Oppfinnelsen angår en fremgangsmåte ved fremstilling av nye h-halogen-l,2a; 6,7P-bismethylen-A -3-ketosteroider med den generelle delformel

Det var hoyst overraskende at foreliggende fremgangsmåtevar gjennomførbar da A 1 » 1+ ' 6-3-ketosteroider (X = H) som er usubstituerté
i ^-posisjonen bare reagerer ved A^-dobbeltbindingen under dannelse av de tilsvarende 1,2-methylenforbindelser selv ved bruk av et stort overskudd av methylenerende reagenser (tysk patent nr..1072991),
DAS nr. 1096353 og tysk patent nr. 1183500). Fagmannen kunne derfor heller ikke forutse at A 1 ' h ' 6-3-ketosteroidet som på carbonatomet C- h var substituert med fluor, klor eller brom, samtidig ville methyleneres i 1,2-posisjonen og 6,7-posisjonen. 1,2a; 6,7P~bis- ■ methylenforbindelser er hittil ikke blitt beskrevet.
Methyleneringen skjer ved hjelp av kjente fremgangsmåter, f.eks. ved bruk av dimethylmethylensulfoniumoxyd. Dette reagens kan med en vannfri base frigjores fra et dimethylsulfoxydmethosalt i et aprotisk opplosningsmiddel, som f.eks. dimethylsulfoxyd eller dimethylformamid. Dimethylmethylensulfoniumoxyd bringes til å reagere med det umettede keton uten isolering ved temperaturer av - ho til 100°C, fortrinnsvis ved værelsetemperatur.
Methyleneringen kan også skje ved pålagring av diazomethan på A"1"- og A^-dobbeltbindingen og ved påfolgende termisk eller katalytisk spaltning av pyrazolinringen. Den termiske spaltning utfores fortrinnsvis ved 220-250°C, hensiktsmessig under hoyt vakuum, og i nærvær av en inert beskyttelsesgass, som f.eks. nitrogen. Den kata-lytiske spaltning skjer ved innvirkning av sure katalysatorer, som fluorborsyre, bortrlfluoridetherat og fortrinnsvis perklorsyre i organiske opplosningsmidler, som f.eks. i aceton, og fortrinnsvis ved værelsetemperatur. Spaltningen kan også skje ved anvendelse av hbytkokende baser, som f.eks. kinolin eller anilin.
De steroider som benyttes som utgangsmaterialer, kan i ringene
C og D være substituert med f.eks. halogenatomer, alkyl-, hydroxyl-, acyl-, alkyloxy- og/eller acyloxygrupper. De kan også inneholde keto-grupper og/eller dobbeltbindinger, men de får ikke inneholde umettede ketongrupper.
De ifolge foreliggende fremgang311 åte fremstilte 1,2a; 6,70-bismethylensterolder kan på grunn av sine verdifulle farmakologiske egenskaper benyttes som legemiddel eller på grunn av deres spesifikke reaksjonsevne som nye utgangsmaterialer ved fremstilling av verdifulle hormonstoffer. Bismethylenderivatene av hydroxyprogesteron-esterne oppviser f.eks. en overraskende sterk gestagen og ovulasjons-hemmende virkning med bare ubetydelig antiandrogen bivirkning. Deetterfolgende tabeller viser de nye forbindelsers overlegne virkning sammenlignet med de kjente gestagener 6-klor-A I. ' £-pregnadien-17a-ol-3,20-dion-17-acetat (I) og l7a-ethinyl-A<l4>'-ostren-l7P-ol-3-on-17-acetat (II). Som eksempel, på forbindelser fremstilt ifolge foreliggende fremgangsmåte ble i tabellene ^-klor-1,2a ; 6 ,7|3-bismethylen-A1+-pregnen-l7a-ol-3,20-dion-17-acetat (III) benyttet.
■ Den gestagene virkning ble undersokt ved hjelp av den kjente Clauberg-test. I tabell 1 angis den nodvendige minimumsmengde (terskelverdi) for oppnåelse av en positiv virkning.

Ovulasjonshemningen ble fastslått ved hjelp av rorinspeksjon. I tabell 2 angis den daglige dose som etter administrering hindret ovulasjon hos 50 % av dyrene (hunrotter).
Det fremgår av tabellene at den nye bismethylenforbindelse III har en minst 3-10 ganger sterkere virkning enn de kjente gestagener I og II.
Eksempel 1
Til en opplosning av 2,92 g trimethyloxosulfoniumjodid og 690 mg natriumhydrid (50 %- lg ol^esuspensjon) i 55 ml dimethyl sulfoxyd tilsettes under nitrogenstrbm 1,77 g ^--klor-A1 androstatrien-17^-ol-3-on. Etter en reaksjonstid av 30 minutter ved værelsetemperatur og under omroring innrores det hele i svakt eddiksurt isvann, og det utfelte bunnfall avsuges, vaskes noytralt og torkes. Etter omkrystallisering fra isopropylether fås 910 mg M—klor-l,2a; 6,7^-bismethylen-A 11-androsten-17|3-ol-3-on med et smeltepunkt av 197-197,5°C.

