyd som kan brukes, er metylenklorid (det foretrukne), etylacetat, aceton, kloroform, metyl-etylketon og etylendiklorid. Mengden av polyetylenoksyd i oppløsningen er fortrinnsvis mellom 0 og 10% (vekt/volum)
og mest foretrukket mellom 2 og 8% (vekt/volum). Poly-
etylenoksydet i oppløsI ningsmiddelet påføres I ved dypping, sprøyting eller lignende i et kort tidsrom, f.eks. ifra 5
til 50 sekunder. Etter at polyetylenoksydojppløsningen er blitt påført den belagte overflaten, ble oppløsningsmiddelet fordampet, fortrinnsvis ved lufttørking. De gjenværende spor av oppløsningsmiddel fjernes ved at man herder belegget, fortrinnsvis ved en temperatur mellom 50 og 100°C, f.eks.
i en ovn i et tidsrom jpå fra 5 til 30 minutter. Alle andre trinn i foreliggende fremgangsmåte utføres ved vanlig romtemperatur .

Hensikten med herdingen som med fordel utføres i nærvær av en vannholdig gass såsjom vanlig luft, er å binde isocyanatforbindelsene sammen slik at det dannes et Ltabilt ikke-reaktivt nettverk som binder det hydrofile polyetylenoksyd. Isocyanatgruppene reagerer med vann under hjerdingen og gir
et amin som raskt reagerer med andre isocyanatgrupper, og det dannes en urea-tveIirrbinding.
Ifølge foreliggende oppfinnelse har man overraskende funnet
at det er mulig samtidig å redusere den lave friksjonen på
den hydrofile overflaten, bedre beleggets festeevne og forkorte de nødvendige| reaksjonstider og herdetider, ved å bruke visse additiver jtil oppløsningen av isocyanat og/eller oppløsningen av polyetylenoksyd. Slike egnede additiver innbefatter forskjellige kjente katalysatorer for isocyanat-herding. Disse katalysatorene kan oppløsesj enten i isocyanat-oppløsning eller i polyetylenoksydoppløsning, men er fortrinnsvis oppløst i sistnevnte. Forskjellige typer aminer er spesielt fordelaktige,j f.eks. forskjellige kiaminer, men man kan også bruke trietylenamin. Man anvender!fortrinnsvis et alifatisk amin som larlseg fordampe under tørking og de herdingstemperaturer som brukes etterpå, og som dessuten er ikke-toksisk. Eksempler på egnede aminer er N,N<1->dietylen-diamin, heksametylendiåmin, etylendiamin, paradiaminobenzen, 1,3-propandiol-para-aminobenzosyrediester, diaminobicyklo-oktan og trietanolaminL Mengden av katalysator i poly-etylenoksydoppløsningen ligger fortrinnsvis mellom 0,1 og 50 vekt-% i forhold til mengden av polyetylenoksyd, fortrinnsvis mellom 0,1 og 10 vekt-%. Noen av de ovennevnte aminer, da spesielt diaminene, kan også reagere med isocyanat og derved bidra til tverrbindingen av isocyanatforbindelsene som gir den ønskede sterke bindingen mellom det hydrofile belegg og polymeroverflaten.
Videre har det overraskende vist seg å være mulig å redusere den lave friksjonen på den hydrofile overflaten enda mer ved at man oppløser en polymer i isocyanatoppløsningen. Eksempler på egnede polymerer i så henseende er polyestere, polyvinylforbindelser såsom polyvinylklorid og polyvinylacetat, polyuretaner, polyisocyanater og kopolymerene av disse. Disse ellers i alt vesentlige inerte polymerer antar man gir den overraskende reduserte friksjonen på overflaten i alt vesentlig fordi det hindrer en uønsket diffusjon av ikke tverrbundne isocyanatforbindelser over i polyetylen-oksydlaget. En videre årsak til den lave friksjon kan være at tilsetningen av en polymer øker beleggets elastisitet. Mengden av oppløst polymer i isocyanatoppløsningen ligger egnet mellom 0,5 og 20 vekt-% av oppløsningen, fortrinsnsvis mellom 2 og 10 vekt-%.
