Abstract

La presente invención se refiere a métodos de cultivo de una célula de mamífero y varios métodos que utilizan estos métodos de cultivo. Un método para cultivar una célula mamífero adherente, dicho método está caracterizado porque comprende: (a) proporcionar un matraz de agitación que contiene una célula de mamífero adherente dispuesta en un primer medio de cultivo líquido que ocupa de 20% a 30% del volumen del matraz de agitación y contiene una pluralidad de microportadores a una concentración de 1.0g/L a 15.0g/L; (b) incubar el matraz de agitación durante un primer periodo de tiempo a 35°C hasta 39°C con una agitación rotativa de 85 revoluciones por minuto (rpm) a 125 rpm, y después de las primeras 48 a 96 horas del primer periodo de tiempo, en cada periodo de 24 horas posteriores, (i) retirar de manera continua o periódica un primer volumen del primer medio de cultivo líquido que está sustancialmente libre de microportadores del matraz de agitación, en donde el primer volumen es de 10% a 85% del volumen del primer medio de cultivo líquido; y (ii) añadir al matraz de agitación un segundo volumen de un segundo medio de cultivo líquido, en done el primer y segundo volúmenes son iguales; (c) incubar el matraz de agitación después de que la concentración de células alcance la densidad de células diana durante un segundo periodo de tiempo de 2 días a 7 días, de 32°C a 39°C con agitación rotativa, y en cada periodo de 24 horas, realizar las etapas (b) (i) y (b) (ii) , en donde el primer y segundo medio de cultivo líquido utilizados en la etapa (b) son de un tipo sustancialmente diferente de los utilizados en la etapa (c); y (d) incubar el matraz de agitación durante un tercer período de tiempo superior a 2 días de 35°C a 39°C con la agitación rotativa, y en cada período de 24 horas, realizar las etapas (b) (I9 y (b) (ii) , en donde el primero y segundo medio de cultivo líquido utilizados en la etapa (c) son del mismo tipo que los utilizados en la etapa (d).