Eksempel 2
1,5 g ^-klor-A<1>'^'<6->androstantrien-17P-ol-3-on-17-acetat blandes med en etherholdig diazomethanopplosning (fremstilt fra 15 g nitrosomethylurinstoff, 110 ml ether og 80 ml ko % kalilut) og får henstå i 7 dager ved værelsetemperatur i en lukket beholder. Det overskytende opplosningsmiddel avtrekkes derpå forsiktig under vakuum. Den gjenværende rest av bis-pyrazolin-forbindelsen opploses i 60 ml aceton og tilsettes dråpevis 3 ml 70 % perklorsyre under omroring. Etter at gassutviklingen er opphort, innrores det hele i en 10 ganger så stor mengde isvann, det utfelte bunnfall filtreres, tas opp i methylenljprid, vaskes med vann og torkes over natriumsulfat. Etter inndampning til torrhet kromatograferes resten over silicagel. Det fås 350 mg ^-klor-1,20; 6^p-bis-methylen-A^-androsten-17p-ol-3-on-17-acetat med et smeltepunkt av l8l,5-l82°C. Etter omkrystallisering fra isopropylether stiger smeltepunktet
til 191,5 - 193°C.

Eksempel
5,5 g li-klor-A<1>'<l+>'<6->pregnatrien-l7a-ol-3,20-dion-17-acetat omsettes på samme måte som beskrevet i eksempel 2, med den tilsvarende mengde etherholdig diazomethanopplosning under dannelse av bis-pyrazolin-forbindelsen. Etter sur spaltning og påfblgende kromatografering fås 1,1 g klor-1,2a; 6,7j3-bismethylen-A -pregnen-17a-ol-3,20-dion-17-acetat med et smeltepunkt av 277-278°C etter omkrystallisering fra eddiksyreester.

Eksempel h
1,2 g ^-klor-A<1>'^'<6->pregnatrien-17a-ol-3,20-dion settes til en opplosning av 2,3<*>4- g trimethyloxosulfoniumjodid og ^70 mg natriumhydrid (50 % suspensjon i olje) i ko ml dimethylsulfoxyd, og det hele omrbres i 90 minutter ved værelsetemperatur under nitrogen. Reak-
sjonsblandingen innrores i svakt eddiksurt isvann, og det utfelte bunnfall avsuges, vaskes og torkes. Etter omkrystallisering fra isopropylether/methylenklorid fås 510 mg ^f-klor-l,2a; 6,7J3-bismethylen-A -pregnen-17a-ol-3,20-dion med et smeltepunkt av 251-256 C. UV: £ 2l?l = 11.800..
Eksempel 5
.1,<!>+ g U-brom-A1'^'<6->pre<g>natrien-17a-ol-3,20-dion-17-acetat omsettes som beskrevet i eksempel 2, med etherholdig diazomethanopplosning, og den resulterende bis-pyrazolin-forbindelse spaltes under sure betingelser. Det isolerte, råe h-brom-1,.2a; 6,7P-bismethylen-A Ll<->pregnen-17a-ol-3,20-dion-17-acetat renses ved kromatografering over silicagel og omkrystallisering.
Eksempel 6
3 g lf-klor-A<1>'<l+>'^-pregnatrien-17a-ol-3,20-dion-17-capronat omsettes som beskrevet i eksempel 2, og opparbeides videre. Det fås 735 mg M--klor-l,2a; 6,7P-bismethylen-A<Ll>'-pregnen-17a-ol-3,20-dion-17-capronat i form av olje.
Eksempel 7
a) For fremstilling av utgangsmaterialet settes i lopet av 10 minutter og ved -30°C en opplosning av 565 mg klor i 10,5 ml propion-syre langsomt og dråpevis under omroring til en opplosning av 3,2 g 17,20; 20,21<->bismethylendioxy-A1'<l+>'<6->pregnatrien-3,ll-dion i 50 ml dimethylformamid. Reaksjonsblandingen får så henstå i 2 timer ved -30°C og settes deretter til isvann. Det utfelte bunnfall ekstraheres med methylenklorid, og ekstrakten vaskes suksessivt med en natriumsulfatopplbsning, natriumbicarbonatopplosning og vann og tbrkes over natriumsulfat. Etter omkrystallisering av råproduktet fra eddiksyreester fås 1,3 g H-klor-17,20; 20,21-bis-methylendioxy, A1»1+)^_pregnatrien-3,ll-dion med et smeltepunkt av 2<l>+7-2<1>+9°C (dekom-ponering).
b) Ifolge oppfinnelsen opploses 1,3 g W-klor-l7,20;20,2l-bis-methylendioxy-A ' ' -pregnatrien-3,ll-dion i 6 ml methylenklorid og
settes ved.værelsetemperatur til en opplosning av 1,3 g trimethyloxosulfoniumjodid og 290 mg natriumhydrid (i form av en 50 #-ig olje-suspensjon) i 17 ml dimethylsulfoxyd under omroring og tilfbrsel av nitrogen. Det hele omrbres så i ytterligere 2 timer under de samme betingelser. Derpå helles«den mbrkfarvede reaksjonsopplbsning i eddiksurt isvann og ekstraheres med methylenklorid, og methylenklorid-
oppløsningen vaskes med natriumbicarbonatopplosning og vann, torkes over natriumsulfat og inndampes. Råproduktet kromatograferes over silicagel. Ved eluering med pentan/11 % aceton fås 396 mg ^-klor-17,20;20,2l~bismethylendioxy-l,2a; 6,7P-bismethylen-A -pregneh-3,11-dion som etter omkrystallisering fra^ hexan/aceton smelter ved 276-277°C.

- Fremgangsmåte ved fremstilling av<*>+-halogen-l,2a; 6,7P~bismethylen-A -3-ketosteroider med den generelle delformelhvor X er et fluor-, klor- eller bromatom,karakterisertved at 1,2- og 6,7-dobbeltbindingen i et tilsvarende ^-halogen-1 U Ax» ' -3-ketosteroid methyleneres.