Det fremstilte hydrofile belegg inneholder tilsynelatende
en betydelig mengde av delvis fritt bevegbare polyetylen-oksydkjeder. Det har således vært mulig å vise at belegget kan kompleksbinde en vesentlig mengde av elementært jod (sammenlign med eksempel 5), noe som også er tilfelle med fritt polyetylenoksyd. Et slikt jodholdig hydrofilt og antibakterielt belegg er fordelaktig for mange medisinske formål, f.eks. for urinkatetere som skal settes inn i urinveiene i lengre tidsrom, og hvor det ellers lett oppstår infeksjoner. Det jodholdige belegget fremstilles egnet ved en avsluttende dypping i en Kl/I^-oppløsning inneholdende minst 1 vekt-% jod, fulgt av tørking, eventuelt etter at belegget er blitt vasket i et oppløsningsmiddel.
De følgende eksempler illustrerer oppfinnelsen.
Eksempel 1
En pentamer av toluendilsocyanat av cyanurattypen (med navnet "Desmodur IL"; Bayer AG) ble oppløst i metylenklorid til en konsentrasjon på 2% (vekt/volum). Oppløsningen inneholdt også en mindre mengde butylacetat. Et urinveis-PVC-kateter ble dyppet i denne oppløsningen i 30 sekunder. Kateteret ble henlagt for tørking ved romtemperatur i 30 sekunder, hvoretter det ble dyppet i en oppløsning av 6% (vekt/volum) polyetylenoksyd (type WSRN10; Union Carbide) i metylenklorid i ca. 5 sekunder. Kateteret ble så tørket ved romtemperatur i 60 sekunder, og så i 20 minutter ved 70°C over en bolle fylt med vann. Kateteret ble så avkjølt til romtemperatur i løpet av 20 minutter, og ble så vasket i vann. Kateteret hadde en glatt eller sleip, men godt tilfestet overflate.
Eksempel 2
Det ble fremstilt en oppløsning inneholdende 0,5% (vekt/ volum) av isocyanat "Desmodur IL" (se eksempel 1) og 5%
(vekt/volum) av en kopolymer av PVC og polyvinylacetat (20% vinylacetat/80% PVC) som er betegnet "LONZA CL 4520"
i metylenklorid. Et PVC-kateter ble dyppet i denne opp-løsningen i 15 sekunder og ble så tørket ved romtemperatur i løpet av 30 sekunder. Det ble så dyppet i en oppløsning inneholdende 6% (vekt/volum) polyetylenoksyd (type WSRN10; Union Carbide; med en ca. midlere molekylvekt på 100.000)
og 0,23% (vekt/volum) trietylendiamin i metylenklorid. Kateteret ble tørket i 6 0 sekunder ved romtemperatur og så
i 20 minutter ved 70°C over en bolle fylt med vann. Det ble så avkjølt og til slutt vasket i vann. Kateteret hadde en glatt og adherende overflate.
Eksempel 3
Et lateksgummikateter ble svellet i metylenklorid i ca. en halv time. Kateteret ble så tørket ved romtemperatur i 60 sekunder og så dyppet 30 sekunder i en oppløsning inneholdende 6% (vekt/volum) av "Desmodur L2291" (et trimerisert heksametylendiisocyanat av biurettypen som er fremstilt av Bayer AG) i metylenklorid. Kateteret ble tørket ved romtemperatur i 60 sekunder og ble så dyppet i en opp-løsning inneholdende 6% (vekt/volum) polyetylenoksyd (type WSRN10; Union Carbide) i metylenklorid. Kateteret ble så tørket i 60 sekunder ved romtemperatur og så i 20 minutter ved 70°C over en bolle fylt med vann.
