Description
Nærværende oppfinnelse vedrører nye kosmetiske hudpleiemidler eller oppløsningsmiddel for terapeutisk aktive stoffer for behandling av patologiske hudtilstander hvilket middel omfatter en vannholdig fase hvor urea og et salt er oppløst, og eventuelt en dispers lipidfase i en mengde på 0 - 50 vekt-% av det totale middel, fuktemiddel eller emulgatorer, aminosyrer og andre tilsetningsmidler. Preparatene er som sådanne velgjørende ved behandling av visse abnorme hudtilstander, men de kan også anvendes som oppløsningsmidler for terapeutisk aktive stoffer for behandling av patologiske hudtilstander.
Den vesentlige egenskapen hos de nye hudpleie<p>reparatene
er at de utøver en oppmykende virkning på hudens hornlag og fremmer den normale deskvamasjonen av horncellene. Hornlaget på hendenes hud blir ofte tørr og ru om vinteren, og dette gjelder også personer hvis hender kommer i kontakt med vaske- - løsninger. Slike forandringer av hudens egenskaper kan man betrakte som mindre avvikelser fra det normale og ikke som hudsykdommer. Det nye preparatet er å betrakte som et kosmetisk middel som anvendes for å unngå slike tilstander hos huden.
Preparatet kan også anvendes som et oppløsningsmiddel for spesielle substanser som er spesifikt virksomme overfor hudsykdommer, f.eks. kortison, hydrokortison og andre steroid-forbindelser, f.eks. fluocinolon-acetonid og triamcinolon-acetonid. Andre aktive substanser som kan oppløses i preparatene ifølge oppfinnelsen omfatter antimykotiske og antibiotiske substanser, tjære og tjære-produkter såvel som substanser som filtrerer bort skadelig stråling i sollys.Den oppmykende virkningen som utøves av de nye hudbehandlingspreparatene på hornlaget muliggjør at de aktive substansene peneterer inn i huden med en større hastighet og på en mer fullstendig måte.
De nye preparatene for kosmetiske formål
kan blandes med vanlig forekommende tilsetningsmidler i kosmetiske preparater, som f.eks. parfymer og lignende.
Den forskning som ledet til nærværende oppfinnelse var basert på det forhold at hornlaget i normal hud skiller seg fra hornlag som er fortykket eller som har blitt hardt eller som er patologisk forandret, ved at det normale hornlaget inneholder en viss mengde hygroskopiske substanser, mens hornlag som er fortykket eller som har blitt hardt eller patologisk forandretinneholder en betydelig mindre mengde hydroskopi ske substanser.
Anvendelsen av urea som hudbehandlingsmiddel med en oppmykende innvirkning på hornlag som er fortykket eller som har blitt hardt har tidligere blitt foreslått. Urea har blitt anvendt i vannlosning eller i form av en krem (emulsjon av typen olje-i-vann), men visse ulemper var forbundet med dette, og disse berodde i hovedsak på det forhold at urea i vannlosning ikke er stabilt men spaltes under dannelse av karbondioksyd og ammoni-akk. I en ureaholdig krem forer spaltningen av urea til en fase-separasjon. Den ureaholdige kremen er således ikke hold-bar.
Norsk patent 128.897 beskriver et kosmetisk middel eller
bærer for kosmetiske og farmasøytiske preparater som er karakterisert ved at det inneholder en vandig oppløsning av urea og melkesyre og eventuelt en dispers lipidfase. Ifølge eksemplene 4 og 6 er det også tilsatt natriumklorid idet forholdet urea:natriumklorid er 20:1.
Hudbehandlingspreparatene eller opplosningen for hudbehandlingsmiddel ifblge oppfinnelsen inneholder som obligatoriske komponenter urea og et salt i vannlosning.
