Claims (24)
1. Een werkwijze voor het bepalen van de functionele activiteit van het MRP2 en/of MRP3 uitstroompad (efflux pathway) van een menselijke of dierlijke patiënt, welke werkwijze 5 omvat: (i) het bepalen van het gehalte van een galzuurderivaat in het bloed van bedoelde menselijke of dierlijke patiënt na een voorafbepaald tijdsinterval na het inbrengen van een hoeveelheid van het galzuurderivaat in de patiënt; en (ii) het toepassen van de bepaling zoals verkregen in stap (i) voor het aanduiden van 10 de functionele activiteit van het MRP2 en/of MRP3 uitstroompad van de patiënt.
2. Een werkwijze volgens conclusie 1 waarbij de werkwijze een verdere stap omvat van het toepassen van de verkregen bepaling voor het verkrijgen van een numerieke waarde, waarbij de numerieke waarde de functionele activiteit aanduidt van het MRP2 en/of MRP3 uitstroompad van de patiënt.
3. Een werkwijze volgens conclusies 1 of 2, waarbij de verkregen bepaling wordt vergeleken met ten minste één standaard teneinde de functionele activiteit van het MRP2 en/of MRP3 uitstroompad van de patiënt aan te duiden.
4. Een werkwijze volgens één van de voorafgaande conclusies, waarbij het gehalte aan galzuurderivaat in het bloed van de menselijke of dierlijke patiënt wordt bepaald na twee 20 of meer voorafbepaalde tijdsintervallen na het inbrengen van een hoeveelheid van het galzuurderivaat in de patiënt.
5. Een werkwijze volgens conclusie 4, waarbij de werkwijze verder de stap omvat van het bepalen van de helling van een log-lineaire grafische weergave van de verkregen bepalingen tegen de tijd.
6. Een werkwijze volgens conclusie 4, waarbij de werkwijze verder de stap omvat van het bepalen van een gebied onder een plasma-uitstroomkromme afgeleid uit de verkregen bepalingen.
7. Een werkwijze volgens één van de voorafgaande conclusies, waarbij het galzuurderivaat een merker bevat, zoals een kleurstof of een fluorescerende stof.
8. Een werkwijze volgens één van de voorafgaande conclusies, waarbij de werkwijze verder de stap omvat van het verschaffen van ten minste één bloedmonster dat de lever van de patiënt heeft gepasseerd en welk monster verzameld is na een voorafbepaald tijdsinterval na toediening van het galzuurderivaat aan de patiënt, waarbij het (de) bloedmonster(s) worden verwerkt ter verkrijging van bloedplasma of bloedserum.
9. Een werkwijze volgens conclusie 8, waarbij het (de) bloedmonter(s) worden verwerkt door middel van centrifugatie.
10. Een werkwijze volgens conclusies 8 of 9, waarbij bloedeiwitten worden afgescheiden uit het bloedplasma/bloedserum. -30-
11. Een werkwijze volgens één van de voorafgaande conclusies, waarbij het galzuurderivaat intraveneus aan de patiënt wordt toegediend.
12. Een werkwijze volgens één van de voorafgaande conclusies, waarbij het galzuurderivaat omvat: (a) a steroïdegroep met (i) ten minste één substituent gekozen uit de 5 groep bestaande uit een 3-hydroxysubstituent, een 7-hydroxysubstituent en een 12- hydroxysubstituent, en (ii) een carboxylgroep gehecht door middel van een amidekoppeling aan een zijketen van de steroïdegroep; en (b) een actieve groep welke op de lever is gericht, welke actieve groep gehecht is aan een a-koolstofatoom ten opzichte van de carboxylgroep.
