Description
Diagnostisk middel til påvisning av innholdsstoffene i helblod.
Påvisningen av innholdsstoffene i helblod har hittil krevet
en meget omstendelig, kostbar og tidskrevende fremgangsmåte som bare kan gjennomføres i et dertil innrettet laboratorium og med innarbeidet fagpersonale. Herunder må forst serumet eller plasmaet utvinnes av helblodet idet forst i dette kan stoffene av interesse bestemmes, ellers ville de sterktfarvende egenskaper av hemoglobin-
et forstyrre fremgangsmåter som på en forenklet måte muliggjor påvisning av glucose eller urea i helblod (U.S. Patent nr. 3.O92A65, Britisk Patent nr. 922.665). Prinsippet ved disse nye fremgangs-
måter består deri at en testpapirstripe utenpå overtrekkes med en semipermeabel vegg (av forskjellige cellulosederivater), påforer
man helblod på en slik stripe, går blodplasmaet med innholdsstoffene gjennom den semipermeable film og forer til farvereaksjon i papiret, mens de rode blodlegemer blir tilbake på filmen.' Etter en bestemt tid må det rode blodkoagulat vaskes av for at man kan se reaksjonsfarven i papiret.
Det har nu vist seg at det på vesentlig enklere og mere okonomisk måte lykkes å fremstille utmerket brukbare diagnostiske midler til påvisning av innholdsstoffene i helblod, når man gjor en sugedyktig bsrer (spesielt filtrerpapir), som er impregnert med spesielle reagenser og indikatorer,:hydrofobt ved hjelp av vanlige hydrofoberingsmidler. Dette resultat er fullstendig overraskende. Det kunne.ikke forutsees at et filtrerpapir som er gjort hydrofobt, overhodet kan fuktes så meget med serumet at reaksjonen kan inntre, mens de rode blodlegemer hindres i å trenge inn i papiret ved hydrofoberingen, slik at de kan avspyles fullstendig med vann eller kan strykes av fullstendig med vatt.' Ved ikke-hydrofoberte teststriper trenger de rode blodlegemer nemlig så dypt inn i filtrer-papiret at de hverken kan strykes av eller vaskes av. Ved sin in-tense egenfarve forstyrrer de imidlertid de egentlige farvereaksjoner så ettertrykkelig at man ikke mere kan avlese omslaget.
Egnede midler til hydrofobering er naturlige eller syntet-iske fett, oljer eller voks og spesielt paraffiner, siliconer og blandinger derav. Vanligvis kan man hydrofobere de vanlige teststriper etterpå. Man får imidlertid pgså pålitelige og meget brukbare teststriper når man påforer indikatorfarvestoffet sammen med hydrofoberingsmidlet, men adskilt fra de ovrige reagenser, etterpå på papiret.
Foruten filtrerpapir kan også andre sugedyktige bærere, som asbestpapir eller små trepinner,, anvendes for fremstilling av de ifolge oppfinnelsen hydrofoberte diagnostiske midler, men porene :i treet virker imidlertid leilighetsvis forstyrrende da de rode blodlegemer samler seg i dem og bare1vanskelig kan strykes eller vaskes av. ^
Ved de diagnostiske midler ifolge oppfinnelsen kommer alle ide prover som er av betydning ved blédundersokelser (spesielt som i "screening tests") på tale, f.eks. glucose-, galactose-, fenylalanin-, serumcholinesterase-, urea-, urinsyrebestetnmelser, og også prover på fosfataser, transaminaser, dehydrogenaser, trypsin, histidin, osv.
Vesentlige fordeler ved de ifolge oppfinnelsen hydrofoberte, diagnostiske midler overfor alle tidligere for undersøkel-sen av helblod foreslåtte tester, er den helt enkle behandling og fremstilling da ingen omfintlige og dyre spesialovertrekk eller forseglinger er nddvendig.
I de efterfolgende eksempler er beskrevet nogen hydrofoberte teststriper ifolge oppfinnelsen såvel som deres fremstilling.
Eksempel 1
Fremstilling av en teststripe som er egnet for påvisning av glucose i helblod:

Impregneringsopplosning II:
Med impregneringsopplosning I og II impregneres efter hverandre filtrerpapir fra Schleicher & SchQll nr. 2312, torres og skjæres opp i striper på 5 mm bredde.
Fukter man en slik stripe med en dråpe blod som man ; f.eks. har tatt fra fingeren eller oret, lar blodet innvirke et minutt på strimmelen og derpå spyler blodet av med vann, får man blå farvereaksjoner som er avhengige av glucoseinnholdet i blodet og straks kan bedommes ut fra en farvesammenligningsskaia.
Eksempel 2
Fremstilling av en testremse egnet til påvisning av galactose i helblod:

Impregneringsopplosning II;
Filtrerpapir Schleicher & Schiill nr. 2312 ble efter
hverandre impregnert med impregneringsopplosningene I og II, tor-ret og skåret opp. Galactose-teststrimlene viser ved påfbring av galactoseholdig blod analogt med Eksempel 1 ved en blå farvereaksjon galactoseinnholdet i blod.
Eksempel 3
I stedet for den i Eksempel 1 anvendte impregneringsopplosning II ble anvendt en opplosning av folgende sammensetning:
Forovrig skjer fremstillingen av testremsene som i Eksempel 1 beskrevet, og de erholdte produkter viser de samme egenskaper.
Eksempel 4
Fenylalanin-testpapir:
Filtrerpapir impregneres med L-aminosyreoxydase, per-oxydase og o-tolidin sammen med en 0,2 molar fosfatpuffer med pH 6,5. Det torrede papir efterimpregneres derpå med en opplosning av 4 % paraffin eller 2 % silicon-impregneringsmiddel i methylen-klorid. Oet på denne måte erholdte fenylalanin-testpapir viser ved påforing av fenylalaninholdig blod analogt med Eksempel 1 ved en blå farvereaksjon innholdet av L-fenylalanin i blodet, som kan bedbmmes ved hjelp av en farvetabell.
Claims (1)
- Diagnostisk middel til påvisning av innholdsstoffene i helblod, bestående av en sugedyktig bærer, fortrinsvis filtrerpapir, som er impregnert med spesielle reagenser og indikatorer, karakterisert ved at den sugedyktige bærer er hydrofobert med vanlige hydrofoberingsmidler.
