Abstract

La invención se refiere, en términos generales, al campo de la edición genómica y proporciona ADN recombinasas, que recombinan de manera eficiente y específica secuencias genómicas con especificidad de objetivo mediante la fusión de monómeros de recombinasa. Más específicamente, la invención proporciona un método para generar una proteína de fusión para la edición eficiente y específica del genoma, que comprende un complejo de recombinasas que comprende al menos una primera enzima recombinasa, una segunda enzima recombinasa y al menos un enlazador, en donde dicha primera enzima recombinasa y dicha segunda enzima recombinasa reconocen específicamente un primer medio sitio y un segundo medio sitio de un sitio objetivo aguas arriba y/o un sitio objetivo aguas abajo de una recombinasa; en donde dicha primera enzima recombinasa y dicha segunda enzima recombinasa están interconectadas a través de un enlazador; y en donde dicho enlazador comprende o consiste en un oligopéptido. La invención se refiere adicionalmente a proteínas de fusión generadas con este método. La invención también describe recombinasas por diseño, que catalizan la inversión de una secuencia de ADN presente en las regiones int1h en el cromosoma X humano. La invención se refiere adicionalmente a moléculas de ácido nucleico que codifican para dichos ADN recombinasas y proteínas de fusión, así como al uso de dichas proteínas de fusión, ADN recombinasas y moléculas de ácido nucleico en una composición farmacéutica.