Oppfinnelsen er basert på den oppdagelse at kombinasjonen av urea og et salt ved påforing på huden utover en overraskende synergistisk effekt med hensyn til å holde på vann. Denne synergistiske effekt fremgår klart av kurvene i den vedlagte tegning. Disse kurver har man fått ved å avsette den prosen-tuelle vannopptagningen av torre stykker av hornlag fra menneskehud, som er behandlet med et preparat ifolge oppfinnelsen og som derefter har blitt torket i en atmosfære med 76% relativ fuktighet og en temperatur på 22°C, mot tiden.
Nærmere bestemt ble torre stykker av hornlag fra menneskehud dyppet i testlosningen som inneholdt prepararene ifolge oppfinnelsen og sammenligningspreparater som inneholdt de individuelle komponentene. Losningsoverskuddet ble fjernet med filterpapir, og hornlag-stykkene ble torket i en eksikator (silisiumdioksyd) til konstant vekt (ca. 24 timer). De torre forbehandlede stykkene av hornlag ble derefter plassert i et klimaskap som ble holdt ved 76% relativ fuktighet og ca. 22°C. Prøvestykkene ble veiet i tidsintervaller på 0,5, 1, 2, 4, 6 og 17,5 timer.
De på denne måten erholdte kurvene på figurene 1 og 2 viser
at behandling av hornlag fra mennekse med løsninger som inneholder urea og et salt hver for seg forårsaker en liten vannopptagning i hornlaget tilsvarende opptil ca. 5% vanninnhold efter 6 timer, mens man ved behandling med en løsning som inneholder kombinasjonen urea og salt får en vannopptagning efter 6 timer som tilsvarer et vanninnhold på mer enn 20% og et endelig vanninnhold på 25 - 35%.
Dette økte vanninnhold i hornlag som er behandlet med preparater ifølge oppfinnelsen gjør de tørre hornlagstykkene myke og bøyelige, og dette er ønskelig og nødvendig for en fremgangs-rik behandling av forskjellige hudforstyrrelser av ovennevnte slag.
Av figur 3 i foreliggende søknad fremgår at den vannop<p>takende virkning for urea og natriumklorid i forholdet i henhold til eksemplene 4 og 6 i norsk patent 128.897 og en konsentrasjon på ca. 10% ligger under 3%, mens derimot vannbindingsevnen for produktene ifølge foreliggende oppfinnelse, dvs. ved en konsentrasjon på minimum 10% og i forholdet 1:5 til 5:1 er over 7%, dvs. mer enn dobbelt så stor.
Figuren viser den vannbindende evne hos flass behandlet med en urea- natriumkloridoppløsning. Abscissen angir forholdet mellom natriumklorid og ureakonsentrasjon.

Dette anskueliggjør den tekniske effekt som oppnås ifølge foreliggende oppfinnelse.
Evnen til preparatene ifolge oppfinnelsen med hensyn til å oke vanninnholdet i hudens hornlag gjor dem også egnede som opplosningsmidler for legemidler, som på grunn av hudens okede vanninnhold kan holdes i opplost eller delvis opplost tilstand, og som derved kan penetrere hudlagene.
Karakteristisk for det foreliggende hudbehandlingsmiddel er
at den vandige løsning eventuelt inneholder Na2S20^ og/eller Na2HP0^ som ytterligere salt, har en pH-verdi på 2-8 samtidig med at konsentrasjonen av urea + total saltmengde er 10-40 vekt-% av den vandige løsning og at vektsforholdet urea/total saltmengde i den vandige løsning ligger innen området 5:1 - 1:5.
Det foretrekkes spesielt å holde forholdet urea/salt ved ca.
1:1.
I mange tilfeller anvendes det nye preparater uten noen lipid eller dispers fase, f.eks. i fuktig bandasje. Man kan også impregnere tekstiler med en vannlosning som inneholder urea og salt og innpakker produktene i omslag som forsegles slik at ingen fuktighet dunster bort for anvendelsen.
De nye preparatene kan også anvendes i form av aerosolbesprut-ningsmidler, f.eks. skumaerosoler.
Det er ofte hensiktsmessig å anvende de nye preparatene i form av oljer, kremer, emulsjoner eller dispersjoner, hvorved preparatene inneholder en storre eller mindre mengde av en konvensjonell lipid substans som dispers fase.