Eksempel 4
Innersiden av en to meter lang slange av PVC (indre diameter 3 mm, ytre diameter 4,5 mm) ble vasket med forskjellige opp-løsninger og tørkemidler i følgende rekkefølge og følgende tidsrom;
5% (vekt/volum) av "Desmodur IL" (se eksempel 1) oppløst i metylenklorid i ca. 30 sekunder; gassformig nitrogen ved romtemperatur i 30 sekunder, 2% (vekt/volum) av polyetylenoksyd (type WSRN10, Union Carbide) oppløst i metylenklorid og i 10 sekunder, gassformig nitrogen ved romtemperatur i 60 sekunder.
Slangen ble så plassert i en ovny og gassformig nitrogen ble ført gjennom slangen i 20 minutter. Temperaturen på ovnen var 70°C. Det nitrogen som ble brukt, ble først boblet gjennom vann. Til slutt ble slangen tatt ut av ovnen og vann ført gjennom slangen i en time. Innsiden av den således behandlede slangen hadde et hydrofilt belegg.
Eksempel 5
Et urinkateter av lateksgummi ble dyppet i 30 sekunder i en oppløsning inneholdende 5% (vekt/volum) av "Desmodur IL"
(se eksempel 1) i metylenklorid. Kateteret ble så tørket ved romtemperatur i 60 sekunder og så dyppet i en opp-løsning inneholdende 5% (vekt/volum) polyetylenoksyd (type WSRN10) i metylenklorid. Kateteret ble så tørket i 60 sekunder ved romtemperatur og så herdet i 20 minutter ved 70°C over en bolle med vann.
Etter avkjøling ble kateteret vasket i vann og så dyppet i en mettet vannoppløsning av kaliumjodid som var mettet med elementært jod. Til slutt ble kateteret vasket i vann og henlagt for tørking ved romtemperatur. Kateteret hadde en brun farge, og kjemisk analyse viste at det inneholdt jod.
Claims (8)
1. Fremgangsmåte for på en polymeroverflate å anbringe et hydrofilt belegg som har lav friksjonskoeffisient når det fuktes med en vannbasert væske, karakterisert ved at man på polymeroverflaten påfører en oppløsning inneholdende mellom 0,05 og 40% (vekt/volum) av en forbindelse som inneholder minst to uomsatte isocyanatgrupper pr. molekyl, fordamper oppløsningsmiddelet, påfører en oppløsning inneholdende mellom 0,5 og 50% (vekt/volum) av polyetylenoksyd på den således behandlede polymeroverflaten, hvoretter man fordamper oppløsningsmiddelet fra sistnevnte oppløsning og herder belegget ved forhøyet temperatur.
2. Fremgangsmåte ifølge krav 1, karakterisert ved at det påføres en oppløsning av polyetylenoksyd eller av isocyanat som inneholder en katalysator som akselererer herdingen av isocyanatet.
3. Fremgangsmåte ifølge krav 2, karakterisert ved at det påføres en oppløsning hvor katalysatoren består av et amin, fortrinnsvis et diamin eller et trietylendiamin.
4. Fremgangsmåte ifølge ethvert av kravene 1-3, karakterisert ved at det påføres en iso-cyanatoppløsning som inneholder en polymer som reduserer diffusjon av isocyanat før belegget herdes.
5. Fremgangsmåte ifølge krav 4, karakterisert ved at det påføres en isocyanat-oppløsning hvor polymeren er valgt blant polyestere, polyvinylforbindelser, polyuretaner, polyisocyanater og kopolymerer derav.
6. Fremgangsmåte ifølge krav 1, karakterisert ved at herdingen utføres i nærvær av en
vannholdig gass.
7. Medisinsk artikkel for insetting i kroppen, karakterisert ved at artikkelen har et hydrofilt belegg fremstilt ifølge ethvert av kravene 1-6.
8. Medisinsk artikkel ifølge krav 7, karakterisert ved at den er et kateter.
