Abstract

La invencion se refiere a un metodo para identificar una o mas regiones dentro de un genoma de un organismo de interes que median la expresion de uno o mas genes de interes. El metodo comprende identificar organismos de interes primero y segundo, el primer organismo de interes caracterizado por exhibir una respuesta medible a un estimulo ambiental, o exhibir de otra manera un fenotipo asociado con expresion diferencial de genes asociada con un proceso de interes. El segundo organismo de interes esta caracterizado por la falta de o por no exhibir una respuesta tan intensa al estimulo que la observada dentro del primer organismo de interes, o que exhibe un fenotipo diferente que segrega cuando se compara con el primer organismo de interes, o una combinacion de ambas situaciones. Los organismos de interes primero y segundo son cruzados para producir una poblacion de progenie segregada y se extrae ARN de cada organismo de la progenie segregada. El nivel de expresion de genes para uno o mas genes de interes que estan asociados con la respuesta a un estimulo ambiental, o proceso de interes, es cuantificado. Un mapa de eslabones de la progenie segregada usando o uno o mas marcadores es preparado, y se determina la relacion entre dichos uno o mas marcadores en el mapa de eslabones y la expresion de genes del uno o mas genes de interes y se identifican uno o mas sitios de rasgos cuantitativos (QTL). Este metodo tambien se refiere a identificar uno o mas QTLs asociados con uno o mas genes de interes en la progenie segregada que son sometidos a un estimulo ambiental deseado. A mayor abundamiento, este metodo puede ser usado para la identificacion de uno mas QTLs correspondientes a un factor de transcripcion o cualquier factor que controla la expresion de uno o mas genes de interes, y uno o mas genes ubicados en uno o mas QTLs pueden ser aislados y caracterizados.
