Abstract

Un método para determinar una secuencia de aminoácidos de un polipéptido, que incluye: poner en contacto una primera y segunda muestras que contienen el polipéptido con una primera proteasa (por ejemplo, tripsina) y una segunda proteasa (por ejemplo, Tryp-N), respectivamente, para producir un primer y segundo conjuntos de fragmentos de péptidos digeridos; fragmentar los conjuntos de fragmentos de péptidos digeridos para producir conjuntos de iones de péptidos fragmentados; seleccionar pares de iones de péptidos de los conjuntos de iones de péptidos que difieren en masa en una masa de un residuo de aminoácido arginina o lisina; asignar un tipo de ion (ion de péptido de extremo N terminal o de extremo C terminal) a los pares seleccionados de iones de péptidos de los dos conjuntos; seleccionar una escalera de masa de los iones de péptidos del mismo tipo en cualquier conjunto de iones de péptidos con masa incremental por la masa de uno o más residuos de aminoácidos, y ensamblar los residuos de aminoácidos identificados de la escalera de masa para determinar la secuencia de aminoácidos del polipéptido de interés.