Fremstillingen av emulsjoner eller dispersjoner er velkjent innen fagkretser. Nærvær av et salt i den vannholdige fasen kan forårsake vanskeligheter ved fremstillingen av emulsjoner på grunn av fase-separasjon. Det å utvelge en funksjonsdyktig emulgator eller et dispersjonsmiddel er imidlertid noe som ligger innenfor rammen av faglig dyktighet. Eksempler på anvendbare emulgatorer for emulsjoner, som inneholder uorganiske salter, er monoglyceridene av fettsyrer, såsom monolaurinsyre-glycerid og monomyristinsyreglycerid. Et foretrukket preparat ifolge oppfinnelsen inneholder vannfasen i kombinasjon med en emulgator av oven angitt type som eneste lipide substans,
som derved virker som et middel som gir vannfasen en viskos konsistens som egner seg for applisering på huden, dvs. at det virker som et fortykningsmiddel.
Lipidinnholdet er vanligvis 5-50 vekts-% av det totale preparatet inkludert emulgatorer, fortrinnsvis 10 - 40 vekts-%.
Ytterligere komponenter som eventuelt kan være nærværende i de nye hudbehandlingspreparatene ifolge oppfinnelsen er folgende: a) Aminosyrer som forekommer i menneskehudens hornlag eller derivater derav, f.eks. glycin, serin, ornitin, citrullin, arginin og spesielt betain; b) overflateaktive midler (emulgatorer) som er nærværende når hudbehandlingspreparatene inneholder en dispers lipid-f ase; c) fuktemiddel som letter preparatenes spredning og penetrering i huden.
Som oven angitt kan et hudbehandlingspreparat ifolge oppfinnelsen inneholde en lipid-fase, men dette er ikke av vesentlig betydning. Når et hudbehandlingspreparat ifolge oppfinnelsen inneholder en lipid fase foretrekker man imidlertid at vannfasen er den kontinuerlige fasen, og den lipide fasen utgjor den disperse fasen, dvs. preparatene består fortrinnsvis av en emulsjon av typen olj e-i-vann.
Som lipidkomponenter kan man anvende forsåpbare, men også ikke forsåbare li<p>ider og naturlige produkter som inneholder forsåpbare og.også ikke fprsåpbare lipider,
f.eks. ullfett. Man kan også anvende hydrokarboner, f.eks. parafinolje, petrolatum eller andre petroleumsprodukter som vanligvis anvendes i oljer eller kremer.
Hudbehandlingspreparatene ifolge oppfinnelsen kan videre inneholde tilsetningsmidler som vanligvis forekommer i kosmetiske preparater.
Kliniske prover på dermatologiske pasienter med forskjellige typer av torr. hud har bekreftet den velgjbrende effekten som preparatene ifolge oppfinnelsen har.
Oppfinnelsen skal i det folgende anskueliggjøres nærmere ved hjelp av eksempler.

I preparatene ifølge eksemplene 1-5 kan også andre salter anvendes med gunstig resultat, f.eks. MgCl2 og NaHPO^.
Claims (2)
1. Kosmetisk hudpleiemiddel eller oppløsningsmiddel for terapeutisk aktive stoffer for behandling av patologiske hudtilstander hvilket middel inneholder urea og NaCl i vandig løsning og eventuelt en dispers lipidfase i en mengde på 0 - 50 vekt-% av det totale middei, karakterisert ved at den vandige løsning eventuelt inneholder ^2820.3 og/eller Na2HP04 som ytterligere salt, har en pH-verdi på 2 - 8 samtidig med at konsentrasjonen av urea + total saltmengde er 10-40 vekt-% av den vandige løsning og at vekstforholdet urea/total saltmengde i den vandige løsning ligger innen området 5:1 - 1:5.
2. Middel ifølge krav 1, karakterisert ved at vektsforholdet urea/total saltmengde er omtrent 1:1.