13. Een werkwijze volgens één van de voorafgaande conclusies, waarbij het 10 galzuurderivaat de algemene formule (I) heeft, E NV^(CHön f 12 \ \ I i | 1 iy 0 cööh A D B i C waarin A gekozen is uit de groep bestaande uit a-OH en β-ΟΗ; B gekozen is uit de groep bestaande uit a-H en β-Η; C gekozen is uit de groep bestaande uit -H, a-OH en β-ΟΗ; of 15 waarin B en C samen een dubbele binding vormen; D gekozen is uit de groep bestaande uit -H, a-OH en β-ΟΗ; E gekozen is uit de groep bestaande uit -H, a-OH en β-ΟΗ; L een koppelende groep is; J een gekleurde of fluorescerende groep is; en n 0 of 1 is.
14. Een werkwijze volgens conclusie 13, waarbij J is of omvat een fluoresceïne, rhodamine of andere fluorescerende groep.
15. Een werkwijze volgens conclusies 13 of 14 waarbij the koppelende groep N- getermineerd is aan het einde ervan dat gehecht is aan de actieve groep J, en L gekozen is uit de groep bestaande uit:-(CH2)nNH, waarin n is 3 of 4, -(CH2)4NH-(CH2)3NHC(=NH)NH-, en -(CH2)2-CH(OH)CH2NH-.
16. Een werkwijze volgens conclusie 13 of conclusie 14, waarbij de groep -NH-CH(COOH) -L- in de algemene formule(l) afgeleid is van een verbinding gekozen uit de groep bestaande uit S-adenosylhomocysteïne, S-adenosylmethiodine, S-aminoimadazool-4-carboxamide, asparagine, cadaverine, cystamine, citrulline, diaminopimelinezuur, 2,4- -31 - diaminoboterzuur, cysteamine, glutamine, 3-hydroxykynurenine, kynurenine, putrescine en negamycine.
17. Een werkwijze volgens conclusie 12, waarbij de steroïdegroep van het galzuurderivaat gebaseerd is op een zuur gekozen uit de groep bestaande uit galzuur (cholic 5 acid), chenodeoxygalzuur, deoxygalzuur, hyodeoxygalzuur, hyogalzuur, α-, β- of ω- murigalzuur, nor-galzuren, lithogalzuur, 3 β-hydroxycholenoïnezuur, ursodeoxygalzuur en allogalzuur (5a-cholan-24-oic-zuur).
18. Een werkwijze volgens één van de conclusies 1 tot en met 6 of 8 tot en met 11, waarbij het galzuurderivaat een cholyl-lysyl-fluoresceïne (CLF) is met de formule: σΟΟΟΗ ^vf/\ Γ T 1 M * ï! ji J 1 O COOH s HO OH 10
19. Een werkwijze volgens conclusie 18, waarbij het galzuurderivaat een zout is van cholyl-lysyl-fluoresceïne.
20. Toepassing van een galzuurderivaat voor de bepaling van de functionele activiteit 15 van het MRP2 en/of MRP3 uitstroompad van een menselijke of dierlijke patiënt.
21. De toepassing van een bepaling van MRP2 en/of MRP3 functionele activiteit verkregen onder toepassing van de werkwijze volgens één van de conclusies 1 tot en met 18 voor het bepalen van de leverfunctie van een menselijke of dierlijke patiënt.
22. Een werkwijze voor het bepalen van de functionele activiteit van het MRP2 en/of 20 MRP3 uitstroompad van een menselijke of dierlijke patiënt in hoofdzaak zoals hierin beschreven.
23. Toepassing van een galzuurderivaat voor het bepalen van de functionele activiteit van het MRP2 en/of MRP3 pad van een menselijke of dierlijke patiënt, in hoofdzaak zoals hierin beschreven.
24. Een werkwijze voor het bepalen van de functionele activiteit van het OATP1B3 opnamepad van een menselijke of dierlijke patiënt, welke werkwijze omvat: -32- (i) het bepalen van het gehalte van een galzuurderivaat in het bloed van bedoelde menselijke of dierlijke patiënt na een voorafbepaald tijdsinterval na het inbrengen van een hoeveelheid van het galzuurderivaat in de patiënt; en (ii) toepassing van de bepaling zoals verkregen tijdens stap (i) voor het indiceren van 5 de functionele activiteit van het OATP1B3 opnamepad van de patiënt.
