Abstract

The automatic determination of triglycerides in serum is carrid out by (a) dividing the serum into a sample and a blank; (b) subjecting both to enzymatic hydrolysis; (c) adding the reagents necessary for converting the hydrolysis product (glycerol) into pyruvate (including a light-absorbing substance which reacts with pyruvate) to both the sample and the blank, except that the reagent which converts the glycerol into the next reaction intermediate is omitted in the blank; and (d) carrying out photometric measurements simultaneously on the sample and blank after a suitable reaction time. The use of a blank overcomes the problems associated with the fact that serum already contains pyruvate and substances which absorb at the relevant wavelength (including triglycerides.).
